AIM: To investigate biological effect of endothelin-1 (ET-1) on differentiation of cardiomyogenic cells (CGCs) induced from rabbit bone mesenchymal stem cell (BMSCs) in vitro. METHODS: The BMSCs from shaft of femur of Japan rabbits were isolated and propagated for cell culture. Cultured BMSCs were randomly divided into six groups: Group Ⅰ: the control group (uninduced group); Group Ⅱ: the ET-1 group, added ET-1 (30 nmol/L) into culture medium; Group Ⅲ: 5-aza group, added 5-aza (10 ?mol/L) into culture medium; Group Ⅳ: 5-aza+ET-1 group, after inducing with 5-aza for 3 weeks, ET-1 was added into culture medium. This group was divided into three parts-Ⅳ 1、Ⅳ 2、Ⅳ 3, and separated to add 10、 30、 50 nmol/L ET-1. During the cell culture, growing and differentiation of BMSCs were observed. After inducing for 4 weeks, the differentiation rate and the diameter of cardiomyogenic cells were calculated; Western-blot was employed to analyze the expressions of GATA-4 protein and phosphorylation level. The expression of ?-MHC mRNA was assessed by RT-PCR. Immunohistochemistry staining of Troponin-I and ultrastructure observation of induced cardiomyogenic cells were also completed simultaneously. Objective To investigate the resistance rate of Pseudomonas aeruginosa to several commonly used antibiotics,as well as the prevalence status of three species of integrons in multi-drug resistant Pseudomonas aeruginosa from 2005 to 2006 in our hospital,and to identify the genetic structure of class Ⅰ integron in in our hospital.Methods Drug resistance slip method was applied to detecting the resistant rates of 100 isolates of Pseudomonas aeruginosa to ciprofloxacin,cefotaxime,gentamicin and imipenem.Boiling method was adopted to abstract DNA template of 100 isolates of Pseudomonas aeruginosa,and PCR assay adopted to identify the prevalence rate of three species of integrons and class Ⅰ integron structure.Results The resistant rates of 100 isolates of Pseudomonas aeruginosa to ciprofloxacin,cefotaxime,gentamicin,cefoxitin,tobramycin and imipenem were 32%,45%,82%,37%,39% and 43% respectively.The prevalence rates of three species of integrons was as follows:class Ⅰ 53%,Ⅱ 21% and Ⅲ 5%.Sequence analysis displayed there existed box gene dfrA17 and aadA2 in variable region of class Ⅰ integron.Conclusion The resistant rates of Pseudomonas aeruginosa to four drugs are elevated,especially to gentamycin.Multi-drug resistance may be partly due to prevalence of class Ⅰ integron.

Objective To study anti-inflammatory, analgesic and immunomodulatory effect of total flavone in culture of Saussurea medusa Maxim and the wild one. Methods Different pain, inflammatory and immune models were made to investigate the anti-inflammatory, analgesic and immunomodulatory effect of total flavone in culture of Saussurea medusa Maxim and the wild one such as paw edema in mice, ear edema in rats and so on. Results There are obvious analgesic effect in pain caused by heat in total flavone in culture of Saussurea medusa Maxim and the wild one, and they have obvious restraining effect on paw edema in mice and ear edema in rats as well as on the function of transformation of lymphocyte T and B in rats. Moreover, there is no obvious difference between total flavone in culture of Saussurea medusa Maxim and the total flavone of the wild one. Conclusion Total flavone in culture ofSaussurea medusa Maxim and the wild one possess anti-inflammatory, analgesic and immunomodulatory effect, and there is no obvious difference between them.

Objective To evaluate the clinical efficacy and safety of Jiefujing lotion in the treatment of patients with vulvi- tis and vaginitis(due to downward flow of damp-heat).Methods A randomized,double blind,and multi-center clin- ical trial was performed with Jie' eryin lotion as positive parallel control.Results No obvious difference was observed in general curative effect and symptom-sign curative effect between the two groups(P＞0.05),neither obvious side ef- fects was observed.Conclusion Jiefujing lotion is safe and effective in the treatment of vulvitis and vaginitis due to downward flow of damp-heat.

Objective To explore the thoracic CT manifestations and pathologic basis of pulmonary Wegener granulomatosis (WG),in order to improve the diagnostic accuracy.Methods Eighteen patients with pulmonary WG confirmed by pathology were collected.All the patients underwent CT scan and enhanced scan with 64-slice volume CT.CT features and clinical symptoms were retrospectively reviewed.Results The pathological manifestations of 18 cases with pulmonary WG were miscellaneous,including necrotizing granuloma infiltrated with inflammatory cells,intrapulmonary vasculitis,parenchymal necrosis,hemorrhage of pulmonary alveolus,microabscess and so on.Thoracic CT manifestations:multiple nodules were seen in 7 cases,multiple masses were detected in 4 cases,cavitations showed in 10 cases,peripheral wedge-shaped shadows showed in 2 cases,ground-glass attenuation and consolidation showed in 2 cases,bronchial wall thickening in 2 cases,solitary pulmonary nodule or mass in 3 cases,pleural effusion was seen in 5 cases.All the nodules,masses and cavitations demonstrated inhomogeneous and ring enhancement on contrast-enhanced images.Some patients presented with varied kinds of the above lesions,which could transform each other and became more or less.Conclusions Pulmonary WG has many pathological manifestations,resulting in the variety of the thoracic CT manifestations.The main manifestations are multiple nodules and multiple masses,necrotic cavitations.The feeding vessel sign to a nodule or mass and ring-shade cavity are relatively characteristics to diagnose pulmonary WG.The correct diagnosis could be established on the bases of understanding of the special developments and pathology of the lesions.

ObjectiveTo investigate the changes and values of serum high-sensitivity C-reactive protein (hs-CRP) and cardiac troponin I (cTnI) in patients with chronic heart failure.MethodsEighty patients with chronic heart failure (heart failure group) were divided into NYHA heart function Ⅲ grade ( 40 patients) and Ⅳ grade (40 patients).Eighty healthy people were included in control group.The levels of serum hs-CRP and cTnI were measured and compared.ResultsBefore treatment,the levels of hs-CRP and cTnI in heart failure group [ ( 11.56 ± 2.72) mg/L,(0.46 ± 0.11 ) μ g/L] were significantly higher than those in control group [ (2.31 ± 0.56) mg/L,(0.04 ± 0.13 ) μg/L ] (P ＜ 0.01 ).The levels of hs-CRP and cTnI in heart function Ⅳ grade patients [ ( 13.07 ± 4.31 ) mg/L,(0.57 ± 0.05 ) μg/L] were significantly higher than those in heart function Ⅲ grade patients [(10.04 ±3.12) mg/L,(0.35 ±0.09) μg/L](P＜0.01).After treatment,the levels of hs-CRP and cTnI in heart function Ⅲ grade and Ⅳ grade patients were significantly decreased compared with those before treatment(P＜ 0.01 ).ConclusionsThe levels of hs-CRP and cTnI in chronic heart failure patients are related to the inflammatory activities and myocardial damage.hs-CRP and cTnI may be effective factors to reflect the severity of heart failure.

Objective To analyze the outcomes and the prognostic factors of allogeneic peripheral blood stem cell transplantation (allo-PBSCT) for acute lymphoblastic leukemia (ALL).Method From Feb.2002 to Feb.2014,a total of 95 patients with ALL were treated with alloPBSCT in our hospital.Of these,73 cases obtained the first CR (CR1),11 cases obtained late CR,7 patients were in relapse and 3 patients suffered from primarily refractory disease (PRD) before transplant.The median age was 26 (4-57) years.Conditioning regimens including total body irradiation (TBI)/ etoposide/semustine/cyclophosphamide or busulfan/semustine/cyclophosphamide were used.Matched sibling transplantation was performed on 68 patients,and matched unrelated donor transplantation was performed on 27 patients.Combination of CsA,MTX and low-dose,short-course mycophenolate mofetil was used for graft-versus-host disease (GVHD) prophylaxis.The average fellow-up was 57 months.Result Hematopoietic reconstitution was achieved in all 95 patients.Five-year estimate of overall survival (OS) was 54.3％,disease free survival (DFS) was 51.2％,relapse rate (RR) was 30.2％ and transplant-related mortality (TRM) was 24.0％.The 5 year OS and DFS were significantly longer in patients with CR1 than in late CR and relapse/PRD patients before allo-PBSCT (P＜0.001).There was no significant difference in OS between the two different conditioning regimens.Multivariate analyses revealed that Ⅱ-Ⅳ aGVHD and cGVHD were correlated with higher TRM,CR1 before allo-PBSCT and TBI were associated with a lower RR,and non Ⅱ-Ⅳ aGVHD and CR1 before allo-PBSCT were favorable factors which were associated with OS and DFS.In the patients with DFS≥1 year after allo-PBSCT,DFS and OS were shorter in patients with cGVHD (P =0.008).Conclusion Allo-PBSCT in adult ALL patients should be performed in CR1.Severe acute and chronic GVHD are not associated with improved survival.

Objective: To investigate constitutional and phys ical stamina status in soldiers with different adaption periods in Tibetan areas . Methods: Two hundred and eithty-two male soldiers from a barr acks at an elevation of 3 600 m were divided into 2 groups: the veteran group ( n=143) in Tibet for 1.45±0.22 year, and the recruit group (n=139) in Ti bet for 0.33±0.00 years. The test items included body weight, cirumference, st a nding long jump, chin-up, 100 m dash and 3 000 m jogging, all were evaluated ac cording to the national military standard, which were Comprehensive Evaluation o f Health in Troops, and Examination and Evaluation of the Physical Stamina of So ldiers. Results: The physical stamina indexes of both the vetera ns and the recruits were up to the national military norm on the whole, ranking as moderate. The veteran group showed no significant difference in standing long jump and 100 m dash,（P<0.05), but obvious lower level in chin-up and 3 00 0 m jogging（P<0.01）, compared with the national military norm. The recr ui t group showed significant lower level in chin-up, 100 m dash, 3 000 m jogging as compared with the national military norm（P<0.01）, and also significant lower level in chin-up and 3 000 m jogging （P<0.05 or P<0.01) as compa red with the veteran group. Conclusion: The physical stamina of both the veterans and the recruits meet the basic national military requirements , ranking as moderate. The soldiers who have been in service for over 1 year hav e better explosive force, but they need more tolerance and aerobic exercises. Th e newly recruited need more exercise to raise tolerance to the hypoxic environme nt in plateau areas so as to shorten the adaption time to high altitude.

Objective To evaluate the relationship between lifestyle and development of Alzheimer's disease(AD).Methods Two hundred and thirty-eight AD patients(102 males and 136 females) and 476 healthy controls(204 males and 272 females) were recruited from Ningbo communities into this 1 ∶ 2 matched case-control study.Mini-Mental State Examination(MMSE) and Clinical Dementia Rating(CDR) were required to fill in.Chi-square test and conditional logistic regression were used for data analysis.We adopted Epidata 3.1 to establish the database and did statistical analysis by SPSS15.0.The count data were analysis by Chi-square test,meanwhile multiple factors analysis by conditional Logistic regression analysis.Results Through single factor analysis we found thatcigarette ≥20/day(F=8.687,P=0.003),children visiting(F=22.721,P＜0.05),friendship(F=16.784,P＜0.05),family gathering(≥1 times/week)(F=8.198,P=0.004),working after retirement(F=33.099,P＜0.05),travel(F=16.784,P＜0.05),social activities(F=24.919,P＜0.05),physical exercises(F=24.404,P＜0.05),eating pickles or pickled products(F=6.662,P=0.01),saturated fatty acid intake(F=23.069,P＜0.05),daily consumption of fruits and vegetables(F=8.401,P=0.004),chess(F=17.365,P＜0.05),reading(≥30 min/d)(F=36.390,P＜0.05),using computer(F=8.688,P=0.003) were related AD,the difference was statistically significant(P＜0.05).In multiple factors analysis,social activities,chess games or joke,travelling,working after retirement,reading,physical exercises,friendship,family gathering,saturated fatty acid intake and daily consumption of fruits and vegetables were risk factors of AD(odds ratio(OR) values were 0.571(0.342-0.753),0.623(0.343-0.889),0.686(0.461-0.942),0.534(0.326-0.714),0.276(0.175-0.438),0.538(0.336-0.738),0.585(0.385-0.765),0.466(0.316-0.745),0.527 (0.368-0.787) and 0.482(0.316-0.665),respectively; constant terms:OR=0.526).Conclusion A positive and leisure lifestyle and health reasonable diet could effectively reduce the risk of AD.
