1.Changes in serum osteopontin level and its clinical significance in carcinogenesis and development of gastric cancer

Objective To reveal the change tendency of serum osteopontin(OPN) in carcinogenesis and development of gastric cancer(GC), and explore the usefulness of serum OPN in the diagnosis of GC and the differential diagnosis of early GC and advanced GC.Method Serum OPN levels were detected by enzyme-linked immunosorbent assay, including 70 advanced GC, 24 early GC, 47 dysplasia, 42 intestinal metaplasia, 46 chronic superficial gastritis patients and 38 normal controls.Results Serum OPN level in GC group (57.03 ng/ml) was significantly higher than those in other groups.Serum OPN level in advanced GC group (66.05 ng/ml) was significantly higher than that in early GC group (41.93 ng/ml), serum OPN level in early GC group was significantly higher than those in dysplasia, intestinal metaplasia and chronic superficial gastritis, and serum OPN level in the latter three groups was significantly lower than that in normal controls (P
2.Effect of HELICOBACTER PYLORI infection on apoptosis and proliferation along with P53 gene expression in gastric epithelium

Purpose:To investigate the relationship between HELICOBACTER PYLORI (Hp) and gastric carcinoma, as well as the possible carcinogenic mechanism by Hp. Methods: DNEL technique and immunohistochemical staining were used in the research to study the state of apoptosis, proliferation and p53 gene expression. Total 100 gastric mucosal biopsy specimens, including normal mucosa, Hp negative and Hp positive gastric precancerous lesions as well as gastric carcinomas were studied. Results:There were several apoptotic cells in the superficial epithelium and a few proliferative cells within neck of gastric glands, and no p53 protein expression in normal mucosa. In gastric carcinoma, apoptotic cells accounted for 1.62%, while proliferative cells 4.99%, and P53 protein significantly increased. in metaplasia, the apoptotic index (AI), proliferative index (PI) and positivity of p53 expression in Hp positive group were higher than those in normal mucosa ( P
3.Efficacy of continuous glucose monitoring system combined with insulin pump in diabetic pafients aged 60 years and over

Objective To observe hypoglycemic efficacy of continuous glucose monitoring system (CGMS)combined with continuous insulin infusion(CSII)in the elderly diabetic patients aged 60 years and over.Methods A total 0f 100 elderly diabetic patients were randomly divided into two groups.50 were treated with CGMS or self-monitoring blood glucose(SMBG)and CSII.and whose insulin dose was adjusted based on results of monitoring,and the other 50 with SMBG and insulin pump as controls.whose insulin dose was adiusted based on the results of SMBG for three days.After two weeks.all the patients were monitored by CGMS for blood glucose.Results Mean blood level of glucose and mean amplitude of glycemic excursions(MAGE)were significantly lower in CGMS+CSII group than those in controls[(6.6±2.3)mmol/L vs.(7.5±2.1)mmol/L,and(3.9±0.9)mmol/L vs.(5.1±0.6)mmol/L,respectively,P<0.05 1.Average insulin dose used was significantly lower in CGMS+CSII group than that in controls [(0.64±0.21)U/kg vs.(0.82±0.41)U/kg,P<0.05],and duration of hypoglycemia was less in the former than that in the latter[(20±3)min vs.(40± 9)min,P<0.05 ].Conclnsions CGMS combined with CSII can decrease blood glucose.glucose excursions.average dose of insulin and duration of hypoglycemia in the elderly diabetic patients,as well as prevent occurrence of hypoglycemia.

Microarray technology are being performed more widely than ever before on many areas in lifescience,although the technology is still evolving,the challenge of performing a microarray experiment is no longer in the data generation,but in extracting useful information and utilizing it to get the results with biological meanings.Some methods and tools used for expressional microarray data mining based on previous work were summarized.These methods include gene clustering,GO analysis,regulating pathway analysis,and related algorithm.We hope this can be helpful for those researchers who are implementing expressional microarray for biological analysis.
5.Changes of vasocative intestinal peptide neurons in the myenteric plexus of gastric antrum in rats with se-vere acute pancreatitis

Objective Patients with severe acute pancreatitis ( SAP) always suffer decrease in gastrointestinal motility , even gastrointestinal failure.The article aimed to investigate the changes of vasoactive intestinal peptide (VIP) in the myenteric plexus of the gastric antrum in rats with severe acute pancreatitis ( SAP) complicated by gastrointestinal dysmotility . Methods Twenty Sprague Dawley rats were randomly allocated into two groups:control group and SAP group .Acute pancreatitis was induced by intraductal infu-sion of 5%sodium taurodeoxycholate .24 h after operation , records were made on pathological evaluation of pancreatic lesion , amylase, C-reactive protein ( CRP) , gastric emptying rate and the percentage of VIP neurons in the total neurons . Results Compared with the control group, the gastric emptying rate in the SAP group was significantly lower ([31.49 ±3.96]% vs [70.99 ±1.82]%).The score of pancreatic lesion, amylase, CRP and the percentage of VIP neurons was significantly higher ([14.63 ±1.06]score vs [1.60 ± 0.52]score, [2061.25 ±434.86]U/L vs [360.20 ±30.29]U/L, [140.10 ±17.07]μg/mL vs [35.71 ±6.43]μg/mL, [22.45 ± 4.10]%vs [10.72 ±1.92]%)(P<0.05). Conclusion Plas-ticity took place in VIP neurons in the myenteric plexus of gastric an-trum in rats with SAP complicated by gastrointestinal dysmotility , which was connected with neural mechanism underlying SAP compli-cated by gastrointestinal dysmotility .
6.Periodontal ligament cells from miniature swine grow on a hydroxyapatite scaffold

BACKGROUND:Periodontal tissue engineering technology provides new ideas and new ways for periodontitis-induced bone defect repair. OBJECTIVE:To develop a culture model for the periodontal ligament cells of miniature swine, which was constructed with hydroxyapatite, to investigate the biocompatibility with hydroxyapatite. METHODS:Periodontal ligament cells from miniature swine were harvested by using tissue explant method. Immunofluorescence was used to detect the expression of stromal cel antigen 1 in the periodontal ligament cells of miniature swine. The third passage cells were co-cultured with a three-dimension hydroxyapatite scaffold, and the biological characteristics of the cells were observed under a scanning electron microscope at days 1, 3, 7 of co-culture. RESULTS AND CONCLUSION:The pirmary miniature swine periodontal ligament cells grew wel , and they were positive for stromal cel antigen 1. Under the scanning electron microscope, the periodontal ligament cells of miniature swine grew wel on the hydroxyapatite scaffold at days 1, 3, 7 of co-culture. These prove that the miniature swine periodontal ligament cells, which can be separated using tissue explant method and cultured successful y in vitro, can grow wel on the hydroxyapatite scaffold.
7.Construction of student-centered learning system for undergraduate clinical training in otolaryngology-head and neck surgery

The author proposed student-centered learning system in otolaryngology-head and neck surgery for undergraduate clinical training after exploration and intentions.Four mutual impacted frames were built including integration of teaching philosophy,visualization of training methods,diversification of educational targets and interaction of training courses.Endoscopic navigated learning and multimedia aided training were applied,respective teaching purposes were set and various clinical training courses were introduced to students in their learning of otolaryngology,which were believed to help develop more medical talents with higher comprehensive qualities and better clinical skills.
8.Efficacy and safety of Fosfomycin trometamol in elderly diabetic patients with acute urological tract infections

Objective To evaluate the efficacy and safety of Fosfomycin trometamol(FMT)versus Norfloxacin(FPA)in elderly diabetic patients with acute urological tract infections.Methods From Jan.2012 to Mar.2013,eighty-six patients with uncomplicated and complicated urinary tract infection were randomized into FMT group(n =43)and FPA group(n =43).Patients in both groups were balanced with regard to baseline urinary tract symptoms,fever,leukocytosis,positive urinary culture and disease type(P>0.05).Patients received FMT 3 g daily and Norfloxacin 0.4 g twice daily for a week.Extended treatment was given to 35 patients not cured within a week.Total treatment course should not exceed two weeks.Patient's symptoms,signs,complete blood count,urinalysis,urine culture,pharmacological sensitivity tests,renal and liver function tests were performed.Results After one week of FMT treatment,28(65.1%)cases were cured and 10 cases were improved(1 was with abnormal urinalysis,9 had positive urine culture).In total,in 38(88.4%)patients treatment was as effective;In FPA group,23(53.5%)were cured and 7 cases were improved(7 had positive urine culture).In total,there were 30(69.8%)patients classified as effective.Difference was statistically significant(P<0.05 =.After 2 weeks of FMT treatment,38(88.4%)cases were cured and 2 patients were improved(1 was with abnormal urinalysis,1 had positive urine culture).In total,40(93.0%)patients were classified as effective.In FPA group,25(58.1%)cases were cured and in 8 patients were improved(8 had positive urine culture).In total,there were 33(76.7%)patients classified as effective;Difference was statistically significant(P<0.05 =.Bacterial clearance rate in FMT and FPA group was 88.9% and 72.7%,respectively.Difference was statistically significant(P<0.05 =.The adverse reaction rates were 9.3% and 14.0%,respectively.Difference was not statistically significant(P>0.05).Conclusions FMT is an effective and safe antibiotic in the treatment of urinary tract infections in elderly diabetic patients.
9.Study on correlation between glucose excursions and oxidative stress in the elderly type 2 diabetic patients

ObjectiveTo investigate the correlation between glucose excursions and oxidative stress in the elderly type 2 diabetic patients. MethodsFifty five elderly type 2 diabetic patients (32 males and 23 females) were recruited. All the patients were submitted to continuous glucose monitoring system (CGMS) for three days. According to the result of CGMS monitoring ,the patients were divided into two groups: high glucose excursion group (30 cases) and low glucose excursion group (25 cases). In high glucose excursion group, the therapy was adjusted for 4 weeks based on the CGMS monitoring. After the treatments, the specimens of blood were collected again to detect the related indexes. Results(1) There were no differences in age, blood pressure, fasting blood glucose and glycosylated hemoglobin between the two groups (all P>0.05). The stardard deviation of blood glucose was higher, and the maximun and average amplitude of glycemic excursion were significantly lower in high glucose excursion group than in low glucose excursion group (t= 5. 4620,5. 9416,3. 8281, all P<0.05) ;(2) Compared with low glucose excursion group, plasma concentration of 8-iso-PGF2a was obviously higher in high glucose excursion group[(57.56 ± 3.86)ng/L vs.(34. 21±3. 82) ng/L, t= 18. 221, P=0. 0000)] . (3) Stepwise regression analysis showed that the standard deviation of blood glucose was involved in regression model (β= 1. 959, P= 0. 013).ConclusionsIn elderly patients with type 2 diabetes mellitus, glucose excursion is related with oxidation stress, which suggests that the glucose excursion may be the risk factor for oxidation stress.
