1.Thalidomide inhibits the expression of hypoxia-inducible factor-1?in synovial tissues of rats with collagen-induced arthritis
Chinese Journal of Rheumatology 2001;0(04):-
Objective To observe the therapeutic effect of thalidomide(Thd)on rheumatoid arthritis and its relation with the expression of hypoxia-inducible factor-1?(HIF-1?).Methods Female SD rats were randomly divided into two groups:a normal group and a model group. Each group was further divided into three subgroups:the control group, the vehicle group and the Thd group. The model group was subjected to collagen-induced arthritis. The Thd group and the vehicle group were given Thd(600 mg/kg)once daily and its vehicle DMSO respectively. The profile of arthritis was assessed by observing the clinical symptoms and histopathology and a qualitative clinical score of arthritis. The expression of HIF-1?in synovial tissuses was further detected by Western blot and immunohistochemistry technics. Results Compared to the control group with arthritis, clinical signs and pathological changes of arthritis were improved one and two weeks after ad- ministration of Thd respectively, and the qualitative clinical score of arthritis was evidently lowered. The ex- pression of HIF-1?in the inflammed synovial tissuses was found to be enhanced(P

Objective To analyze the effect of implementing traditional Chinese medicine (TCM) clinical pathway on family sickbed patients. Methods Those family sickbed patients with herpes zoster or facial neuritis were involved in this study.Their medical records and data of ‘Family sickbed quality control scale’ were collected,in which there are methods of TCM clinical pathway, decoction of the Chinese Medicine, ingredients and treatment procedures of the topically used TCMs,and acupuncture treatment procedures were documented. The family sickbed days, prognosis, informed rate and satisfaction rate were analyzed before and after TCM clinical pathway treatment. Results In the treatment group, the family sickbed days (11.67± 1.91)d and referral rate 12.20% (10/82) were significantly lower (P< 0.01) than those in the control group [(1 3.88 ± 2.61) d,50.00% (41/82) respectively].The rate of cure87.80% (72/82),the rate of informed patient 97.56% (80/82) and the rate of satisfaction91.46% (75/82) were significantly higher(P< 0.01)than those in the control group [50.00% (41/82)、65.85% (54/82)、50.00% (41/82) respectively].Conclusion The clinical pathways for family sickbed patients with herpes zoster or facial neuritis were effective.

AIM: To investigate the effect of atorvastatin ( AT) on the release of endothelial microparticles (EMP) and myocardial apoptosis in the rats with myocardial infarction .METHODS: SD male rats (n=24) were ran-domly divided into 3 groups:sham operation ( sham) group , myocardial infarction ( MI) group and MI+AT group. The rat model of acute myocardial infarction was prepared by coronary artery ligation .At 2 h and 24 h after modeling , the pe-ripheral blood was collected to detect creatine kinase-MB (CK-MB) and cardiac troponin T (cTnT).The circulating levels of EMP were measured by flow cytometry .The myocardial apoptosis was detected by terminal deoxynucleotidyl transferase -mediated dUTP nick end labeling (TUNEL) assay.RESULTS: At 2 h after modeling, the level of CK-MB was signifi-cantly increased in MI group compared with sham group , and the level of EMP and the myocardial apoptotic rate were sig-nificantly increased in MI group and MI +AT group compared with sham group .At 24 h after modeling , the level of EMP was significantly increased in MI group compared with sham group .The levels of CK-MB, cTnT, EMP and the myocardial apoptotic rate were significantly decreased in MI +AT group compared with MI group .Moreover , the level of CK-MB in MI group was significantly increased at 24 h compared with that at 2 h after modeling .The levels of CK-MB, cTnT and EMP were significantly decreased in MI +AT group at 24 h compared with those at 2 h after modeling .CONCLUSION: Ator-vastatin may reduce the level of EMP and the myocardial apoptotic rate in the rats with acute myocardial infarction , indica-ting that atorvastatin plays a role in protecting endothelium .

Objective: To investigate the effect of oxLDL and VitE on the expression of CD40 and CD40 ligand(CD40L) in cultured human monoc ytes. Methods: The expression of CD40 and CD40L on monocytes su rface were measured by indirect immunorescence technique in combination with flo w cytometry. Results: Low concentration of oxLDL(≤200 μg/L) significantly increased the expression of CD40 and CD40L in a dose and time dep endent manner. High concentration (>200 μg/L)of oxLDL markedly reduced the exp ression of CD40 and CD40L. When VitE was added, it significantly reduced the ex pression of CD40 and CD40L on monocytes surface induced by oxLDL in a dose-depe ndent manner. Conclusion:It is an important mechanism that the high expression of CD40 and CD40L induced by oxLDL may be contributed to the for mation of atherosclerosis. Antioxidan VitE can partially inhibit the high expres sion of CD40 and CD40L on monocytes surface induced by oxLDL.

Objective: To investigate the effects of hyaluroni dase (HAase) and hyaluronan (HA) on proliferation of vascular endothelial cells and its mechanism. Methods: The cultured bovine aortic endothel ial cells (BAEC) were treated with HAase or HA. Cell proliferation rate was dete cted by MTT assay. The expression of CD44 and DNA content of the cells were meas ured by flow cytometry (FCM). Results: HAase (50 μg/ml) stimula ted cell proliferation [(50.10±1.23)% vs control, P<0.01], incre ased S phase cell rate and induced the expression of CD44, but HA (100 μg/ml) i nhibited cell proliferation and the expression of CD44. Conclusion: HAase may degrade antiangiogenic HA of extracellular matrix, which may stim ulate proliferation of endothelial cells and enhance the curative effect of grow th factors to myocardial ischemia.
