2.Role of interleukin-4 receptor in renal fibrosis following renal ischemia-reperfusion injury in mice

Objective To evaluate the role of interleukin-4 receptor (IL-4R) in renal fibrosis following renal ischemia-reperfusion (I/R) injury in mice.Methods Twelve male wild type BALB/C mice and 12 IL-4Rα gene-knockout mice,aged 8-10 weeks,weighing 20-30 g,were used in the study.The mice of either type were divided into 2 groups (n =6 each) using a random number table:sham operation group (group S) and group I/R.In group I/R,renal I/R was induced by occlusion of the right renal artery for 1 h with atraumatic microclips followed by 2 weeks of reperfusion.The right renal artery was only isolated in group S.At 2 weeks of reperfusion,blood samples were taken from the orbital vein for determination of the concentrations of serum blood urea nitrogen (BUN) and creatinine (Cr).The renal tissues were obtained,and the renal fibrosis area was measured by Sirius Red staining.The expression of fibronectin (FN),collagen Ⅰ (COL-Ⅰ) and α-smooth muscle actin (α-SMA) in renal tissues was detected by immunofluorescence.The expression of signal transducer and activator of transcription 6 (STAT6) and phospho-STAT6 in renal tissues was determined by Western blot.The ratio of phoshop-STAT6 to STAT6 was calculated to reflect the phosphorylation of STAT6.Results Compared with group S of wild type mice,the serum BUN and Cr concentrations and renal fibrosis area were significantly increased,the expression of FN,COL-Ⅰ and α-SMA in renal tissues was significantly up-regulated,and the phosphorylation of STAT6 in renal tissues was significantly increased in group I/R of wild type and IL-4Rα KO mice (P<0.05).Compared with group I/R of wild type mice,the serum BUN and Cr concentrations and renal fibrosis area were significantly decreased,the expression of FN,COL-Ⅰ and α-SMA in renal tissues was significantly down-regulated,and the phosphorylation of STAT6 in renal tissues was significantly decreased in group I/R of IL-4RαKO mice (P<0.05).Conclusion The mechanism of renal fibrosis following renal I/R injury is partially related to IL-4R,and IL-4R results in renal fibrosis through promoting activation of STAT6 signaling pathway in mice.
3.Study on Cleavage in vitro of HPVll/E2 Gene by Specific Ribozyme

In order to study the pathogenesis of HPVl1 and seek for a therapeutic approach of the disease caused by HPV1l. The HPVl1/E2 644bp was amplified by PCR with HPVll plasmid DNA. pGEM-T Easy was used as vector and a clone pTV-644 was obtained.The inserted DNA sequencing was carried out after selecting and identification. According to the hammerhead structure described by Symon's, the possible secondery cleavage sites on HPVll/E2 mRNA were analyzed and the secondery structure of substrate was predicted by computer and ribozyme, excluding analogous sequence of substrate combined with ribozyme were found in the mRNA.The hammerhead ribozyme of RZ2777 against HPV1l/E2 mRNA was seleted to carry out cleavage reaction in vitro. Results of the experiment showed that 644bp substrate derived from HPVll/E2 can be cleavaed site-specifically by ribozyme in vitro. The cleavage activity showed over 85% by choosing the optimun reaction condition, which was not affected by two cis-ribozymes on both 3′-and 5′-ends released by self cleavage,but two flan-king sequences of target RNA influenced the cleavage activity. Results demonstrated that the ribozyme will become a highly effective and specific therapy against HPVll infection.
4.Effect of Clinic Treatment by Tropicamide Esculin-digitalisglycosides Combination Eye Drops on Low Myopia of Children

Objective To investigate the effects of tropicamide and esculin-digitalisglycosides combination eye drops on low myopia of children (age ranging from 6 to 14). Methods Eighty children with 160 eyes of low myopia in the outpatient department of ophthalmology of our hospital were chose from July 2010 to April 2013, then the patients were equally divided into four groups:the control, esculin-digitalisglycosides eye drops,tropicamide,and tropicamide and esculin-digitalisglycosides combination groups. The naked vision and myopia correction of each group were compared before and after treatment for two weeks, and the treatment effects were compared among four groups. Results (1) The results showed there were no significant changes before and after treatment in the control, esculin-digitalisglycosides eye drops and tropicamide groups ( >0.05); (2) On the contrast, the tropicamide and esculin-digitalisglycosides combination have significantly improved naked vision and myopia correction after 2 weeks ( <0.05);(3) Prior to the treatment,there was no significant difference among four groups (<0.05);(4) After treatment,group under tropicamide and esculin- digitalisglycosides combined was statistical different from the other groups treated with other three eye drops ( <0.05);However, there was no statistical difference among groups in these three eyedrops. Conclusion Esculin-digitalisglycosides combined with tropicamide was the most effective treatment for low myopia of children.
5.Use of Antimicrobials in Non-surgical Departments

OBJECTIVE To investigate and analyze the application of antibiotics in non-surgical departments in our hospital.METHODS To make a survey of the use of antimicrobials in non-surgical departments from a total of 548 randomly selected medical records during Jan-Mar 2007,the data of reasonable use of antibacterials were obtained after evaluation and analysis.RESULTS Of the 548 patients in our study,229(41.79%)used antimicrobials.The use of antimicrobials for infection prevention was in 46 cases(20.09%),for infection treatment was in 183 cases(79.91%).From them 95 cases(41.48%)used a combination of two and more antimicrobials.The most widely used antibacterials were penicillins,cephalosporins,and quinolones.CONCLUSIONS The use of antimicrobials is generally reasonable,and meets the safe,effective,and economical goal in our hospital.
6.Antibiotics Usage in Outpatients

OBJECTIVE To investigate the use of antibiotics in our Outpatient Department during 2006-2007,and to analyze and evaluate the trend and rationality.METHODS The data about consumption quantity and consumption sum of antibiotics in our Outpatient Department during 2006-2007 were retrieved by means of computer management software in the department of pharmacy for the analysis of the clinical use of antibiotics using the method of defined daily dose(DDD).RESULTS Fluoro quinolones,cephalosporins and plant antibiotics have dominated the first places among the antibiotics used in our hospital,and the consumption of cephalosporins kept increasing year by year.CONCLUSIONS The use of antibiotics in our hospital should be further standardized to ensure the drug using safe,efficient and economic in our hospital.
7.Rational Usage of Antibiotics Prophylaxis in Department of Obstetrics and Gynecology During Perioperative Period

OBJECTIVE To promote clinical rational use of antibiotic prophylaxis through surveying it in Obstetrics and Gynecology Department (OG) by pharmacists. METHODS Data of 223 cases using of antibiotic prophylaxis in OG were retrospectively analyzed. According to unreasonable use of antibiotic prophylaxis,the clinical pharmacists put out reasonable measures and directed treatment in succedent 206 patients. RESULTS The rate of unreasonable use of antibiotic prophylaxis in 223 cases was 100% and rate of infection after operation was 9.87%. After giving intervention,the rate of unreasonable use of antibiotic prophylaxis and infection were reduced to 3.88% and 1.46%,respectively (P

Objective Horizontal rotating bioreactor is a kind of advanced bioreactor,which is the basic and important facility in tissue engineering.In this paper,we developed a controlable horizontal rotating bioreactor system,which can supple choiceness environment condition for cells culture.Methods System used PWM modulation to drive DC-motor,used sensor to detect and control the temperature of bioreactor reactor,and the system connected with PC to remote watch test and control.The system primarily achieved distributed control.Results The biology characteristic of the cells that were cultivated by this system improved greatly.Conclusion This practices of realization controlable horizontal rotating bioreactor system have bigger value in applications.
9.Application of virtual touch tissue quantification technique in staging of chronic kidney disease

Objective To investigate the application value of virtual touch tissue quantification (VTQ) technique in staging of chronic kidney disease (CKD). Methods VTQ examination was performed on a random sample of 96 CKD patients at different clinical stages and 45 healthy volunteers (control group). Shear wave velocity (SWV) of renal parenchyma was measured and compared. Correlation between SWV and clinical stage and glomeru-lar filtration rate (GFR) of CKD patients was analyzed. Results The SWV value of renal parenchyma decreased gradually, along with the progression of CKD disease and impairment of renal function. There was no significant difference(P>0.05)between the control group and the CKD1 group ( P=0.061), the CKD2 group and the CKD3 group (P = 0.238), and the CKD4 group and the CKD5 group (P = 0.624). The difference of SWV between the remaining groups was statistically significant(P < 0.05). SWV of renal parenchyma was negatively correlated with CKD stage (r =-0.65, P < 0.001), which means the higher the stage of CKD was associated with the lower the SWV. SWV was positively correlated with GFR (r = 0.73, P < 0.001), which means the lower the GFR was related to the lower the SWV. Conclusion VTQ technique can noninvasively reflect the changes of renal parenchyma elas-ticity and evaluate the impairment of renal function in CKD patients. It can provide a new quantitative evaluation index for the clinical staging of CKD.
10.THE NUTRITIONAL EVALUATION AND CLINICAL TRIALS OF A TOTAL ENTERAL NUTRIENT PREPARATION

The composition, nutritional evaluation and clinical trials of a nutritionally complete enteral preparation were presented. It was composed of glucose oligosaccharides, emulsified fat, solubilized proteins, twelve vitamins, certain minerals and trace elements. Rats were forced to negative nitrogen balance by means of protein-free diet. Body weight and serum total protein declined accordingly. When the feed was changed to this preparation in one group, and a routine one with equal nitrogen content in another, nitrogen balance was turned to positive, body weight increased, serum total protein returned to normal in both groups. The increments of nitrogen balance and body weight were, however, higher significantly in the group fed with the preparation than that with routine diet. The biological value, net protein utilization and protein efficiency ratio of the solubilized proteins in the preparation were higher than casein in the routine feed. Twenty surgical patients having the preparation as the sole source of nutrients could take around 8.37MJ(2000 kcal)a day. Nitrogen balance turned from negative to positive in an average time of 3.6 days. Body weight, serum total protein, serum albumin were all increased. Fourty patients took this as dietary supplement and the similar results were observed too. Clinical effects were excellent in all cases, especially for healing of surgical wounds and fistula. Incidence of diarrhea was 3.3%.
