OBJECTIVE:To explore the effectiveness and safety of pranoprofen combined with sodium hyaluronate in the treat-ment of moderate and severe dry eyes. METHODS:180 patients with moderate and severe dry eyes were divided into observation group and control group by random number table method, with 90 cases in each group. Control group was given Sodium hyaluro-nate eye drops, one drop each time, tid, and received physical therapy as cleaning eyelid, hot compress and glandulae tarsales mas-sage. Observation group was additionally given Pranoprofen eye drops, one drop each time, tid. A treatment course lasted for 2 weeks. The monocular corneal fluorescence staining score,break-up time of tear film(BUT),dry eye symptom scores,ShirmerⅠtest (SIT) before and after treatment, clinical efficacy and ADR were compared between 2 groups. RESULTS:2,4 weeks after treatment, monocular corneal fluorescence staining scores and dry eye symptom scores of 2 groups were significantly lower than be-fore treatment;BUT and SIT were significantly longer than before;those indexes after 4 weeks of treatment were significantly bet-ter than after 2 weeks of treatment;those indexes of observation group after 2,4 weeks of treatment were significantly better than those of control group at the same time, with statistical significance (P<0.05). The total effective rate of observation group was 94.44%,which was significantly higher than that of control group(78.89%),with statistical significance(P<0.05). Both groups suffered from eye irritation symptom to different extent, and no other severe complications was found. There was no statistical sig-nificance in eye irritation symptom score between 2 groups(P>0.05). CONCLUSIONS:For moderate and severe dry eyes,prano-profen combined with sodium hyaluronate can effectively control ocular inflammation and improve tear film stability. It shows defi-nite therapeutic efficacy and good safety.

AIM: To construct a recombinant lentivirus RNAi vector carrying cytochrome C oxidase gene to obtain the titer of the lentiviral stock for investigation of the expression in the eukaryotic cell and the affection of the COX gene silencing in the eukaryotic cells. METHODS: According to the DNA of the cytochrome C oxidase gene, we designed and synthesized complementary single-strand DNA oligos, annealed the single-stranded oligos to generate a ds oligo, cloned the ds oligo into pENTR/U6 to obtain an entry clone; An LR recombination reaction was performed between the pENTR/U6 entry construct and pLenti6/BLOCK-iT-Dest to generate expression construct, the 293FT cell line was cotransfected with pLenti6/BLOCK-iT expression construct, and the viral packaging mix, viral supernatant was harvested to determine the titer. RESULTS: The DNA sequence of interest clone to the vector was constructd to generate an entry clone and an expression clone successfully, which were proved by sequence determination. A vector producing cell line 293FT was established, and the titer for transfection was obtained. Western blotting analysis demonstrated that COX shRNA expression construction could suppress the expression of MTCOX-I. CONCLUSION: A lentivirus RNAi vector containing cytochrome C oxidase gene was successfully constructed.

Objective Biomechanical analyses of straight techniques for Chinese male skaters during the 1 500-m race were conducted to determine the influence of motion techniques on sports performance. Methods Thirty-four male skaters participating in the 1 500-m National Speed Skating Championship race were selected as research subjects.Kinematic characteristics were collected using three-dimensional fixed-point video analysis. The correlations between the periodic characteristics, motion phase characteristics, moment characteristics, and periodic velocity were analyzed. Results The period distance(r=0.560),gliding distance(r=0.554),and push-off distance(r=0.512)showed strong positive correlations with the periodic velocity. The trunk range of motion(ROM)(r=0.651),ankle angle(r=0.434),and ankle ROM(r=0.446)showed moderate or high positive correlations with the gliding velocity. The trunk angle(r=-0.427)showed a moderate negative correlation with the gliding velocity.Additionally, the trunk angle(r=0.673),hip extension angle(r=0.804),abduction angle(r=0.560),and abduction angle speed(r=0.566)were highly positively correlated with the push-off velocity. The push-off angle(r=-0.605)showed a high negative correlation with the push-off velocity. Conclusions The velocity of a straight line in 1 500-m speed skating depends on the dancing distance.Greater gliding and push-off distances result in higher velocities. The trunk and ankle angles significantly influence the gliding velocity, and appropriately increasing the trunk ROM can effectively improve the gliding velocity. The trunk, hip, and push-off angles are key determinants affecting the push-off velocity, and actively extending the hip and pushing-off can effectively enhance the push-off velocity.
