1.Effect of enamel matrix proteins on gene expression in human bone marrow stromal cells analysed by cDNA microarray

Objective To investigate the effect of enamel matrix proteins (EMPs) on gene expression in human bone marrow stromal cells (hBMSCs) by cDNA microarray. Methods hBMSCs were cultured in vitro by whole bone marrow technique. Those stimulated with EMPs at a concentration of 200 ?g/mL in vitro were classified as test group, and those normally cultured were served as control. Afer culturing for 5 days, a cDNA microarray containing 4 096 genes was used to detect and analyze the gene expression. Four differentially-expressed genes in cDNA microarray were determined by real-time PCR for validation of the microarray data. Results Twenty-seven genes of hBMSCs were differentially expressed in the presence of EMPs, among which 18 were up-regulated and 9 down-regulated. The result of real-time PCR was in accordance with that of the cDNA microarray. Conclusion cDNA microarray technique can screen the differentially-expressed genes in hBMSCs treated with EMPs, which lays a foundation for the research of molecular mechanism of EMPs in inducing the regeneration of periodontal tissue.

IL-6 and IL-17 levels in saliva and serum were measured by ELISA in 116 cases of chronic periodontitis(CP)in Mulei district of Xinjiang,including 62 cases of Han and 54 of Kazak.Health controls included 50 subjects of Han and 45 of Kazak.IL-6 and IL-17 levels in serum and saliva were higher in CP groups than in the controls(P <0.05),and correlated with the severity of CP(P <0.05),in Kazak CP group were higher than in Han CP group(P <0.05).

Objective To investigate the diagnostic value of a double action MR contrast agent——gadobenate dimeglumine (Gd-BOPTA) for focal liver lesion and biliary system disease.Methods Articles about Gd-BOPTA in CNKI and PubMed for the past few years were searched and the value of Gd-BOPTA in the diagnosis of focal liver lesion and biliary system disease was summarized.Results For focal liver lesion,Gd-BOPTA not only can reveal blood supply of the lesion,but also reveal the hepatocellular functional status in the lesion.For biliary system,biliary excretion of Gd-BOPTA can be used to evaluate the anatomic structure of bile duct,function of gallbladder and biliary system disease.Conclusions Gd-BOPTA has an important value in the diagnosis of focal liver lesion and biliary system disease.Gd-BOPTA may have wider applications in the future.
5.Preparation and in vitro evaluation of chitosan nanoparticles containing 5-[125Ⅰ] Iodo-2'deoxyuridine

Objective To prepare the chitosan nanoparticles loading 5-[125Ⅰ] Iodo-2'-deoxyuridine (125Ⅰ-UdR-CS-DLN) at 100-200 nm in diameter, and analyze the characteristic of drug sustained-releasing and tumor targeting. Methods Orthogonal experimental design and One-way analysis were applied to optimize the preparation of 125Ⅰ-UdR-CS-DLN using tripolyphosphate cross-linking. Dynamic dialysis was utilized to investigate the in vitro releasing characteristics of the nanoparticles. The tumor targeting effect of the nanoparticles was observed with laser confocal microscopy. Results The optimal conditions for preparing the nanoparticles at particle diameters (70. 39 ± 5.12 ) nm (PDI 0. 16 ± 0. 012 ) were 1 g/L of CS, 2 g/L of TPP, stirring rate 600 r/min, relative molecular mass of CS 3 × 103. The TEM results showed that the exterior of the nanoparticles was spheroid, with a uniform and fine dispersivity. The optimized condition with the initial 125Ⅰ-UdR concentration of 2. 96 MBq/ml at pH5 provided the highest loading capacity (1253. 55 MBq/g) and the highest entrapment rate (42. 35% ). The in vitro releasing curves of 125Ⅰ-UdR-CS-DLN followed Higuchi equation, shown a characteristic of long-acting preparation.Laser confocal microscopy observations approved that the tumor cells uptake of FITC-CS-nanoparticles were significantly more than that of normal cells. Conclusions Chitosan nanoparticles loading 125Ⅰ-UdR at diameters range 127. 81 ± 15. 25 nm (PDI 0. 240 ± 0. 035 ) were successfully prepared with the optimized conditions, and showed a characteristic of sustained-releasing and tumor targeting. The chitosan-based nanotechnology provided a new and efficient approach for the application of 125Ⅰ-UdR in intracellular radiotherapy for tumor.
6.Three new xanthones from Garcinia xanthochymus

To study xanthones from the barks of Garcinia xanthochymus, the constituents were isolated by normal-phase and reverse-phase silica gel column chromatography from the EtOAc extract. Their structures were elucidated by spectral analysis. Three new xanthones were purified and identified as 1,2,5-trihydroxy-6-methoxyxanthone (1), 1,4,6-trihydroxy-5-methoxyxanthone (2), 1,2,7-trihydroxy-4-( 1,1-dimethylallyl) xanthone (3).
7.Rabbit nucleus pulposus cells induce the differentiation of mesenchymal stem cells after co-culture in a three-dimensional environment

BACKGROUND: In the field of intervertebral disc tissue engineering, seed cells primarily consist of nucleus pulposus cells (NPCs) and mesenchymal stem cells (MSCs). NPCs have been known to have several shortcomings: limited source, inconvenient collection, poor proliferative capacity, and difficult in vitro culture.OBJECTIVE: To investigate the proliferation of MSCs after co-culture with NPCs in alginate beads-simulated three-dimensional environment.DESIGN, TIME AND SETTING: A cell observation was performed at the Laboratory of Orthopedics, Tongji Hospital, Tongji Medical College of Huazhong University of Science and Technology between December 2007 and October 2008.MATERIALS: Six healthy New Zealand rabbits of 4 months old were provided by the Laboratory Animal Center, Tongji Medical College of Huazhong University of Science and Technology and were used for the present study.METHODS: Rabbit MSCs were isolated and purified using density gradient centrifugation and adherence methods. Rabbit NPCs were isolated and purified using collagenase Ⅱ digestion and adherence methods. Following liposome-mediated green fluorescent protein tranfection and G418 screening, MSCs of passage 3 were cultured either alone (control group) or with NPCs at a ratio of 1:1 (co-culture group) in alginate beads. After 14 days of culture, alginate beads were dissolved and MSCs were collected by flow cytometry sorting.MAIN OUTCOME MEASURES: The expression of collagen Ⅱ and aggrecan in MSCs was detected by RT-PCR and Western blot techniques.RESULTS: mRNA and protein expression of collagen Ⅱ and aggrecan was observed in the co-culture group, but not in the control group, after 14 days of culture.CONCLUSION: MSC differentiation towards nucleus pulposus-like cells can be induced by co-culture with NPCs in a three-dimensional environment.
8.Effects of different acupuncture and moxibustion treatments on cervical spondylotic radiculopathy:A systematic evaluation

OBJECTIVE:To evaluate the effect and safety of conservative treatment for treating cervical spondylotic radiculopathy by cochrane evaluation systems.DATA SOURCE:CBM(from January 1978 to October 2008),CNKI(from January 1978 to October 2008)and VIP(from January 1989 to October 2008)databases were retrieved by computer and partial documents were searched by hands.DATA SELECTION:Randomized controlled trial was adopted in course of acupuncture and moxibustion treatment for cervicel spondylotic radiculopathy and its quailty was assessed.Dafa source was randomized and semi-randomized controlled.Some evaluations such as random method-selection bias,allocation concealment-selection bias,blind method-performance bias and measurement bias,exclusion.lose to follow-up,withdraw-attrition bias were chosen.MAIN OUTCOME MEASURES:①Abdominal acupuncture needles were superior to the conventional method's efficiency.②Electro-acupuncture plus cupping with barbed floc were superior to the conventional method's efficiency.③Abdominal acupuncture were superior to traction's efficiency.④Cervical traction,electro-acupuncture combined with point injection and massage were superior to traction's efficiency.⑤Traction,electro-acupuncture,massage,TDP-guided were superior to traction,physiotherapy,TDP combined with exercise therepy's efficiency.⑥Vinegar iontophoresis and electro-acupuncture were superior to simple electro-acupuncture's efficiency.⑦Long's approach acupuncture was superior to simple acupuncture's efficiency.⑧Adverse reaction.RESULTS:A total of 1416 subjects were selected from 7 randomized controlled trials.All the studies have selection bias,measurement bias,loss of the high probability of bias,the quality and grade of"C"grade.And only one study proved that the efficiency of the experimental group was better than that of the control group.Study 1 showed that there was insufficient evidence suggested that acupuncture treatment of abdominal cervical spondylotic radiculopathy was efficient than conventional acupuncture.Study 2 showed that there was insufficient evidence suggested that electro-acupuncture plus cupping therapy with barbed floc was efficient than conventional acupuncture to cure cervical spondylotic radiculopathy.Study 3 showed that there was insufficient evidence suggested that acupuncture treatment of abdominal cervical spondylotic rediculopathy was efficient than traction.Study 4 showed that the cervical traction,electro-acupuncture combined with acupoint-injection treatment of cervical spondylotic radiculopathy was efficient than traction combined massage therapy.Study 5 showed that there was insufficient evidence suggested that traction,electro-acupuncture,massage,TDP-guided therapy was efficient than traction,physical therapy,TDP combined with exercise therapy.Study 6 showed that there was insufficient evidence suggested thatelectro-acupuncture combined with vinegar iontophoresis treatment of cervical spondylotic radiculopathy was better than the simple electro-acupuncture therapy.Study 7 showed that there was insufficient evidence suggested that the acupuncture therapy using Long's approach was better than simple acupuncture.No adveme reactions appeared to terminate the test in 7 studies.CONCLUSlON:The above seven studies mainly present with low quality,absence of a unified curative method,small samples,difference of follow-up time,iudgment standard and measurement standard.So more samples,multicenter and more regular randomized controlled trial should be performed.
10.Comparison between intravitreal Ranibizumab and Tramicinolone acetonide for macular edema secondary to central retinal vein occlusion

AIM:To compare the efficacy and safety of intravitreal ranibizumab to those of triamcinolone acetonide ( TA ) injection for the treatment of macular edema secondary to central retinal vein occlusion ( CRVO) .
METHODS:This retrospective study included 40 eyes of 40 patients with macular edema associated with CRVO. Twenty patients 20 eyes were treated with intravitreal injection of triamcinolone acetonide (1mg, 0. 1mL), the other 20 patients 20 eyes accepted intravitreal ranibizumab (0. 5mg, 0. 05mL). The change of best corrected visual acuity ( BCVA ) , central macular thickness ( CMT ) , and intraocular pressure ( IOP ) before treatment and at 1, 2wk, 1, 2,3,6mo post-injection in the two groups were observed.
RESULTS:BCVA was improved at 1, 2wk, 1, 2,3,6mo post-injection in the TA group (P<0.05) and ranibizumab group ( P<0. 05 ). No significant difference was found between the two groups ( P > 0. 05 ). CMT decreased significantly within each group ( P < 0. 05 ), and no significant difference between groups was found ( P >0.05). In the TA group, the IOP was significantly higher at 2wk and 4wk than before treatment (P<0. 05). In the ranibizumab group, no elevated IOP was observed at 1, 2wk, 1, 2,3,6mo (P>0. 05). However, the IOP at 1mo was significantly higher in the TA group than that in the ranibizumb group (P<0. 05).
CONCLUSION:Intravitreal ranibizumab is an effective and safe treatment method for macular edema secondary to CRVO. It can effectively improve BCVA and reduce CMT without ocular and systemic complications compared with intravitreal TA.
