1.Rosuvastatin Up-regulating the Expression of Sirt1/NF-κB Signal Pathway and Inhibiting Doxorubicin-induced Cardiotoxicity Injury in Experimental Mice

Objective: To investigate the effects of rosuvastatin for up-regulating the expression of Sirt1/NF-κB signal pathway and inhibiting doxorubicin (DOX)-induced cardiotoxicity injury in experimental mice.
Methods: A total of 30 male C57 BL/6J mice at (4-6) weeks of age were randomly divided into 3 groups. Treatment group, the mice received intra-gastric rosuvastatin for 7 days, followed by intra-peritoneal injection of DOX 15mg/kg to induce the cardiotoxicity injury, and then received rosuvastatin for another 5 days. Model group, the mice received intra-peritoneal injection of DOX 15mg/kg to induce the cardiotoxicity injury, and then received intra-gastric normal saline for the same volume. Control group, the mice received intra-gastric normal saline for the same volume. n=10 in each group. The mice were killed at 12 days after treatment. The pathological change in myocardial tissue was observed by HE staining, the myocardial oxidative stress indexes of malonadehyde (MDA) level and super oxide dismutase (SOD) activity were measure by the
operating kits and the protein expression of Sirt1/NF-κB was examined by immunohistochemistry.
Results: Compared with Control group, Model group had obviously increased levels of MDA, NF-κB and decreased SOD activity, Sirt1 level, all P<0.05;the mice in Model group showed disordered myocardial structure with inlfammatory cell inifltration. Compared with Model group, Treatment group had obviously decreased levels of MDA, NF-κB (while they were still higher than Control group), and increased SOD activity, Sirt1 level, all P<0.05;the mice in Treatment group showed intact myocardial structure with much less edema and lymphocyte inifltration.
Conclusion: Sirt1/NF-κB signal pathway was involved in DOX-induced cardiotoxicity injury in experimental mice, rosuvastatin could protect the injury via up-regulating the expression of Sirt1/NF-κB signal pathway.
2.Relationship of Erxiang Zhitong Capsule between ratio of components and pharmacodynamics

AIM:To stuudy the relationship of the constituents of Erxiang Zhitong Capsule(Rhizoma alpiniae officinarum,Rhizoma cyperi and Radix aucklandiae) and its pharmacodynamics.METHODS:The use of supercritical CO_2 fluid extraction as extraction for three herbs acted as experimental materials.The preparation composed of Rhizoma Alpiniae officinarum extract,Rhizoma Cyperi extract;Radix Aucklandiae extract(3∶3∶2) had obviously antiulcerative,antispasmodic and anti-inflammatory analgesic effects.CONCLUSION: From view of pharmacodynamics view,we gain ratio of components of Erxiang Zhitong Capsule better than that of classical prescription.
3.Expressions of CD34,Delta-like-1 protein in hepatic regeneration tissue of rats after partial hepatectomy

BACKGROUND: When the liver affected severe damage or excalation, liver stem cells rapidly proliferated and differentiated into mature liver cells to repair liver tissue. However, liver regeneration is a complicated multiple signal molecular regulatory process and related genes and proteins that participated in regulation remain unclear. OBJECTIVE: To investigate the expressions of CD34 and Delta-like-1 protein in rat liver regenerated tissues after partial hepatectomy. METHODS: Rat hepatic regeneration models were established after hepatectomy. Expressions of ATPase, proliferating cell nuclear antigen (PCNA), CD34 and Delta-like-1 protein were detected by hematoxylin-eosin staining, enzyme-histochemistry and immunohistochemistry method at various time points. RESULTS AND CONCLUSION: At the early stage of liver regeneration, the expression of ATPase was down-regulated, and the expressions of CD34 and Delta-like-1 protein were up-regulated. At the middle stage, the expression of ATPase was increased to the normal level, and the expressions of CD34 and Delta-like-1 protein were down-regulated. The expression of PCNA protein was continuously up-regulated. These suggest that the expressions of ATPase, CD34 and Delta-like-1 protein were different at different stages of hepatic regeneration, which means that different signal molecules were cooperated during the hepatic regeneration, and Notch-Delta signal pathway takes part in the pathogenesy.
4.Effect of ischemic preconditioning on alterations in regional fluidity of myocardial membrane during cardiopulmonary bypass

Objective To elucidate the effect of ischemic preconditioning(IPC)on regional fluidity of myocardial cell membrane during cardiopulmonary bypass (CPB). Methods Seventy-five felines were randomized into three groups: Group A (CPB group,n =25),in which CPB was conducted without aortic cross-clamping (ACC);Group B (IR group,n =25),with 60min ACC followed by 60min reperfusion,and cardioplegia used during period of ACC;Group C (IPC group,n =25),with protocol similar to that of group B except for three-cycle IPC applied before ACC. Electron spin resonance (ESR) technique was employed to monitor membrane regional fluidity of cardiomyocyte during CPB in all groups. Results Regional fluidity of myocardial cell membrane kept unchanged in CPB group and markedly decreased during ACC and reperfusion periods in IR group. Though the regional fluidity of myocardial cell membrane during ACC and reperfusion periods in IPC group was slightly decreased compared to CPB group,it was significantly higher than that in IR group. Conlusion IPC can protect myocardial cells from ischemia and reperfusion injury by maintaining the fluidity of myocardial membrane during CPB.
5.Comparison of Efficacy between Paroxetine and Estazolam on Chronic Insomnia

Objective: To determine whether paroxetine would be effective in the treatment of patients with chronic insomnia and to compare efficacy of paroxetine and estazolam on chronic insomnia. Methods: 74 patients with chronic insomnia were divided into 2 groups, receiving treatments of paroxetine and estazolam respectively. All of the subjects were assessed with Pittsburgh Sleep Quality Index (PSQI) and sleep diary. Results: 1. On the eighth day of treatment, estazolam group showed improvement, all the indexes of sleep were better than those before treatment and those of paroxetine group; 2. On the fifteenth day of treatment, paroxetine group showed improvement, all the indexes of sleep were better than those before treatment and those of estazolam group, while the indexes of the estazolam group returned to pre-treatment level. 3. At the end of treatments and 3 months' follow up, paroxetine group still showed better effect in sleep indexes than estazolam group and the pre-treatment level. Conclusion: paroxetine is effective in the treatment of chronic insomnia and shows long-term effect.
6.Long-term follow-up of tricuspid valve function by color Doppler echocardiography after mitral valve replacement

Objective To investigate the long-term morphologic and functional changes of the tricuspid valve after mitral valve replacement. Methods The tricuspid valve function was evaluated by Doppler echocardiography in 903 patients who underwent mitral valve replacement during recent 10 years. Tricuspid regurgitation and annular dilatation in various degrees were found in all patients preoperatively. 686 patients underwent Kay annuloplasty or DeVega annuloplasty, 106 patients underwent tricuspid ring annuloplasty, and 201 untreated. Results Among 201 patients who did not undergo tricuspid annuloplasty, severe tricuspid regurgitation developed in 46 during 2 to 3 years follow-up. One out of 16 patients who underwent tricuspid ring annuloplasty developed moderate tricuspid regurgitation 2 years after operation. Of 686 patients who underwent Kay annuloplasty or DeVega annuloplasty, 150 developed moderate or severe tricuspid regurgitation during 3~5 years after operation. Conclusion Tricuspid annular dilatation, right heart impairment and severe pulmonary hypertension are responsible for the development of late tricuspid regurgitation after mitral valve replacement. For the patient who underwent mitral valve replacement ,tricuspid annuloplasty should be performed when annular dilatation was found, even without tricuspid regurgitation and tricuspid ring annuloplasty should be performed in patient with severe tricuspid regurgitation and obvious tricuspid annular dilatation.
7.Research on tissue-engineered heart valve

8.Atlantoaxial surgery through transoropharyngeal approach: an anatomical study

Objective To provide anatomical profile of the cranio-cervical region for the surgical treatment of ventral atlantoaxial lesions through transoropharyngeal approach.Methods Ten fresh specimens of craniocervical region were dissected layer by layer with microsurgical technic in accordance with the transoropharyngeal approach for the surgical treatment of atlantoaxial lesion, with special attention to anatomical stratification of pharyngeal posterior wall, the course of the vertebral artery, anatomical relation between the atlas and the axis, and anatomical parameters pertaining to internal fixation for the atlantoaxial joint. Results Pharyngeal posterior wall consisted of two layers (mucosa and prevertebral fascia) and two spaces (posterior interspace of pharynx and anterior interspace of vertebrae). Through anterior transoropharyngeal approach, the region between the anterior rim of the foramen magnum down to C 3 could be exposed. The distance between the vertebral artery and midline of the atlar and axis was 25.2?2.3mm(20.4-29.7mm) and 18.4?2.6mm(13.1-23.0mm) respectively. The exposed areas of the atlas and axis were 39.4?2.2mm(36.2-42.7mm) and midline was 39.0?2.1mm(35.8-42.3mm), respectively. The distance(a) between the two screw inserting points on the atlas (middle point of C 1 lateral mass) was 31.4?3.3mm(25.4-36.6mm). The vertical distance(b) between the connecting line of two screw inserting points on the atlas and that of two screw inserting points on the axis (3-4mm lateral to the midline of C 2 vertebra) was 18.7?2.7mm(14.9-23.2mm). The odds of a/b ranged from 1.5 to 1.7. Conclusion Atlantoaxial surgery through transoropharygeal approach is safe and feasible. The approach is suitable for internal fixation of anterior atlantoaxial joint, and the design of the plate should be based on the above data.
9.Effect of intact follicle transplantation by punching with syringe needle on eyebrow defect

Objective To explore the effect of the transplantation of intact follicle by punching with syringe needle on defect eyebrow reconstruction.Methods Fifty-two patients with eyebrow defects due to burn,trauma,resection and inappropriate eyebrow trimming were recruited,35 with bilateral defects and 17 with unilateral defect.The scalps strips from the safe donor area or temporal region were taken and made into 1 to 2 cm long hair with intact follicle under 3 fold magnifier.The recipient area was punched with 21G or 22G syringe needle.Then the hair shaft was clamped with mi croforceps and the follicles were transplanted to the defective area.Results Folliculitis were found in two burn patients after the transplantation and cured by alcohol inunction.Hairs of the rest patients transplanted grew well.The direction and appearance were satisfied.The survival rate of hair was more than 90 %.Conclusions It is feasible to restore the partial eyebrow defect by intact follicular transplantation by punching with syringe needle and clamping the hair shaft with microforceps.
10.Mechanism and effect of hDOT1L on the proliferation inhibition of decitabine in THP-1 cells: a preliminary study

Objective To analyze the role of hDOT1L signaling pathway on the proliferation of human acute monocytic leukemia cell line THP-1 cell inhibited by demcitabine, and to explore the other effects except DNA demethylation and the mechanism of hDOT1L signaling pathways involved in leukemia. Methods Methyl thiazolyl tetrazolium (MTT) method was used to detect the influence of decitabine on THP-1 cell proliferation, and trypan blue anti-staining was applied to detect the effect of decitabine on THP-1 cell survival rate, reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression levels of hDOT1L, HOXA9, HOXA10, MLL-AF10 (MLLT10) mRNA, then the methylation level of H3K79 was detected by Western blotting assay. Results Compared with the control group, decitabine inhibited the proliferation of THP-1 cell in a time and dose-dependent manner. The inhibitory effect of 72 h was the highest, and the highest inhibition rate was 56.18%. In addition to 0.5 μmol/L in decitabine treatment group, the other groups were statistically significant (all P<0.05). There were high expressions of hDOT1L, HOXA9, HOXA10, MLLT10 mRNA and H3K79 hypermethylation in the THP-1 cell, meanwhile decitabine reduced the levels of hDOT1L mRNA after 48 h as well as HOXA9, HOXA10, MLLT10 mRNA, and there was a significant difference compared with the control group (all P < 0.01). Decitabine reduced obviously the methylation of H3K79 in cell after 48 h, especially the expression levels of bis and three methyl protein, and these differences were significant compared with the control group (all P< 0.01). Conclusion Decitabine plays its inhibitory effect on the proliferation of THP-1 cells probably through reducing the expression level of hDOT1L and H3K79 methylation level and decreasing the expression levels of key molecules, HOXA9, MLL-AF10, HOXA10, MLLT10 in the hDOT1L signal pathway.
