1.A STUDY ON THE BIOAVAILABILITY OF HEME IRON IN RAT

In the present work, the bioavailability of heme iron has been studied by means of the hemoglobin regeneration method with iron-deficient rats model, which was conducted by feeding a low iron basal diet. Anemic rats were then divided into groups to which basal diets with various iron levels (7, 14, 28 ppm Fe) from heme iron (Group 1,2,3) and ferrous sulfate (Group 4, 5, 6, as a reference standard) were subsequently fed respectively.After 5-week regeneration period, the regenerated hemoglobin levels of the rats of corresponding groups were (1) 9.56→12.25, (2) 9.71→12.02, (3)9.51→ 11.09, (4) 9.74→10.81, (5) 9.66→11.24, and (6) 9.67→12.05, thereby the percentage of relative bioavailability (RBA) of heme iron were 117.7%, 124.8%, and 94.9% respectively. This result showed that the lower level of heme iron has a higher RBA. The reason why the effect of high level of heme iron was relatively lower was not fully understood.
2.Application of Tono-Pen tonometer during Perioperative Period

Objective To introduce the clinical application of Tono-Pen tonometer.Methods The intraocular pressure was detected with Tono-Pen XL tonometer at different time points for sixteen patients went through laparoscopic gynecologic operation,then the statistical analysis of the results was performed.Results In processes of artificial pneumoperitoneum and low position,the intraocular pressure escalated gradually,and after artificial pneumoperitoneum,the intraocular pressure descended rapidly.Conclusion Tono-Pen tonometer is the only tonometer can be used for the patient of general anesthesia until now.
3.Application and Evaluation of OTD Method in Teaching of Rehabilitation Medicine

Objective To evaluate the effect of OTD(observation-teaching-discussion) method in teaching of kinesiology of rehabilitation medicine.Methods 110 students majoring in rehabilitation medicine were divided into two groups,routine teaching group(56 students) and OTD teaching group(54 students),and the teaching effect was evaluated by exam score and questionnaire.Results There were obvious difference in the exam score and questionnaire between two groups(P<0.01).Conclusion In the teaching of medical rehabilitation,OTD method is more advantageous than routine method in bring up students' study interest,the initiative spirit,and the ability of self-study and analyzing.
4.Treatment of great saphenous varicosity by inserting optical fiber straigat into EVLT

Objective To investigate the methods of treating great saphenous varicosity by inserting optical fiber straight into EVLT.Methods Wuhan Linyun Laser Therapeutic System was introduced to treat 205 cases (257 limbs)of great saphenous varicosity.Optical fiber instead of blood vessel sheath and catheter was inserted straight into endovenous laser treatment(EVLT)through arteriovenons permanent needle was adopted.Results Operations lasted 35~55 minutes,averaging 45 minutes.Patients were hospitalized for 5~7 days.Postoperative vis- its were followed for 3~12 months,averaging 6 months.Two months after the operation,appraisal according to the curative effect evaluation criteria was carried on.The cure rate for superficial varicosity is 97.3%(250/257),for lower extremities swelling asthenia is 93.7%(150/160),for skin pigmentation is 91.8%(56/61),and for swelling is 90.9%(20/22).10 cases(3.9%)suffered from skin ambustion.9 cases(3.5%)suffered from shank anesthesia and stabbing pain.15 cases(5.8%)suffered from hypodermic tubercle.The patients had not deep venous thrombosis after the operation and the wounds healed well.Conclusion Inserting optical fiber straight into EVLT through arte- riovenous permanent needle brings about more accurate treatment.It can be operated simply,causing small wounds and less pain.

6. Expression of matrix metalloproteinase-9 in human abdominal aortic aneurysmal tissues

Objective: To study the effects of MMP-9 (Matrix Metalloproteinase-9, MMP-9) in the pathogenesis of abdominal aortic aneurysms (AAAs) by localizing the expression of MMP-9 in the aneurysmal tissues. Methods: By means of immonohistochemistry, the frozen sections (5 μm) with aneurysmal tissues (n = 10) were incubated with MMP-9 antibody-added agents, then the sections were stained and observed under the microscope to localize the expression of MMP-9, which displayed a brown precipitate within the arterial walls. The normal arterial wall tissues (n = 10) and the diseased arterial wall tissues from the arterial occlusive diseases (AODs) (n = 15) were also immunized exactly the same way as control. Results: A quantity of positive granules which appeared within the aortic media showed the strong expression of MMP-9 in the AAAs, with the positive rate reaching 95%(19/20), while no expression of MMP-9 was observed in the normal artery. However, the scattered distributed positive granules were seen within the arterial wall of some cases of the AODs, implying the weak positive expression of MMP-9 in this disease with the positive rate of 26.7% (4/15). There was a significant difference of the expression of MMP-9 within the arterial wall between the AAAs and AODs(P<0.01). Conclusion: High expression of MMP-9 within the aortic media faciliatates the degradation of collagen and elastin fibres and subsequent dilation of the aortic artery , thus playing an important role in the pathogenesis of AAAs. To refrain MMP-9 from enhanced expressing within the aortic wall is of clinical significance in the prevention and treatment of AAAs.

8.A Case Control-study on the Associations between Polymorphism of GSTT1,GSTM1 and Susceptibility for Breast Cancer

Objective To explore the relation of the polymorphism of GSTT & GSTM1 genes with the susceptibility of breast cancer.Methods A hospital-based case-control study was conducted,and 105 normal women as a control group and 100 women cases with breast cancer were recruited.The polymorphism of GSTT1 and GSTM1 were assessed by polymerase chain reaction(PCR).Logistic regression was conducted to evaluate the odds ratio of breast cancer in relation to single,multiple gene and estrogen exposure factors.Results The results showed that significant difference was found in the distribution of GSTT1,GSTM1 between controls and cases.There was a negative correlation between CSTT1 & GSTM1 and the risk of breast cancer with OR of 2.133 and 2.458,respectively.Polygenic model analysis indicated that there was a statistically significant correlation of the vacancy of GSTT1 & GSTM1 with breast cancer.Furthermore,it was also revealed that individual in simultaneous lack of GSTT1 and GSTM1 would run 12 times as high risk as normal controls did(OR=12.338).Results from unconditional logistic regression analysis showed that GSTT1 and GSTM1 were significantly associated with the occurrence of breast cancer.Conclusion The study indicated that the polymorphisms of estrogen-metabolizing genes were related to the risk of breast cancer.
9.Cthrc1 Inhibits Proliferation of Hepatic Stellate Cells in Mice

Background:Collagen triple helix repeat containing protein-1(Cthrc1)has been reported playing an important role in liver diseases,especially in liver fibrosis,however,its effect on hepatic stellate cells proliferation is not fully clear. Aims:To investigate the effect of Cthrc1 on proliferation of hepatic stellate cells. Methods:Recombinant adenovirus vector of Cthrc1 was constructed. After injecting Ad-Cthrc1 through tail vein,mRNA and protein expressions of Cthrc1 were determined by real-time PCR and Western blotting,respectively. Liver fibrosis model was established by bile duct ligation and fed with 3,5-diethoxycarbonyl-1,4-dihydrocollidine(DDC)in mice,respectively. The liver fibrosis mice were injected with Ad-Cthrc1 or Ad-GFP through tail vein. Immunofluorescence was used to determine number of hepatic stellate cells. Results:Recombinant Cthrc1-adenovirus vector was successfully constructed. Real-time PCR and Western blotting showed that mRNA and protein expressions of Cthrc1 were increased in Ad-Cthrc1 group than in control group. HE and Masson staining demonstrated that mice model of liver fibrosis was successfully established. Immunofluorescence showed that overexpression of Cthrc1 inhibited significantly the proliferation of hepatic stellate cells. Conclusions:Recombinant adenovirus vector of Ad-Cthrc1 constructed can express stably in vivo,and inhibit the proliferation of hepatic stellate cells. Therefore,Cthrc1 may become a potential target for treatment of liver fibrosis.
10.Expression of Glucocorticoid Receptor in Peripheral Blood Mononuclear Cells from Patients with Pemphigus

Objective To clarify whether the mRNA expression level of glucocorticoid receptor (GR) in the peripheral blood mononuclear cells (PBMCs) can predict the clinical response to glucocorticoid (GC) in patients with pemphigus. Methods Reverse-transcription polymerase chain reaction (RT-PCR) was applied to determine the mRNA expression of GR? and GR? in PBMCs from 30 patients with pemphigus and 30 healthy volunteers. Results GR? mRNA was detected in all patients and all healthy volunteers, and the expression of GR? mRNA in pemphigus patients was significantly lower than that in healthy volunteers (P = 0.044), but no significant difference was found between patients insensitive and sensitive to glucocorticoid. In contrast, GR? mRNA was detected in 10 of 12 patients insensitive to glucocorticoid, 3 of 18 patients sensitive to glucocorticoid, and 9 of 30 healthy Volunteers.
