Objective To investigate the in vitro effect of low dosage of paclitaxel on normal murine bone marrow-derived dendritic cells(mDCs)and its role in reactivating tumor-pulsed DCs. Methods The concentration of paclitaxel which could induce 30% apoptosis of 3LL cell lines was figured out.mDCs were generated from murine bone marrow precursors.Cell culture insert system was used and four groups were divided as following: mDC,mDC+3LL,mDC+ low dose of paclitaxel,and mDC+3LL with 30% apoptosis induced by low dose of paclitaxel.The phenotypes,chemoattractive function to MIP1? and MIP-3?,and viability in activating allogeneic T cell proliferation of DCs in the four groups were analysed. Results Paclitaxel of 50 nmol/L could induce 30% apoptosis of 3LL,and had protective effects on DCs.It could stimulate the maturation of mDCs by up-regulating the phenotypes of CD11cCD80,CD11cCD86,CD11cCD40 and CD11cCDIab,and could enhance the chemoattractive function to chemokine MIP-3?.Compared with those cocultured with 3LL,DCs pulsed with apoptosis antigen of 3LL cell which was induced by 50 nmol/L paclitaxl up-regulated the phenotype of CD11cCD40,enhanced the the chemoattractive function to MIP1? and MIP-3?,and activated the proliferation of T cells. Conclusion Paclitaxel of 50 nmol/L can stimulate the maturation of DC,and can partially recover the phenotype and function of tumor-pulsed DC.

Objective:To Study the effect of static magnetic field on the proliferation and differentiation of osteoblasts.Methods:In vitro cultured rat calvarial osteoblasts were seeded into 96-well culture plate and exposed to the different magnetic intensity field for 24,48 and 72 h respectively.MTT assay was applied to study the cell proliferation. ALP was measured by a microplate reader.Results:When the magnetic exposure time extended to 48 h or 72 h, the corresponding MTT value of osteoblasts with magnetic treatment of 40 mT or 62 mT increased obviously(P0.05). ALP activity increased after 24 h exposure to static magnetic field, the effect of static magnetic field intensity of 62 mT and 83 mT showed the strongest promotion effect(P

Using site-directed mutagenesis and DNA recombinant technology, GGC fragment was inserted into human IL-18 cDNA. The mutated IL-18 cDNA was constructed into plasmid pPIC9K, then transformed into Pichia pastoris GS115 and efficiently expressed. A Glycine residue was inserted into the recombinant IL-18 between Arg39 and Asp40 to form a RGD motif. The mutated IL-18 was termed IL-18-RGD. The protein was purified with Sephadex G-100. The cell culture of melanoma B16 showed that IL-18-RGD efficiently inhibited B16 tumor growth, IC50 = 8.10?mol/L, but the inhibitory effect of IL-18 was not detected. Both IL-18-RGD and IL-18 showed inhibitory activities on mice loaded B16 in vivo, the inhibitory efffct of IL-18-RGD was stronger than that of IL-18. The inhibitory activities of IL-18-RGD and IL-18 on bFGF induced angiogenesis on chorioallantoic membranes were detected. There was no differences between IL-18-RGD and wild IL-18 in the activity of inducing IFN-? in human PBMC.

Objective To study the relationship of the collection efficiency and physical parameters,such as the height of exhaust hood,baffle,ventilation conditions of these parameters on the effect of control.Methods The computational fluid dynamics software FLUENT was used to form the model,and to maximize production in line with the actual operating conditions for physics experiments,to take air numerical simulation and physical model of re-testing the method of combining.Results Without the lagging conditions,the air flow system should be chosen 1 320 m~3/h,with the lagging conditions,the air flow system should be chosen 1 078 m~3/h.The system in a certain air volume,lower installation height could increase the average wind speed of breathing zone.At a certain suction cover installation height and air flow,the baffle could improve the collection efficiency.Without the baffle,the collection efficiency and control distance(dimensionless)showed a relation of cubic polynomial.Conclusion The physical parameters of suction on the hood may impact the collection efficiency at breathing zone measuring point.

In the process of their application, they finally promoted the capacities for their competition and innovation which were undoubted to help us to establish headship/main forces in the fields of infectious disease precaution and devolvement of new clinical medicine in the future.

Objective:To investigate the effects of TGF-?1 on promoter activity of human transforming growth factor-beta1(TGF-?1) gene.Methods:The fragments with different length of the 5′-end sequence of the human TGF-?1 gene between -1 328 bp to +812 bp were obtained from a healthy male person genomic DNA, and the sequences were fused to chloramphenicol acetyltransferase reporter gene in the pCAT-enhancer plasmid to construct five chimeric recombinant plasmids. These recombinant plasmids were transiently transfected into rat hepatic setellate cell line(nFSC) by FuGENE6 transfection methods. The cells transfected with chimeric recombinant plasmids were cultured on media in the absence or presence of TGF-?1(2 5 ng/ml),CAT activities in transfected cells were tested and compared.Results:The different concentration of TGF-?1 can prevent the proliferation of nFSC, and these effects are dose-dependent; The presence TGF-?1 can stimulate the CAT activity of cells transfected with phTGF0 585?phTGF1 120?phTGF1 423?phTGF1 680?phTGF2 140 up to 2～5 times higher then cells cultured with absence of TGF-?1.Conclusion:The TGF-?1 can stimulate CAT activities in nFSC transfected with phTGF0 585?phTGF1 120?phTGF1 423?phTGF1 680?phTGF2 140. These results suggest that TGF-?1 have an autoregulation effect on TGF-?1 gene.

Objective To investigate the guiding significance of medical image three-dimensional visualization system (MI-3DVS) in precise hepatectomy. Methods The clinical data of 45 patients with hepatic neoplasms who were admitted to the Zhujiang Hospital from June 2008 to September 2010 were prospectively analyzed. The preoperative image data of the liver were three-dimensionally reconstructed by MI-3DVS. According to the distribution of the intrahepatic portal veins and hepatic veins, the liver was divided into different sections,and then tumors can be located within these hepatic segments. The volume percentage of residual liver and volume of liver resected were detected. Evaluation of surgical resectability and surgery simulation were done before operation. Results According to the distribution of the intrahepatic portal veins and hepatic veins, all patients were divided into seven types: 21 patients were with normal type which was the same as Couinaud type, six with nondivided type, 11 with non-divided right liver type, four with non-divided left liver type, one with right hepatic vein type, one with double middle hepatic vein type and one with right posterior vein type. Thirty-nine patients received open hepatectomy, and the volume percentage of the residual liver was 74% ± 17%. Postoperative pathological examination confirmed that all the 39 patients were with hepatocellular carcinoma. Six patients received transcatheter arterial chemoembolization. No severe complications such as acute hepatic failure, bleeding, bile leakage were detected. All patients were followed up for six months, and they survived with or without tumor. Conclusion MI-3DVS has guiding significance in preoperative assessment and perioperative guidance for precise hepatectomy.
