1.New progress in study of inflammatory reflex

Proinflammatory cytokines induced by peripheral immune challenge can activate the inflammatory reflex, which results in negative-feedback control of inflammation .Some recent studies revealed that it is the splanchnic nerve , not the vagus nerve , to constitute the efferent arm of the inflammatory reflex .Further researches are needed to iden-tify the neural construction and regulatory mechanism of the inflammatory reflex , which might be harnessed for the treatment of inflammation and the development of anti-inflammatory drugs .
2.Statistical analysis on papers of Cancer Hospital, Chinese Academy of Medical Science from 2004 to 2013

Objective We analyzed the current status of research publications and its citation in the Cancer Hospital of Chinese Academy of Medical Sciences,aimed to provide information for policy making in the office of research management.Methods We analyzed the number of published,cited frequency and subject classification for the research articles published in the Cancer Hospital,Chinese Academy of Medical Sciences from 2004 to 2013.Results A total of 3749 research papers had been published from during the study period; cited frequency was 12633.Papers were mainly focused on three types of study,including cancer treatment,diagnosis,etiology and prevention.Conclusions Quantity of publications from 2004 to 2013 in Cancer Hospital,Chinese Academy of Medical Sciences were improved significantly.The number of SCIE papers,cited papers and citation frequency increased as well,the number of articles cited and cited overall rate was essentially flat.It suggested that over the past decade academic research capabilities of Cancer Hospital in the field of cancer research continues to improve,however we should identify the existing problems and explore the strategy to improve the research ability.
4.Analysis of application for and granted NSFC projects at Cancer Institute and Hospital of Chinese Academy of Medical Sciences in the 11th five-year plan period

In order to evaluate the overall situation of projects granted by National Natural Science Foundation of China (NSFC) in the 11th five-year plan period,the data about applications and approved NSFC projects at Cancer Institute and Hospital of Chinese Academy of Medical Sciences (CIH CAMS) were analyzed and compared with those of the 10th five-year plan period.It showed that the number of granted NSFC projects increased and the structure of program type maintained during the 11th five-year plan period.The program researchers were younger and had better education background.The quality and quantity of both the General Program projects and Young Scientists Fund projects were at high level.
6.Study on inhibiting expression of IGF1R in hepatocellular carcinoma cells by constructing shRNA eukaryotic expression vectors

Objective To construct short-hairpin RNA (shRNA) eukaryotic express vectors targeting of insulin like growth factor-1 receptor (IGF1R) gene, and to explore the changes of adhesion, invasion and FAK protein expression of MHCC-97 H hepatocellular carcinoma cells with RNA interference. Methods The shRNA oligonucleotide fragments were designed and synthesized based on the sequence of IGF1R mRNA. Double strands were formed after annealing and inserted into pGCsi-U6-Neo-GFP vector. The recombinant was transformed into Stable 3, then plasmids were extracted and identified by restriction enzyme and sequencing analysis. The most effective vectors were selected by RT-PCR and Western blotting after transfecting 293T cells. The best one was used to transform MHCC-97H cells which were selected with G418 to obtain positive colons. The changes of adhesion, invasion and FAK protein expression in MHCC-97H cells were studied. Results The restriction enzyme analysis and sequencing analysis demonstrated that shRNA had been inserted into vectors, and their sequences were the same as the design. It was indicated by RT-PCR and Western blotting that the silencing efficacy of IGF1R was 88%. The ability of adhesion and invasion significantly decreased after IGF1R silencing in MHCC-97H cells, and so was the expression of FAK protein. Conclusion IGF1R pGCsi-U6-Neo-GFP shRNA can significantly decrease the abilities of adhesion and invasion in MHCC-97H cells, and inhibit the expression of FAK protein.
7.Effect of small dose ketamine on efficacy of PCIA with sufentanil after abdominal surgery in aged patients

Objective To evaluate the effect of small dose ketamine on the efficacy of intravenous PCA (POA) with sufentanil after intra-abdominal surgery in aged patients.Methods Sixty ASA Ⅰ orⅡpatienm aged 65-82 yr undergoing elective intra-abdominal surgery under general anesthesia were randomized into 3 groups (n=20 each)according to the composition of PCIA solution:group I sufentanil 200 μg in 200 ml of noilnal saline (group S);group μ sufentanil 200μh+ketmine 100 mg in NS 200 ml(group K1)and group Ⅲ sufentannil 200 μg+ketmine 200 mg in NS 200 ml(group K2).A loading dose of 5 ml wag given at the end of operation.The PCIA setting was as follows:backgound infusion 1 ml/h,bolus dose 2 ml,lockout interval 5 min and 4-hour maximum dose 30 m1.If VAS score(0=no pain,10=womt pain)was≥7,pethidine 25 mg was given iv.The total amount of pethidine given within 48 h after operation and postoperative complications including nausea and vomiting and respiratory depression were recorded.Results Small dose ketamine added to the PCIA solution can significantly reduce the amount of pethidine administered after operation in a dose-dependent manner.Conclusion Small dose ketamine can improve the efficacy of PCIA with sufentanil after intra-abdominal surgery in aged patients with no significant adverse effect.
8.Ethical Issues in the Recruitment of Subjects during Phase I Clinical Trial

Subject recruitment is an important part of clinical researches. It should follow the ethical principles of fairness and representativeness. The recruitment process should pay attention to protecting the privacy of sub-jects. Various recruitment materials must be approved by the ethics committee before use. Whether could recruit el-igible subjects during Phase I clinical trials will have a significant impact on the results, which should attract the relevant persons′attention including the ethics committee, researchers, clinical research associate and the sponsor.
9.The effect of dexmedetomidine on oxidative stress and inflammatory cytokines in tourniquet-induced ischemia-reperfusion injury

Objective To investigate the effects of dexmedetomidine (DEX) on oxidative stress and inflammatory cytokines caused by tourniquet-induced ischemia-reperfusion injury at limbs.Methods Eighty patients who had been scheduled for lower limb operation with tourniquet were assigned equally by sequence number to use or not use DEX (DEX or control group,n =40).Combined spinal-epidural anesthesia was performed in both groups.In the DEX group,DEX intravenous infusion was started immediately after the femoral vein cannulated at a dose of 1 μg/kg for 10 minutes,followed by 0.5 μg/kg · h until the end of operation,whereas the control group received an equivalent volume of 0.9% saline.At 10 min before tourniquet inflation (T1),10 min (T2),30 min (T3) and 60 min (T4) after tourniquet release,femoral venous blood samples were obtained to measure heart rate (HR),mean arterial pressure (MAP),saturation of pulse oximetry (SPO2),serum malondialdehyde (MDA),serum superoxide dismutase (SOD),serum tumor necrosis factor-α (TNF-α) and interleukin-8 (IL-8) levels in both groups.Results There were no significant differences in HR,MAP or SPO2 at all time points between the 2 groups (P > 0.05).There were no significant differences in HR,MAP or SPO2 at all time points within either group (P > 0.05).There were no significant differences in serum MDA,SOD,TNF-α and IL-8 levels at T1 between the 2 groups (P> 0.05).The serum MDA,TNF-α and IL-8 levels were significantly lower and the serum SOD level significantly higher in the DEX group than in the control group at T2,T3 and T4,respectively (P <0.05).In both groups,the serum MDA,TNF-α and IL-8 levels were significantly higher and the serum SOD level significantly lower at T2,T3 and T4 than at T1,respectively (P < 0.05).Conclusion Dexmedetomidine can reduce the oxidative stress and inflammatory cytokine level which are caused by tourniquet-induced ischemia-reperfusion injury.
10.Study on the expression of nitric oxide synthase 1(NOS1)and mRNA after rat cerebral contusion

The experimental right cerebral contusion model was established in rats.To observe the relationship between the expression of Nitric oxide synthase 1(NOS1)and time elapsed after cerebral contusion,the expression of Nitric oxide synthase 1(NOS1)and mRNA was studied by immunohistochemistry and in situ hybridization at different intervals after cerebral injury.The results indicated that a positive relationship existed between the expression of NOS1 and NOS1 mRNA and the intervals elapsed after brain injury.This method also can be used to distinguish antemortem and postmortem injury rat′s cerebral contusion.So that,the expression of NOS1 and NOS1 mRNA is of great value for timing of brain injury.
