1.Research advances in drug transporters and multidrug resistance
China Oncology 2000;0(06):-
Multidrug resistance (MDR) is the most common cause in the failure of cancer chemotherapy. Drug transporters play an important role in multidrug resistance in cancer. Activation of drug pumps which belong to ATP-binding cassette (ABC) transporters leads to the efflux of drug via cell membrane.Three subgroups of ABC transporters are involved in the transport of cytotoxic drugs. These transporters include ABCB subgroup,ABCC subgroup and ABCG subgroup. In this review, the author discussed the recent research advances in the ABC transporters associated with MDR.

Objective To compare the effects of target-controlled infusion (TCI) of remifentanil (R) in various doses in combination of TCI propofol (P) on hemodynamic response to tracheal intubation. Methods Fifty ASA Ⅰ - Ⅱ patients (18 male, 32 female) aged 100 bpm) . The TCI system comprised Graseby 3500 infusion pump controlled by computer program Rugloop which included Marsh and Minto pharmacokinetic parameters. Results Induction time (from start of TCI R + P to LOC) was significantly longer in group RO and R2 than that in group R4, R6 and R8 ( P

Objective To assess the value of joint detection of serum cysteine proteinase inhibitors C (sCys-C),urinary kidney injury molecule 1 (uKIM-1),urinary neutrophil gelatinase-associated lipocalin(uNGAL) and urinary interleukin 18 (uIL-18) for early diagnosis of acute kidney injury (AKI) in critically ill patients. Methods A total of 256 adult patients who stayed Intensive Care Unit for 24 hours in the Third People's Hospital of Liaocheng between Aug 2011 and Dec 2012 were enrolled.According to Kidney Injury Net(AKIN) work, the patients were divided into non-AKI group and AKI group (including state 1,2 and 3).The concentrations of urine NGAL,KIM-1,IL-18 and serum sCys-C were measured. The diagnosis value of four biomarkers joint detection and single detection for AKI were analyzed with the receiver operating characteristic (ROC) curve and the area under curve (AUC).Results (1) The levels of uNGAL,uKIM-1,uIL-18 and sCys-C were higher in patients with AKI than the patients with no AKI (P < 0.01).(2) The area under curves of uNGAL,uKIM-1,uIL-18,sCys-C and joint detection were 0.742,0.871,0.803,0.703,0.925 respectively.(3) The sensitivity and specificity of parallel tests and serial tests of four biomarkers were 97.9%,62.8%,64.3% and 96.2% respectively. There were significant differences of sensitivity or specificity between single test and joint tests. Conclusions The urine NGAL,KIM-1,IL-18 and serum Cys-C are sensitive indexes for the early diagnosis of acute kidney injury.Joint detection has high value for early diagnosis of AKI.

At present, an effective detecting method for brain function impairment for the patients with the glioma is urgently needed in clinic, because it may help us understand its pathogenesis. This paper proposes a method of combining diffusion tensor tracing technology and 'small world' network. It utilizes the degree of brain function network to study complex network topological properties of the patients with the glioma in temporal lobe area. The experimental results showed that the brain networks of the patients with the glioma of different grades were destroyed compared with those of the normal persons, but the destruction degree is independent of the tumor grades. The distribution of functional connections is index truncated power-law accompanied by significant heterogeneity. Meanwhile, the stronger functional areas of information in the glioma have transferred and there exists lack of language function area and sensory function area.

AIM: To observe the effect of crocetin on the apoptosis and the changes of its related regulating proteins caspase-3 and Bcl-2 expression induced by hydrogen peroxide (H_2O_2) in cultured cardiomyocytes. METHODS: Changes of cellular morphology were detected under microscope. Apoptosis rates of the cells were analyzed by PI staining with flow cytometry. Expressions of caspase-3 and Bcl-2 proteins in the cells were determined by immunofluorescence with flow cytometry. RESULTS: In the concentrations used, more severe morphological changes with higher apoptosis rate of the cultured myocardial cells were seen in each H_2O_2 group than that in control group. When treated with 1?10 -4 mol?L -1 H_2O_2, the caspase-3 was increased and Bcl-2 protein decreased remarkably in the cells. But each dosage of crocetin, especially the highest one (5?10 -5 mol?L -1, P

Objective: To evaluate the clinical profile of target controlled infusion based anesthesia using remifentanil and propofol. Methods: 16 ASA I II patients undergoing elective laparoscopic cholecystectomy were enrolled. Remifentanil was set at 7?g?L -1 as target and propofol at 3 mg?L -1 throughout the whole procedure. The hemodynamics during induction of anesthesia and recovery profiles were recorded. Arterial blood samples for analysis of remifentanil were taken 15min after infusion, 20 min after infusion and at time of emergence. Results: After induction of anesthesia, systolic blood pressure (SBP) decreased from (144?27) mm Hg to (101?18) mm Hg ( P 0.05). SBP, MBP and HR remained stable after intubation for 3min. No patient showed haemodynamic stress to tracheal intubation. Times from stopping administration of anesthetics until full spontaneous respiration, eye opening, tracheal extubation, orientation and discharging from the postanesthetic care unit (PACU) were (12?6), (9?4), (13?6), (15?5) and (19?7) min respectively. Measured drug values of remifentanil were (4.6?9.5) ?g?L -1 , (6.6?11.5) ?g?L -1 , (1.2?8.7) ?g?L -1 respectively. Conclusion: Remifentanil/propofol TCI based anesthesia achieved the optimal hemodynamic stability during anesthesia induction and maintenance, and better recovery profile from anesthesia. Measured drug values of remifentanil showed a considerable interindividual variation and more lower than the set target.

Five known compounds have been isolated from the culture of an endophytic fungus Fusarium sp., which was originated in a seagrass Spartina alterniflora. Their structures have elucidated by TLC comparision with standard and spectral methods as ergosterol (Ⅰ), ergosterol peroxide (Ⅱ) cinnamic acid (Ⅲ), para-hydroxyphenylpropionic acid (Ⅳ) and beauvericin (Ⅴ), respectively.
