T cell immunoglobulin mucin (TIM) family plays a key role in regulating immune responses. In this study, the interactions of human TIM family with apoptotic cells were evaluated in order to provide a foundation for further study on the roles of human TIM genes in apoptosis. Nine kinds of pEGFP-N1 eukaryotic expression vectors containing different lengths of the three members of human TIM genes for the expression of TIM-EGFP and the vectors for the expression of TIM-Fc fusion proteins were constructed. It was found that human TIM proteins could recognize and bind to apoptotic cells directly, but not to viable cells. The interactions of sTIM-1-EGFP, sTIM-3-EGFP and sTIM-4-EGFP with apoptotic cells were blocked by TIM-1-Ig, TIM-3-Ig and TIM-4-Ig fusion proteins respectively. In addition, human TIM proteins mediated the recognition of apoptotic cells and bound to apoptotic cells directly via the IgV domains. In conclusion, the TIM family may play a key role in the regulation of apoptosis. Our data also suggest that human TIM proteins probably serve as novel proteins for the detection of the early cellular apoptosis.

The structure and the blood supply of perichondral ring of human being and rabbits in developing period were studied by microangiographic and histological methods. Perichondral ring is situated around the epiphyseal disc and metaphysis and consists of osseous ring, zone of mesenchymal cell, which contains fairly abundant blood vessels derived from the metaphyseal periosteum and perichondrium. The perichondrium is thick and also contains abundant vessels which connect epiphyseal and metaphyseal circulations in the extra-osseous and part of which passes through the zone of mesenchymal cell to supply the epiphyseal disc. Role of perichondral ring in the development period of bones were discussed.

BACKGROUND: Loss of sacral slope has been shown to be an important anatomic basis for low back pain. The effect of sacral slope changes after lumbar fusion on pain relief remains unclear.OBJECTIVE: To analyze the variations of sacral slope and clinical significance at the early stage after posterior lumbar interbody fusion at L4-5.METHODS: Sixty patients with herniation at disc levels L4-5 and spinal stenosis (n=38) or lumbar spondylolisthesis at L4 (n=22) undergoing posterior lumbar interbody fusion were enrolled. All patients were followed up for 12-24 months to compare the sacral slop at baseline and postoperatively. The clinical outcomes were evaluated by MacNab criteria, and its correlation with age, sex and sacral slop variations was analyzed.RESULTS AND CONCLUSION: At 12-24 months postoperatively, the sacral slope in the two groups was significantly improved than that at baseline (P < 0.05), but there was no significant difference in sacral slop between two groups (P > 0.05). The age, sex and sacral slop variations showed no significant effects on the early clinical outcomes (P > 0.05).These results suggest that posterior lumbar interbody fusion can markedly improve the sacral slope in patients with spinal stenosis and lumbar spondylolisthesis at early period. Furthermore, age, sex, and sacral slope variations all show no obvious effect on postoperative early functional recovery.

Borderline personality disorder (BPD) is characterized by emotional regulation, interpersonal relationship, self-image and impulse control instability. It is a mental disorder with high morbidity, high mortality and difficult treatment. Psychotherapy is the main treatment for this disease. However, current mainstream psychotherapy, such as dialectical behavior therapy (DBT), has certain limitations, so it is necessary to find a more convenient and effective treatment method. Acceptance and commitment therapy (ACT), as the representative therapy of the third generation of cognitive therapy, aims to make individuals accept inevitable pain, find their own value and commit to action, and finally improve their psychological flexibility to live a meaningful life. ACT has been used in the treatment of a variety of mental disorders, and the effect is significant. By expounding the pathogenesis of BPD and the theoretical model of ACT, this paper analyzed the rationality and feasibility of ACT intervention in BPD from the pathological model and treatment model of ACT. Finally, the research results of ACT treating BPD in recent years were summarized. The results show that ACT provides a promising method for the treatment of BPD patients, however, more studies are needed to prove its use in this group and its specific mechanism.

Background Antigen retrieval method is the key of improving the successful rate of immunohistochemical assay in paraffin sections. To study an available method of antigen retrieval is a goal to achieve both good immunochemistry result and preserving retinal proteins.ObjectiveThe aim of present study is to investigate tyrosin digestion,high-temperature heat pressure, water bath heating and microwave repair in antigen retrieval for retinal tissues.MethodsRetinal tissue was isolated and obtained from clean Chinchilla rabbits.Four hundreds retinal paraffin sections were prepared.Four kinds of antigen retrieval methods for retinal tissue including tyrosin digestion,high-temperature heat pressure, water bath heating and microwave repair were used respectively. The depigmentated retinal paraffin section without antigen retrieval was used as control. The positive rates of expression of CRALBP,Rhodopsin and opsin proteins were evaluated and compared among four kinds of antigen retrieval methods by immunochemistry.ResultsCRALBP,Rhodopsin and opsin protein was positively expressed in cytoplasm of retinal pigment epithelial cells and the outer segment of photoreceptor respectively. No significant difference was found in the positive expression rates of these three proteins among four kinds of antigen retrieval methods (P>0.05),but the differences in tissue integrity and background staining were statistically significant (P<0.01).The structural damage of retina included loss and pucker of scalera, crack of nucleus and abnormal background stain in high-temperature heat pressure method, water bath heating method and microwave retrieval method. However, stable CRALBP,Rhodopsin and opsin protein expression and strain effectiveness, clear background without unspecific staining and integrated tissue were seen in tyrosin digestion method.ConclusionDuring the clinical pathology analysis of retinal tissue, the application of tyrosin digestion in antigen retrieval could obtain a better effectiveness.

OBJECTIVE: Adopting methods of cell culture to explore the effects and mechanisms of Jianpi Bushen Huoxue Prescription (JPBSHXP), a traditional Chinese compound herbal medicine for strengthening spleen, reinforcing kidney and activating blood circulation, in inhibiting hematopoietic cells apoptosis in a mouse model of aplastic anemia (AA). METHODS: Blood serum of AA mice was made from an AA mouse model. Blood serums containing different traditional Chinese compound herbal medicine were made from rats after intragastric administration of JPBSHXP and its related decoctions, respectively. Bone marrow cells of normal mice were incubated by these blood serums for 24 hours, respectively. The apoptosis of the bone marrow cells were assayed by flow cytometry and transmission electron microscopy (TEM). RESULTS: It was indicated that the bone marrow cells of normal mice incubated with blood serum of AA mice displayed typical apoptosis. The apoptosis rates of bone marrow cells of the AA mice incubated by blood serum containing different traditional Chinese herbal medicine were decreased. The effect of Bushen (reinforcing kidey) Recipe was better than Jianpi (strengthening spleen) Recipe and Huoxue (activating blood circulation) Recipe, while the effect of JPBSHXP was the best. TEM results showed that the effect of Bushen Recipe was better than that of the Jianpi Recipe and the Huoxue Recipe, while the effect of JPBSHXP was the best. CONCLUSION: JPBSHXP and its related decoctions can significantly decrease the apoptosis rate of bone marrow mononuclear cells of the AA mice. It is inferred that JPBSHXP can promote bone marrow hematogenesis.
