1.Aspero saponins phospholipid preparation and absorption in rats' intestine in situ

AIM: To prepare the asperosaponin—phospholipid compound to improve the bioavailability of asperosaponin in rat intestine in situ. METHODS: Ratio of asperosaponin to phospholipid was chosen as index to screen the optimal proportion based on the orthogonal design.Rat intestine perfusion was use to compare aspersaponin absorption under with and without phospholipid. RESULTS: Asperosaponin—phospholipid compound showed the markedly improvement in absorption capacity and absorption rate. CONCLUSION: The experiment recommends that asperosaponin in the combination with phospholipid is better than only used asperosaponin concerning bioavailability.
2.Role of zinc-α2-glycoprotein in lipid metabolism in cancer cachexia

Zinc-α2-glycoprotein (ZAG), a new adipokine, plays an important role in the development of cancer cachexia by decreasing lipid content in body via promoting lipid mobilization and utilization. The lipolytic effect of ZAG is achieved by increasing the activity of hormone sensitive lipase, activating β3 adrenergic receptors,and up-regulating cyclic adenosine monophosphate. In addition, ZAG increases adiponectin expression and decreases leptin expression in adipocytes and adipose tissue. The ZAG expression is regulated by glucocorticoid, peroxisome proliferator-activated receptor gamma agonist rosiglitazone, tumor necrosis factor-α and many other factors.ZAG can potentially be applied as a tumor marker, and may become a new target in the treatment of cancer cachexia.
3.Effect of Cooking Stoves Improved in Controlling Fluorosis Caused by Burning Coal

Objective To know the implementary circs and short-dated effects of cooking stoves improvement in reducing fluoride level in the endemic fluorosis areas and to offer the background information and scientific basis for continuing the development of cooking stoves improvement in reducing fluoride level. Methods To do the surveys in the households, 769 families were selected and investigated from 38 000 peasant families in the program, by cluster sampling with 2% sample rate and 70 families were randomly selected from them, the fluoride levels in the air, foodstuff, non-staple food was determined. Results 98.7% of the families in the cooking stoves improvement areas could rightly use the improved stove. The fluoride levels in corn and hot peppers were greatly improved in county A compared with that in 2000, but the fluoride levels in the air, foodstuff, non-staple food still exceeded the standard limit, not all reached the national standard. Conclusion The headstream interventions measure in coal-burning type endemic fluorosis areas is effective. Controlling the fluoride pollution of capsicum and tea is necessary, the production methods must be improved. The health education and management should be strengthened for reducing fluoride level in the investigated areas.
4.Impact of Fractal Dimension of Osmanthus Fragrans Leaves Induced by Different Levels of Formaldehyde Air Pollution

AIM: To compare the dissolution of asperosaponin Ⅵ between the ultra-micro powder and the fine powder of Radix Dipsaci.METHODS : The real contents,in vitro release and releasing rate of asperosaponin Ⅵ were determined by HPLC for the ultra-micro powder and the fine powder.RESULTS: In the ultra- micro powder and the ordinary powder,the real content of asperosaponin Ⅵ were 4.87%,4.74%,respectively; in vitro release in 1 h were 48.2 mg/g,47.5 mg/g,respectively; releasing rate parameter T_(0.9) were 0.23 min,10.41 min,respectively.CONCLUSION: The ultra- micro porphyrization could not influent the real content and in vitro release of asperosaponin Ⅵ in Radix Dipsaci.But it could improve the releasing rate of asperosaponin Ⅵ.
6.Toxicity of Radix Aconitii on Focal adhesion signal pathway of mice

Objective To study the toxicity and its mechanism of Radix Aconitii from genetic and molecular levels.Methods According to the requirements of ICH,the acute toxicity experiment was carried out under the condition of SPF by ig administration of the decoction of Radix Aconitii to mice.Gene expression profiling was used to describe whole-genome of five organs in mice,the related data were analyzed by using bioinformatics statistics,such as Cluster,GO,and Pathway,and the results were validated through quantitative PCR.Results The effects of Radix Aconitii on the key genes of the Focal adhesion pathway of mice,such as ECM,FAK,Cdc42,were remarkably.Conclusion The reason why Radix Aconitii produces the toxicity is probably that Radix Aconiti could cause the toxicity of Focal adhesion signal pathway through influencing the key genes.

8.The Effects of Thalidomide on Angiogenesis, Growth and Metastasis of Human Hepatocellular Carcinoma in Nude Mice

Objective To obsrve the effects of thalidomide on angiogenesis, growth and metastasis of human hepatocellular carcinoma (HCC) in nude mice. Methods The model bearing HCC was set up in 24 nude mice, and then the rats were randomly divided into therapy group and control group, each containing 12 animal. Thalidomide (200mg/kg/d) dissolved in 0.5% CMC suspension was administered intraperitoneally in the therapy group, and equivalent 0.5% CMC was administered in the control group. Mice were sacrificed at the 30th day after administration, the size and weight of tumors and metastatic condition in liver and lung were determined, levels of CD34 and VEGF mRNA in tumor tissues were detected by immunohistochemical and semi-quantitative RT-PCR, respectively, and microvessel density (MVD) in tumor tissues was also counted. Objective To select a practical and reliable method in cleaning and disinfecting endo-scope, and to investigate the efficacy of endoscope disinfection. Methods A study was done on the disinfection efficacy using or not using enzyme cleaning plus 2% glutaraldehyde immersion, and disinfection efficacy of automated washer/disinfectors for endoscopes at tertiary hospitals in Beijing. Results In 301 cases of en-doscopes cleaned with enzyme ( Epizyme rapid multi enzyme cleanse) and immersed in 2% glutaraldehyde for 7 minutes, culture was negative in 283 cases (94. 02% ) ; in 155 cases without enzyme cleaning but with immersion for 7 minutes in 2% glutaraldehyde, culture was negative in 137 cases (88. 39 % ). Objective To study the effects of three kinds of endoscopic therapy on gastric varices. Methods In this study, the efficacies of endoscopic band ligation ( EVL) , endoscopic injection of tissue adhesives (histoacryl) and sclerotic agent (EIS) for gastric varices were assessed.
