1.Comparison of Effects of Radix Astragali and Triperygium Glucosides on Glomerular Mesangial Cells Proliferation and Interleukin-6 Secretion

[Objective] To compare the effects of Radix Astragali (RA) and triperygium glucosides (TG) on glomerular mesangial cells (GMC) proliferation and interleukin-6 (IL-6) secretion. [Methods] GMC were cultured with in-vitro cell culture technology. Serum containing RA and serum containing TG were prepared with serum pharmacological methods. GMC proliferation was detected by methylthiazolyltetrazolium (MTT) assay and IL-6 level by enzyme-linked immunosorbent assay (ELISA) to evaluate the effect of low-, moderate- and high-dosages of RA-containing serum and TG-containing serum under the condition of GMC over-proliferation. [ Results] Different dosages of RA-containing serum and TG-containing serum inhibited GMC proliferation and IL-6 secretion in a dose-effect manner, and the inhibitory effect of high- and moderate-dosage TG and high-dosage RA was stronger. [Conclusion] Under the condition of in-vitro GMC over-proliferation, RA has the similar inhibitory effects on GMC proliferation and IL-6 secretion with TG. The combined treatment of RA and TG can reduce the dose of TG and probably has an action in increasing effect and reducing toxicity.
3.ARIMA models to predict new-diagnosing cases of pneumoconiosis in Nanjing.

OBJECTIVEThe primary goal of our study was to evaluate and predict the epidemiological trend of pneumoconiosis.
METHODSWe established a new database on new-diagnosing cases of pneumoconiosis in Nanjing during 1955-2007. The database was analyzed by using SAS9.1.3 statistical software, Data sequence was stabilized by using the process of differencing. Goodness of Fit Test verified that the residual-error sequence was white noise sequence. We determined the Autoregressive Integrated Moving Average models (ARIMA models) as an appropriate model. By Taking advantage of back-substitution model, we predicted new cases of pneumoconiosis during 2008-2012. We then compared the predicted value with the actual value to test and verify the predicting function.
RESULTSWe finally had chosen out ARIMA (2, 1, 0) models to fit the original sequence, which led to the results that the observed values are basically comparable with the predicted values. The past 5-year predicted-value was similar to the actual value. We then built a new model by new cases during 1955-2012, predicting that the trend of pneumoconiosis in the next 5 years will tend to approach plateau with approximately 10∼13 new cases per year.
CONCLUSIONARIMA models is suitable for fitting large sample series of new diagnosed pneumoconiosis over the years and for the predicting the incidence of pneumoconiosis.

4.Investigation of nutrition and feeding status among children under 2 years in Beijing.

OBJECTIVETo investigate the nutrition status, breast feeding and complementary feeding among children under 2 years of age in Beijing.
METHODSA stratified cluster randomly sampling method was used. After having got the compliance a questionnaire survey was carried out to 422 children < 2 year-old in total 18 districts of Beijing.
RESULTSThe average infant birth weight was 3325 g, the rate of weight (2500 g and > or = 4000 g were 2.8% and 13.0% respectively. The rate of exclusive breast-feeding, partial breast-feeding, bottle-feeding of infant under 4 months was 56.6%, 33.9% and 9.5% respectively. The average weaning time was 7.4 months. The rate of complementary feeding of infant up to 6 months was 99.4%. The average ages for addition of complementary feeding such as cereal, vegetable and fruit, egg, oil, nutrients in rural areas were later than those in urban.
CONCLUSIONIt is important to promote breast feeding and improve complementary feeding by education and intervention.

6.Value of measuring ankle brachial index for diagnosing peripheral arterial disease in Chinese patients with high cardiovascular risk.

OBJECTIVETo evaluate the value of measuring ankle brachial index (ABI) for diagnosing peripheral arterial disease(PAD) compared with conventional digital subtraction angiography (DSA) as the reference standard.
METHODSA total of 383 consecutive inpatients (245 male, mean age 64.1 +/- 11.7 years) underwent both conventional DSA and ABI measurements.
RESULTSThe rate of statin intervention was 90.9%, ACEI 69.2%, antiplatelet 96.6% and beta-blockers 67.9%. The intravascular stenosis was classified into six degrees: normal, < 30%, 30% - 49%, 50% - 69%, 70% - 89% and > or = 90%. Compared to the traditional gold standard (DSA) in diagnosis PDA, the ABI value decreased in proportion to the severity of PAD (the ABI value was 1.08 +/- 0.11, 1.05 +/- 0.16, 0.99 +/- 0.17, 0.66 +/- 0.24, 0.55 +/- 0.28 and 0.54 +/- 0.00 respectively in the six ranks). There was a significant correlation between DSA and ABI in diagnosis PAD.
CONCLUSIONABI measurement is an accurate and reliable non-invasive alternative to conventional DSA in the assessment of lower extremity arteries in patients with peripheral arterial disease.

7.Rapid identification for Mycobacterium fortuitum with RNA isothermal transcription-mediated amplification and real-time detection assay

The aim of the study is to establish and evaluate a RNA isothermal transcription-mediated amplification and realtime detection assay (RIARD-MF) for the identification of Mycobacterium fortuitum in clinical isolates.RNA probes and specific primers of reverse transcription and amplification for T7 promoter were designed based on the sequence of M.fortuitum 16S rRNA.The isothermal successive cycles of amplification were performed for real-time detection by using T7 RNA polymerase at 42 ℃.Five non-mycobacterium strains,20 Mycobacterium strains and 259 clinical strains were detected by the established assay to evaluate the specificity and sensitivity,and the results were compared with those of PCR sequencing.In the test of 5 non-mycobacterium strains and 20 Mycobacterium strains,only M.fortuitum was positive,and the remaining 24 strains of bacteria were negative,which was consistent with PCR gene sequencing.The sensitivity and specificity of RIARD-MF reached 60 CFU/mL and 100%.In the test of 259 strains of clinical isolates,5 strains were identified to be M.fortuitum,the remaining 254 strains were not identified to be M.Fortuitum,which was also consistent with PCR gene sequencing.Both the specificity and sensitivity reached up to 100% in the detection of clinical isolates.It suggested that the RIAR-DMF established in this study is a specific,sensitive,accurate and rapid method for the identification of M.Fortuitum and it may be hopeful for rapid identification of M.fortuitum in clinical isolates.
8.Clinical and Pathologic Features of Multifocal and Multicentric Breast Cancer in Chinese Women: A Retrospective Cohort Study.

PURPOSE: This study aims to analyze the clinical-pathological characteristics of multifocal and multicentric breast cancer (MMBC) in Chinese women. METHODS: Sixty-seven cases with MMBC were randomly collected and reviewed at seven hospitals in representative districts of China during 1999 to 2008. RESULTS: The incidence of MMBC in breast cancer in China was 1.75%. Compared to those with unifocal breast cancer, women with MMBC were more likely to have larger tumor size, lymph node metastasis (59.70% vs. 45.62%) and stage III to IV (46.26% vs. 21.10%). The peak age at onset of MMBC was 40 to 49 years old and has been gradually increasing during 1999 to 2008. Most of the MMBC women were treated with surgery and adjuvant therapy. CONCLUSION: In China, the incidence of MMBC in breast cancer is significantly lower than that in Western countries. Compared to unifocal breast cancer, MMBC is biologically more aggressive. Most MMBC women underwent mastectomy, instead of breast conservation surgery.

9.Impact of zearalenone on male fertility: An update.

Zearalenone (ZEA) is a nonsteroidal estrogen-like mycotoxin widely distributed in maize, wheat, rice and other cereals with its derivants. It also presents in meat or dairy products or even in the aquatic ecosystem via rain, and thus can affect human health. ZEA affects the body function in various ways. On the one hand, it can disturb the synthesis of estrogen and its combination with the receptor, influence the reproductive ability via the estrogen signaling pathway, and cause the dysfunction of the reproductive systems. On the other hand, it can disturb the synthesis of DNA and proteins and result in lipid peroxidation and cytotoxicity by inducing the apoptosis of germ cells. It is known that exposure to different doses of ZEA can affect the female reproductive system by increasing the apoptosis of germ cells and inducing germ cell prematurity, sexual precocity, endocrine disorder, reproductive cycle disorder, and so on. But studies of its influence on the male reproductive system are relatively rare, especially about its unique male-related action mechanisms. This review presents an overview of the studies on the mechanisms of ZEA affecting male fertility and the phenotype changes in the male reproductive system after exposure to ZEA, hoping to provide some new ideas for the protection of human fertility.
10.Analysis of measles immunity level in persistent populations in Beijing, 2012.

OBJECTIVETo analyze the measles immunity level of persistent population in Beijing.
METHODSA total of 2125 objects from 10 age groups, who had been living in Beijing for over 6 months, were selected from urban and rural areas in Beijing in 2012. Demographic characteristics, history of measles and vaccine immunization were investigated by questionnaire. 5 ml blood sample of each subject was collected, and the Measles IgG antibody was measured by ELISA assay.
RESULTSPositive rate of measles antibody was 84.71% (1800/2125) and standardized positive rate was 88.07% . Median of antibody was 960.46 IU/L. Positive rate and median of measles antibody were significantly different between population from different age groups (χ(2) = 341.60, P < 0.01; H = 216.27, P < 0.01). Antibody positive rate and median were lowest in the <1 year age group, which were separately 43.06% (90/209) and 185.80 IU/L; and highest in the 1-4 (97.31% (181/186) and 2448.81 IU/L) and 5-9 years age group (96.46% (218/226) and 1910.72 IU/L). The range of antibody positive rate and median in adults of ≥ 15 years were 81.98%-90.14% and 744.38-1474.84 IU/L. Antibody positive rate and median in persistent population, which were separately 82.45% (883/1071) and 899.82 IU/L, were lower than those in migrant population, which were 87.00% (917/1054) and 166.19 IU/L, respectively (χ(2) = 8.51, P < 0.01;U = 538 704.00, P < 0.01). Antibody positive rate and median in population with vaccination history, which were separately 91.95% (891/969) and 1443.11 IU/L, were higher than those population without vaccination history and people whose history unknown (32.95% (57/173) , 127.33 IU/L; 86.67% (852/983) , 923.73 IU/L). The difference showed statistical significance (χ(2) = 399.92, P < 0.01; H = 202.11, P < 0.01).
CONCLUSIONAmong the persistent population in China, measles antibody level among the children aging 1-9 years old was high enough to prevent outbreak and epidemic of measles. However, we should try our best to strengthen the measles antibody level among the babies younger than 1 year old and the migrant population aging between 15 and 40 years old.
