1.Technique for recycling used oxygen-18 water
Academic Journal of Second Military Medical University 1985;0(05):-
Objective:To study the recycling technique for used nuclear-medicine tracer oxygen-18 water. Methods:A small-scale purification device for wasted oxygen-18 water was built.Six processes, including cotton core filtration, active carbon adsorption, ultraviolet oxidation,anti-osmosis filtration, ion exchange, and simple evaporation, were included in the device.Totally 10.5 kg experimental samples were evenly divided into 21 portions.Three portions of the samples were treated by each of the above 6 processes for 5 cycles by dosing pump. The electrical conductivity,TOC(total organic carbon), enrichment of oxygen-18 water, and the material consumption, were statistically analyzed after each recycling. Results:Samples were greatly consumed in the anti-osmosis filtration process and the enrichment of oxygen-18 in the ion exchange process dropped markedly;therefore, the above 2 processes were ruled out for the recycling.Though sample consumption was higher in the initial steps of cotton core filtration and active carbon adsorption, but was lower in the latter steps.Samples were hardly consumed in ultraviolet oxidation and simple evaporation;meanwhile, these processes had little influence on enrichment of isotope oxygen-18.The suitable processes for wasted oxygen-18 water purification were cotton core filtration, active carbon adsorption, ultraviolet oxidation, and simple evaporation. The 4 suitable steps were used for 1500 g samples. The conductance value of the resultant samples(1341 g)was(2.22?0.03)?S/cm,the TOC was(0.86?0.01)mg/L,and the enrichment was (32.44?0.04)atom%(dropped by 2.56%),with the sample consumption rate being 10.6%.Conclusion:Cotton core filtration, active carbon adsorption, ultraviolet oxidation, and simple evaporation can basically meet the demand for recycling oxygen-18 water.

Objective To discuss surgical techniques during microendoscopic discectomy (MED) in the treatment of lumbar intervertebral disc herniation complicated with posterior longitudinal ligament calcification and osteophyte of posterior lumbar border. Methods The self-made arc-like annular knife and L-shape introducer were used during MED in the treatment of 115 patients. Results All the patients were followed for 12~30 months (mean, 22 months). According to the MacNab criteria, excellent or good results were achieved in 109 patients, fair results in 5 patients, and poor in 1, the rate of excellent or good results being 94.8% (109/115 ). No ruptured spinal dura mater, nerve root injuries, or infection of intervertebral space occurred. No conversions to open surgery were required. Conclusions Use of self-made instruments and corresponding modified techniques during MED can effectively manage the problems of posterior longitudinal ligament calcification and osteophyte of posterior lumbar border, which broadens the surgical indication and improves the efficacy and safety.

Objective To analyze the correlation between the polymorphism of PTPN22 gene (R620W and R263Q sites)and pulmonary tuberculosis(TB)in Chinese Han population and to investigate the environmental factors associated with pulmonary TB. Methods A case-control study was conducted on 235 patients with pulmonary TB and 251 healthy subjects. The single nucleotide polymorphisms(SNP)of PTPN22 gene at R620W and R263Q sites were detected by the assay of polymerase chain reaction and re-striction fragment length polymorphism(PCR-RFLP). A questionnaire was designed to gather information about tuberculosis-associated environmental factors. Univariate and multivariate logistic analysis were con-ducted. Results Genotype frequencies of PTPN22 R620W(C1858T)SNP were 233(99. 15% ,CC),2 (0. 85% ,CT),0(0% ,TT)in patients with pulmonary TB and 240(95. 62% ,CC),11(4. 38% ,CT), 0(0% ,TT)in healthy subjects. There was a difference with the distribution of PTPN22 C1858T allele be-tween patients with pulmonary TB and healthy subjects[0. 43% vs 2. 19% ,P = 0. 01,odds ratio(OR)=0. 19,95% confidence interval(CI)= 0. 07-0. 35]. Genotype frequencies of PTPN22 R263Q(G788A) were 218(92. 77% ,GG),17(7. 31% ,GA),0(0% ,AA)in patients with pulmonary TB and 248 (98. 71% ,GG),3(1. 29% ,GA),0(0% ,AA)in healthy subjects. The frequencies of G788A allele in patients with pulmonary TB were higher than those in healthy subjects(3. 62% vs 0. 60% ,P﹤0. 01,OR=6. 03,95% CI=2. 12-18. 38). Conclusion The results of this study suggested that the R263Q GG geno-type of PTPN22 gene was associated with the susceptibility to TB in Chinese Han population.

To investigate the effects of metallothionein (MT) on isolated rat heart, 16 Wistar rats were randomly divided into 2 groups. In control group (group C), distilled water was injected intraperitoneally and 24 h later isolated hearts were perfused with Langendorff and stored at 4 degrees C for 3 h with histidine-tryptophan-ketoglutarate (HTK) solutions, and then isolated hearts were perfused for 2 h by Langendorff. In experimental group (group E), 3.6% ZnSO(4) was injected intraperitoneally, 24 h later isolated hearts were perfused by Langendorff and stored at 4 degrees C for 3 h with HTK solutions, and then the isolated hearts were perfused for 2 h with Langendorff. MT content, the recovery of hemodynamics, myocardial water content (MWC), lactate dehydrogenase (LDH) and creatine kinase (CK) leakage, adenosine triphosphate (ATP) and malondialdehyde (MDA) content, superoxide dismutase (SOD) activity, myocardial cell Ca(2+) content, Ca(2+)-ATPase activity of mitochondria ([Ca(2+)-ATPase](m)) and its Ca(2+) content ([Ca(2+)](m)), synthesizing ATP activity of mitochondria ([ATP](m)), and the ultrastructure of cells were examined. There were a significant increase in group E in hemodynamic recovery, ATP content, SOD activity, [Ca(2+)-ATPase](m) activity, [ATP](m) activity, and substantial reduction in MWC, LDH and CK leakage, MDA content, myocardial cell Ca(2+) content, [Ca(2+)](m) content, and the ultrastructural injury were obviously milder than that of group C. This study demonstrated that MT has protective effects on isolated rat heart.

Journal of Huazhong University of Science and Technology (Medical Sciences) 2002;22(2):107-8
To study the effects of different pH HEPES-KH reperfusate solution on immature myocardial protection, isolated perfused Langendorff model from immature rabbit hearts were developed formed. Control group (C) was perfused only with pH 7.4 HEPES-KH solution for 90 min. Ischemia/reperfusion group (group I/R) was perfused with pH 7.4 HEPES-KH solution before ischemia or after ischemia. Experimental group (group E), after ischemia, was perfused with pH 6.8, pH 7.1 and pH 7.4 HEPES-KH solutions for 5 min, 5 min, and 20 min, respectively. The left ventricular function recovery, MWC, LDH and CK leakage, MDA, ATP content, and SOD activity were determined. Our results showed that the left ventricular function recovery, ATP content and SOD activity in group E were higher than those of group I/R (P < 0.05). MWC, MDA content, LDH and CK leakage in group E were lower than those of group I/R (P < 0.05). These findings suggested that pH paradox might be one of important mechanisms for immature myocardial ischemia-reperfusion injury, and acidic perfusate, at the beginning of reperfusion, might attenuate pH paradox and ameliorate functional recovery in isolated perfused immature rabbit hearts.
