1.Clinical Research of Qizhong Granules for Preventing Upper Respiratory Tract Infection In the Elderly

Objective To evaluate the effect of Qizhong Granules(QG) on preventing upper respiratory tract infection(URTI) in the elderly through clinical observation.Methods One hundred and forty-nine qualified aged patients were randomized into 3 groups: group A(N =50) received QG,group B(N =49) received Jade Screen Powder(JSP),and group C(N =50) was the blank control group.The treatment lasted 7 continuous days.We observed the incidence of URTI as well as the changes of symptoms and signs on the 7th and 14th day of treatment.Results(1) URTI occurred in 1(2.00%),2(4.08%) and 11(22.00%) patients of groups A, B and C,respectively.The differences between groups A and C,and those between groups B and C were significant(P

Objective:To investigate the influence of p38MAPK inhibitor SB203580 on osteoblast MC3T3-E1 cells under high glu-cose concentration.Methods:MC3T3-E1 cells were exposed to glucose at 5.5 mmol/L,25.5 mmol/L and 25.5 mmol/L combined with SB203580 respectively.Cell proliferation was analysed by MTT assay,calcified nodules were quantitated by Alizarin Red S stai-ning.Osteogenesis gene(OCN,Runx2)mRNA level was examined by realtime PCR.Results:High glucose decreased cell prolifera-tion(P<0.05),ALP activity(P<0.05),calcified nodule formation and osteogenesis gene expression of MC3T3-E1 cells.SB203580 reversed the down-regulation of osteogenic markers under high glucose,increased the ALP expression but decreased cell proliferation. Conclusion:SB203580 is antagonistic to P38MAPK in the regulation of preliferation and differetiation of MC3T3-E1 cells.
3.Pathological and clinical effects of preoperative arterial infusion chemotherapy on the breast cancers

Objectives:To investigate the pathological and clinical effect of preoperative arterial infusion chemotherapy on breast cancers.Methods:Twenty patients with breast carcinoma received regional arterial angiography by Seldinger's procedure followed by arterial infusion chemotherapy.Sixteen patients who didn't undergo preoperative chemotherapy were selected as controls.All the operation specimens were analyzed by the same pathologist.Results:Histological analysis of the two groups revealed the following results:① cancer tissue necrosis increased in the arterial chemotherapy group;②karyopyknosis,karyorrhexis coagulation and necrosis of cytoplasm around the vascular vessels as well as interstitial edema were found in the tumor tissue,invasion of inflammatory cells,intimal proliferation thrombus and inflammation of vessels could also be seen.All the changes were much severe in the infusion chemotherapy group than in the controlled group;Conclusions:Histological changes are significant after preoperative arterial infusion chemotherapy for breast carcinoma.

Objective In order to further study the role of gap junction(GJ) in growth control; and explore the possible mechanisms for the inhibition of GJ formation. Method A monoclonal antibody,called ND6, which is specific for MIP(major intrinsic protein)in the plasma membrane of eye lens fiber cells in chicken,was injected into the right eyes of chicken embryos at development stage 20, while the left eyes were not injected and served as controls. The size of the lenses was measured 24 hours after the ND6 treatment; the protein phosphorylation of the lenses, including intact and homogenized lenses was also analyzed 24 hours after the ND6 treatment. 5.Prevention and management of perioperative complications of carotid endarterectomy with 318 cases

Objective To introduce the experience of prophylaxis and management of the complications of carotid endarterectomy (CEA) during perioperative period because those complications can not be avoided yet.Methods During Oct 2000 to Jan 2013,318 patients were performed CEA.There were 213 men and 105 women with an average age of 74.8 years ranged from 48 to 85y.There were 186 patients in the left,120 patients in the right,and 12 patients in two sides.Simply,CEA are performed in 166 patients,CEA and patch in 140,and 12 patients were performed bypass with the autogenously great saphenous vein after the lesions removal.Results The perioperative complications were preoperative stroke in 3 cases,hypotension during operation in 16,after-operation deaths in 3,hyperperfusion syndrome in 18,postoperative severe cerebral infarction in 1,hoarseness in 14,and hematoma in 28.Conclusions Perioperative complications of CEA can be decreased by cautious surgical procedure and postoperative observation.
6.Cell type specificity of signaling: view from membrane receptors distribution and their downstream transduction networks.

Studies on cell signaling pay more attention to spatial dynamics and how such diverse organization can relate to high order of cellular capabilities. To overview the specificity of cell signaling, we integrated human receptome data with proteome spatial expression profiles to systematically investigate the specificity of receptors and receptor-triggered transduction networks across 62 normal cell types and 14 cancer types. Six percent receptors showed cell-type-specific expression, and 4% signaling networks presented enriched cell-specific proteins induced by the receptors. We introduced a concept of "response context" to annotate the cell-type dependent signaling networks. We found that most cells respond similarly to the same stimulus, as the "response contexts" presented high functional similarity. Despite this, the subtle spatial diversity can be observed from the difference in network architectures. The architecture of the signaling networks in nerve cells displayed less completeness than that in glandular cells, which indicated cellular-context dependent signaling patterns are elaborately spatially organized. Likewise, in cancer cells most signaling networks were generally dysfunctional and less complete than that in normal cells. However, glioma emerged hyper-activated transduction mechanism in malignant state. Receptor ATP6AP2 and TNFRSF21 induced rennin-angiotensin and apoptosis signaling were found likely to explain the glioma-specific mechanism. This work represents an effort to decipher context-specific signaling network from spatial dimension. Our results indicated that although a majority of cells engage general signaling response with subtle differences, the spatial dynamics of cell signaling can not only deepen our insights into different signaling mechanisms, but also help understand cell signaling in disease.

7.The accuracy analysis of fully-guided mucosa-supported template based computer-assisted implant surgery

Objective :
The purpose of this study is to determine the accuracy of implant placement in the edentulous jaw using computer planning and fully-guided mucosa-supported surgical template by Simplant software.
Methods:
63 implants were placed in 9 patients (11 fully edentulous jaws), 26 implants were placed in upper edentulous jaw and 37 implants in lower edentulous jaw. Preoperatively, first, a cone beam CT was required for patients with radiographic template and radiographic template respectively. Therefore, the data of CBCT was inputted in Simplant software by DICOM format, followed by virtual implant planning. Hereafter, a mucosa-supported surgical template was designed by dentist and made by Masterilise company to allow implant placement using the template as a guide. To investigate the accuracy of implant placement, a postoperative CBCT scan was obtained and matched to the preoperative scan. The accuracy of implant placement was validated three-dimensionally including divations of implant shoulder, apical point, axial angulation and depth.
Results:
The survival rate of 63 implamts for a 6 month to 10 year observation period was 100%. The mean divation of implant shoulder was 0.73 ± 0.53 mm, implant apical point was 1.15 ± 0.62 mm, implant depth was 0.95 ± 0.64 mm and implant axial angulationwas 4.10 ° ± 3.23°.
Conclusion
Divation between virtual and actual implant was existed and it should be considered preoperatively when virtual implant was planned to avoid injuring anatomic structure and keeping surgery safely. Correct manipulation during implant operation are helpful to decrease the divation of implant placement.
8.Observation of dendrite osteocytes of mice at different developmental stages using Ploton silver staining and phalloidin staining.

OBJECTIVE:
To assess the value of Ploton silver staining and phalloidin-iFlour 488 staining in observation of the morphology of osteocyte dendrites of mice at different developmental stages.
METHODS:
The humerus and femurs were harvested from mice at 0 (P0), 5 (P5), 15 (P15), 21 (P21), 28 (P28), and 35 days (P35) after birth to prepare cryo-sections and paraffin sections. HE staining of P35 mouse femur sections served as a reference for observing osteocytes in the trabecular bone and cortical bone. The humeral sections at different developmental stages were stained with Ploton silver staining to observe the morphology of osteocytes and canaliculi, and the canalicular lengths in the cortical and trabecular bones of the humerus of the mice in each developmental stage were recorded. The cryo-sections of the humerus from P10 and P15 mice were stained with phalloidin iFlour-488 to observe the morphology of osteocytes and measurement of the length of osteocyte dendrites in the cortical bone.
RESULTS:
In the trabecular bone of the humerus of P0-P15 mice, Ploton silver staining only visualized the outline of the osteocytes, and the morphology of the canaliculi was poorly defined. In P21 or older mice, Ploton silver staining revealed the morphology of the trabecular bone osteocytes and the canaliculi, which were neatly arranged and whose lengths increased significantly with age (P21
CONCLUSIONS
Mouse osteocyte dendrites elongate progressively and their arrangement gradually becomes regular with age. Ploton silver staining can clearly visualize the morphology of the osteocytes and the canaliculi in adult mice but not in mice in early stages of development. Phalloidin iFlour-488 staining for labeling the cytoskeleton can be applied for mouse osteocytes at all developmental stages and allows morphological observation of mouse osteocytes in early developmental stages.
