1.Antitumor activity and toxicityin vivo of iron-fluouracil complex

BACKGROUND:Previous research indicated that iron-fluorouracil-phenanthroline complex has good antitumor activity in vitro, which can inhibit the proliferation of human cancer cels. OBJECTIVE:To detect the antitumor activity and toxicity of iron-fluouracil-phenanthroline complex, [Fe(5-Fu)2(Phen)SO4],in vivo. METHODS:A total of 40 Kunming mice were randomly divided into four groups, which were intraperitoneally injected with 72, 102.9, 147, 210 mg/kg [Fe(5-Fu)2(Phen)SO4] and the half lethal dose of the complex was detected. One day after the establishment of mouse S180 sarcoma models, the model mice were randomly divided into eight groups, and administered with the intraperitoneal injection of 15 mg/kg (low dose group), 30 mg/kg (middle dose group), 60 mg/kg (high dose group) [Fe(5-Fu)2(Phen)SO4], normal saline (negative control group), cisplatin (positive control group), 5-fluorouracil, iron-salt and phenanthroline, respectively. The injection was done once a day, lasting for 7 days. The weight of sarcomas, body weight, the main organ coefficient and histopathological changes of the main organs were detected. RESULTS AND CONCLUSION: The half lethal dose of [Fe(5-Fu)2(Phen)SO4] was 103.9 mg/kg. Compared with the negative control group, high dose group, positive control group and 5-fluorouracil could significantly inhibit the growth of the tumor (P< 0.05 orP< 0.01), and the effect of high dose group was the most obvious (P < 0.01). Compared with cisplatin, 60mg/kg [Fe(5-Fu)2(Phen)SO4] had a weaker inhibitory effect on the kidney, but higher inhibitory effect on the liver, spleen and thymus, indicating the complex has a lower nephrotoxicity, but stronger immunotoxicity and hepatotoxicity than cisplatin.
2.Vertebral bodies implanted with biodegradable reticulated balloon and calcium phosphate cement:Changes in the vertebral biomechanics

BACKGROUND:It has become a focus to look for new vertebral body fil ing materials which have the biomechanical property, biological activity and low cement leakage rate.
OBJECTIVE:To investigate the biomechanical characters and cement leakage rate of the vertebral bodies implanted with biodegradable reticulated bal oon and calcium phosphate.
METHODS:Thirty-two vertebral bodies from pigs were randomly divided to four groups. For A group, 2.5-3.0 mL polymethacrylate cement was injected into the body through a unilateral thoracic pedicle pathway;for B group, 2.5-3.0 mL calcium phosphate cement were injected by the same protocol;for C group, the biodegradable reticulated bal oons fil ed with 2.5-3.0 mL calcium phosphate cement were implanted;D group, including normal vertebral bodies, was designed as controls. Leakage of bone cement was observed in each group. The load-shift curves were recorded by an electronic universal testing machine (SCHENCK RSA-250).
RESULTS AND CONCLUSION:The stiffness and strength of A group were significantly higher than those of D group (P<0.05), and the stiffness and strength of B group were statistical y lower than those of D group (P<0.05). The stiffness and strength of C group, otherwise, were similar with those of D group (P>0.05). Cement leakage rate of C group was lower than that of A or B group (P<0.05). Vertebral bodies implanted with biodegradable reticulated bal oons may lead to similar biomechanical characters as the normal vertebral bodies and reduce the cement leakage rate.
3.Clinical efficacy of combined hemihepatectomy for hilar cholangiocarcinoma

Background and purpose: Because of the aggressive nature of hilar cholangiocarcinoma and the absence of effective adjuvant therapy, surgical radical resection offers hilar cholangiocarcinoma patients the only choice. Research focus include preoperative assessment, the use of preoperative biliary drainage, the range of hepatic resection, and the range of lymphadenectomy. To investigate the clinical experience and efifcacy of combined hepatectomy in the treatment of hilar cholangiocarcinoma. Methods: Two hundred and seven patients with hilar cholangiocarcinoma treated surgically in the First Afifliated Hospital of Kunming Medical University form Jan. 2007 to Oct. 2013 were retrospectively analyzed. Results:Of the 207 patients, 125 patients who received radical resection (R0 resection) and the curative resection rate was 60.4%. One hundred and iffty-six cases were treated in combined hepatectomy group, 51 cases in non-hepatectomy group, the rate of R0 resection was 70.5%in hepatectomy group and 29.4%in non-hepatectomy group, and the difference was signiifcant (P<0.01). Two patients died perioperatively, the main postoperative complications included hepatic function insufifciency and bile leakage. One hundred and seventy-two patients were followed up, the median survival time of the 102 patients who received R0 resection was 45 months, and the 1, 3, 5 year survival rates were 96.1%, 59.1%and 17.2%. The median survival time of the 70 patients who received R1-2 resection was 26 months, and the 1, 3 year survival rates were 81.3%and 19.2%, and none of the patient survived for over 5 years. The survival rate of patients who received R0 resection was signiifcantly higher than those who received R1-2 resection (χ2=39.121, P<0.01). In the hepatectomy group was awarded the R0 resection in patients with postoperative 1, 3, 5 year survival rate was 97.8%, 63.9% and 18.0%, in non-hepatectomy group received R0 resection in patients with postoperative 1, 3, 5 year survival rate was 83.3%, 20.8%and 8.3%. There were signiifcant differences in the postoperative survival rate between both group (χ2=5.988, P=0.014). Conclusion:Radical excision is the key to improve the long term survival. Combined hemihepatectomy and standardized lymph node resection has signiifcantly improved the radical resection rate and the efifcacy of treatment for hilar cholangiocarcinoma.
4.The experimental study of the effect of dexamethasone,lidocaine and contrast medium on the activity of collagenase

Objective To study the effects of hormone,anesthetie and contrast medium on the activities of collagenase lysis.Methods Nuclear tissues were divided into equal amount for different groups and same units of collagenase were used for the lysis.The only difference in the control group from experimental groups was that there were no dexamethasone,lidocaine or omnipaque but existing in experimental groups.Twenty four hours later,the concentrations of the hydroxyproline were determined in different groups and the data were analyzed by statistical software with computer.Results 1.The concentrations of hydroxyproline in the dexamcthasone group,lidocaine group and omnipaque group were significantly lower than that of control group,especially that of lidocaine group,P value was<0.05 or 0.01.2. The concentrations of hydroxyproline in the dexamethasone+lidocaine group,dexamethasone+omnipaque group,lidocaine+omnipaque group and dexamethasone+lidocaine+omnipaque group were significantly lower than that of control group;and simultaneously lower in dexamethasone group,lidocaine group,omnipaque group respectively;P value was also<0.05 or 0.01.Conclusion Dexamethasone,lidocaine and omnipaque can individually inhibit the activity of collagenase at different degrees,so they shouldn't be used together with collagenase in treating the lumber disc herniation.
5.Rapidly Progressive Glomerulonephritis Induced by Antithyroid Agents in Children

Objective To explore the clinical characteristic of rapidly progressive glomerulonephritis(RPGN) induced by the antithyroid agents[propylthiouracil(PTU) and methimazole(MMI)].Methods The analysis was made according to the drug histories,clinical manifestation,serology investigations,renal biopsy in 2 children.Results Two cases presented with gross hematuria,proteinuria and renal dysfunction.One case with antineutrophil cytoplasmic antibody(ANCA)-positive and vasculitis lesion with cresent in the renal biopsy;another presented with mesengial proliferation and cresent.They both had been diagnosed RPGN induced by antithyroid agents for 5 years because of Graves disease.The treatment included that PTU and MMI were withdrawn and then combined prednisone with mycophenolate mofetil(MMF)were applied.One year after treatment,the urine analysis and renal function returned to normal.Conclusions The antithyroid agents may induce RPGN.A early diagnosis and immunosuppressive therapy can improve its prognosis of the disease.
6.Changes of Serum IgE and Tryptase in Anaphylactic Shock Rats.

OJECTIVE:
To explore the changes of serum IgE and tryptase caused by anaphylactic shock rats and discuss the relation to PMI and preservative environment of corpse and specimen.
METHODS:
Rats were used for establishing anaphylactic shock models and randomly divided into room temperature group, refrigeration group, frozen group, manual hemolysis group, specimen preservation group. And the control group was also established. The blood samples were collected after rats were sacrificed. The degree of hemolysis was graded according to the color of the upper layer of the serum. The mass concentration of IgE and tryptase in each group was detected by ELISA.
RESULTS:
The levels of serum IgE and tryptase in anaphylactic shock dead rats were higher than that of the control group. Room temperature and frozen made obviously differences on the levels of serum IgE and tryptase with various PMI. The levels of serum IgE and tryptase in refrigeration group showed relatively stable. The levels of serum tryptase and IgE were elevated with differently increasing hemolysis. The levels of serum IgE and tryptase showed no obvious changes during the specimen kept under different temperature conditions for 25 days.
CONCLUSION
Serum IgE and tryptase obviously increased in anaphylactic shock rats. However, the levels were influenced by PMI and environmental temperature, especially under the conditions of room temperature and frozen.

7.Effect of serum from acute myocardial infarction rat on inducing rat bone marrow mesenchymal stem cells into cardiomyocytes

AIM: To induce rat bone marrow mesenchymal stem cells (BMSC) into cardiomyocytes and investigate the influence of serum coming from acute myocardial infarction (AMI) rat on the procedure. METHODS: The passage 3 BMSC were divided into six groups: groupⅠwas control group; groupⅡwas induced with 5-azacytidine; group Ⅲ was induced with 5-azacytidine and serum from AMI rat; group Ⅳ was induced with 5-azacytidine and serum from normal rat; group V and group Ⅵ were induced with serum from AMI rat or normal rat. The cardiac troponin T, GATA-4 and desmin were detected 30 days after induction. RESULTS: After inducing by 5-azacytidine, 5-azacytidine and two kinds of serum, some cells in the three groups differentiated into cardiac like cells. The expressions of cardiac troponin T, GATA-4 and desmin were positive in cells differentiated from BMSC. The troponin T expression in control group and group inducing by AMI serum alone were negative but GATA-4 and desmin expressed weakly. Some cells induced with 5-azacytidine and serum were slowly beating 2 weeks after induction, but the cells induced with 5-azacytidine alone was not beating.CONCLUSION: Serum from AMI can not induce BMSC to differentiate into cardiomyocytes, but it promotes BMSC differentiate into cardiomyocytes induced by 5-azacytidine and facilitate the differentiated cells to mature.
8.Effects of a combined regime of auricular-plaster and body acupuncture in treatment of cervical spondylosis of vertebral artery type.

OBJECTIVETo study effects of a combined regime of auricular-plaster and body acupuncture in treatment of cervical spondylosis of vertebral artery type and make a preliminary revelation of the mechanism.
METHODSNinety-two patients were randomly divided into 2 groups, the treatment group (n = 56) received the combined regime of auricular-plaster and body acupuncture, and the control group (n = 36) received treatment with body acupuncture. Clinical symptoms and signs, therapeutic effect and some indexes about vertebrobasilar hema-kinetics and hema-rheology were investigated before and after treatment.
RESULTSThe treatment group was better than the control group in the clinical overall effective rate (89.29%) and the clinically control rate (17. 85%), and in improving the following indexes, including dizziness and headache, the vertebrobasilar volume and rate of blood flow etab and IR (P < 0.05).
CONCLUSIONSA combined regime of auricular-plaster and body acupuncture ameliorates not only main signs but also some indexes about vertebrobasilar hema-kinetics and hema-rheology. This treatment is an effective therapy for cervical spondylosis of vertebral artery type both in Malaysia and in China.

9.Genetic diversity analysis of Andrographis paniculata in China based on SRAP and SNP.

In order to reveal genetic diversity of domestic Andrographis paniculata and its impact on quality, genetic backgrounds of 103 samples from 7 provinces in China were analyzed using SRAP marker and SNP marker. Genetic structures of the A. paniculata populations were estimated with Powermarker V 3.25 and Mega 6.0 software, and polymorphic SNPs were identified with CodonCode Aligner software. The results showed that the genetic distances of domestic A. paniculata germplasm ranged from 0. 01 to 0.09, and no polymorphic SNPs were discovered in coding sequence fragments of ent-copalyl diphosphate synthase. A. paniculata germplasm from various regions in China had poor genetic diversity. This phenomenon was closely related to strict self-fertilization and earlier introduction from the same origin. Therefore, genetic background had little impact on variable qualities of A. paniculata in domestic market. Mutation breeding, polyploid breeding and molecular breeding were proposed as promising strategies in germplasm innovation.

10.Advancement of Salvia miltiorrhiza on reversing tumor resistance and chemotherapy sensitization

At present, chemoresistance is one of the leading causes of chemotherapy failure but there is no effective drugs or strategies to reverse the resistance of chemotherapy. In recent years, the anticancer effect of Salvia miltiorrhiza receives wide attention of researchers. The major bioactive constituents in Sal-via miltiorrhiza can be divided into two groups: liposoluble com-ponents ( tanshinoneⅠ, tanshinoneⅡA, dihydrometanoneⅠ and cryptotanshinone, etc. ) and hydrophilic components (salvianol-ic acid A, salvianolic acid B and protocatechualdehyde, etc. ). Recent studies have found that these components not only play different anti-tumor activities, but showed significant reversal re- sistance effects on drug-resistant tumor cells. Besides, with the combination of chemotherapy drugs, they have significant attenu-ated toxicity and synergistic effect. In this paper, we summarize the researches pertaining to the effects of Salvia Miltiorrhiza on the reversal of drug resistance and chemotherapy sensitization in recent years, providing references for the development of Salvia miltiorrhiza as multidrug resistance reversal agents and its ration-al application in clinic.
