1.Application of St George's respiratory questionaire in evaluating the life quality of community patients with chronic obstructive pulmonary disease

Objective To study the value of St George's respiratory questionaire(SGRQ)in evaluating the life quality of community patients with COPD.Methods The SGRQ score,PEF% were collected from 35 patient with COPD.Paired t test was performed to evaluate the sensitivity of SGRQ to changes of disease severity.Correla- tion coefficients were calculated to evaluate the validity.Results Three parts of the SGRQ score and the total score were significantly correlated with the changes of SGRQ(P<0.001),SGRQ was more sensitive than PEF in evalu- ating the changes of disease severity.The three part of the scale and the total scale were significantly correlated with PEF%.Conclusion SGRQ is a valid,sensitive and feasible method in measuring the quality of life in community patients with COPD.
2.Expression of CD44_S and CD44_(V6) in human ameloblastoma

Objective: To study expression of CD44 S and CD44 V6 in ameloblastoma (AB) and odontogenic keratocyst (OKC) . Methods: S-P method was used to detect CD44 S protein in 77 cases of AB (32 of primary, 39 of recurrent,6 of malignant), 19 cases of OKC and 12 cases of oral normal mucosa;and CD44 V6 protein in 61 cases of AB (24 of primary , 31 of recurrent and 6 of malignant), 19 cases of OKC and 12 cases of oral normal mucosa. Results: Expression of CD44 S and CD44 V6 were detected in cell membrane of stratum spinosum and stratum basale in normal oral mucosa. In 19 cases of OKC, loss of expression of CD44 S was detected in 10 cases, and loss of integrity of CD44 V6 expression was detected in 15 cases. Loss ration of expression and abnormal expression ratio of CD44 S in AB were 27.3% and 63.6%, respectively. That of CD44 V6 in AB were 11.5% and 50.8% respectively. 3.Value of magnetic resonance spectroscopic imaging in differential diagnosis of prostate cancer in central gland and benign prostatic hyperplasia nodus

Objective To explore the metabolic characteristic of prostate cancer (PCa) in central gland with magnetic resonance (MR) spectroscopic, and evaluate the value of MRS in the differential diagnosis of benign prostatic hyperplasia nodus. Methods MR images were performed in 38 cases with prostate disease by 3.0T MR, 11 cases with PCa in central gland and 27 cases with benign prostatic hyperplasia nodus. All the cases were scanned by routine, then by the combined MRS. MRS findings were reviewed in 27 cases with benign prostatic hyperplasia (BPH) and 11 cases with PCa (3 in central gland origin, 8 with large tumor invading both peripheral zone and central gland). (Cho+Cr)/Cit ratios of PCa and BPH were retrospectively measured, (Cho+Cr)/Cit of PCa voxels were compared with that of BPH voxels. Results Significantly higher choline levels and lower citrate levels were observed in central gland of PCa compared with BPH. In the glandular BPH region, the amount of Cit was high; while in the stromal region, the Cit and Cho level was much lower. The average (Cho+Cre)/Cit values of PCa and BHP were 1.94±1.43 and 0. 83±0. 28 respectively, the difference in ratio between both was statistically significant (P<0. 01). Conclusions The combined use of MRI and MR spectroscopy is propitious for differentiating prostate cancer in central gland and benign prostatic hyperplasia nodus and for increasing the diagnostic accuracy of prostate cancer.
4.Effects of stromal cells derived from the normal prostate on the glycolysis of prostate cancer cells.

OBJECTIVETo investigate the effects of prostate stromal cells from different zones of normal prostate tissue on the growth of prostate cancer cells and their action mechanisms.
METHODSWe extracted stromal cells in the fresh normal prostatic tissue derived from the peripheral zone (PZ) or transitional zone (TZ), amplified them in vitro, and used the supernatants of the cells as conditioned media to culture hormone-resistant prostate cancer DU145 cells. We measured the growth curve of the tumor cells using the CCK8 method, determined the number and viability of the cells by trypan blue staining, evaluated their invasiveness by scratch test, and detected the effects of the stromal cells on the key enzymes in the glycolysis of the tumor cells by Western blot.
RESULTSThe conditioned medium with the PZ-derived stromal cells promoted, while that with the TZ-derived stromal cells inhibited the growth of the tumor cells. The former significantly increased, while the latter markedly decreased the expressions of the key enzymes hexokinase 2 (HK-2), pyruvate kinase 2 (PKM-2), lactate dehydrogenase (LDHA), and pyruvate dehydrogenase (PDH) in the glycolysis of the tumor cells.
CONCLUSIONProstate stromal cells from different zones exert different influences on the growth of tumor cells, which may be associated with their different effects on the glycolysis of tumor cells.

5.Mechanisms of Effects of Indol-2,3-dione on Quail Experimental Atherosclerosis

OBJECTIVE:To study the possible mechanisms of the effects of indol-2,3-dione (MW147) on experimental atherosclerosis (AS). METHODS: A total of 120 male quails were randomly divided into six groups: normal control group, model group, lovastatin (79.5 mg?kg-1) positive control group, and MW147 (20, 60, 120 mg?kg-1) groups. The normal control group was fed on normal diet, while the other 5 groups were fed on high lipid diet and treated ig with corresponding drugs for eight weeks. Then the lipid levels including TC, TG, L-DLC and H-DLC in serum and tissues, and the total superoxidedismutase (T-SOD), glutathione peroxidase (GSH-Px), total antioxidant capacity (T-AOC) and malondialdehyde (MDA) were determined. Meanwhile the tissues of aorta and liver were observed under light microscope. Objective To realize K space image reconstruction by using VC++ and Matlab in the development of MRI software.Methods VC++ called FFT(Fast Fourier Transform Algorithm) function of matlab math dlls to realize transforming the data from k space to real space.Results Image reconstruction was realized successfully for the clinical demand.The development circle and programming workload were greatly reduced.Conclusion The mixed programming by VC++ and Matlab can reduce the developing process and improve the reliability of the software.
8.Effect of Vaccinium vitis procyanidin on regulation of glioma cell growth

AIM:To explore the effect of Vaccinium vitis procyanidin on the growth of glioma cells .METH-ODS:Glioma C6 cells were cultured and divided into control and 10, 20 and 40μg/L Vaccinium vitis procyanidin groups . The influence of Vaccinium vitis procyanidin on the growth of C 6 cells was measured by MTT assay and the observation un-der inverted microscope .The apoptotic rate was detected by Annexin V/PI staining .The protein expression of Bcl-2 and Bax was determined by immunocytochemistry .The protein levels of Bcl-2, Bax and caspase-3 were also examined by West-ern blotting .RESULTS:The growth of C6 glioma cells was inhibited by Vaccinium vitis procyanidin at concentrations of 10, 20 and 40 μg/L.The growth was significantly inhibited in 40 μg/L Vaccinium vitis procyanidin group at 24 h and 48 h, and in 20 and 40 μg/L Vaccinium vitis procyanidin groups at 72 h (P<0.01).The density of the cells was decreased when the concentration of Vaccinium vitis procyanidin increased .The apoptotic rate was increased when the concentration of Vaccinium vitis procyanidin increased either .The expression of Bcl-2 was decreased and Bax was increased after 10, 20 and 40 μg/L Vaccinium vitis procyanidin treatments .The ratio of Bax/Bcl-2 was increased when the dose of Vaccinium vitis pro-cyanidin increased (P<0.05 or P<0.01).The expression of Bcl-2 was decreased (P<0.01), and Bax and caspase-3 were increased after 10, 20 and 40 μg/L Vaccinium vitis procyanidin treatments .The ratio of Bax/Bcl-2 was increased when the dose of Vaccinium vitis procyanidin increased (P<0.01).CONCLUSION:Vaccinium vitis procyanidin inhibits the growth of glioma cells by down-regulating Bcl-2 protein and up-regulating Bax protein to activate caspase-3, thus indu-cing apoptosis .
9.Protective Effects of Inonotus Obliquus Polysaccharide on Liver Injury Induced by Anti-tuberculosis Drugs in Mice

Objective To study the protective effect and mechanism of Inonotus obliquus polysaccharide on liver injury induced by isoniazid( INH) combined with rifampicin( RFP) in mice. Methods Forty-eight mice were randomly divided into six groups ( control, model, huganpian, Inonotus obliquus polysaccharide at low, middle, and high dose ) . The mice were administered orally with isoniazid and rifampicin simultaneously except the control. After 2 h,the control and model groups were administered with normal saline,and others were treated with huganpian and Inonotus obliquus polysaccharide,respectively,once a day for 2 weeks. The liver index and serum levels of ALT and AST,malondialdehyde(MDA),superoxide dismutase(SOD) and glutathione peroxidase( GPx) were measured. Pathology examination of liver tissue was performed. Results The activities of ALT and AST, liver index, content of MDA in liver homogenate of mice treated with Inonotus obliquus polysaccharide and huganpian decreased obviously, while the liver SOD activity increased. Histopathological exzamination showed that Inonotus obliquus polysaccharide alleviated the degeneration and necrosis of hepatic cells. Conclusion Inonotus obliquus polysaccharide shows protective effects on mice with hepatic toxic injury induced by isoniazid and rifampicin,which maybe due to its antioxidation effect.
10.Evaluation of renal vascular in living donors before transplantation using dynamic contrast enhanced MR angiography

Objective To explore whether dynamic contrast-enhanced MRA (DCE MRA) can provide an effective assessment of renal vascular in living donors before transplantation.Methods Thirty five healthy living renal donor candidates were scanned on MR system before transplantation.After injection of Gd-DTPA 1 ml in vein, a test-bolus scan was used to get the time delay of Gd-DTPA reaching renal artery.Then, a 3D T1-weighted fast low-angle shot sequence (3D FLASH) was performed in the coronal plane.The 3D FLASH scan would repeat four times with an inter-phase of 10 seconds.Thus, the imaging of the renal arterial, venous and collecting systems were got.Two radiologists observed renal arteries and veins on original imaging and MIP reconstructed imaging.The quality of MR angiography was evaluated on a fivepoint scale and the vascular anatomy or variations of the arterial and venous systems were recorded, using intraoperative findings as a standard of reference.Results The quality for all MRA was good or very good for the most of living renal donors.Among 70 renals, several variations of vascular were found, including 5 left accessory artery, 9 right accessory artery, 3 left proximal arterial branch and 6 right proximal arterial branch.Among 70 renal veins, 1 right accessory veins and 2 left varieocele were observed.One small accessory artery of right kidney was missed with DCE MRA, but identified by operation.Conclusion DCE MRA was noninvasive tool for evaluation of the renal vasculature and variations with high accuracy.It would be a good modality in preoperative evaluation of living renal donors.
