1.The Clinical Study on Chronic Pelvic Inflammation with Combined TCM and WM Methods

[Objective]To investigate the therapeutic effects of treating chronic pelvic inflammation with combined traditional Chinese medicine (TCM) and western medicine (WM). [Methods]500 patients in hospital were divided randomly into two groups:control group and treating group.Each group included 250 cases.The contral group were treated with anti-by veins for one week.the treating group were treated with antibiotics by vein and herbs Yupeng Tang by enema.The change of symptoms,signs and Ultra-B were observed before and after treatment.[Results]56 cases were cured ,102 cases had remarked effect ,91 cases had therapeutic effect,the total effective rate was 99.6% in treating group.However ,20 cases were cured,41 cases had remarked effect,135 cases had therapeutic effect ,the total effective rate was 78.4% in control group. Objective:To study the effects of Jiaweitaohechengtang on expression of?-SMA protein in hepatic fibrosis rats induced by CCl4.Methods:Hepatic fibrosis models were induced by CCl 4 .All the experimental rats were divided into six groups:normal control group,model control group,colchicine positive treatment group,jiaweitaohechenqitang groups with high- dosage,medium-dosage and low-dosage.Drugs were given to rats for 8 weeks by intragastric administration.Scores of hepatic fibrosis were assessed with semiquantitive scoring system(SSS)through tianlanghong staining.Protein expression levels of ?-smooth muscle actin(?-SMA)were determined with immunohistochemical steining method.Results:SSS in model control group were higher than those in normal control group,but lower than those in three treatment groups with jiaweitaohechengqitang. Compared with nornal control group,the protein expression levels of?-SMA in liver tissues in model control group increased markedly,but decreased when it was compared to jiaweitaohechengqitang groups.There was a significant difference between model control group and three treatment groups.Conclusion:Jiaweitaohechengqitang has therapeutic effects on hepatic fibrosis.The mechanism may be ralated to the inhabitition of the activation of HSC.
3.Effect of MAPKK/MEK-MAPK/ERK Signal Transduction Pathway in Rat Neural Stem Cells

Objective To incubate the rat neural stem cells with specific inhibitor (PD98059) of MAPKK/MEK to clarify the effect of MAPKK/MEK-MAPK/ERK signaling pathway in neural stem cells (NSC).Methods NSCs derived fiom E15-16 rats were isolated and cultured.After treated with different concentration of PD98059,they were subjected to the detection of cell proliferation,Nesn'n, BrdU and β-tubulin-Ⅲ by WTS-8 assay,immunofluorescent staining or Western Blot respectively.Results PD98059 had an effect on the survival,proliferation and differentiation of NSC at a concentration-dependent manner.The viability of the NSC was significantly decreased than the control (P<0.05),whereas the numbers of BrdU-positive and β-tubulin-Ⅲ positive cells were notably fewer than the control (P<0.05) after incubated with the higher concentration of PD98059.The ERK expression was blocked by PD98059 at a concentration-dependent manner.Conclusion The MAPKK/MEK-MAPK/ERK signaling pathway played a vital role in the survival, proliferation and differentiation of NSC.
4.Influence of acupoint application on lipid metabolism in elderly diabetics with hyperlipidemia

ObjectiveTo observe the clinical effect of acupoint application on lipid metabolism in elderly diabetics with hyperlipidemia.Methods60 elder diabetics were divided into two groups.In the drugs group,patients were purely treated with simvastatin or lipanthyl.In the acupoint group, patients received acupoint application.The serum total cholesterol(TC), triglyceride (TG), low-density lipoprotein (LDL-C), high-density lipeprotein (HDL-C), aPoprotein A(ApoA) and apoprotein B(ApoB) ere measured respectively.Adverse events were observed.ResultsThe levels of TC、TG、LDL-C、ApoB were significantly dechned(all P <0.01 ~0.05).However,the rates of reduce lipid had no significant difference in two groups(P >0.05).The levels of HDL-C and ApoA were increased in the drugs group,but the homologous levels in the acupoint group showed no significantly change(all p > 0.05).The adverse reaction in the acupoint group was significantly lower than that in the drugs group(P <0.05).ConclusionAcupoint application could decrease the blood lipid spectrum in the senile diabetics with hyperlipidemia.This therapeutics could reduce adverse drug reactions.
5.The Role of Proliferation and Differentiation of Preadipocyte on High Fat Diet-induced Obesity and Prevention of Obesity through Exercises in Rats

Objective To explore the influences of proliferation and differentiation of preadipocytes on high fat diet-induced obesity and obesity prevention through exercises in rats.Methods Forty male Sprague-Dawley rats were divided into a normal diet control group (C,n=8),a normal diet with exercises group (E,n=8),a high fat diet control group (H,n=14) and a high fat diet with exercises group (HE,n=10).Group C and group H kept sedentary,while group E and group HE underwent treadmill exercises at about 75%VO2max level.After 12 weeks of intervention,the body weight,epididymal,perirenal and retroperitoneal fat mass were recorded,and the total fat mass was determinated.Stereology method was used to calculate the number and average diameter of fat cells.Western Blotting was conducted to measure the expression of PPARγ and C/EBPα protein in adipose tissues.Results (1) The body weight and total fat mass of group H were significantly higher than those of group C (P<0.01).Perirenal,retroperitoneal fat and total fat mass of group E and HE were significantly lower than those of group C and H respectively (P<0.01).(2) The total fat cell number of group H was significantly higher than that of group C.The average diameter of fat cells in perirenal and retroperitoneal fat pads of group E were significantly lower than that of group C (P<0.05,P<0.01),while that of group HE in epididymal,perirenal and retroperitoneal fat pads was significantly lower than that of group H (P<0.01).(3) Compared with group C,the expression of PPARγ protein of group E and H increased significantly in epididymal,perirenal and retroperitoneal fat pads (P<0.01).The expression of C/EBPα protein of group H was significantly higher than that of group C in epididymal and perirenal fat pads (P<0.01),the expression of C/EBPα protein of group E was significantly higher than that of group C in perirenal and retroperitoneal fat pads (P<0.01),while the expression of C/EBPα protein of group HE was significantly lower than that of group H in perirenal fat pads(P<0.01).Conclusion The proliferation and differentiation of preadipocyte was enhanced in the development of high fat diet induced obesity and exercises to prevent obesity,but its role and mechanisms were different.The high fat diet increases the number of fat cells which is a compensatory mechanism for the body to adapt to fat accumulation,while exercises might promote cell renewal and decrease the average size of fat cells which is an adaptive mechanism to improve fat storage.
6.Study on biocompatibility of hydroxyapatite/high density polyethylene (HA/HDPE) nano-composites artificial ossicle.

This study was aimed to evaluate the biocompatibility of Hydroxyapatite/High density polyethylene (HA/ HDPE) nano-composites artificial ossicle. The percentage of S-period cells were detected by flow cytometry after L929 cells being incubated with extraction of the HA/HDPE nano-composites; the titanium materials for clinical application served as the contrast. In addition, both materials were implanted in animals and the histopathological evaluations were conducted. There were no statistically significant differences between the two groups (P >0.05). The results demonstrated that the HA/HDPE nano-composite artificial ossicle made by our laboratory is of a good biocompatibility and clinical application outlook.

7.Effect of CT guided implantation of 125Ⅰ radioactive particles in superficial malignant tumors

Objective To investigate the short-term clinical effect of CT guided 125Ⅰ radioactive particle therapy in superficial malignant tumor. Methods The clinic data of 28 patients with metastatic superficial malignant tumor in our hospital were analyzed retrospectively.All patients were treated with CT guided 125Ⅰ radioactive particle therapy.The short-term effects,1 year survival rate and 1 year progression free survival rate of the patients were compared.Results Objective remission rate(ORR)and disease control rate(DCR)after 6 months were 92.86% and 100.00%.1 year overall survival and 1 year progression free survival were 96.43%(27/28)and 82.14%(23/28), respectively.The median overall survival and median progression free survival were 26.978 months (95%CI:22.558-31.399)and 16.932 months (95 % CI:14 .471-19.393).There were 27 cases of 0-Ⅱ degree adverse reactions,1 case of grade Ⅲ adverse reactions and no grade Ⅳ adverse reactions.No signs of 125Ⅰ radioactive particle translocation,vascular embolism and vascular rupture were found. Conclusion 125Ⅰ radioactive particle treatment of superficial malignant tumor has a definite short-term curative effect,with overall survival and progression free survival longer and higher safety,which can be considered in clinical application.
8.Supervillin regulates convergence and extension movements via Wnt/β⁃catenin pathway in zebrafish embryos

Objective :
To explore the role and molecular mechanism of Supervillin in zebrafish embryo development.
Methods :
Clustal Omega and DNAman software were used to analyze and compare the homology of amino acid sequences of Svil and SVIL in zebrafish and human. The expression pattern of Svila (subtype of Svil protein in zebrafish) during early embryonic development was analyzed by RT⁃PCR and in situ hybridization. Svila expression in zebrafish was inhibited by injecting morphinos ( MO) , and morphological changes of embryos were observed. The expression and nuclear localization of β ⁃catenin protein were detected by Western blot , and the expression of Wnt/β⁃catenin target gene was detected by RT⁃PCR.
Results :
During the early embryonic development of zebrafish , Svila was maternally expressed and showed an upward trend with the development process. The expression of Svila was reduced by MO , and the development of zebrafish embryos was distorted and the body axis was bent , which might be related to the blocked movement of concentrated extension of embryos. Further studies showed that Svila expression affected β⁃catenin nuclear transport and Wnt/β⁃catenin signaling pathway activation.
Conclusion
Svila regulates the concentrated extension movement of zebrafish embryos by activating the Wnt/β⁃catenin signaling pathway.
