2.Effect of oxidized low density lipoprotein on CD36 expression in macrophage

Objective: To investigate the effect of oxLDL on CD36 expression and cholesterol influx in THP-1 macrophage. Methods: THP1 macrophage were treated with 50mg/L oxLDL for 0,12,24,36,48 h respectively.Oil red O staining was used to observe the intracellular lipid droplets.\labeled Cholesterol influx was determined by FJ-2107P type liquid scintillator.CD36 mRNA and protein level were determined by reverse transcription-polymerase chain reaction(RT-PCR) and Western blotting respectively. Results:We found that oxLDL elevated CD36 in both protein and mRNA levels and increased cholesterol influx in a time-dependent manner.The levels of cholesterol influx were 23.5%、(27.8%、)39.7%、44.1% and 49.7% respectively. Conclusion: oxLDL may upregulate CD36 expression and increase cholesterol influx.
3.Establishment of a rat chronic asthma model and its evaluation

This study is to establish a rat chronic asthma model. Sensitive SD rats were selected through histamine challenge. The asthmatic groups were sensitized by ih and ip with OVA, aluminium hydroxide gel and inactivated bacillus pertussis on day 1 and 14. From day 21, acute asthmatic group was aerosolized 1% OVA for 1 week, chronic asthmatic group was aerosolized 0.1% OVA for 12 weeks. The control groups received saline as the substitution of OVA. Twenty four hours after the last provocation, physiological monitoring equipment was used to detect the pulmonary function, then the rats were sacrificed. Bronchoalveolar lavage fluid (BALF) was collected to calculate the ratio of different inflammatory cells. ELISA was used to detect total IgE and OVA-specific IgE in serum. Microscopy was conducted to observe the histopathology of lung stained with haematoxylin and eosin staining. Collagen fibers were detected using Picric acid-Sirius red staining technique. The optical density at 610 nm of extractive from locus caeruleus was detected by passive cutaneous anaphylaxis (PCA). The results showed that the asthmatic characteristics were significantly developed in model groups, but not in control groups. Chronic asthmatic group had significantly higher indexes than acute asthmatic group, including the thickness of airway smooth muscle and bronchial basement membrane, and goblet cell hyperplasia, the area of collagen in airways, A610 of extractive from locus caeruleus, the concentration of total IgE and OVA-specific IgE in serum. However, inflammatory cell infiltrate in lungs and the percentage of eosinophils of white blood cells in BALF were lower in chronic asthmatic group than those in acute asthmatic group. Respiratory rate and respiratory flow showed no significant difference in both model groups. In conclusion, the rat chronic asthma model is established by the way in this study, which is comparable to the physiopathologic characteristics of human asthma.
4.Optimization on expression condition of IL-1ra-Fc? fusion gene in rats and its immunogenicity analysis

Aim To investigate the optimal condition for expression of pCI-neo-IL-1ra-Fc? plasmid for gene therapy and its immune response in rats.Methods Different doses(0.25,0.5,2.5 mg?kg-1)pCI-neo-IL-1ra-Fc? plasmid were instilled into lungs of rats through tracheas,then expressions of IL-1ra-Fc? fusion protein were detected at different interval times(d 4,d 7,d 20)by RT-PCR and Western blot.Through immu-nohistochemistry analysis of rat lungs,those results of optimal condition were validated.Dynamic changes of rat serum IgG level,including IgG2a and IgG1 were detected by indirect ELISA.Results IL-1ra-Fc? could be detected at 20th day after instilling pCI-neo-IL-1ra-Fc? plasmid in all dose groups.The expression level in 2.5 mg?kg-1 groups was higher than that in other two groups.IL-1ra-Fc? protein could not be expressed at 4th day,but increased at 7th day and reached maximum at 20th day after instillation.IL-1ra-Fc? gene could be efficiently expressed in rat lungs by immunohistochemistry analysis.Humoral immunity of rats was induced by pCI-neo-IL-1ra-Fc? plasmid.IgG2? expression was predominant in IgG subsets,showing that the humoral immunity was Th1-like immune response.Conclusion The optimal condition is established for gene therapy with pCI-neo-IL-1ra-Fc? plasmid which can induce Th1-like immune response in rats.
5.Effects of 5-Aza-2'-deoxycytidine on proliferation of human gastric cancer BCG-823 cells and the expression of HOXA5 gene

Objective To explore the effects of 5-Aza-2'-deoxycytidine (5-Aza-CdR) on the proliferation of BCG-823 human gastric cancer cells and the expression of HOXA5.Methods The methylation status of the promoter of HOXA5 was measured by methylmion specific PCR (MSP).The BCG-823 cells were treated with different concentrations of 5-Aza-CdR,and then the changes of expression and methylation status of HOXA5 gene were detected by quantitative real-time polymerase chain reaction (QRT-PCR),Western-blot,and MSP.Cells proliferation was assessed by methyl thiazolyl tetrazolium (MTT) assay.Results (1) Different methylation status of HOXA5 gene promoter was detected in BCG-823 cells.The mathylation rate of HOXA5 gene promoter were reduced after treatment with 5-Aza-CdR,and also were negatively related to the concentration of 5-Aza-CdR (F =438.307,P < 0.01).(2) Compared to the control groups,the expressions of HOXA5 mRNA and protein were increased after treatment with 5-Aza-CdR,with statistical significance (P < 0.05).(3) The proliferation rate of BCG-823 cells was significantly inhibited (P <0.05).Conclusions The methylation status of HOXA5 promoter was detected in BCG-823 cells.5-AzaCdR is able to inhibit BCG-823 cells growth in vitro,which might be related to the expression of HOXA5.It may be a new way to treat gastric cancer.
6.EARLY FLUID REPLACEMENT IN SEVERE BURN INJURY

The results of early resuscitation of 104 adult patients with BSA more than 50%were studied retrospectively. Although the mean amount of fluid replaced during 48h postburn was similar to the amount calculated with our formula, there existed significant individual differences. Therefore it seems not necessary to set up a rigid fluid replacement plan. To ensure adequate tissue perfusion, the fluid replacement formula might be modified as follows: 2 ml/kg/BSA%, with urinary output 30-40 ml/h, in the first 24h; and 1.5ml/kg/BSA%, with urinary output 40-50ml/h, in the second 24h. It should be emphasized that resuscitation should be started as early as possible,and adequate amount of fluid replacement is especially important during the first 2-3h postburn.There was no obvious relationship between the incidence of visceral complications and the total amount of fluid replaced during the resuscitation. Available data indicated that the amount of fluid calculated on the basis of our formula neither increased the incidence of early pulmonary edema nor influenced its development.As far as prevention of pulmonary edema was concerned, it did not seem justifiable to restrict the amount of resuscitation fluids. It was also noted that fluid therapy alone would not prevent entirely the development of postburn renal insufficiency.
7.A retrospective study of 548 cases of erectile dysfunction treated by penile prosthesis implantation

Objective To evaluate the different penile prosthesis for the management of erectile dysfunction. Methods Of the 548 cases,melleable prosthesis has been implanted in 228(41.6%),one piece inflatable prosthesis(Dynaflex) in 45 (8.2%),two piece inflatable prosthesis (Mentor) in 2 and three piece inflatable prosthesis in (AMS 700 CXM) 273 (50.2%). Results The overall complication rate has been 5.1%(28/548),nonmechanical in 8 (1.5%) and mechanical complication in 20 (3.6%). The mechanical complication rate of AMS malleable 600 and AMS 700 CXM were 1.3% (3/228) and 5.5% (15/273) respectively.The cylinder leakage of AMS 700 CXM was 3.7% (10/273). Conclusions The mechanical function and concealment have been significantly improved with AMS 700 CXM whereas improvement of the cylinder quality is yet needed.
8.Effect of acetylcysteine on the inflammation and oxidative stress in elderly patients with chronic obstructive pulmonary disease

Objective To investigate the effect of acetylcysteine on the inflammation and oxidative stress in elderly patients with chronic obstructive pulmonary disease(COPD).Methods Sixty patients with COPD were divided into two groups by random mechanical sampling method (30 cases each group).The control group was treated with conventional therapy and the treatment group added acetylcysteine for 8 weeks.The levels of tumor necrosis factor- t (TNF- α ),interleukin-6 (IL-6),interleukin -8 (IL-8),superoxide dismutase (SOD),malondialdehyde (MDA) were monitored and analyzed before and after treatment.Results The levels of SOD,MDA were (51.08 ± 7.80),(75.09 ± 8.03) μ U/L and (7.13 ± 0.89),(4.51 ±0.61 ) μ mol/L before and after treatment in the treatment group,and (52.12 ± 7.31 ),(65.16 ± 8.01 ) μ U/Land (7.11 ± 0.87),(6.21 ± 0.78 ) μ mol/L in the control group.There were significant differences before and after treatment in two groups (P<0.01 or <0.05),and there were significant differences after treatment between two groups (P < 0.05).The levels of IL-6,IL-8 and TNF- α were significant differences before and after treatment in the treatment group [ ( 18.25 ± 7.24) ng/L vs.(29.02 ± 6.70) Ng/L,( 115.28 ± 13.76 ) ng/Lvs.(148.99 ± 16.61) ng/L,(20.43 ±3.92) ng/L vs.(32.32 ± 8.18)ng/L] (P <0.05).But there was nosignificant difference before and aftertreatment in the control group [ (25.25 ± 7.55 )ng/L vs.(28.82 ± 7.54)ng/L,( 136.27 ± 12.97) ng/L vs.( 150.21 ± 17.52) ng/L,(28.43 ± 3.92) ng/L vs.(32.56 ± 8.78) ng/L] (P>0.05),there were significant differences after treatment between two groups (P <0.05).Conclusions Acetylcysteine can inhibit inflammation and e hminate free radicals in plasma.Acetylcysteine has beneficial effects on COPD.
9.Present situation and progress of dose verification in radiotherapy.

The dose verification methods in advanced radiotherapy are elaborated in this paper. The usage and application results for various dosimeters in dose verification are explained. As a theoretical method, Monte Carlo simulation, which has been developed greatly in recent years based on the technical progress in computer science, can be also used in dose verification with unique advantages. On the other hand, the principle of dose verification on proton and heavy-ion therapy is discussed briefly. Finally, the evaluation criteria for verification and the future development for dose verification are presented.
