1.Protective effect of nano carbon tracer on parathyroid function in the surgery of thyroid cancer

Objective To investigate the effect of nano carbon tracer for protecting parathyroid function in the surgery of thyroid cancer.Methods A total of 178 patients with thyroid cancer admitted in our hospital from Jun.2014 to Mar.2016 were collected and divided into the control group and the observation group according to the random number table method,89 cases in each group.The control group received routine surgery,while the observation group received nanocarbon suspension during surgery.The levels of serum parathyroid hormone and serum calcium in the 2 groups were measured at 3 days after surgery,and the patients with low parathyroid hormone,normal parathyroid hormone and low calcium were counted.Results The rate of ormal serum calcium in the observation group was 91.01%(81/89),significantly higher than 67.42%(60/89) in the control group (P< 0.05).The rate of normal parathyroid hormone in observation group was 94.38% (84/89),significantly higher than 64.04% (57/89) in the control group (P<0.05).Conclusion The nano carbon tracer is helpful for protection of parathyroid function in the surgery of thyroid cancer.
2.Construction and evaluation of the luciferase reporter vector from 3'-UTR of mice MIA3 gene

Objective To construct MIA3 psicheck2 wild-type and mutant vectors targeting miR-374b,and provide the previous guarantee for the dual luciferase reporter assay.Methods The amplification primer was firstly designed according to mice MIA3-3'UTR sequence information,mice whole blood genomic DNA was taken as the template for PCR amplification of MIA3-3'UTR sequence,and the PCR product was cloned into psicheck2 dual luciferase reporter vector.Then,mutant primer was designed to mutate the MiR-374b seed sequence target TATTATA into AAATTAT so as to construct mutant vector.At last,the vector enzyme digestion evaluation and sequencing method was used to evaluate the constructed vectors.Results It could be seen from the analysis of agarose electrophoresis that the PCR amplification size of vector was consistent with the theoretical size.DNA sequencing evaluation showed that the MIA3-3'UTR-WT vector had been constructed successfully.The construction of mutant vector has successfully mutated the MiR-374b seed sequence target TATTATA into AAATTAT.Conclusion The successful construction of the vector will lay a foundation for the further evaluation on whether there is an actual binding site between the miR-374b and the chondrogenic differentiation-related target gene MIA3.
3.Quality and quantity classification models of Fructus Amomi applying electronic nose with multiple mathematical statistics methods

Objective Fructus Amomi(Sharen) is derived from the dry ripe fruit of Amomum villosum Lour., A.villosum Lour. var. xanthioides T.L. Wu et Senjen and A.longiligulate T.L.Wu, which is widely utilized for its clinic effects on digestive system. However, Fructus Amomi from different species and habitats, possessing different quality, is difficult to identify. In this study, we aim to develop a simple, rapid and reliable method for authentication of Fructus Amomi. Methods Twenty-five batches of samples of Fructus Amomi were collected and electronic nose was introduced into analyzing their odor with multiple mathematical statistics methods. Na?ve bayes network (NBN), radical basis function (RBF) and random forest (RF) were applied to establish different classifiers while BestFirst+CfsSubsetEval (BC) was used to screen the attributes for searching sensor array with higher contributions. Results Firstly, after attribute-screening via BC, the established discriminative models via NBN, RBF and RF could successfully identify genuine and non-genuine samples, presenting correct judging ratios of 78% and 84% through ten-fold cross validation and external test set validation, respectively. Besides, quantity predictive models were constructed as well. In case of content of bornyl acetate, one of the effective components in Fructus Amomi, values were higher than 3.5 mg/g and lower than 1.8 mg/g with sensor response of 0.04 and 0.03, respectively. Conclusion In this paper, quality discriminative model and quantity predictive model of Fructus Amomi were established via electronic nose and multiple mathematical statistics methods. It indicates that electronic nose could be a promising method for quality evaluation of Chinese material medica.
4.Identification of Sulfur Fumigated Chrysanthemi Flos with Electronic Nose Technology

This study was aimed to identify Chrysanthemi Flosbefore and after sulfur fumigation based on its different odour by the electronic nose technology.It was expected to explore a new method for the Chrysanthemi Flos identification according to the odour.The electronic nose technology was used in the detection of peak response values of Chrysanthemi Flos on sensor array.The principal component analysis (PCA) and 10 machine learning (ML) ways were used in the analysis of response values and establishment of optimized identification models.The results showed that there was a significant difference in the odour between sulfur fumigated Chrysanthemi Flos and non-sulfur fumigated ones.The identification models were successful with high correct judge rate by PCA and 6 ML ways including BF Tree,J48 and Random Tree.It was concluded that the electronic nose technology can be used for the accurate identification of sulfur fumigated Chrysanthemi Flos and non-sulfur fumigated ones.The electronic nose technology combined with multiple ML methods can be introduced in the quality evaluation of Chrysanthemi Flos.It provided more ideas for the application of electronic nose in data mining for traditional Chinese medicine (TCM) studies.
5.Methodological Research on TLC Identification of Radix Mirabilis himalaica

This study was aimed to establish the TLC identification method of Radix Mirab ilis himalaic a. The β-sitosterol and daucosterol were used as the reference substances. The single-factor test was used. A variety of factors which affected TLC were systematically investigated to filter out the best TLC conditions for identification of different batches of medicines. The results showed that the best TLC conditions were as follows: silica gel G plates, extraction solvent (methanol), reagent (5% sulfuric acid in ethanol), extraction method (ultrasonic extraction with methanol), ex-tracted time (30 min), the agent (petroleum ether-ethyl acetate-acetone (5:2:1)) and sample volume (6 μL). It was concluded that the method, which had high separation degree, was reproducible and simple. It can be used as the quality control of Radix Mirab ilis himalaic a.
6.Content Determination of Trigonelline in Wild and Cultivated Himalaica Mirabilis of Different Districts

Objective To compare differences and similarities of the content of trigonelline in Himalaica mirabilis of either wild or cultivated materials from different places. Methods An HPLC method was established to determine the content of trigonelline in Himalaica mirabilis. ZORBAX XDB-CN column (4.6 mm×250 mm, 5 μm) was used, with mobile phase of acetonitrile-0.03%acetic acid (85∶15), flow rate of 0.8 mL/min, detection wavelength of 265 nm, determination wavelength of 360 nm, and column temperature of 30 ℃. Results Regression calculation was made on peak area with the reference solution concentration, and then got the regression equation A=23.409C-26.398, r=0.999 8. Trigonelline showed good linear relation with peak area among the range of 2.004-200.400 μg/mL. The average recovery of trigonelline was 99.57%, RSD=1.11%. Conclusion There was no significant difference in the content of trigonelline of either wild or cultivated materials from different places. This study laid the foundation of application of the cultivated Himalaica mirabilis.
7. Therapeutic effects of curcumin on rats with knee osteoarthritis induced by monoiodoacetic acid

Objective: To investigate the therapeutic effects of curcumin on sodium iodide-induced osteoarthritis (OA) rats. Methods: Thirty-six rats were randomly divided into three experimental groups: control group, model group (OA+DMSO) and treatment group (OA+DMSO + curcumin). A knee OA rat model was established in the right knee joint cavity of male Sprague-Dawley rats with a single injection of monoiodoacetic acid (MIA). The control group was injected with the same volume of normal saline. Three days later, the treatment group was treated with intraperitoneal injection of curcumin, and the model group was injected with the same volume of DMSO. The rats in each group were observed for 2 weeks and 3 weeks after the swelling of the knee joint and the hind paw retraction test. The knee joint cartilage specimens were taken, and the femoral condyle and tibial plateau were observed under a dissecting microscope and the Pelletier scoring was performed. Sections were frozen and subjected to HE, toluidine blue, safranin O staining and Mankin scoring. Results: The model group had severe cartilage defects, rough surface, synovial hyperplasia with inflammatory changes, and osteophytes. The articular cartilage of the curcumin group was obviously repaired, and the cartilage surface was relatively smooth with no obvious hyperplasia and callus. There was a significant difference between the model group and the treatment group in Pelletier score and Mankin score (P<0.05). Conclusion :Curcumin can slow the degradation of articular cartilage, repair damaged articular cartilage, and reduce inflammation. This experiment provides an experimental basis and evidence for curcumin as a potential clinical drug for the treatment of knee osteoarthritis.

Objective To evaluate the efficacy and safety of tacrolimus extended-release (Tac-ER) in the early stage after kidney transplantation. Methods Clinical data of 68 recipients undergoing kidney transplantation from 34 pairs of renal allografts were retrospectively analyzed. Two recipients who received bilateral kidneys from the same donor were treated with Tac-ER (Tac-ER group) and tacrolimus immediate-release (Tac-IR) (Tac-IR group) as one of the basic immunosuppressant. The changes of tacrolimus dosage and blood concentration, intra-patient variability (IPV), renal function, incidence of acute rejection, recipient and allograft survival rates and adverse events were statistically compared between two groups. 9.Research on the application of PBL case library in integrated teaching of circulation system

To evaluate the advantages of using PBL case library in teaching circulatory system diseases to clinical medicine undergraduate students, a PBL case library was established and applied in teaching practice in the reform of circulatory system teaching. The PBL case library achieved the characteristics of combining theories with cases, morphology with functions, and basic knowledge with clinical knowledge. The PBL case library also realized the informatization, query, and update of cases. Preliminary application showed that the median practical score of students in the case library group was 94.00 points, which was significantly higher than the 92.00 points in the control group ( P=0.005). The average lesson preparation time for teachers in the case library group was (5.00±1.00) hours, which was significantly shorter than the (6.89±0.42) hours in the control group ( P<0.001). The difficulty score for lesson preparation among teachers in the case library group was significantly lower than that of the control group [(1.89±1.05) vs. (3.22±0.44), P<0.001]. However, there were no significant difference in theoretical scores and student satisfaction with teachers and courses. These results suggest that the construction of case library can improve practical teaching effectiveness and enhance the efficiency of lesson preparation for teachers.
10.Relation of perceived social support to mental health in prison police: a moderated mediating analysis

BackgroundThe mental health level of the prison police is relatively low, so finding innovative ways to improve the mental health of them is of great significance for the safety of prison supervision and the implementation of peaceful China initiative. ObjectiveTo explore the relationship between perceived social support, perceived stress and psychological resilience with mental health of prison police, and to provide references for improving their mental health. MethodsIn March 2022, 424 policemen working in a male prison in a western province were selected by cluster sampling method, and investigated with the Perceived Social Support Scale (PSSS), Chinese Perceived Stress Scale (CPSS), Connor-Davidson Resilience Scale (CD-RISC) and General Health Questionnaire 20 (GHQ-20), then Process 4.2 was used employed to verify the mediating role of perceived stress as well as the moderating role of psychological resilience in the relationship between perceived social support and mental health. Results①Male subjects scored higher on GHQ-20 than female subjects (t=2.095, P<0.05). ②CPSS score was negatively correlated with PSSS and GHQ-20 scores (r=-0.670, -0.703, P<0.01), and GHQ-20 score showed a positive correlation with PSSS and CD-RISC scores (r=0.580, 0.693, P<0.01). ③Perceived social support positively predict mental health (β=0.154, 95% CI: 0.133~0.175, P<0.01). ④Perceived stress played a mediating role in the relationship between perceived social support and mental health, and the mediation effect size was 0.087, accounting for 88.78% of the total effect (95% CI: 0.064~0.112, P<0.01). ⑤Psychological resilience played a moderating role in the second half (perceived stress→mental health) of the mediating path of "perceived social support→perceived stress→mental health"(
