AIM:The different effect of bivalent immunoglobulin Yolk(IgY) was evaluated against snake venom between intragastric administration and intraperitoneal injection in mice with cobra or viper envenomation.METHODS:The venom of naja and viper was injected alternately into the leghorn hen.Bivalent anti-snake venom IgY was extracted by water dilution. The concentration of bivalent IgY in plasma was observed in indirect ELISA assay after bivalent anti-snake venom IgY taken orally. The gastric emptying function test was used for determining optimization time after gastric administration of IgY.The protective effect of bivalent anti-snake venom IgY was compared between intragastric administration and intraperitoneal injection in mice with cobra or viper envenomation.RESULTS:Bivalent anti-snake venom IgY was extracted from eggs laid in 28-42 d after the first immunization. The titers of Bivalent IgY against cobra and viper venom were 1:12 800 and 1:6 400.At the time of 2.5-3.5 h after bivalent anti-snake venom IgY was taken orally in three concentrations(75 mg,150 mg,300 mg?0.5 mL-1?20 g-1 BW),the gastric evacuation rate of mice was above 68.9%,with the plasma concentration of bivalent IgY in peak. The survival time of mice envenomation with snake venom was extremely prolonged(P

Objective To study antidepressant effects of Epimedium brevicornum extracts. Methods(Behavioral) despair models of male mice, tails suspension test(TST),and forced swimming test(FST) were used to evaluate the effects of E.brevicornum extracts on behavioral, monoamine oxidas(MAOA) and monoamine oxidase B(MAO-B) activities in brain and liver tissue, and MDA level in liver tissue of mouse.Reserpine antagonistic model was also used to investigate possible antidepressant mechanisms of E.brevicornum extracts. Results The extracts of E.brevicornum(25,50, 100,and 200 mg/kg) significantly reduced the duration of murine immobility in TST and FST,inhibited MAO-A and MAO-B activities in brain and liver of mice, and reversed the elevated liver MDA level in mice in TST.There was no significant amelioration in the decreases of body temperature in mice of Reserpine antagonistic model.Conclusion(E.brevicornum) extracts possesses the definite antidepressant properties.

Objective To observe the effect of humanistic nursing care in rehabilitation ward. Methods 440 inpatients in rehabilitation ward were randomly divided into the observation group(228 cases, treated with humanistic nursing)and control group(212 cases, treated with traditional nursing).40 cases in each group received the investigation of patients' satisfaction to the service. Results The satisfactory rate of the patients in the control group to the attitude of the service, the nursing skill and health education missionary were 72.5%,75% and 67.5% respectively, but that of the patients in the observation group were all 100%,there was a significant difference between two groups(P<0.01).Conclusion Humanistic nursing care in rehabilitation ward can improve nursing qualification and increase patients' satisfactory rate.

Objective To investigated the effects of long term use of very low protein diet(VLPD,0 3 g?kg-1?d-1) treatment on patients with chronic renal insufficiency without essential amino acids(EAAs) or related ketoacids supplement. Methods Thirty seven patients with established severe chronic renal failure (CRF)[Scr(588 2?123 5)?mol/L, Ccr(9 77?3 48)ml?min-1?(1 73m2)-1]were divided into 2 groups according to their actual protein intake: 20 patients with protein intake of (0 33?0 04)g?kg-1?d-1 were used as VLPD group, while other 17 CRF patients whose protein intake was 0 6 g?kg-1?d-1 were served as low protein diet group (LPD). Results All patients in VLPD group showed good compliance to this very low protein diet, and no one presented signs of protein malnutrition during the observation. The concentrations of serum albumin and transferrin were maintained in normal ranges during the follow up period despite the transferrin levels in both groups gradually decreased as time went on. The serum concentration of transferrin was higher in VLPD patients than that in LPD patients at the end of study (P

Objective To compare the biochemical and clinical features of antimitochondrial antibody (AMA) positive and negative primary biliary cirrhosis (PBC),and to compare the sensitivity of both the detection of M2 antibody using enzyme linked immunosobent assay (ELISA) with antigen expressed from human sources, and the detection of AMA using conventional indrect immunofluorecence for diagnosing PBC.Methods AMA and M2 antibodies were tested in 63 patients with diagnosed PBC.Results Serum AMA positive rate in PBC patients was 81%(51/63) compared with 100%(63/63) of M2 positive rate.Antinuclear and anti smooth muscle antibodies(ANA and ASMA) were more frequently positive in the AMA negative patients (83% vs 35%,and 42% vs 4%, P

AIM: To study the effect of caffeine on the large conductance calcium activated potassium (K_(Ca)) channels by patch-clamp technique on smooth muscle cells enzymatically isolated from the porcine coronary artery (PCASMC),and to investigate the effect of ryanodine on K_(Ca) being activated by caffeine.METHODS: Using the single channel patch-clamp technique, single PCASMC was isolated by collagenase, the activity of single K_(Ca) channel was recorded in porcine coronary artery smooth muscle cells.RESULTS: Caffeine (0.1-10 mmol/L) enhanced the open probability (Po) of K_(Ca) channels in a dose-dependent manner in the intracellular side of inside-out patches and its effect was almost completely abolished by washout. Caffeine decreased the mean close time markedly, but had no effect on the amplitude of K_(Ca) channels. However, ryanodine (10-40 μmol/L) decreased Po of K_(Ca) channels activated by caffeine in a dose-dependent manner in cell-attached patches. The mean open time also decreased.CONCLUSION: Caffeine directly activates K_(Ca) channels of porcine coronary artery smooth muscle cells in inside-out patches, the activity of single K_(Ca) channel is inhibited by ryanodine indirectly in cell-attached patches.

Objective:To investigate the effects of TGF-?1 on promoter activity of human transforming growth factor-beta1(TGF-?1) gene. Methods:The fragments with different length of the 5′-end sequence of the human TGF-?1 gene between -1 328 bp to +812 bp were obtained from a healthy male person genomic DNA, and the sequences were fused to chloramphenicol acetyltransferase reporter gene in the pCAT-enhancer plasmid to construct five chimeric recombinant plasmids. These recombinant plasmids were transiently transfected into rat hepatic setellate cell line(nFSC) by FuGENE6 transfection methods. The cells transfected with chimeric recombinant plasmids were cultured on media in the absence or presence of TGF-?1(2 5 ng/ml),CAT activities in transfected cells were tested and compared. Results:The different concentration of TGF-?1 can prevent the proliferation of nFSC, and these effects are dose-dependent; The presence TGF-?1 can stimulate the CAT activity of cells transfected with phTGF0 585?phTGF1 120?phTGF1 423?phTGF1 680?phTGF2 140 up to 2~5 times higher then cells cultured with absence of TGF-?1.Conclusion:The TGF-?1 can stimulate CAT activities in nFSC transfected with phTGF0 585?phTGF1 120?phTGF1 423?phTGF1 680?phTGF2 140. These results suggest that TGF-?1 have an autoregulation effect on TGF-?1 gene.
