1.Effect of neoadjuvant chemotherapy on gastric cancer based on FOLFOX7 or XELOX

Objective To investigate the effect of neoadjuvant chemotherapy on gastric cancer based on FOLFOX7 or XELOX,so as to achieve evidence of clinical practice.Methods From January 2005 to June 2010,a retrospective analysis was performed for those patients with gastric cancer and underwent surgery.The patients were divided into neoadjuvant chemotherapy group and conventional chemotherapy group according to the chemotherapy method.The clinical efficacy,Ro resection rate and survival rate were evaluated in two groups.Results A total of 185 cases were included in neoadjuvant chemotherapy group (FOLFOX7 or XELOX,113 cases) and conventional chemotherapy group (72 cases).There was no significant difference in general status between two groups (P > 0.05).Complete remission was in 48 cases,partial remission was in 51 cases,stable was in 14 cases,progression of disease was in 0 case in neoadjuvant chemotherapy group,and the effective rate was 87.6% (99/113).Complete remission was in 30 cases,partial remission was in 25 cases,stable was in 15 cases,progression of disease was in 2 cases in conventional chemotherapy group,and the effective rate was 76.4%(55/72).There was significant difference in the effective rate between two groups (P< 0.05).The R0 resection rate in neoadjuvant chemotherapy group was higher than that in conventional chemotherapy group [60.2% (68/113) vs.38.9% (28/72)],and there was significant difference between two groups (P< 0.01).There was no significant difference in survival rate between two groups (P > 0.05).Conclusion Neoadjuvant chemotherapy can reduce the tumor stage and improve the R0 resection rate,but has no obvious advantage in improving the survival rate during follow-up period.

4.Evaluation of heat-activated and common nickel-titanium wire for orthodontic treatment

BACKGROUND: Heat-activated nickel-titanium (HANT) wire is a new type of arch wire in orthodontic treatment, characterizing by well memory alloy ability and uneasily deformation. However, environmental temperature may affect its performance.OBJECTIVE: To compare the effects of HANT and common nickel-titanium wire in clinical application.METHODS: A total of 80 patients who have accepted and finished orthodontic treatment in the First Affiliated Hospital of Sun,Yat-sen University from July 2006 to November 2008 were randomly divided into two groups. The common group was treated with common arch wire in aligning and leveling stages, while the HANT group was treated using HANT in aligning and leveling stages. Tooth extracted cases were matched to non-tooth extracted cases. Indicators, such as time span for alignment, total treating time, times for wire breaking, times for bracket shedding, and times for return visits, were measured.RESULTS AND CONCLUSION: There was significant difference in treating time and times for wire breaking between the two groups (P < 0.05), but there was no significant difference in the times for bracket shedding. Anaphylactic response, toxic reaction,and other adverse reactions were not observed in the HANT group. Therefore, HANT wire can simplify orthodontic operation,shorten operation time beside the chair, save the total treating time, and reduce chances of shedding brackets and breaking wires.
5.The effects of γ-IFN on the expression of HLA-G mRNA in primary cultured astrocytoma cells

Objective To study the effects of γ-IFN on the expression of HLA-G mRNA in primary cultured astrocytoma cells. Methods Different concentrations of γ-IFN were added to primary cultured cells, and HLA-G mRNA were detected by RT-PCR. Results After γ-IFN treatment, HLA-G mRNA can not be determined from HLA-G originally negative astrocytoma cells. The expression of HLA-G are up - regulated in all original HLA-G positive astrocytoma cells in a dose-dependent manner. Conclusion γ-IFN can increase the ex-pression of HLA-G gene in the primary cultured astrocytoma cells which HLA - G are originally positive.
6.Enhancing effect of CpG on sensitivity of Lewis lung cancer to X-ray radiation in mice

Objective: To explore the role of cytosine-phosphate-guanine oligodeoxynucleotide(CpG ODN)in enhan- cing the radiosensitivity to X-ray in mouse with Lewis lung cancer.Methods: The tumor-bearing mouse model was in- duced by injevting Lewis lung cancer cells into the right infra-axillary dermis.Thirty-two C57BL/6J mice were evenly ran- domized into 4 groups.Group A: the control group;Group B: the X-Ray radiation group;Group C: the CpG group; Group D: the CpG plus X-Ray radiation group.Group B was treated with X-Ray radiation only(3 Gy/F,on day 1,3,5, 8,10,and 12;the total dose was 18 Gy);group C was administered with CpG ODN 0.05 mg on day 1,3,5,8,10, and 12;group D was administered with CpG ODN 6h before X-ray radiation.The tumor growth and tumor growth delay (TGD)were observed in all groups.Meanwhile,the pathological change of the tumor tissue was observed with H-E staining method and the apoptosis of tumor cells were examined with the method of TUNEL.Results: The Lewis hmg cancer-bearing model was successfolly established in mice.The tumor volumes of the treatment groups were smaller than that in lhe control group(P
8.Epidemiological study of malignant tumor among rubber workers in Hainan Province.

OBJECTIVETo investigate the prevalence of malignant tumor and its influencing factor among rubber workers in Hainan Province of China, and to study the high incidence of cancer and type of work.
METHODSA retrospective cohort study on registered workers over 1 working age among partial rubber factories in Hainan Province in the period from 2005 to 2010.
RESULTThere was a remarkable difference in the incidence between nature exposed rubber group and non-exposed group (χ(2) = 52.13,P < 0.01). Occupational exposure induced a high risk in malignant tumors (OR = 2.47, P < 0.05). The incidence of tumor may be associated with occupational exposure: the longer occupational exposure time, the higher was the incidence of tumor (χ(2) = 11.40, P < 0.05).
CONCLUSIONSThe occupational exposure in rubber workers can induce a high incidence of malignant tumor.

9.Biological implication of PTEN gene expression in human gastric cancer and related molecular mechanisms.

OBJECTIVETo study the role of PTEN gene involved in the biological behavior of human gastric carcinoma cells and underlying molecular mechanisms.
METHODSGastric carcinoma cell line, SGC7901, was transfected with plasmid PBP-PTEN and stable high PTEN expression clones were selected by Western blot and cell immunohistochemistry screening. Cell proliferation rate and apoptosis index of transfected cells were investigated by growth curve analysis, colony-formation assay and flow cytometry (FCM). Expressions of vascular endothelial growth factor (VEGF), matrix metalloprotease-2 (MMP-2) and MMP-9 proteins in cell culture supernatant and cytoplasm were determined by ELISA, gelatin zymogram, Western blot and cell immunohistochemistry.
RESULTSStable clone with high level expression of PTEN was successfully established (PTEN-SGC7901). Cell doubling time of PTEN-SGC7901 was longer than that of the control cells (P < 0.05). The size and colony-forming efficiency of PTEN-SGC7901 cells deceased compared with those of the control. The relative colony-inhibition efficiency of PTEN-SGC7901 to SGC7901 (naïve untransfected) and PBP-SGC7901 (control vector transfected) cells were 69.8% and 64.8%, respectively. PTEN-SGC7901 clone had more cells at G1 phase (P < 0.05) compared with that of the control. However, the apoptosis index did not show significant differences among the three groups (P > 0.05). There were significantly less VEGF and MMP-9 protein expressions in the PTEN-SGC7901 culture supernatant and cytoplasm (P < 0.05). In contrast, the MMP-2 expression among three cell groups had no significant difference (P > 0.05).
CONCLUSIONSPTEN expression suppresses the growth and proliferation of gastric carcinoma cell SGC7901, possibly through an inhibition of the expressions of VEGF and MMP-9.

10.Real-time UV imaging of chloramphenicol intrinsic dissolution characteristics from ophthalmic in situ gel.

In this paper, chloramphenicol was selected as a model drug to prepare in situ gels. The intrinsic dissolution rate of chloramphenicol from in situ gel was evaluated using the surface dissolution imaging system. The results indicated that intrinsic dissolution rate of chloramphenicol thermosensitive in situ gel decreased significantly when the poloxamer concentration increased. The addition of the thickener reduced the intrinsic dissolution rate of chloramphenicol thermosensitive gel, wherein carbomer had the most impact. Different dilution ratios of simulated tear fluid greatly affected gel temperature, and had little influence on the intrinsic dissolution rate of chloramphenicol from the thermosensitive in situ gel. The pH of simulated tear fluid had little influence on the intrinsic dissolution rate of chloramphenicol thermosensitive in situ gel. For the pH sensitive in situ gel, the dissolution rates of chloramphenicol in weak acidic and neutral simulated tear fluids were slower than that in weak alkaline simulated tear fluid. In conclusion, the intrinsic dissolution of chloramphenicol from in situ gel was dependent on formulation and physiological factors. With advantages of small volume sample required and rapid detection, the UV imaging method can be an efficient tool for the evaluation of drug release characteristics of ophthalmic in situ gel.
