2.Ultrasonic features of triple-negative breast cancer

Objective To investigate the ultrasonic features of triple-negative breast cancer (TN-BC).Methods Ultrasonographic findings of 299 patients with pathologically confirmed breast cancer were analyzed retrospectively.Patients were divided into TNBC group (46 cases) and non-triple-negative breast cancer(NTNBC) group (253 cases) according to the expression of estrogen receptor (ER),progesterone receptor (PR),and human epidermal growth factor receptor 2 (HER2) that were determined with immunohistochemical staining.Each patient was ultrasonically analyzed.Results The ultrasonic images showed that TNBC group had a greater proportion in the mass with regular shape,clear boundary,or microlobulated relative to NTNBC group (P < 0.01).Calcification was significantly less in TNBC than NTNBC (P <0.01).Eight (17.3%)of 46 Cases of TNBC had BI-RADS sonographic features that favored the diagnosis of a benign condition.Conclusions Some of sonographic criteria for TNBC are more likely to be associated with benign lesions,ultrasound-guided biopsy should be recommended for such lesions.
3.Experimental study on electrical impedance properties of human hepatoma cells.

The AC impedance of human hepatoma SMMC-7721 cells were measured in our laboratory by Agilent 4294A impedance analyzer in the frequency range of 0.01-100 MHz. And then the effect of hematocrit on electrical impedance characteristics of hepatoma cells was observed by electrical impedance spectroscopy, Bode diagram, Nyquist diagram and Nichols diagram. The results showed that firstly, there is a frequency dependence, i.e., the increment of real part and the imaginary part of complex electrical impedance (δZ', δZ"), the increment of the amplitude modulus of complex electrical impedance (δ[Z *]) and phase angle (δθ) were all changed with the increasing frequency. Secondly, it showed cell volume fraction (CVF) dependence, i. e. , the increment of low-frequency limit (δZ'0, δ[Z*] 0), peak (δZ"(p), δθ(p)), area and radius (Nyquist diagram, Nichols diagram) were all increased along with the electric field frequency. Thirdly, there was the presence of two characteristic frequencies: the first characteristic frequency (f(c1)) and the second characteristic frequency (f(c2)), which were originated respectively in the polarization effects of two interfaces that the cell membrane and extracellular fluid, cell membrane and cytoplasm. A conclusion can be drawn that the electrical impedance spectroscopy is able to be used to observe the electrical characteristics of human hepatoma cells, and therefore this method can be used to investigate the electrophysiological mechanisms of liver cancer cells, and provide research tools and observation parameters, and it also has important theoretical value and potential applications for screening anticancer drugs.

Objective To investigate the effect of expression of pituitary tumor transforming gene(PTTG) on the expression of inhibitory of vascular endothelial growth factor(VEGF).Methods The constructed(recomibinant) vector,pcDNA3.1-PTTGas that contained full-length antisense PTTG,was transfected into the cholangiocarcinoma cell line QBC939 in different quantity.After 48h incubation,the variation of expressions of PTTG and VEGF mRNAs and proteins were observed by RT-PCP and Western-blot method.Results(After) successful transfecting the recomibinant vector into QBC939,the mRNAs and proteins of PTTG and VEGF were all inhibited.Linear correlation analysis showed positive correlation between descendent extents of PTTG and VEGF(P
5.Efficacy of precise hyperthermic intraperitoneal perfusion chemotherapy in the treatment of advanced pancreatic cancer

Objective To investigate the efficacy of precise hyperthermic intraperitoneal perfusion chemotherapy (HIPEC) in the treatment of advanced pancreatic cancer.Methods Thirty-six patients with advanced pancreatic cancer were analyzed retrospectively.Eighteen patients who received HIPEC combined with radiotherapy and chemotherapy were assigned as the treatment group and the other 18 patients who received chemotherapy and radiotherapy were assigned as the control group.Recent curative efficacy,Karnofsky Performance Status (KPS) score,postoperative complications and survivals between the two groups were analyzed,respectively.Results Significant differences were found between two groups in total short-term effective rate (P < 0.05).The total short-term effective rate of treatment group and control group were 66.67% (12/18) and 27.78% (5/18),respectively.The increment of KPS score of treatment group was significantly higher than that of control group (P < 0.05).There was no significant difference between two groups in postoperative complications (P > 0.05).The median overall survival time (OS) of treatment group was 11 months (7 ~ 31 months),and the median OS of control group was 7 months (4 ~ 18 months).The survival of the treatment group was longer than the control group (P < 0.05).Conclusions HIPEC treatment improved significantly the survival and life quality of advanced pancreatic cancer patients.With acceptable morbidity and mortality rates,HIPEC regime was an effective treatment modality for patients with advanced pancreatic cancer.
6.LRIG1 expression and its possible suppressor role in oral verrucous carcinoma

Objective:To explore leuncine-rich repeats and immunoglobulin-like domains protein 1 (LRIG1) expression in oral verrucous carcinoma(OVC) and its possible mechanism of tumor suppression.Methods:Paraffin specimens of OVC (n =15) and oral squamous cell carcinoma(OSCC,n =30) and corresponding adjacent normal tissues,and 15 normal mucosa tissues(NM) as control from injured patients were collected.The expression levels of LRIG1 and Bcl-2 were examined using immunohistochemical SP method.The correlation between the expression levels of LRIG1 and Bcl-2 was determined using two tailed Pearson's correlation.Results:LRIG1 was significantly lower in OVC tissue than that in NM tissue,but higher than in OSCC tissue.The expression levels of Bcl-2 in OVC were significantly higher than that in NM,but lower than in OSCC.The expression level of LRIG1 was negatively correlated with expression level of Bcl-2 in NM,OVC and OSCC tissue.Conclusion:LRIG1 may inhibit the expression of Bcl-2 and the development of OVC.
7.Cole-Cole Mathematical Model Analysis on Impedance Spectrum in Rat Erythrocyte Suspension

Objective To establish binomial equation Cole-Cole mathematical modeling methods and the parameters in rat erythrocyte impedance spectra,as well as to discuss the sources of the ? and the ? dispersion.Methods The impedance of erythrocyte suspension was measured with Agilent 4294A impedance analyzer within the frequency range of 0.01~100 MHz.The residual error of the fitting curve was determined with non-linear numerical calculation of Cole-Cole equation.Then the Cole-Cole mathematical model parameters of rat erythrocyte were established with the Bode and the Nyquist plots.Results The Cole-Cole mathematical model parameters of rat erythrocyte impedance spectrum: the high-frequency limit of impedance R∞=63/?cm,the 1st relaxation impedance increment ?R1=59/?cm,the 1st characteristic frequency fC1 =2.4 MHz,the 1st relaxation dispersion angle ?1=0.07,the 2nd relaxation impedance increment ?R2=48/?cm,the 2nd characteristic frequency fC2 =370 MHz,the 2nd relaxation dispersion angle ?2=0.22.Conclusion The ? and ? relaxation behavior of rat erythrocyte impedance spectrum can be expressed with the two-type Cole-Cole mathematical model.The ? dispersion is formed from the capacitive response of cell membrane and the ? dispersion from frequency response of intracellular hemoglobin.
8.Small RNA interference-mediated gene silencing of TREK-1 potassium channel in cultured astrocytes.

This study was aimed to examine the effect of TREK-1 silencing on the function of astrocytes. Three 21-nucleotide small interfering RNA (siRNA) duplexes (siT1, siT2, siT3) targeting TREK-1 were constructed. Cy3-labeled dsRNA oligmers were used to determine the transfection efficiency in cultured astrocytes. TREK-1-specific siRNA duplexes (siT1, siT2, siT3) at the optimal concentration were transfected into cultured astrocytes, and the most efficient siRNA was identified by the method of immunocytochemical staining and Western blotting. The proliferation of astrocytes tranfected with TREK-1-targeting siRNA under hypoxia condition was measured by fluorescence-activated cell sorting (FACS). The results showed that TREK-1 was expressed in cultured astrocytes. The dsRNA oligmers targeting TREK-1 could be transfected efficiently in cultured astrocytes and down-regulate the expression of TREK-1 in astrocytes. Moreover, the down-regulation of TREK-1 in astrocytes contributed to the proliferation of astrocytes under hypoxia condition as determined by cell cycle analysis. It was concluded that siRNA is a powerful technique that can be used to knockdown the expression of TREK-1 in astrocytes, which helps further investigate the function of TREK-1 channel in astrocytes under physicological and pathological condition.

Freshwater crabs and snails were collected from Ninghai County in Zhejiang Province, and examined respectively for Paragonimus metacercariae and cercariae. Among 97 freshwater crabs found, the prevalence was 11.3% (11/97) with a mean intensity of 1 metacercariae per crab. It was 10.2% (5/49) and 20.2% (4/20) in the groups weighted 5-15 g and 15-25 g respectively, with an average intensity of 1, and no metacercariae were found in weight group of 25-35 g. Objective To explore the value of elastography score and strain rate ratio in the diagnosis of small breast malignant focus. Methods We retrospectively analyzed 22 patients with breast small malignant foci less than 10 mm. Ultrasound characteristics were summed up in breast small cancer. Results On elastogram, 2 patients scored 3, 14 scored 4 and 6 scored 5.The average strain rate ratio of all foci was 4.76, and there was correlation between it and elastography scores. Conclusion Ultrasonic elastography has important value in the diagnosis of breast small cancer.
