1.Expression of angiopoietin-2 in esophageal squamous cell carcinoma and its significance

Objective To analyze expression of angiopoietin-2 (Ang-2) protein in human esophageal squamous cell carcinoma (ESCC),and to investigate the relationship between Ang-2 protein and clinicopathologic factors.Methods 60 cases of ESCC,5 cases of latero-carcinoma tissues and 30 cases of normal tissue were immunohistochemically stained using Ang-2 monoclonal antibodies with PV-9000 non-biotin detection system.The positive expression rate and expression level of Ang-2 were evaluated.Results Ang-2 was mainly expressed in cytoplasm.The positive expression rates in normal epithelium,latero-carcinoma tissues and ESCC were respectively 3.33 % (1/30),40.00 % (2/5),53.33 % (32/60).Compared ESCC with normal epithelium and latero-carcinoma tissues,Ang-2 positive expression rate was significantly higher (P < 0.05).Among ESCC,expression of Ang-2 was positively correlated with the tumor infiltration depth (rs =0.621,P =0.002),and inversely correlated with tumor differentiation status (rs =-0.309,P =0.018).Conclusions Ang-2 may play an important role in the carcinogenesis and development of ESCC.Tumor differentiation degree,infiltration depth and TNM staging are the independent prognostic indicators for ESCC patients.Combinative detecting the expression of Ang-2 may provide more accurate information in early diagnosis and predicting the prognosis of ESCC.
2.Construction and identification of eukaryotic expressed plasmid vector pLenti6/V5- DEST-Cx43 in human

Objective To construct eukaryotic expressed plasmid vector pLenti6/V5- DEST-Cx43 in human for further study on bioinformative communication between myocardial cell, embryonic development of heart and differentiation of myocardial cell. Methods Cx43 cDNA fragments were obtained from plasmid p18-T by restriction enzyme BamH1、Xho1, after the agarose gel electrophoresis was performed, the Cx43 cDNA was retrieved. pENTR11 were extracted by enzyme BamH1、Xho1, then connected with Cx43 cDNA by T4 DNA ligase. LR reaction was performed and Cx43 cDNA were cloned into Lentiviral vector pLenti6/V5-DEST. Results Agarose electrophoresis and sequent examination, identified that Cx43 cDNA was cloned into Lentiviral vector pLenti6/V5-DEST. Conclusion The present experiment demonstrate a successful cloning of human Cx43 cDNA into the Lentiviral vector pLenti6/V5-DEST.
3.IMMUNOHISTOCHEMICAL STUDY OF THE EXPRESSIONS OF EGF AND EGFR DURING TOLERANTION CYTOPROTECTION OF GASTRIC MUCOSA UNDER STRESS

SD rats were exposed to single or repeated water immersion restraint stress (WRS) for 4 h every other day for up to 6 days, during which the extent of gastric mucosal lesions was evaluated grossly and histologically. The expressions of EGF and EGFR in gastric mucosa were assayed by immunohistochemistry. The results showed that single exposure to WRS led to haemorrhagic lesions, but tolerance developed following repeated exposures to WRS. Histological evaluations showed that single exposure to stress conditions led to crater like haemorrhagic necrosis almost extending to the muscularis mucosae. The submucosa was apparently congestive and edematous. Following repeated WRS, however, mucosal haemorrhagic necrosis was significantly attenuated and submucosal edema and congestion apparently lessened. Active regeneration and proliferation of mucosal cells were observed at the same time. The expressions of EGF and EGFR could be detected in normal gastric mucosa. EGF was expressed in cytoplasma mainly in the regenerative zone with weaker expression at the basal portions of the gastric glands. EGFR was mainly distributed on the membrane of cells in the regenerative zone. Single WRS led to significant decreased in the expressions of EGF and EGFR. their expressions were absent in the necrotic region. After repeated WRS, enhanced expression of EGF and its receptors could be observed not only in mucosal cells of regenterative zone but also in other areas including the lumen of gastric glands. The results suggested that single WRS could cause severe gastric mucosal lesions, but gastric mucosa became tolerant to repeated WRS by way of toleration cytoprotection. Gastric adaptation was accompanied by active regeneration of mucosal cells mediated by EGF binding with its receptor, EGFR.
4.REGULATING ROLE OF ENDOGENOUS NITRIC OXIDE IN GASTRIC MUCOSAL BLOOD FLOW AND TOLERANT CYTOPROTECTION UNDER STRESS

To determine the role of GMBF in gastric mucosal tolerant cytoprotection under stress and its possible regulator, SD rats were exposed to repeated WRS, during which L NAME , a non selective NOS inhibitor, or L Arg, a substrate for NO synthesis, was administered to inhibit or promote the synthesis of NO, GMBF was measured using LDF 3 Flowmeter, NO level in gastric mucosa was monitored by Griess reaction, and gastric mucosal lesions were evaluated by UI. The results showed that gastric tolerant cytoprotection was accompanied by increased GMBF and NO level in gastric mucosa. Inhibition of endogenous NO synthesis by L NAME worsened mucosal lesions induced by WRS. After repeated WRS, adaptive increase of GMBF was abolished and NO content in gastric mucosa significantly reduced. In contrast, enhancement of endogenous NO synthesis by L Arg attenuated mucosal erosions produced by WRS and GMBF, NO content in mucosa increased. Good relationships between the changes in GMBF, UI, NO content in mucosa were found. It suggested that GMBF might play an important role in gastric mucosal tolerant cytoprotection. Endogenous NO might be one of its regulators. Inhibition of its synthesis delayed the induction of tolerant cytoprotection, while enhancementpromoted it.
5.Effects of caspases in indomethacin-induced gastric mucosal cell apoptosis

Objective To determine the changes of caspase 3 and 8 genes and protein expressions during indomethacin (IND) induced gastric mucosal cell apoptosis in vivo . Methods Healthy male SD rats were treated intragastrically with four different doses of IND. The rats were killed 3 hours after IND administration and the TUNEL technique was applied to detect mucosal cell apoptosis. The change of caspase 3 mRNA expression was detected by in situ hybridization and RT PCR techniques and the changes of caspase 3, 8 protein expression were monitored immunohistochemically. Results In the rats of control group, TUNEL assay revealed that only a few apoptotic cells. Oral administration of IND resulted in the appearance of massive TUNEL positive cells. Computer aided image analysis showed that the mean pixels in 30~120mg/kg groups were 6 3 , 8 0 , 12 6 and 17 1 fold that in control group ( P

7.Epidemiological analysis of HIV infection among voluntary blood donors in Liuzhou city

Objective To understand the epidemiological characteristics of HIV infection in voluntary blood donors in Liuzhou city to provide the basis for the blood donation recruit launched by the blood collection and tranfusion service agencies and reduce the risk of transfusion transmitted HIV.Methods The epidemiological investigation on the voluntary unpaid blood donors infected with HIV during 2008-2012 was performed for analyzing the infection characteristics.Results HIV infection in blood donors in Liuzhou city had no statistical difference between different years (P >0.05);the differences in different sexes,age groups,married and unmarried persons had statistically significant differences (P <0.01 );the education background less than middle school ac-counted for 52.94%;56.21% of the infectors were the sexual transmission.Conclusion HIV infection in voluntary unpaid blood donors in Liuzhou city is dominated by young adult men and the persons with the education level less than junior high school,and the heterosexual transmission is predominant.The donor recruitment work should pertinently exclude the persons of high-risk be-havior and collect blood in low risk population as far as possible.
8.The effect of early enteral nutrition on gastric acid in severe brain injury patients

Objective: To evaluate the effect of early enteral nutrition(EEN) on gastric acid secretion in patients with severe brain injury.Methods: 168 patients with severe brain injury(GCS≤8)were divided randomly into two groups.84 patients were given EEN by naso-gastric tube as EEN group and 84 patients were treated with parenteral nutrition(PN)as PN group.Gastric juice pH and upper gastrointestinal hemorrhage were monitored on admission,the 3rd,5th and 7th day after admission.Results: The gastric juice pH was significantly lowered in EEN group.Conclusion: EEN could neutralize gastric acid and increase the gastric juice pH in patients with severe brain injury.
9.Influence of L-NAME and L-Arg on gastric mucosal tolerant cytoprotection under stress

Results Gastric tolerant cytoprotection was accompanied by increased GMBF and NO levels in gastric mucosa. Inhibition of endogenous NO synthesis by L NAME worsened mucosal lesions induced by WRS. After repeated WRS, adaptive increase of GMBF was abolished and NO content in gastric mucosa significantly reduced. In contrast, enhancement of endogenous NO synthesis by L Arg attenuated mucosal erosions caused by WRS. GMBF and NO content in mucosa increased. After 4th WRS, mucosal lesions could be negligible. Conclusion By regulating GMBF, endogenous NO might play an important role in the gastric mucosal tolerant cytoprotection under stress. Inhibition of NO synthesis delayed the induction of tolerant cytoprotection, while increase NO synthesis will ptomote the induction of tolerant cytoprotection.
10.The diagnostic value of symptom index in gastroesophageal reflux-induced chronic cough

Objective To explore the diagnostic value and optimal cut-off point of symptom index (SI) in gastroesophageal reflux-induced chronic cough (GERC).Methods The recordings of multichannel intraluminal esophageal impedance and pH monitoring were retrospectively analyzed in 118 patients with suspicious GERC.SI for all the refluxes,acid reflux and non-acid reflux was calculated respectively by analyzing the temporal association between detected reflux and cough recorded on diary card.Based on the favorable response to the anti-reflux therapy,the diagnostic value for GERC of SI was evaluated and compared with that of the symptom association probability (SAP).Results GERC was definitely determined in 100 patients (84.7%).When SI for all the refluxes was used for the diagnosis of GERC,the cut-off point of ≥45% had the highest diagnostic efficacy,with the sensitivity of 56.0%,the specificity of 83.3%and Youden index of 0.393.SI for acid or non-acid reflux had the same optimal cut-off point of ≥30% and presented with the similar efficacy in the diagnosis of acid or non-acid GERC.Compared with SAP of ≥75%,SIforall the refluxes of ≥45% had a lower sensitivity (56.0% vs 75.0%,x2 =7.988,P=0.005),a higher specificity (83.3% vs 44.4%,x2 =5.900,P =0.015) and the comparable positive or negative predictive value in the diagnosis of GERC.The diagnostic accuracy for GERC was further improved when combining SI for all the refluxes with SAP.Conclusion SI for all the refluxes has a diagnostic value similar to SAP and its optimal cut-off point for GERC may be ≥45%.
