2.Genetics comparison on the Yersinia pestis strains of 3 kinds of ecotypes in the Sanjiangyuan area, Qinghai Province

Objective To compare the genetic polymorphism of 34 Yersinia pest strains of 3 kinds of ecotypes(Microtus fiscis,Qilian Mountain and Qinghai-Tibet Plateau ecotype)that isolated from the Sanjiangyuan area in Qinghai Province and Shiqu County in Sichuan Province,in order to know the ecotype relationship among the strains.Methods Thirty-four strains were amplified using the application of random amplified polymorphism DNA(RAPD)and were detected with agaroge gel electmphoresis.Results Amplified products by ngarose gel electrophoresis method showed same stripes in 31 strains,only 3 strains had slight differences.Conclusion The Microtus fuscas,Qilian Mountain and Qinghai-Tibet Plateau ecotype strains isolated from Sanjiangyuan area are genetically homologous.
3.Epidemiological analysis of the human plague from 2001 to 2011 in Qinghai Province

Objective To analyze human plague from 2001 to 2011 in Qinghai Province and to provide a scientific basis for formulating prevention and control measures.Methods Using the descriptive epidemiological methods,epidemiological field survey data and medical records of each case of human plague were collected from 2001 to 2011 in Qinghai Province.Human plague was judged in accordance with the Plague Diagnostic Criteria (WS 279-2008).Results From 2001 to 2011,human plague was reported 14 times,with incidence of 38 cases,17 dead and death rate was 44.74% in Qinghai Province.Epidemic areas mainly distributed in the 12 townships of 9 counties.Prevalent season was from May to October,September and October accounted for 57.89% (22/38).There were cases of Tibetan herders and Han farmers,accounting for 76.32% (29/38) and 23.68% (9/38),respectively;onset age from 5 to 67 years,mainly around the age of 20-45 [68.42% (26/38)].The most prevalent clinical types were pneumonic and septicemic plague and initial case was caused by actively contact with infected plague animals.Conclusions Qinghai human plague is mainly caused by approaching the plague infected animals,human plague in Qinghai Province is on the rise,the risk of long-distance transmission of the plague is significantly increased.

5. Comparison on pharmacodynamic differences between Simiao Pill with crude Atractylodis Rhizoma and bran stir-baked Atractylodis Rhizoma

Objective: To study the pharmacodynamic differences between Simiao Pills with crude Atractylodis Rhizoma and bran stir-baked Atractylodis Rhizoma on the treatment of adjuvant arthritis. Methods: Wistar rats were randomly divided into blank control group (normal saline), model control group (normal saline), Tripterygium wilfordii glycosides (TWG) group (9.45 mg/kg), low-and high-dose (1.080 and 4.320 g/kg) Simiao Pills (crude Atractylodis Rhizoma) group, low-and high-dose (1.080 and 4.320 g/kg) Simiao Pills (stir-baked Atractylodis Rhizoma with bran) group. Except the blank control group, the other groups were modeled to adjuvant arthritis with complete Freund's. Paw edema value, spleen and thoracic gland indexes, serum interleukin-1β (IL-1β), and nitric oxide (NO) were observed. Results: Both Simiao Pills with crude Atractylodis Rhizoma and bran stir-baked Atractylodis Rhizoma could inhibit paw edema, decrease spleen index, advance the thoracic gland index, and decrease IL-1β and NO. The Simiao Pills with crude Atractylodis Rhizoma showed better effects. Conclusion: Both Simiao Pills with crude Atractylodis Rhizoma and bran stir-baked Atractylodis Rhizoma show a certain therapeutic effect on adjuvant arthritis, but the crude Atractylodis Rhizoma is in favor of the efficacy of Simiao Pills.
6.Analysis of clinical manifestations and emergency operations of Crohn disease:a report of 26 cases

Objective To discuss the clinical features,diagnosis and operative treatment in Crohn disease(CD)characterized by acute abdomen.Method The clinical data of 26 CD cases testified by pathology after emergency operations from April 1997 to February 2009 were retrospectively analyzed.Results In 26 cases of acute abdomen,17 cases characterized by mechanical obstruction,6 cases characterized by peritonitis,3 cases characterized by hemorrhage of digestive tract.Eight cases(30.8%)were diagnosed before operation,and 13 cases(50.0%)were diagnosed during operation,totally 80.8%(21/26).All patients received surgical operations as following:resection and anastomosis in 21 eases,resection and fistulation in 4 cases,ileotransverso-stomy in 1 case.Two cases died after operation,and others discharged including 1 case received second operation within 5 days.Conclusion It is difficult to be diagnosed before operation when CD characterized by acute abdomen,and appropriate emergency operations are needed according to the particular cases.

9.Meta-analysis of disodium cantharidinate injection combined with transcatheter arterial chemoembolization in the treatmemt of hepatocellular carcinoma

Objective To evaluate the efficacy and safety of disodium cantharidinate injection combined with transcatheter arterial chemoembolization(TACE) compared with TCAE alone in the treatment of hepatocellular carcinoma(HCC) by meta-analysis.Methods Databases of Pubmed, CNKI,Wangfang and VIP were searched electronically, and the randomized controlled trials about disodium cantharidinate injection combined with TACE in the treatment of HCC were included.Meta-analyses were conducted after the quality of the included studies was assessed.Results Seven eligible studies that included a total of 562 HCC patients were identified in the present meta-analysis.The combined results showed that disodium cantharidinate injection combined with TACE in the treatment of HCC, compared with TACE alone, could increase effective rate ( RR =1.31, 95%CI 1.10-1.56) and clinical benefit rate (RR =1.20, 95%CI 1.04 -1.39), improve the life quality (RR =1.60, 95%CI 1.26 -2.03) and improved the 1, 2 year survival (RR =1.57 with 95%CI 1.28 -1.93 and RR =2.08 with 95%CI 1.46 -2.97, respectively).Conclusion Meta-analysis indicates that the efficacy of disodium cantharidinate injection combined with TACE is superior to TACE alone for the patients with HCC.
10.Progress on glioblastoma multiforme treatment with chimeric antigen receptor T-cells

Glioblastoma multiforme (GBM) is the most malignant form of glioma, and its treatment through traditional surgery combined with chemotherapy and radiotherapy has limited efficacy. Chimericantigen receptor T-cells (CAR-T) are recombinant receptors for antigen, which, in a single molecule, redirect and mediateantigen recognition, T-cell activation, and, in the case of second-generation chimeric antigen receptors (CARs) costimulation (CD28 or 4-BB), augment T-cell functionality and persistence. CARs are the focus of attention in emerging treatment options for GBM. This article mainly introduces the development process of CAR-T therapy and the recent success of adoptive transfer of CAR-T cells. Effective targets of the treatment of GBM with CAR-T according to this research are discussed as well. Some of the most extensively studied targets on GBM, especially interleukin-13 receptor α chain variant 2, epidermal growth factor receptor-Ⅷ(EGFRⅧ), human epidermal growth factor receptor 2 (ErbB2), and ephrinA2 receptor (ErbA2), and the different characteristics of each kind of alloantigen-specific CAR-T cells, are the basis for CAR-T therapy and indicate their different characteristics or utilities and the prospect of further clinical research. The discovery of selective expression of interleukin-13 receptor alpha 2 in glioma cells more than 20 years ago prompted the clinical trial of CAR-T therapy in stage I GBM tumors, and the therapy was proven safe and effective. EGFRⅧ is a neoantigen presenting only in cancer cells and glioblastoma stem cells. Its presence is correlated with poor prognosis, and a phase Ⅰ/Ⅱ trial is ongoing at different institutes. ErbB2-specific CARs were also expressed in human Tcells.Adoptive transfer of EphA2 (or ErbB2)-specific T cells resulted in the regression of glioma xenografts. Thus, target-specific CAR-T immunotherapy may be a promising approach for the treatment of different target-positive GBM. Finally, we summarize the application value and challenge of CAR-T cell therapy in the treatment of GBM.
