1.Effect of early rehabilitation intervention on postoperative functional rehabilitation of artificial femoral head replacement

Objective To explore the effect of early rehabilitative intervention on postoperative functional rehabilitation of artificial femoral head replacement,to provide the basis for clinical practice.Methods Sixtyfour patients received artificial femoral head replacement were divided randomly into the rehabilitative group and the control group with 32 patients in each group.The patients in the control group received routine rehabilitation exercises,while the rehabilitative group received rehabilitative intervention after 3 hours of operation.The incidence of complications was compared after 2 weeks,and Harris scores were compared after 6 months.Results The incidence of complications in the rehabilitative group was significantly lower than the control group.Harris scores of the rehabilitative group were significantly better than the control group.Conclusions Early rehabilitative intervention can decrease the incidence of complication and accelerate the functional recovery of affected limbs after artificial head replacement.
2.Relationship Between Optimal Time Duration to Use Peripheral Vein Catheter and Bacterium Colonization in Summer Season

OBJECTIVE To study the relationship between how long to use peripheral vein catheter and bacterium colonization,then to make an approach to the optimal time duration of using peripheral vein catheter in summer season from infection prevention viewpoint.METHODS To divide the patients into four groups according to the different time duration of using peripheral vein catheter,and select 20 samples at random in every group.Make bacterial culture from skin(before sterilization,after sterilization and withdrawal of needles) and the catheter,and compare the colony number and strain distribution.RESULTS There was bacterium colonization on punctured skin after using peripheral vein catheter for a long time.The longer it was,the higher the positive rate was.After 96 hours there was bacterium colonization in vein catheter(10%) and after 120 hours positive rate was obviously higher(40%).CONCLUSIONS After using peripheral vein catheter for 96 hours in summer season,there is bacterium colonization in catheter,increasing the risk for infection,so the optimal time duration of using peripheral vein catheter in summer is 72-96 hours.
3.Study of RHD genes polymorphism among RhD negative Han indivduals in Xuzhou area

Objective To investigate the RHD gene polymorphism among the RhD negative Han individuals in Xuzhou.Methods The RhD antigen phenotypes were detected by routine serological techniques,and were identified with indirect antiglobulin test (IAT).The genotypes of RHD were detected by using sequence specific primer polymerase chain reaction(SSP-PCR).Results A-mong 110 RhD negative individuals,there were 5 cases carring RHD positive gene,47 cases carring RHD negative gene,22 cases carring RHD-CE(2-9)-D gene,17 cases carring DVI Ⅲ gene,2 cases carring weak D15 gene,and 17 carring DEL-1227A gene,re-spectively.Conclusion The RhD negative Han individuals present complex RHD gene polymorphism in Xuzhou region,and variant alleles such as RHD-CE(2-9)-D,DVI Ⅲ,DEL-1227A are given priority.
4.The relationship between gait features at usual speed and falls among community-dwelling elderly people

Objective To explore gait features at usual speed of community-dwelling elderly people,and its relationship with history of falls in the past one year.Methods According to history of falls in the past one year,135elderly people in Hefei City were recruited and divided into fallers(23 cases) and non-fallers(112 cases).Self-designed questionnaires were completed by all participants to collect data including gender,age,frequency of exercise,BMI,Barthel Index,MMSE,fear of fall and so on.Gait parameters were measured by IDEEA LifeGait when participants walked for 10 meters at usual speed.Results Between fallers with non-fallers,the time of Timed Up and Go Test had statistically significant differences(P<0.05).Gait analysis results showed that swing power,foot fall,preswing angle,speed had statistically significant differences between two groups (all P<0.05).Conclusion When community-dwelling elderly people are walking at usual speed,the shorter of the time of Timed Up and Go Test,the weaker of swing power,foot fall and pre-swing angle and the slower speed is,the more possible falls will occur.
5.Comparison of two protein extraction methods from velvetantler

Objective To compare optimal protein extraction method of velvet antler ( VA ) and explore the repairmen activity of velvet antler proteins ( VAPs ) on 1-methyl-4-phenylpyridinium ( MPP +) damaged nerve cell.Methods VAPs was obtained from fresh velvet antler by homogenization and micronization method respectively.The protein content of VAPs was measured by Bradford and the molecular weight was identified by SDS-PAGE.Scanning electron microscope (SEM) was perfomed to observe the ultrastructure of VAPs.Neuroblastoma cell line (SH-SY5Y) damage was induced by MPP +, and activity of each compound was compared by the proliferation of damaged cell detected by MTT method.Results The yield of micronization method of VAPs was higher than homogenization method and with better characters both in SDS-PAGE and SEM.Activity detection indicated that VAPs at the concentration of 125 μg/mL by homogenate method could significantly increase the proliferation rate of damaged SH-SY5Y cells.By contrast, VAPs by micronization method had more effective effect at 62.5μg/mL, and proliferation rate could reach 25.45% .Conclusion Micronization method does not only acquire more protein but also has cell proliferation activity, is a relatively ideal way of protein extraction of velvet antler.
6.Discovery of Human Leukocyte Antigen HLA-A/C Gene Recombination

Objective To investigate the recombination events occurring between HLA-A and-C loci discovered in two Chi-nese families.Methods HLA class I (HLA-A and-B)and II (HLA-DRB1)alleles low resolution typing were typed by pol-ymerase chain reaction-sequence specific oligonucleotide,(PCR-SSO)and PCR-sequence specific primer (PCR-SSP).HLA class I and II high resolution typing was done by sequencing-based typing (SBT).Then the recombination sites were ana-lyzed by family study.Results The results indicates that the recombination events occurred in one family between A*30∶01/32∶01-C*06∶02 and another family between A*11∶01/26∶01-C*07∶06 during meiosis.The recombination both came from fathers and resulted in new HLA haplotypes that were inherited by the children.Conclusion Two HLA-A/C re-combination events occurring between HLA-A and-C loci have been found in two Chinese families,which help further study the mechanisms of HLA recombination.
7.Comparative analysis of GM260 portable blood glucose meter and AU5821 automatic biochemical analyzer

Objective To compare GM260 portable blood glucose meter and AU5821 automatic biochemical analyzer in order to prove the accuracy of GM260 and its applicability for clinical use.Methods Totally 20 pieces of EDTA-K2 anticoagulative specimens and 23 GM260 meters were numbered,and each specimen underwent examinations by both GM260 and AU5821,then the bias between the two kinds of devices was calculated.Results The maximal bias between GM260 and AU5821 was 0.47 mmol/L and all the meters had the bias between-0.83 and 0.83 mmol/L in case of 5 specimens with the glucose concentration less than 4.2 mmol/L;the maximal bias between GM260 and AU5821 was 18.07% and all the meters had the bias between-20% and 20% in case of 15 specimens with the glucose concentration not less than 4.2 mmol/L;the examination results by GM260 all accorded with industrial standard.The results by GM260 were lower than those by AU5821,and the maximal negative deviation was-13.43%.Conclusion Portable blood glucose meter can only be used for screening,and automatic biochemical analyzer is the preferred device for diabetes diagnosis.
8.Changes of main components of compound Mailuqishen and its anti-tumor effect in H22 tumor-bearing mice

Objective:To determine the changes of the main components after compatibility of compound Mailuqishen(MLQS), and to explore its anti-tumor effect in the H22 tumor-bearing mice.Methods:The contents of ginsenoside and amino acid in MLQS were detected by HPLC, and the content of polysaccharide was detected by phenol-sulfuric acid.A total of 144 female Kunming mice were randomly divided into normal control group, model group, positive control group, low dose of MLQS group, medium dose of MLQS group, high dose of MLQS group, ginseng geng group, ginseng group and antler plate group (n=16).Except normal control group, the mice in the other eight groups were used to establish the H22 tumor-bearing mouse models, then the mice were treated with drugs at next day.The tumor weights, inhibitory rates of tumor, spleen and thymus indexes of the H22 tumor-bearing mice were detected 10 d after administration.The morphological changes of tumor and spleen tissue were examined by HE staining, and the apoptotic rates of H22 tumor cells were tested by flow cytometry.Results:As calculation with the ginseng and antler plate single herb, the contents of ginsenosides, polysaccharides and amino acids in MLQS were significantly higher than those of single herbs (P<0.05).Compared with model group, the inhibitory rates of tumor in various administration groups were significantly increased(P<0.01);the spleen indexes and thymus indexes of the mice in different doses of MLQS groups were significantly increased(P<0.01);the apoptotic rates of tumor cells were markedly increased(P<0.05).Compared with model group, the tumor tissue of the mice in various administration groups was destroyed, the cells were sparse and irregular, and the tumor presented necrotic lesions;the morphology of spleen tissue was normal with discernible fringe, and the lymphocytes arranged densely.Conclusion:The contents of ginsenosides, polysaccharides, and amino acids in compound MLQS are significantly increased compared with those of single herbs, and its anti-tumor effect is stronger than the single herbs.
9.Composition of and drug resistance in bacteria isolated from the lesions of patients with squamous cell carcinoma of the face and head

Objective To analyze the composition of and drug resistance in bacteria isolated from the lesions of patients with squamous cell carcinoma of the face and head.Methods Lesional tissue or discharges were obtained from 246 patients with squamous cell carcinoma of the face and head,and subjected to conventional bacterial culture.The isolated bacteria were identified by VITEK TWO automated microbiology system.Antimicrobial susceptibility testing was carried out by Kirby-bauer method.WHONET 5.3 software was utilized for statistical analysis.Results Totally,294 bacterial strains were isolated,including 168 Gram-negative bacteria (57.1%)and 126 Gram-positive bacteria(42.9%).The bacterial isolates were predominated by Staphylococcus aureus(21.4%),followed by Escherichia coli(20.4%),Staphylococcus epidermidis(18.4%),Klebsiellapneumoniae(15.4%)and Pseudomonas aeruginosa(9.5%).The prevalence was 40%,26.7%,42.9% and 55.6% respectively for extended spectrum β lactamases-producing E.coli and K.pneumoniae,methicillinresistant staphylococcus aureus(MRSA)and methicillin-resistant coagulase-negative S.epidermidis(MRCNS)respectively.P.aeruginosa,E.coli and K.pneumoniae were highly susceptible to imipenem and meropenem,and favorably sensitive to β-lactam and β-1actamase inhibitor combination.No resistance was observed for vancomycin,teicoplanin or linezolid in staphylococci.Conclusions The bacterial isolates from squamous cell carcinoma tissue on the head and neck are predominated by conditional pathogenic bacteria,and the proportion of Gram-negative bacteria is higher than that of Gram-positive bacteria.These isolates seem to be highly resistant to common antibiotics.
10.Crosslinking sodium hyaluronate gel with different ratio of molecular weight for subcutaneous injection: animal experimental study and clinical trials subcutaneous injection.

OBJECTIVETo investigate the biocompatibility and degradation rate of crosslinking sodium hyaluronate gel with different ratio of molecular weight, so as to choose the effective, safe and totally degraded hyaluronate gel for aesthetic injection.
METHODS(1) Compound colloid was formed by cross-linking the divinyl sulphone and sodium hyaluronate with different molecular weight (4 x 10(5), 8 x 10(5), 10 x 10(5), 12 x 10(5)). (2) Healthy level KM mice was randomly divided into two groups to receive hyaluronic acid gel or liquid injection. Each group was subdivided into three subgroup to receive hyaluronic acid with different molecular weight. The biocompatibility and degradation rate, of hyaluronate were observed at 7, 90, 180 days after injection. At the same time, different molecular weight of sodium hyaluronate gel is sealed or exposed respectively under the low temperature preservation to observe its natural degradation rate. (3) The most stable colloid was selected as aesthetic injector for volunteers to observe the aesthetic effect.
RESULTSThe sodium hyaluronate gel with molecular of 4 x 10(5) was completely degraded 90 days later. The sodium hyaluronate gel with molecular of 8 x 10(5) was completely degraded 180 days later. The sodium hyaluronate gel with molecular of 10 x 10(5) was degraded to 90.0% after 180 days. The sodium hyaluronate liquid can be degraded completely within 7 days. The colloid could be kept for at least 12 months when sealed under low temperature, but was totally degraded when exposed for I d. Sodium hyaluronate gel with molecular 10 x 10(5) was confirmed to be kept for at least 6 months in animal experiment and clinical trials.
CONCLUSIONSUnder the same condition of material ratio, the higher the molecular weight is, the lower the degradation rate is. But the liquidity of gel is not good for injection when molecular weight is too large. It suggests that Sodium hyaluronate gel with molecular 10 x 10(5) maybe the best choice in cosmetic injections.
