1.Detection of methylation in exon 1 of the survivin gene in condyloma acuminatum (CA) tissue and its significance

Objective To investigate the relationship between the expression of the survivin gene and CpC methylation in exon 1 of the survivin gene in CA tissue, and to study the expression of survivin protein in CA tissue and its modulation mechanism. Methods Tissue samples were obtained from the CA lesions of 30 patients, normal cervix of 10 female controls, and normal foreskin of 10 male controls. Immunohistochemistry was carried out to detect the expression of survivin protein in these specimens, RT-PCR to measure the mRNA expression of survivin, and methylation specific PCR (MSP) to analyze the methylation status of CpG island in the survivin gene exon 1. Results The positivity rate of survivin protein and mRNA was 90% (27/30) and 93.3% (28/30) in CA tissue specimens, respectively, 5% (1/20) and 5% (1/20) in control tissue specimens, respectively; there was a significant difference between the two groups of specimens in both the parameters (x2 = 35.187, 38.437, both P < 0.01). The demethylation of CpG island in the survivin gene exon 1 was observed in 86.7% (26/30) of the CA tissue specimens and in 15% (3/20) of the control tissue specimens (x2 = 10.865, P < 0.01). There was a positive correlation between the demethylation status of CpG island in exon 1 and the mRNA expression of survivin gene (x2 = 13.929, P = 0.014). Conclusions The expression of survivin protein in CA tissues might be associated with the demethylation of CpG island in exon 1 of the survivin gene, and may play a certain role in the development of CA.
2.Analysis of laboratory external quality control of iodine deficiency disorders in Guizhou province in 1999 -2008

Objective To analyze the assessment results of iodine deficiency disorders(IDD) External Quality Control Laboratories at all levels in Guizhou province and the network operation to further standardize and improve the laboratory, improve network performance, provide reliable laboratory quality assurance for epidemiological surveillance and control of IDD and reliable decision-making. Methods The results of different level of IDD labs in Guizhou that took part in the entire country's IDD lab exo-network quality control examination of 1999 - 2008, which organized by the nation iodine deficiency disorder reference lab were analyzed. Results In the past 10 years, except the provincial laboratory examination results of urinary iodine in 2000 was failed, the other results were all qualified in the rest 9 years;iodized salt examination results were qualified. The urinary iodine laboratory response rate of Guizhou provinces and municipalities(state, district), from 1999 to 2002, were 88.9%(8/9), 66.7%(6/9), 77.8%(7/9), 66.7%(6/9), respectively, and the rate was stable at 100% from 2003 to 2008. Qualifying rate reached 100% in 2007, the remaining 9 years were 33.0%(3/9) - 88.9%(8/9). The iodized salt laboratory response rates were 100%(9/9) in 2000 - 2008. The pass rates were 77.8%(7/9), 88.9%(8/9),77.8%(7/9) from 2001 to 2003, 88.9%(8/9) in 2007, and the remaining 5 years 100%. Response rate of iodized salt laboratory at the county level that participated in the External Quality Control were 66.7% (20/30), 90.0%(27/30), 80.0% (24/30), 96.7% (29/30) from 2000 to 2003, respectively, and 2006 was 96.7% (29/30), and the remaining four years were all 100% (30/30). The pass rates in 2000 - 2008 were between 53.3% (16/30) -93.3%(28/30). Conclusions The accuracy of test results of external quality controls and the normal operation of the network at all levels of laboratories is closely related to the IDD laboratory conditions, detection techniques, and the degree of attention of relevant departments and professional bodies.
3.Pathogen Infection after Liver Transplantation:A Clinical Study

OBJECTIVE To analyze the prevalence of pathogen infection and the drug sensitivity of pathogen after liver transplantation so as to provide the reference to the effective clinical prevention and control of pathogen infection and drug resistant strains.METHODS A retrospective survey in 155 patients who underwent 158 orthotopic liver transplantation was carried out.RESULTS The incidence of pathogen infection after liver transplantation was 25.9%,the major pathogens were coagulase-negative Staphylococcus,then Pseudomonas aeruginosa and Acinetobacter.CONCLUSIONS It is very important to prevent pathogen infection and use antibiotic reasonably.Selection of effective antibiotics referring to the etiologic data and resistance of pathogen can help to prevent the drug resistant strains.
4.CT manifestations of pediatric mesenchymal hamartoma of liver

Objective To explore the CT manifestations of pediatric mesenechymal hamartoma of liver (MHL).Methods Clinical data of 15 cases with MHL confirmed by surgery and pathology were retrospectively analyzed.All children were performed with CT scans including plain and enhanced scans,and the imaging features were analyzed.Results All the lesions were solitary,and 9 masses located at the right lobe of liver,4 located at the left lobe,the others located at both right and left lobe.The maximum diameter of lesions was from 5 cm to 30 cm,with an average of (11.52±6.84) cm.Ten cases were cystic and solid mixed,5 cases were solid.After contrast administration,the solid component and the septa of the masses showed enhancement while no enhancement was observed in the cystic component.Two cases had the spot-like calcification.Conclusion MHL has some special characteristics in the CT scan before surgery,which should be differentiated from other cystic and solid tumors of live.Some MHLs can be diagnosed when combined with the clinical information and CT images.
5.Clinical Features and Influential Factors of Therapeutic Efficacy of Reflux Esophagitis in Elderly Patients with Type 2 Diabetes Mellitus

Background:The prevalence rate of diabetes mellitus ( DM ) associated with gastroesophageal reflux disease ( GERD)is increasing recently,and the relationship between DM and GERD has become a research hotspot. Aims:To study clinical features and influential factors of therapeutic efficacy of reflux esophagitis( RE)in elderly patients with type 2 DM(T2DM). Methods:Two hundred elderly patients with RE diagnosed by gastroscopy from March 2011 to October 2013 were enrolled,and divided into T2DM associated with RE( T2DM)group and RE group. The clinical features, endoscopic findings and therapeutic efficacy of the two groups were analyzed,and the influences of DM course and control of blood glucose on efficacy of RE were also analyzed. Results:The main manifestations of RE were extraesophageal symptoms,typical esophageal symptoms were less common. The overall efficacy rate decreased with the increase of endoscopic grade of RE,however,extraesophageal symptoms improved significantly. With the prolonging of DM course,the endoscopic grade of RE was increased,and the control of blood glucose was worse. The overall therapeutic efficacy was significantly reduced with the prolonging of DM course in patients with well controlled blood glucose. Conclusions:Extraesophageal symptoms are common,endoscopic grade of RE is worse and blood glucose is difficult to control in elderly T2DM patients associated with RE. DM course and status of blood glucose control have influences on the therapeutic efficacy of RE.
6.The Effect of Dexamethasone on the Chemosensitivity of Colon Carcinoma Cells to L-OHP and 5-Fu Treatment

Objective: To examine the expression of glucocorticoid receptor in human colon cancinoma tis-sues and call lines and to explore the survival of colon cancer cell lines treated with dexamethasone alone or in combination with 5-Fu and L-OHP in a way of dexamethasone pretreatment or co-administration in vitro.Methods: The expression of glucocorticoid receptor was detected in 61 cases of colon cancer tissue samples and 4 types of colon cancer cell lines by immunohistochemistry. Apoptosis was detected by Hoechst33342 staining and flow cytometry. MTT assay was employed to detect the chemosensitivity of colon carcinoma cells to L-OHP and 5-Fu with dexamethasone pretreatment for 24 hours or co-administretion. Results: Positive GR expression was found in 57.3% colon cancer tissue samples and in Lovo and HCT-116 cell lines, not in HT-29 and SW-480. Apoptosis was detected in GR-expressed Lovo and HCT-116 cells at 72 hours after 1×10~(-4)mol/L Dex treatment, and the rates of apoptosis were higher than those in the control groups without Dex (P<0.01),GR-negative cells, HT-29 and SW-480 even treated with 1 × 10~(-4)mol/L Dex for 72 hours. Pretreatment and co-administration for Lovo cells with 1×10~(-4)mol/L Dex could decrease the IC50 of L-OHP from 13.7±1:1.3μg/mL to 5.9±0.6μg/mL and 4.8±0.7μg/mL, respectively. IC50 of 5-Fu was decreased from 72.2±8.1 μg/mL to 21.1±4.1μg/mL and 18.6±4.0μg/mL, respectively. Conclusion: There is expression of glucocorticoid receptor in part of colon carcinoma tissue samples and cell lines. Apoptosis does occur in GR-expressed Lovo and HCT-116 cells induced by dexamethasone in vitro. Pretreatment for 24h and co-administration with Dex can increase the chemosensitivity of Lovo cells to L-OHP and 5-Fu.

[Objective] To observe the effect of Shuhou Kangyan Decoction (SKD), a liver-kidney nourishing, Qi-strengthening and blood-activating, wind-dispelling and heat-clearing prescription mainly composed of Radix Astragali, Radix Angelicae Sinensis, Fructus Lycii, Radix Rehmanniae, Radix Saposhnikoviae, Rhizoma Atractylodis Macrocephalae, etc. , on interleukin-2 (IL-2) and soluble IL-2 receptor (SIL-2R) in serum and tears of patients with cataract after operation. [Methods] Forty-seven cases of cataract after ultrasonic emulsification plus artificial lens implantation were randomized into two groups. Group A (n = 26) was treated with routine operation and eye drops and group B ( n = 21) with SKD additionally and other 15 healthy aged people ( n = 15) receiving physical examination in the out-patient department served as normal controls. [Results] Post-operative serum SIL-2R level was higher in groups A and B than that before operation ( P 0.05). 8.Growth activity of osteoblast on a novel strontium incorporated calcium sulfate.

OBJECTIVETo investigate the growth activity of osteoblast on a novel strontium incorporated calcium sulfate and make comparison with normal calcium sulfate material.
METHODSOsteoblast was inoculated on samples and cell proliferation was measured on the 1st, 3rd, 5th days, and the activities of ALP and osteocalcin were observed on the 5th day. And microcosmic morphology of osteoblast was observed by scanning electron microscopy(SEM).
RESULTSOsteoblast grows robustly on tested material. Cell quantity on the surface of novel material was obviously higher than normal calcium sulfate material (P < 0.05). The activity of ALP and osteocalcin on novel material was 57.8% and 40.2% higher than on normal calcium sulfate material respectively (P < 0.05). On strontium incorporated surface, osteoblast spread well. Cells were polygonal with abundant cytoplasm and the morphology was active.
CONCLUSIONStrontium incorporated calcium sulfate can sustain robust growth activity of osteoblast, which is promising to be used for bone substitute materials.

9.Identification of liposoluble constituents in Yushu tablets by UPLC-ESI-IT-TOF/MS.

In order to identify the chemical constituents of Yushu tablets comprehensively, we studied the chemical constituents of CHCl3 extract from Yushu tablets by the ultra performance liquid chromatography-electrospray ionization-ion trap-time of flight mass spectrometry (UPLC-ESI-IT-TOF/MS). It showed that there were more than 100 compounds separated, and forty-nine peaks among these were identified on the basis of high resolution mass spectrometry data and literature data reported. Determination of twelve peaks was further confirmed by standard substances. These components assigned to the different plant sources mainly included phenylpropanoids, triterpenoids, quinones and m-trihydroxybenzene compounds. By analyzing the chemical components of CHCl3 extract from compound Chinese medicine Yushu tablets comprehensively, this study provided the foundation for studying chemical components, pharmacodynamic substance and quality control of Yushu tablets.

10.Value of transrectal ultrasound in diagnosing middle ureteral calculi

Objective To explore the value of transrectal ultrasound (TRS) in diagnosing middle ureteral calculi.Methods Patients who were suspected as middle urinary calculi by clinicians underwent transabdominal ultrasound (TAS) and transrectal ultrasound in the First Affiliated Hospital of Guangzhou Medical University and Guangzhou Hospital of Integrated Traditional and West Medicine from 2011 to 2012. The diagnosis were finally confirmed by a variety of methods .Results Forty patients were eventually diagnosed as middle ureteral calculi by X-ray,computer tomography (CT),surgery,intravenous pyelography (IVP)imaging and magnetic resonance imaging (MRI).The other 5 cases were diagnosed as ureteral tumor , ureteral cystis(confirmed by MRI)and oppressed ureteral by gynecology cystis (confirmed by surgery).TRS detected 37 of 40 middle ureteral calculi(92.5%).TAS detected 8 of 40 middle ureteral calculi(20.0%). The detection ratio of TRS was higher than that of TAS and the difference was statistically significant (P=0.00).TAS could display ureter across iliac blood vessels expansion in all 40 patients.Conclusions Using TRS in diagnosing middle ureteric calculi is quick , convenient with high sensitivity and worthy of wide application.TAS can display ureter across iliac blood vessels expansion and can help to diagnosis of middle ureteral calculi.
