2.The expression and significance of PCNA,CDK4 and p16INK4a in nonneoplastic epithelial disorders of vulvar

Objective To study the expression and significance of proliferation cell nuclear antigen (PCNA) ,cyclin dependent ki‐nase 4(CDK4) and p16INK4a in nonneoplastic epithelial disorders of vulvar(NNEDV) and explore the correlation of them .Methods Fifty‐two vulvar lesion samples(lesion group) and 52 vulvar skin surrounding the lesions samples(lesion‐adjacent group) were col‐lected from NNEDV patients ,and lesion group include 27 samples with squamous hyperplasia of vulvar(VSH) ,15 samples with li‐chen sclerosus of vulvar(VLS) and 10 samples containing both lesions(mixed‐lesion) .25 vulvar skin samples were collected from patients receiving perineal repair operation as control group .The expression of PCNA ,CDK4 and p16INK4a was detected by immuno‐histochemistry assay .Results The expression of PCNA in lesion group was significant higher than those in lesion‐adjacent group and control group (P< 0 .05) ,while there was no significant differences among VSH ,VLS and mixed lesion ,and there was no sig‐nificant difference between lesion adjacent group and control group (P> 0 .05) .The expression of CDK4 in lesion group and lesion‐adjacent group were significant higher than that in control group (P < 0 .05) ,while there weren′t significant differences among VSH ,VLS and mixed‐lesion (P> 0 .05) ,and there was no significant difference between lesions group and lesion‐adjacent group (P> 0 .05) .The expression of p16INK4a in lesion group was significant lower than those in lesion‐adjacent group and control group (P< 0 .05) ,while there was no significant differences among VSH ,VLS and mixed‐lesion (P> 0 .05) ,and there was no significant difference between lesion‐adjacent group and control group (P> 0 .05) .About PCNA and CDK4 ,there was a positive correlation in lesion group (P< 0 .05) and there were no correlations in other groups (P> 0 .05) .About PCNA and p16INK4a ,there was a negative correlation in lesion group (P< 0 .05) and there were no correlations in other groups (P > 0 .05) .About CDK4 and p16INK4a ,there were no correlations in all groups (P> 0 .05) .Conclusion expression of PCNA ,CDK4 and p16INK4a may be related of abnormal in‐creasing of cell proliferation and acceleration the process of cell cycle ,which may be applied to molecular target for treatment .
3.The herbal-partitioned moxibustion for primary dysmenorrhea and it's impact on reproductive endocrinal function of patients.

OBJECTIVETo explore the efficacy and the reproductive endocrinal mechanism of herbal-partitioned moxibustion in the treatment of primary dysmenorrhea.
METHODSOne hundred and seventy-one cases of primary dysmenorrhea were randomized into an herbal-partitioned moxibustion group (group A), an starch-partitioned moxibustion group (group B) and an acupuncture group (group C), 57 cases in each one. In the group A, moxibustion isolated with herbal medicine was applied to Shenque (CV 8). In the group B, moxibustion isolated with starch was used at Shenque (CV 8). In the group C, acupuncture was given at Sanyinjiao (SP 6). The changes of estradiol (E2), progesterone (P) and prostaglandin levels (PGF2alpha) were observed before and after treatment, and the therapeutic effects were compared among the 3 groups.
RESULTSThe therapeutic effect in the group A was better than those in the other two groups [compared the cured rate: 89.8% (44/49) vs 60.0% (30/50), 60.4% (32/53), both P < 0.05]. In the group A, E2 level [(110.99 +/- 12.90) pg/mL vs (83.94 +/- 8.91) pg/mL, P < 0.05] and PGF2alpha level [(24.58 +/- 3.01) pg/mL vs (14.34 +/- 1.48) pg/mL, P < 0.01] were decreased and P level was increased [(4.65 +/- 0.68) ng/mL vs (6.68 +/- 0.95) pg/mL, P < 0.05]. In the group B and C, PGF2alpha level were reduced. Concerning to the regulating of E2 and PGF2alpha levels, the results in the group A were better than those in the group B and C [(-30.16 +/- 10.20) pg/mL vs (10.79 +/- 15.01) pg/mL, (22.81 +/- 12.22) pg/mL; (-13.10 +/- 2.40) pg/mL vs (-6.52 +/- 1.88) pg/mL, (-3.14 +/- 1.19) pg/mL, (see text) P < 0.05]. Concerning to the regulation of P level, the results in the group A and B were better than that in the group C (all P < 0.05).
CONCLUSIONThe herbal-partitioned moxibustion achieves the significant efficacy on primary dysmenorrhea, which could be related to regulating the reproductive endocrinal level. It decreases E2 and PGF2alpha levels and increases P level.

4.Ultra-mini percutaneous nephrolithotomy for the treatment of renal or upper ureteral stones (report of 32 cases)

Objective To investigate the efficacy and safety of ultra-mini percutaneous nephrolithotomy for the treatment of renal or upper ureteral stones.Methods The data of 32 patients diagnosed as renal or upper ureteral stones were retrospectively reviewed,who underwent ultra-mini percutaneous nephrolithotomy from June to October,2015,including 29 kidney stones,3 upper ureteral stones,of which 21 single stone,7 multiple stones,and 4 staghorn stones.There were 8 cases with inferior calyx stones,10 with renal pelvis stones,3 with upper ureteral stones,10 with renal pelvis stones plus inferior calyx stones and 1 with renal pelvis stones plus upper calyx stones.The mean stone size was (20.1 ±7.6) mm (ranging from 10 to 41mm).The mean Hounsfield unit (HU) was (1 125.9 ±225.9) Hu (ranging from 520 to 1 550Hu).In this cohort,13 cases had mild hydronephrosis,2 moderate hydronephrosis,1 severe hydronephrosis while no hydronephrosis identified in the other 16 patients.One case had concomitant kidney and ureter duplication malformation,and 1 case had residual stones after laparoscopic nephrolithotomy.Results All of the patients were treated by single tract UMP.Among them,middle calyceal puncture was performed in 9 cases,and the other 23 cases underwent lower calyceal puncture,including 4 cases of intercostal puncture,and 28 subcostal puncture.The mean operating time was (30.3 ± 15.0) min (ranging from 10 to 90 min).The mean postoperative hospital stay was (1.9 ± 1.0)days (ranging from 1 to 5 days).The mean hemoglobin decrease was(14.6 ±8.4)g/L (ranging from 1 to 46 g/L).No analgesics were used.Peri-operative complication rate was 3.13% (1/32) with 1 case of collection system perforation.There was no fever,severe bleeding,urinary extravasation,pleural injury,or blood transfusion.The SFR was 93.75% (30/32) and 96.88% (31/32) on the first day and the first month after the operation,respectively.Conclusions UMP could be a safe and effective method for the treatment of renal or upper ureteral stones,especially for inferior calyceal stones less than 2cm.It offers high stone clearance rate,minimally invasiveness,quick recovery,short length of hospital stay and improved quality of life.
5.Interactional effects of corticosterone and stress in the formation of PTSD-like memory impairment rats

Objective To investigate the interactional roles corticosterone and stress played during the PTSD-Like memory impairment.Methods First we established the model of PTSD-like memory using three levels of electric shock:0 mA,0.8 mA and 1.4 mA.The freezing time percent of each group were calculated 24 hours later in order to find which group of rats obtained the PTSD-like memory,and generalizing tests was used to verify it.After the model was established,rats received adrenalectomy (ADX) were operated bilaterally in order to investigate the effect of corticosterone and fed with corticosterone in the drinking water 7 days before fear conditioning.These rats were divided into four groups:control group (no corticosterone supplement,0 mA shock),Cort group (corticosterone supplement,0 mA shock),shock group (no corticosterone supplement,1.4 mA shock),Cort-shock group (corticosterone supplement,1.4 mA shock).A two-way factorial analysis of variance was used to determine whether there was a significant interaction between corticosterone and shock.Results The freezing time percent of group 0.8 mA raised compared with the group 0 mA,whereas the freezing time percent of group 1.4 mA showed reversely and the generalizing effects appeared,compared with group 0.8 mA.In two-way factorial analysis design,the freezing time percent of Cort-shock group significantly increased (P<0.05) compared with other groups,and there was no significant variance among the other groups.Corticosterone (F=6.464,P<0.05) and stress (F=53.419,P <0.01) played parts in the formation of PTSD-like memory impairment,and they interacted with each other (F=11.580,P<0.05).Conclusion PTSD-like memory impairment can be induced on rats with high (1.4 mA) electrical shock,and they have interactional effect with each other in the process.

10.Analysis of HPV infection genotype spectrum in female cervical cells

Objective To investigate the distribution of 39 kinds of human papillomavirus(HPV)infection genotypes in female cervical cells and its clinical significance.Methods 39 types of HPV DNA were extracted from 434 samples of female cervical cells. The gene amplification combined with the gene chip technique was adopted to detect 39 kinds of HPV genotype.And the clinical da-ta of the patients were analyzed.Results Among 434 samples of female cervical cell,175 cases were HPV positive,the total HPV infection rate was 40.32%(175/434).Among them,105 cases were the single type HPV infection with the positive detection rate of 24.19%(105/434)and 70 cases were the multiple types HPV infection with the positive detection rate of 16.13%(70/434).Among single type HPV infection,31 cases were the HPV18 infection with the positive detection rate of 17.71%(31/175),which was the main HPV infection type;followed by HPV16 in 12 cases with the positive detection rate of 6.86%(12/175)and HPV52 in 11 cases with the positive detection rate of 6.29%(11/175).Among the multi-type HPV infection,each 2 cases were HPV 6+54,HPV 18+52,HPV 51+68 infection respectively,each accounted for 2.86% of the multi-type HPV infection,which were the main infection types.Conclusion HPV 16,18,52 and HPV 6+54,HPV 18 +52 and HPV 51 +68 are the main HPV infection genotypes of fe-male cervical cells.The gene amplification combined with the gene chips technique is a method suitable for clinically conducting the HPV genotyping diagnosis and the molecular epidemiologic research of HPV infection.Along with the increase of detected HPV genotypes,the HPV infection rate is also increased,its genotypes combinations trend towards diversification.
