2.Association of early prognosis and risk factors about early neurological deterioration in aged patients with cerebral hemorrhage in basal ganglia

Objective To study the association of early prognosis and risk factors about early neurological deterioration (END) in aged patients with cerebral hemorrhage in basal ganglia. Methods One hundred and thirty-nine aged patients with cerebral hemorrhage in basal ganglia were selected. The patients were divided into END group (59 cases) and non-END group (80 cases), the clinical data were retrospectively analyzed. The patients were followed up 3 months after discharge, the prognosis was evaluated by modified Rankin scale. Logistic regression analysis was used to determine the independent risk factors of END. Results The incidence of END was 42.4%(59/139). The incidence of unfavourable prognosis in END group was significantly higher than that in non-END group:78.0%(46/59) vs. 30.0%(24/80), and there was statistical difference (P<0.05). The score of Canadian stroke scale (CSS) at admission in END group was significantly lower than that in non-END group: (5.2 ± 2.1) scores vs. (6.9 ± 1.7) scores, but the white blood cell count at admission, hematoma volume and brain ventricle hemorrhage rate were significantly higher than those in non-END group: (10.7 ± 2.9) × 109/L vs. (7.9 ± 2.4) × 109/L, (17.4 ± 14.9) ml vs. (11.2 ± 10.5) ml and 42.4%(25/59) vs. 18.8%(15/80), and there were statistical differences (P < 0.05). There were no statistical differences in sex, age, systolic blood pressure, diastolic blood pressure, body temperature, random blood glucose, fibrinogen, activated partial thromboplastin time, prothrombin time and the incidences of hypertension, smoking history, stroke, diabetes mellitus, hyperlipidemia between 2 groups (P>0.05). The Logistic regression analysis results showed that brain ventricle hemorrhage and white blood cell count at admission were independent risks factor for END in aged patients with cerebral hemorrhage in basal ganglia(P < 0.05 or < 0.01). Conclusions The aged patients with cerebral hemorrhage in basal ganglia will be prone to END, the brain ventricle hemorrhage and white blood cell count at admission are independent risks factor for END, and the early prognosis is poor.
3.Effects of 3 carbohydrates on CAP10 promoter activity of Cryptococcus neoformans capsule-associated gene

Objective:To study the effects of glucose,mannose and galactose on CAP10 promoter activity of Cryptococcus neoformans capsule-associated gene.Methods: Yeast cells were transfected with plasmid containing a 951 bp length of 5′ upstream flanking sequence of CAP10 coding region and a reporter gene,chloramphenicol acetyl transferase(CAT);the transfectants were treated with different concentrations(10,20,40,60,80 mg/ml) of glucose,mannose and galactose.CAT activity was assessed by ELISA method and CAT activities of different groups were compared.Results: Different concentrations of glucose and mannose had no obvious influence on CAT activity;different concentrations of galactose had obvious influence on CAT activity and the influence was positively dependent with its dose within the experimental concentration range.Conclusion: Glucose and mannose have no obvious effect on the activity of CAP10 promoter;galactose has obvious inductive effect on activity of CAP10 promoter,suggesting that CAP10 gene might be related with galactose metabolism.
4.Study on Extracting with Supercritical CO_2 Fluid from Polyrhachis vicina Roger and Analysis on the Chemical Constituents by GC-MS

Objective To study the optimum parameters of the supercritical CO2 fluid extraction of Polyrhachis vicina Roger and chemical constituents of ant oil. Methods The experiments were designed with the uniform design. Three factors are extraction pressure, temperature and time. GC-MS was applied for analyzising. Results The optimum condition was obtained:the extraction pressure is 36 MPa, the temperature of extraction is 40 ℃ and extraction time is 50min. 45 components were identified from 60 chromatographic peaks. The main chemical constituents were (Z)-9-octadecenoicacid (48.278%), hexadecanoicacid (17.574%), (Z)-9-hexadecenoi (4.912%), cholesterol (2.957%). There are some linear paraffin in ant oil. Conclusion This method can extract the ant oil from Polyrhachis vicina Roger quickly and efficiently. This experiment provide foundation for exploitation and utilization of ant resource.
5.Drying process of Qingxiang Tablets by spray-drying method

AIM: To optimize the spray-drying process of Qingxiang Tablets. METHODS: Excipients were selected in terms of hygroscopicity and yield rate of dried powders with several excipients added;then,the process parameters,such as the relative density of fluid extracts,intake temperature,and the input speed of liquid feedstock were optimized by orthogonal experiment,in which the transfer rate of peoniflorin,a constituent in the formulation,and dried powder yield rate were set up as the evaluation index. RESULTS: The optimal processing conditions were obtained and validated as follows: relative density of liquid feedstock was 1.12 mg/mL(containing 6%?-CD),the input speed was 10 mL/min and intake temperature was at 190 ?C. CONCLUSION: The optimal process is stable and the experimental results provide the basis of industrial production.

7.Insulin-like growth factor 1 promotes in vitro proliferation of chondrocytes in traumatic arthritis

BACKGROUND:The early damaged chondrocytes are susceptible to de-differentiate and exert unstable phenotype during the in vitro culture, thus needing some growth factors.
OBJECTIVE:To observe the promotion effect of insulin-like growth factor 1 on the in vitro proliferation of chondrocytes in adult rabbits with traumatic arthritis.
METHODS:Traumatic arthritis models of adult rabbits were established by using the modified Hulth method. After the models were successful y established, the distal femur and proximal tibia were harvested under sterile conditions, the chondrocytes were cultured. The cultured cells were divided into two groups:control group was cultured with Dulbecco’s modified Eagle’s medium containing 10%fetal bovine serum, while experimental group was cultured with Dulbecco's modified Eagle’s medium containing 100μg/L insulin-like growth factor 1. The effect of insulin-like growth factor 1 on the proliferation of chondrocytes in adult rabbits with traumatic arthritis was determined through the cytomorphology, cellcounting, and cellactivity.
RESULTS AND CONCLUSION:The chondrocytes in adult rabbits with traumatic arthritis were successful y cultured, the majority of cells were mini-cells, presenting smal fusiform, round or polygonal shape. Hematoxylin-eosin staining showed that the number of cells in experimental group was higher than that in control group. MTT assay found that the absorbance of cells in experimental group was greater than that in control group (P<0.01). Our findings indicate that, insulin-like growth factor 1 can promote the in vitro proliferation of chondrocytes in adult rabbits with traumatic arthritis.
8.Induction of immune tolerance using kidney-bone marrow transplantation: Does it promote chimerism formation?

BACKGROUND: Infusion of hemopoietic stem cell from donors can promote the chimeric formation and induce specific immunologic tolerance in the allograft recipients. However, the pretreatment for cell transplantation has great toxicity to recipients. So immunosuppressant combined bone marrow infusion is introduced to anti graft versus host reaction. OBJECTIVE: Based on microchimerism, to study the security and associativity of chimera formation induced by kidney-bone marrow transplantation and immunologic tolerance.DESIGN, TIME AND SETTING: The contrast observation was performed at the department of urinary surgery, The Third People's Hospital of Zhengzhou City from January 1998 to December 2005.PARTICIPANTS: According to ABO/Rh blood type and HLA matching, 96 female patients with chronic renal failure and waiting for kidney transplantation were divided into 2 groups, In the combination group, patients received kidney combined bone marrow transplantation; the other uremia patients received the other kidney of cadavers were served as control group. The donors were 48 healthy males. METHODS: Bone marrow of donors was collected simultaneously with kidney obtain and preserved with cryoprotectant at -198 ℃ in nitrogen canister. After kidney transplantation, large dose of anti-human lymphocyte immune globulin were used for 2 weeks, then (0.9-2.5)×10~8/kg mononuclearcell was reinfused. PCR-SRY was used to identify donor derived cell-chimerism. Lymphocyte subgroup of recipients was determined by blood test; and interleukin 10 was measured by enzyme linked immunosorbent assay; in addition, the mass concentration of tumor necrosis factor α and tumor necrosis factor β was detected. MAIN OUTCOME MEASURES: Chimerism, lymphocyte subsets and cytokines were detected at various time points following transplantation. Simultaneously, the transplantation results and complication status of recipients were observed. RESULTS: The positive rate of chimera in the combination group was greater than that of the control group (P < 0.05). The 3-year follow-up showed that incidence differences of acute rejection between recipients with positive chimera and recipients with negative chimera had significance (13%, 35%, P < 0.05). There was no graft versus host disease occurred in the combination group. CONCLUSION: Kidney-bone marrow transplantation can augment chimerism in early postoperative period, and significantly reduce the rate of acute rejection, which is safe and beneficia1to induce specific immunologic tolerance in the renal allograft recipients.
9.The influence of ginkgo dipyridolum injection on blood-rheological and coagulation function in pationts with AECOPD

ObjectiveTo evaluate the influence of ginkgo dipyridolum injection on blood-rheological and coagulation function in pationts with AECOPD.Methods78 cases of patients with AECOPD were divided into group A (38 cases)and group B (40 cases)by mean of digital random table method.The routine treatment including oxygen inhalation,spasmolytics and anti-infection was given to patients in both groups while ginkgo dipyridolum injection (30 ml/d)was additionally added to patients in group B for two weeks.The blood-rheological and coagulation function were evaluated.ResultsAs for blood-rheological after the treatment,whole blood high shearing viscosity(5.25 ± 1.24)mPa · S,low shearing viscosity (11.12 ±2.43) mPa · S,plasma viscosity (2.06± 0.14 ) mPa · S and hematocrit (45.52 ± 2.78) % in the group A indicated significant differences compared to those in group B [ (4.83 ± 1.42)mPa· S,(8.78± 3.02) mPa · S,(1.73 ±0.21) mPa · S,(39.05 ± 3.41) %],(P<0.05); as for coagulation function after the treatment,PT (13.14± 1.31 ) S,APTT (30.85±5.24)S,FIB (4.99±1.04)S,D-D (1.42±0.23)mg/L in the group A indicated significant differences compared to those in group B [ (14.78 ± 3.13) S,(36.67 ± 8.12)S,(3.81 ± 0.42) S,(0.84 ±0.39) mg/L],(P<0.05).ConclusionFor the patients with AECOPD,the ginkgo dipyridolum injection can decrease hood-viscosity,and obviously improve hypercoagulabale state.
10.Assessment of left ventricular dyssynchrony and its relationship with cardiac function in dilated cardiomyopathy by two-dimensional strain parameters

Objective To explore value of two-dimensional speckle tracking imaging (2D-STI) strain parameters in assessment of left ventricular (LV) dyssynchrony and its relationship with cardiac function in patients with dilated cardiomyopathy (DCM).Methods Totally 25 patients with DCM and 25 age-matched normal controls were enrolled.The LV time to peak longitudinal strain (Tls) and time to peak circumferential strain (Tcs) were measured by 2D-STI,from which the standard deviations and maximal differences of the Tls in all 18 segments,6 segments at apical 4-chamber,3-chamber and 2 chamber views (Tls-SD,Tls-SD-4,Tls-SD-3,Tls SD-2,Tls dif,Tls-dif-4,Tls-dif-3,Tls-dif-2) were calculated,as same as the standard deviations and maximal differences of Tcs in all 18 segments,6 segments at mitral valve,papillary muscle,apical short-axis views (Tcs-SD,Tcs-SD-M,Tcs-SD-P,Tcs-SD-A,Tcs-dif,Tcs-dif-M,Tcs-dif-P,Tcs-dif-A) were calculated.Results Compared with the control group,the global and segmental strain parameters of the DCM group increased significantly (all P<0.05).Except for Tcs-dif-P and Tcs dif,the residual strain parameters were negatively correlated with LV ejection fraction (all P<0.05).Except for Tcs-dif-M,Tcs dif-P and Tcs-dif,there was a positive correlation between strain parameters and E/e in DCM group (all P <0.05).Conclusion In patients with DCM,LV dyssynchrony exists both in global and regional,which may aggravates the LV function damage.2D-STI strain parameters can objectively evaluate LV dyssynchrony and its relationship with cardiac function in patients with DCM.
