1.Radioimmunoguided surgery of colorectal cancer by ~(125)I-labeled monoclonal antibody CL58

ObjectiveTo evaluate 125I-labeled monoclonal antibody(McAb) CL58 in radioimmunoguided surgery(RIGS) for patients with colorectal cancer(CC).Methods 125I-McAb-CL58 was injected submucously into pericancerous site under endoscope in 29 CC patients. Intraoperative radioimmunodetection on targets(T) of tumors, regional lymph nodes and incisional margins, was carried out using a portable gamma-detecting probe. The T/NT ratio of 3∶1 was taken as the lowest threshold value of positivity for primary lesions, wall infiltration and lymph node metastasis. Lymph nodes were stained immunohistochemically for micrometastasis. Results The sensitivity for primary lesions was 93 1%. The specificity for tumor margin was 95 5%. 2.Clinical study on neoadjuvant chemotherapy for phase II B breast cancer

Objective To investigate clinical effects of neoadjuvant chemotherapy(NAC)for phase II B breast cancer.Methods From Jan.2002 to Nov.2004,330 female patients with phase II B unilateral breast cancer were enrolled in the study and they were randomly divided into 2 groups.Group A was NAC group and 152 cased were enrolled in this group,among whom 78 cases had left-side cancer and 74 cases had right-side cancer.The mean age of group A was 42.6 years(ranging from 32 to 73 years).Group A were given beth taxane-and anthracycline- based chemotherapy,according to the protocol of group A.After 4 cycles of chemotherapy,patients in group A underwent conservative breast surgery or radical mastectomy according to the result of chemotherapy.Group B,the non-NAC group,had 178 cases in it,among whom 98 had left-side cancer and 80 had rightside cancer.The mean age of group B was 41.4 years(ranging from 31 to 72 years).Group B also underwent conservative breast surgery or radical mastectomy according to tumor size and axillary node status.According to standards of International Union against Cancer(UICC),the primary focus and axillary node status in NAC group were analyzed before and after chemotherapy.Results Group A had a total effective rate of 91.45%.Breast conservation rate in group A and group B was 44.70%and 21.90%respectively.The overall 3 and 5 year survival rate in group A was 96.05%and 75.00%respectively.which was significantly higher than that in group B.Conclusion NAC is effective in reducing clinical stage of phase II B breast cancer,which increases breast conservation rate and postoperative survival rate.

4.Inductive Effect of Ginsenosides on Differentiation of Rat Marrow Mesenchymal Stem Cells into Myocardial Cells in Vitro

[Objective] To observe the in-vitro inductive effect of ginsenosides (GS) on differentiation of rat bone marrow mesenchymal stem cells (MSC) into myocardial cells. [Methods] Marrow stromal cells were isolated from adult SD rats, and then were cultured and cloned by density gradient method and adhesive cultivation to prepare the primary MSC. The cell surface antigens of CD34 and CD44 were detected with flow cytometer to identify MSC. After subculture, the 8th generation of continuous MSC were induced by GS (250 mg/L) and 5-aza (5-azacytidine, 10 ?mol/L) and GS + 5-aza (250mg/L and 10 ?mol/L respectively) for 24, 48 and 72 hours to differentiate to myogenic cells. Meanwhile, a blank control group was set to compare the effect. After the induction, myocardial cell percentage was worked out after examining MSC count and positive cells count under the phase contrast microscope. The specific proteins of Desmin and cardiac troponin I (cTnI) in myocardial cells were detected by immunohistochemistry method, and the cardiac-specific gene expression of ?-MHC (myosin heavy chain) and ?-MHC in myocardiocytes was examined by reverse transcription-polymerase chain reaction (RT-PCR) analysis. [Results] The cultured marrow stromal cells grew well, spindle in shape, and their CD44 expression was positive, indicating that marrow stromal cells differentiated to MSC. After induction with GS and 5-aza, MSC differentiated to myocardial cells, and the myocardial cell percentage was 38% in GS group, 35% in 5-aza group and 39% in GS + 5-aza group, the difference being insignificant. The expression of Desmin, cTnI and MHC in MSC was positive, and MSC were spindle-shaped and looked like fibroblast, indicating that MSC differentiated to myocardial cells. [Conclusion] Ginsenosides can induce MSC to differentiate to myogenic cells in-vitro, and this will supply evidence for cell transplantation.

6. Chemical constituents from Selaginella doederleinii and their bioactivities

Objective: To study the chemical constituents from Selaginella doederleinii and their bioactivities. Methods: Column chromatography on silica gel, RP-C18, and Sephadex LH-20 columns was used to separate and purify the chemical constituents. The structures were elucidated by physicochemical properties and spectroscopic analyses. The inhibition of EtOAc fraction on the tumor cells was tested in vivo and in vitro. Results: Eleven compounds were obtained from EtOAc fraction and identified as flavonoids and phenolic acids. These compounds were asamentoflavone (1), robustafavone 4'-methyl ether (2), 5, 5″, 7, 7″, 4', 4‴-hexahydroxy-(2', 8″)-biflavone (3), 7, 4', 7″, 4‴-tetra-O-amentoflavone (4), 5, 5″, 7, 7″, 4', 4‴-hexahydroxy-(2', 6″)-biflavone (5), ginkgetin (6), isoginkgetin (7), quercetin-3-O-α-D-arabinfuranosi (8), nobiletin (9), gallic acid (10), and shikimic acid (11). Two alkaloids, berberine (12) and palmatine (13) were obtained from n-BuOH fraction. The bioassays indicated that EtOAc fraction could inhibit the growth of HeLa and HepG2 in vitro with IC50 values of 0.12 and 0.60 mg/mL, respectively. The tumor-inhibition rate in S180-bearing mice was 43%. Conclusion: Compound 5 is a new natural product, and compounds 3, 6-10, 12, and 13 are isolated from the plant for the first time.
7.Power density analysis on millimeter waves irradiated into cell monolayers in culture dishes.

Biological effects of millimeter waves (MMWs) at the cellular level are explored in experiments using culture dishes containing cell monolayers with MMW irradiated from the underneath. Analysis is carried out for culture dishes with diameters much larger than the MMW wavelength to discover the relation between the portion of the incident MMW power density (PD) irradiated into the cell monolayer and the interfering factors such as the bottom thickness of the culture dish, the MMW wavelength, and the electromagnetic parameters of the dish and culture solution. With the help of the finite-difference time-domain (FDTD) technique, another analysis is performed to display the effect of the dish geometries upon the MMW PD irradiated into the cell monolayer in a specific culture dish with a diameter similar to the MMW wavelength. The results indicate that rigorous analysis, precise measurement and accurate calculation of MMW PD are essential accompaniments for these experiments, and large-diameter culture dishes are more preferred in experiments to small-caliber ones.

8.Effect of inulin-type hexasaccharide on forced swimming tests in mice and rats and DRL 72 s in rats

AIM To determine whether O-β -D-fructofuranosyl-〔(2→1 )-O-β-D-fructofuransyl〕4α-D-glucopyranoside (inulin-type hexasaccharide, IHS), a monomer extracted from the roots of Morinda of ficinalis How, has antidepressant action. METHODS Fo rced swimming tests in mice and rats and differential-reinforcement-of-low-r ate 72 second schedule (DRL 72 s) in rats were used. RESULTS In the forced swimming test in mice, IHS (80 mg*kg-1, po), like the effe ct of clinically effective antidepressant desipramine (10 mg*kg-1, ip), produced significant decrease in immobility time. IHS (20 mg*kg-1,po ) also elicited significant decrease in immobility time in forced swimming test in rats, which was comparable to the effect of desipramine (40 mg*kg-1, po). Moreover, in the DRL 72 s in rats, IHS (5~10 mg*kg-1, ip), s imilar to desipramine (5 mg*kg-1, ip), elicited significant increase in reinforcers. CONCLUSION These findings demonstrate that IHS has antidepressant action and is an effective component extracted from the root s of Morinda officinalis How.
9.Thoracoscopic internal mammary artery harvesting approach in MIDCAB

Objective To analysis of preoperative preparation,operation,character,advantage and deficiency of two non-robotic TIMAH approach in MIDCAB,and to observe the short-term follow up result.Methods 7 male CAD patients with classic unstable angina pectoris,the age ranged from 52 to 75 years,average (63.8 ± 8.5) years,underwent TIMAH and MIDCAB,in which 6 single LAD disease patients and one patient for reoperation with saphenous vein graft (SVG) graft failure to LAD and progressive obtuse marginal(OM) coronary artery disease.These patients were intubated with a double-lumen endotracheal tube,and one-lung ventilation were used to facilitate the procedure.In the approach of two incisions TIMAH for 4 patients,the thoracoscope was placed at the third intercostals space(ICS) on the anterior axillary line,and LIMA was dissected with endo-instruments placed from the two angles of mini-thoracotomy at fifth ICS on the midclavicular line.In the approach of three incisions TIMAH for 3 patients,LIMA was dissected with endo-instruments placed from two ports at the fourth ICS on the anterior axillary line and at the fifth ICS on the midclavicular line,and the thoracoscope was placed at the second or third intercostals space (ICS) on the anterior axillary line.Anastomosis of LIMA and LAD followed through mini-thoracotomy at fifth ICS with the heart stabilizer after TIMAH.SVG graft was used from LIMA to OM in the reoperation patient.Results All patients underwent TIMAH and MIDCAB safely without transferece to stenotomy,only one LIMA was extended with 2 cm SVG for injure at the distal.In 6 single vessel disease patients the length of mini-thoracotomy incision was (6.0 ± 0.9) cm,TIMAH time was (112 ±18) min,operation time was (293 ± 75) min,bleeding volume was (233 ± 52) ml,endotracheal tube time was (14.2 ± 10.7) h,ICU time was (1.8 ±0.4) d and hospital stay time was (10.1 ±6.7) d.All patients were uneventful discharged and with no recurrence of cardiac symptoms in short-term follow up.Conclusion TIMAH can perform safely in both approaches for LIMA prepare to MIDCAB as described before.The minimally invasive procedure need not enlarge incision for LIMA harvesting with good short term results.
10.Research Progress in Treatment of Malignant Tumors by Traditional Chinese Medicine Based on Exosomes

Malignant tumor is still one of the malignant diseases with high morbidity and mortality in the world. Its occurrence and development are influenced by various factors. Exosomes are nanoscale secretory vesicles that play an important role in the occurrence and development of malignant tumors, and have intercellular communication functions. The mechanism of action of traditional Chinese medicine in prevention and treatment of tumors is not yet comprehensive enough. This article discusses the relationship between exosomes and tumor development, relapse, metastasis and drug resistance, and the application of exosomes in the treatment of malignant tumors by traditional Chinese medicine, to provide reference for finding new breakthroughs in the treatment of malignant tumors.
