1.Effect of HELICOBACTER PYLORI infection on apoptosis and proliferation along with P53 gene expression in gastric epithelium

Purpose:To investigate the relationship between HELICOBACTER PYLORI (Hp) and gastric carcinoma, as well as the possible carcinogenic mechanism by Hp. Methods: DNEL technique and immunohistochemical staining were used in the research to study the state of apoptosis, proliferation and p53 gene expression. Total 100 gastric mucosal biopsy specimens, including normal mucosa, Hp negative and Hp positive gastric precancerous lesions as well as gastric carcinomas were studied. Results:There were several apoptotic cells in the superficial epithelium and a few proliferative cells within neck of gastric glands, and no p53 protein expression in normal mucosa. In gastric carcinoma, apoptotic cells accounted for 1.62%, while proliferative cells 4.99%, and P53 protein significantly increased. in metaplasia, the apoptotic index (AI), proliferative index (PI) and positivity of p53 expression in Hp positive group were higher than those in normal mucosa ( P
2.The role of cell apoptosis in glioma chemotherapy evaluation

Objective To study the role of cell apoptosis during the chemotherapy of human gliocytoma in order to get effective suvilliance on the effect of chemotherapy. Methods Gliocytoma cells were isolated and cultured from 40 human gliocytoma samples. Mitochondrial membrance potential (MMP), cell cycle, the level of Bcl-2 and Bax were detected by flow cytometry (FCM) at 24, 48, 72 hours respectively of incubation with Lomustine (CCNU) and Teniposide (VM-26), and the trends were also analysed. Results MMP decreased greatly, the apoptosis part in the cell cycle ananlysis increase, the expression level of Bcl-2 decreased, and that of Bax increase rapidly, while the Bcl-2/Bax ratio decreased. Conclusions CCNU and VM-26 have singnificant effect in gliocytoma chemotherapy on inducing gliocytoma cell apoptosis. VM-26 has more stronger effect on the cell cycle. MMP is the most sensitive and the fastest index in apoptosis detection.

4.Development of moving darkroom and the value of X-rays photographing in the endemic fluorosis areas

Objective To discuss the value of moving darkroom and accompanying X-rays protective shield in endemic fluorosis areas.Methods Using moving and fixed darkrooms,the X-rays photos of the forearms and shanks of the 320 persons were developed in the endemic fluorosis areas,while the time for development and installing-uninstalling films was documented.The films develpoed in the darkroom were evaluated for the quality.Results Among the 320 films develpoed in the moving darkroom,the 268 had fingerprints,47 had scratches,298 were in good quality(93.13%),while in the fixed darkroom,the figure was correspondingly 735,384,227(70.93%);The moving darkroom increased excellent film rate significantly than fixed darkroom(χ2=53.43,P<0.01),The detectable rate of skelelal fluorosis and degree Ⅰ skelelal fluorosis were highter than that in the fixed darkroom did (χ2=10.34,χ2=9.56,P<0.01).Conclusions The films developed in moving darkroom have superior quality and higher diagnostic value,so it is important to use moving X-rays photographing in ecdemic fluorosis areas.
5.Application of paclitaxel during trabeculectomy and its anti-proliferative effects

Background The filtering surgery is the main method of treating glaucoma.Fibrosis of filtering bleb is a key cause of failure of operation.The study about application of anti-scaring drug in filtering surgery is a hotspot.Objective Present study was to investigate the anti-scaring effect of paclitaxel aher trabeculectomy.Methods Thirty-two adult clean domestic rabbits underwent standardized trabeculectomy and randomly divided into 4 groups.Normal saline solution was used beneath the scleral flap during trabeculectomy for 3 minutes in 16 eyes of 8 rabbits as controls.0.3 g/L mitomycin C,0.2 g/L paclitaxel or 0.3 g/L paclitaxel was administered at the same way respectively in other 3 groups.Intraocular pressure (IOP) was measured,and eye numbers with function blebs were compared among 4 groups at the 4th,7th,14th and 28th day after surgery.The opening of filtration tunnel and the number of inflammatory cells were observed by hematoxylin and eosin staining,and proliferation of new collagen fibers was evaluated by Masson trichrome method.This study complied with the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission(version 1988). Results No significant differences were found in the change of lOP among 4 groups before operation(F=0.54,P=0.83)and the 4th day aher operation(F=0.57,P=0.87).The IOP value was statistically lower in 0.3 g/L mitomycin C group,0.2 g/L paclitaxel group and 0.3 s/L paelitaxel group than the normal saline solution group(P<0.05)with the lowest value in 0.3 s/L paclitaxel group in 7,14,28 days after operation(P<0.05).Functional filtering bleb was seen in all the rabbit eyes in the 4th day after operation.In 7,14,28 days after operation,the number of eyes with functional bleb wag evidently more in 0.3 g/L mitomycin C group,0.2 g/L paclitaxel group and 0.3 g/L paclitaxel group compared with normal saline group(P<0.05).The histological examination showed that the infiltration of inflammatory cells in filtering tunnel was much more obvious in normal saline solution group than the other groups with the most mild response in 0.3 g/L paclitaxel group.Masson trichrome revealed that proliferation of new collagen fibers in 0.3 g/L paclitaxel group was significantly decreased in comparison with those in other three groups at the 4th,7th,14th and 28th day after surgery(all P<0.05). Conclusion Paclitaxel can inhibit the inflammatory response and collagen fibrosis and therefore open the filtering tunnel after it be used topically during the glaucoma trabeculectomy.
6.Effect of Intensive Scapular Control Training on Post-stroke Shoulder Pain

Objective To observe the effect of intensive scapular control training on pain, upper limb motor function and activities of daily living (ADL) in patients with post-stroke shoulder pain. Methods 58 patients with hemiplegia after stroke were randomly assigned into routine exercise group (group A, n=27) and intensive scapular control training group (group B, n=31). They were assessed with visual analogue scale for pain, simplified Fugl-Meyer Assessment and modified Bathel Index before and 8 weeks after treatment. Results All the assessment appeared better in group B than in group A after treatment (P<0.05). Conclusion Intensive scapular control training can promote the recovery of motor function and ADL, relieve the pain in patients with post-stroke shoulder pain.
7.Etanercept combined with Tripterygium wilfordii polyglycoside for treatment of rheumatoid arthritis in the elderly: a clinical study.

OBJECTIVETo evaluate the efficacy and safety of etanercept plus Tripterygium wilfordii polyglycoside (TWP) in elderly patients with active rheumatoid arthritis (RA).
METHODSTotally 46 elderly patients with active RA were randomly assigned to the treatment group (22 cases) and the control group (24 cases). All patients received subcutaneous injection of etanercept, 25 mg each time, twice per week. The dosage was reduced to once per week 3 months later. Patients in the treatment group took TWP Tablet (10 mg each time, three times per day), while those in the control group took methotrexate (MTX), 10 mg each time, once per week. The whole course lasted for 24 weeks. Patients' rest pain, tender joint number, swollen joint number, health assessment questionnaire (HAQ), patients' global assessment, physicians' global assessment, erythrocyte sediment rate (ESR), C reactive protein (CRP), rheumatic factor were assessed at week 0, 4, 8, 12, and 24. The curative effect was statistically evaluated by the United States Institute of Rheumatology ACR20, ACR50, and ACR70 improvement criteria. Meanwhile, any adverse event was recorded and evaluated.
RESULTSTotally 41 completed the trial, and 5 dropped off (3 in the treatment group and 2 in the control group). Compared with the control group, there was no statistical difference in ACR20, ACR50, or ACR70 in the treatment group (P > 0.05). Compared with before treatment in the same group, there was some improvement in tender joint number, swollen joint number, visual analogue scale (VAS) for patients' global assessment, VAS for physicians' global assessment, ESR, CRP, and HAQ between the two groups, showing statistical difference (P < 0.05). Compared with the control group in the same phase, there was no statistical difference in the treatment group (P > 0.05). There was no statistical difference in the occurrence of adverse events between the two groups.
CONCLUSIONSEtanercept plus TWP could achieve equivalent therapeutic effect to that of Etanercept plus MTX. The two regimens could improve clinical signs, symptoms, and QOL related to RA. They were well tolerated in the treatment of elderly patients with active RA.

8.Survey of water improvement project to reduce fluoride in Shandong province

Objective To investigate the present situation of water-improving defluoridation project and the distribution of water fluoride in fluorosis areas in Shandong province. Methods In 2005 - 2007, according to "The National and Shandong province Technical Scheme for Endemic Disease Control", the water-improving form,water type and the running status of defluoridation project in 17 cities of Shandong province were investigated, and the water-fluoride of the running projects were determined by F- ion selective electrode. Results A total of 5816projects were built that involved 8776 villages, 110 counties, 17 cities distributed in Shandong province. In which,the drilling of water-improving projects accounting for 94.55% (5499/5816), under-ground water type 97.73%(5684/5816);operating normally accounting for 75.91% (4415/5816), 7246 villages were supplied with the water, beneficiary population 6 946 459 people, non-normal projects accounting for 24.09%(1401/5816), including 1530 diseased villages. There were still 4415 projects running well. The projects with water fluoride lower than 1.0mg/L were 2893, accounting for 65.53% (2893/4415). The water fluoride higher than 1.0 mg/L accounted for 34.47%(1522/4415), > 2.0 mg/L of 548, > 4.0 mg/L of 97, and the maximum value of water fluoride was 9.71mg/L. Conclusions Nearly 1/4 of the water-improving projects in Shandong province are abnormal. The water fluoride of the project of more than 1/3 are exceeding the standard( > 1.0 mg/L) in 4415 projects, and water-fluoride of 97 projects are higher than 4.0 mg/L. More measures should be taken to strengthen the supervision of the projects and monitoring of water fluoride.
9.Comparison of body fluorine levels in Liangshan and Boxing counties of Shandong province from 2007 to 2009

ObjectiveTo find out the condition of endemic fluorosis in Liangshan and Boxing counties of Shandong province,and to provide the basis for future prevention and treatment of the disease.MethodsIn 2007 -2009,100 water facilities per year were selected in each county and the water fluoride was tested in Liangshan and Boxing counties of Shandong province.Ten households were selected and their water fluoride content was measured.Dental fluorosis prevalence and dental fluorosis index of school children aged 8 - 12 were checked in all monitored places(3 villages in each county).Meanwhile,urinary fluoride was detected in children carried dental fluorosis test.ResultsIn 2007 - 2009,the median of urinary fluoride in Liangshan county was 2.35,3.73 and 1.29 mg/L,respectively; dental fluorosis detection rate was 72.73% (88/121),78.08% (57/73),and 70.64% (77/109),and the dental fluorosis index was 1.43,1.76 and 1.34.While in Boxing county,the median of urinary fluoride was 4.50,6.71 and 5.64 mg/L; dental fluorosis detection rate was 95.93% (118/123),99.06% (105/106),94.41%(152/161) and dental fluorosis index was 2.23,2.54 and 2.09,respectively.ConclusionsThe effect of prevention and control of endemic fluorosis in Liangshan county was better than that of Boxing county of Shandong province.Altering water source is an important measure to control endemic fluorosis.More defluoridation project should be carried out in endemic fluorosis areas.
10.Experimental study of K562 cell apoptosis induced by siRNA.

OBJECTIVESTo construct a siRNA expression vector pBCR6 that produces siRNA against bcr/abl mRNA and detect apoptosis rate of K562 cells after pBCR6 transfection.
METHODSTemplate sequence for siRNA was designed, synthesized and inserted into an expression vector pSilencer1.0-U6. Restriction analysis and sequencing were performed to verify the pBCR6 vector. Then pBCR6 was transfected into K562 cells by X-tremeGene Q2. pSilencer1.0-U6 was used as the control. At different time point after transfection, apoptosis rate was determined by Tunel and Annexin V+ PI with FCM.
RESULTpBCR6 was verified by restriction analysis and sequencing. The apoptosis rate of K562 cells markedly increased at 48 and 72 hour after transfected with pBCR6, and increased in a time-dependent manner [the apoptosis rate of transfected K562 cells was (47.80 +/- 1.63)% at 72 hrs, whereas the control group was (6.67 +/- 0.37)%, P < 0.0001] No prominent change in apoptosis rate was found in the control.
CONCLUSIONThe siRNA expression vector against bcr/abl mRNA was successfully constructed. The pilot study showed that pBCR6 could effectively induce K562 cells apoptosis. siRNA may be a new tool for molecular target therapy for chronic myelogenous leukemia.
