1.Computational Identification of microRNAs and Their Targets

The discovery of microRNAs (miRNAs) has introduced a new paradigm into gene regulatory systems. Since inception, computational methods have been an invaluable tool complementing experimental approaches, and many discoveries have been obtained through combination of experimental and computational approaches. The knowledge that has been accumulated about the principles of miRNAs and target recognition were reviewed. The currently available computational methodologies and software for prediction of miRNA and their target genes also have been discussed.
2.A primary study on the oxytocin levels, the cortisol levels and the relationship with severity of depression in the patients with major depression

ObjectiveTo explore the difference of the oxytocin Levels and cortisol levels between major depression patients and normal controls,and the relationship of symptoms severity of major depression with oxytocin levels and cortisol levels.MethodsTwenty 18 ~ 45years old major depression patients and twenty six normal controls were tested by an enzyme-linked immunosorbent assay to evaluate the oxytocin levels and cortisol levels,and the Hamilton Rating Scale for Depression-17 item was used to evaluate the symptoms severity of depression in patient group.ResultsMann-Whitney test showed there were significantly differences in oxytocin level between patients and controls ( ( 213.60 ± 164.29 ) pg/ml vs ( 112.27 ± 101.21 ) pg/ml,z =- 2.306,P =0.021 ),but there were no significantly differences in cortisol level between two groups.Within the depressive sample( (0.131± 0.089) vs (0.107 ± 0.077 ) pg/ul,z =- 0.920,P =0.358 ),the severity of depressive symptom was positive correlated to the value of oxytocin ( r=0.599,P=0.005) and cortisol( r=0.691,P=0.001 ),the value of oxytocin was also positive correlated to the value of cortisol( r =0.743,P < 0.01 ),Multiple liner regression analyses showed that the value of oxytocin is an impact factor of the severity of depression.( adjusted R2 =0.358,F =11.578,P =0.003 ).ConclusionThis study indicated that the value of plasma oxytocin maybe significantly differences between depression and normal controls and the plasma oxytocin level maybe a factor which influence the severity of major depression,the relationship of the cortisol value and the severity of depression can not be confirmed.
3.The study and diagnostic value of MRI sequences in patients with trigeminal neuralgia

ObjectiveTo investigate the capacity and diagnostic value of various MRI seguences in patients with trigeminal neuralgia.Methods MR images of 60 patients with trigeminal neuralgia were analyzed retrospectively.The sensitivity,specificity and diagnostic value of various MRI sequences were evaluated comparing with clinical data and operation results.All patients were scanned with conventional sequences including SE-T1WI,T2WI,FLAIR of head.Among them,9 cases were injected with contrast agent,49 cases were scanned with 3D-TOF and 3D-TSE sequences on cerebellopontine angle additinally.The sensitivity,specificity and accuracy of the 3D-TOF and 3D-TSE sequences were analyzed by using the x2 test.Results Six cases with tumor,3 cases with radiculitis and meningitis,1 case with multiple sclerosis and 1 case with pons infarction were diagnosed by conventional MR sequences.MRI of 49 cases with 3D-TOF and 3D-TSE showed neurovascular compress proved by operation.The sensitivity,specificity and accuracy of 3D-TOF and 3D-TSE were 95.3% (41/43)vs.95.6% (43/45),66.7% (4/6)vs.50.0%(2/4) and 91.8% (45/49)vs.91.8% (45/49),and no significantly difference was found between the two sequences (x2 =0.13,0.19 and 0.17,P >0.5).ConclusionsMRI plays an important role in displaying the causes and diagnosis of trigeminal neuralgia.The conventional head MR sequences should be recommended for diagnosis of secondary TN caused by tumor and inflammation et al,and further 3D-TOF and 3D-TSE with high sensitivity and accuracy should be applied in patients with primary TN to display the neurovascular relationship.
4.Exploring the Rule of the Diagnosis and Treatment of Stroke Based on the Tibetan Medical Theory of White Meridian

Meridians in human body were classified as white meridian and black meridian according to Tibetan medicine.Season and environment,improper diet,toxic heat and trauma were recognized as main reasons damaging the white meridian in Tibetan Medicine,leading to the emerge of white meridian disease induced by Long (one of the three factors) and blood disorder.White meridian disease in Tibetan medicine involved a series diseases,such as many clinical diseases,due to the damage of white meridian system caused by pathogenic factors.Stroke also belonged to white meridian disease.Drugs and treatments were selected based on the nature of disease such as cold and heat,onset,thelocation of disease and the three factors (Chi Ba,Long and Pei Gen).It was the fundamental principle of the treatment rules of white meridian disease in Tibetan medicine,namely,prescribing medication with the rule of diagnosis and treatment,comprehensive analysis of the causes of diseases and mastering the change law of diseases and syndromes in clinic.
5.Dynamic changes of oxidative stress and cytokines in gerbils with nonalcoholic fatty liver disease and their significance

ObjectiveToexplorethedynamicchangesofoxidativestressandcytokinesinMongoliangerbilswith nonalcoholic fatty liver disease ( NAFLD) and their significance.Methods Forty-eight healthy male gerbils were randomly divided into normal group and model group , 24 in each group .Gerbils of the model group were fed with high fat diet while those of the normal group with normal diet .Eight gerbils in each group were killed at the end of 4 w, 8 w and 16 w, respectively .MDA content and SOD , GSH-PX and T-AOC activity in the liver tissue were detected by chemical method, and serum TNF-α, INF-γand IL-10 levels were determined using liquid suspension chip .Results With the development of NAFLD , MDA content in liver increased gradually , and the MDA contents were all significantly higher than those of the normal group ( P<0.01 ); T-AOC level slightly increased , and then decreased , the levels at 4 w and 16 w were markedly decreased compared with those of the normal group (P<0.05);SOD level was significantly increased and then markedly reduced, the level of the model group at 4 w was significantly increased (P<0.05), while that at 8 w and 16 w were significantly decreased (P<0.05, P<0.01).The level of GSH-PX was decreased gradually , the levels at 8 w and 16 w were significantly lower than those of the normal group (P<0.05).With the progression of NAFLD,serum TNF-αand IFN-γwere increased gradually , while the level of IL-10 decreased gradually , and the levels at 8 w and 16 w were significantly lower than those of the normal group ( P <0.05, P <0.01).Conclusions The oxidative stress-related indicators and inflammatory cytokines in the gerbil NAFLD models induced by high fat diet are significantly changed as simple fatty liver develops into steatohepatitis , liver fibrosis and cirrhosis , and participate in the development and progression of NAFLD .
6.Effect of Wutou decoction on depression and anxiety induced by neuropathic pain mice

Objective To observe the effects of Wutou decoction(WTD)on Anxiety and Depression induced by the L5 spinal nerve ligation (SNL) mice. Methods The ICR male mice were randomly divided into sham,SNL,high-dose,medium-dose and low-dose group of Wutou decoction,and Pregabalin group,with 24 in each group. Except the sham group, SNL models were prepared in the remaining groups of mice. After successful modeling, the high, medium, and low dosages were administered with 12.60, 6.30, 3.15 g/kg of Wutou decoction,and pregabalin group was administered with 0.25 g/kg of pregabalin,and the sham group and the model group was given an equal volume of normal saline. All the administration was once per day, continuously last for 21 days. On the 7/14/21thdays after the operation, the depressive and eanxiety behavior were evaluated by the tests of the sugar water consumption, the open field and forced swimming detection. In addition, the number of the positive cells expressing the extracellular signal-regulated kinase (p-ERK) in hippocampus were analyzed on Day 7/14/21 in each group by Immunohistochemistry and the expression of both p-ERK and postsynaptic density protein 95 (PSD95) in the hippocampus were analyzed on Day 21 by Western blot. Results At 7, 14, and 21thday after administration, compared with the model group, the paw withdrawal mechanical threshold of mice(0.76 ± 0.21 vs.0.05 ± 0.03),(0.93 ± 0.33 vs.0.05 ± 0.01),(1.12 ± 0.20 vs.0.04 ± 0.01)in the high dose group of Wutou decoction respectively increased(P<0.01),and sugar consumption(1.86 ± 0.44 g vs. 0.84± 0.23 g), (1.84 ± 0.10 g vs. 1.02 ± 0.18 g), (1.63 ± 0.31 g vs. 0.75 ± 0.23 g) significantly increased(P<0.01),and immobility time 4 min after swimming(128.90 ± 35.27 s vs.180.30 ± 21.81 s),(125.50 ± 23.07 s vs.195.30 ± 27.70 s),(169.50 ± 23.07 s vs.207.50 ± 7.46 s)significantly decreased(P<0.01),and the retention time in the central area of the open field(35.35 ± 7.61 s vs.13.90 ± 2.27 s),(26.26 ± 3.61 s vs.13.08 ± 1.98 s),(24.04 ± 6.57 s vs.10.62 ± 3.38 s)significantly increased(P<0.01).After 21 days,the number of p-ERK positive cells in the high-dose group of Wutou decoction(11.00 ± 4.89 vs.33.67 ± 7.35)was siginificantly lower than that of the model group,and the hippocampus p-ERK protein(0.15 ± 0.09 vs.0.54 ± 0.04)siginificantly decreased,and the expression of PSD95 protein(0.32 ± 0.04 vs.0.57 ± 0.09)siginificantly decreased(P<0.01). Conclusions The Wutou decoction achieved significant anti-depressant and anti-anxiety effects,and regulated p-ERK expression levels of hippocampus in the late stage of NP.
