Objective To elucidate the effect of ischemic preconditioning(IPC)on regional fluidity of myocardial cell membrane during cardiopulmonary bypass (CPB). Methods Seventy-five felines were randomized into three groups: Group A (CPB group,n =25),in which CPB was conducted without aortic cross-clamping (ACC);Group B (IR group,n =25),with 60min ACC followed by 60min reperfusion, and cardioplegia used during period of ACC;Group C (IPC group,n =25),with protocol similar to that of group B except for three-cycle IPC applied before ACC. Electron spin resonance (ESR) technique was employed to monitor membrane regional fluidity of cardiomyocyte during CPB in all groups. Results Regional fluidity of myocardial cell membrane kept unchanged in CPB group and markedly decreased during ACC and reperfusion periods in IR group. Though the regional fluidity of myocardial cell membrane during ACC and reperfusion periods in IPC group was slightly decreased compared to CPB group,it was significantly higher than that in IR group. Conlusion IPC can protect myocardial cells from ischemia and reperfusion injury by maintaining the fluidity of myocardial membrane during CPB.

Two patients underwent liver transplantation of blood type incompatibility were collected from Fuzhou General Hospital of Nanjing Military Area Command of Chinese PLA. Case 1: A male who had primary hepatic carcinoma underwent classic orthotopic liver transplantation; the blood of donor was type A, and the blood of recipient was type O. Case 2: A female having history of type B hepatitis underwent classic orthotopic liver transplantation due to pregnancy combining with severe liver disease and coagulation disorder; the blood of donor was type B, and the blood of recipient was type O. Immunohistochemistry staining was used to observe pathological changes and deposition of various immunoglobulin and complement in two cases following liver transplantation of blood type incompatibility under optic microscope and to investigate diagnostic standard of humoral rejection. The results showed that linear or granular depositions of IgG, IgM, IgA, C4c, C4d, and Clq were found in endothelial cells of hepatic sinusoid, suggesting that IgG and other immunoglobulin exhibited a strongly positively diffused deposition on the endothelial cells of hepatic sinusoid, while expression of C4d and other complements was also found. All those mentioned above could be considered as evidences to prove onset of humoral rejection in transplanted liver tissue.

Objective:To explore the role of γδ T cells in the transdifferentiation of immature dendritic cells(imDC) into osteoclasts(OC). Methods:(1) Peripheral blood mononuclear cells(PBMNC) were cultured with zoledronate(Zol) and recombinant human interleukin-2(IL-2),and PBMNC from healthy volunteers were cultured with granulocyte macrophage colony-stimulating factor (GM-CSF) and recombinant human interleukin-4(IL-4) to differentiate into imDC,which were then cultured with receptor activator nuclear factor к B ligand(RANKL) and macrophage colony-stimulating factor(M-CSF) to differentiate into OC. The purity of γδ T cells, and phenotype changing of OC transdifferentiated from imDC were investigated by flow cytometry. (2) Co-culture system was es-tablished using millicell inserts.γδT cells isolated with immune magnetic bead were placed in the upper compartment and imDC in the lower compartment in the ratio of 10∶1. To explore the role of γδ T cells during differentiation of imDC into OC,tartrate resistant acid phosphatase( TRAP) staining and bone resorption observation staining were used. Tumor necrosis factor-alpha( TNF-α) of supernatant liquid from different cultures was measured using ELISA(Enzyme linked immunosorbent assay) kit. Results:(1) γδT cells can be ex-panded from PBMNC of MM patients, and the production capacity was similar to that of healthy volunteers ( 68. 87%± 20. 94% vs 69. 33%±16. 84%,P>0. 05 ) . ( 2 ) OC could be transdifferentiated from imDC when cultured with RANKL and M-CSF. ( 3 ) The number of TRAP+ multinuclear cell and the absorption area of dentine were significantly lower in the group of imDC indirectly co-cultured with γδ T cells than in the group of control imDC(5.67±0.58 vs 28.33±2.08,4.97%±4.3% vs 28.47%±12.8%, respectively). (4) Under the circumstance of γδ T cell-imDC indirect coculture,TNF-α got significantly higher. Conclusion: γδ T cells might inhibit the transdifferentiation of imDC into OC.γδ T cells-based immunotherapy is expected to be a new treatment for myeloma bone disease.

Purpose To analyze clinicopathologic and prognostic features in 9 cases of children endocapillary proliferative glomerulone-phritis with hepatitis B virus antigen deposition ( HBV-ECPGN) . Methods Retrospective analysis of demographic information, clini-cal manifestations, laboratory parameters, pathological and prognostic features was carried out for 9 cases of HBV-ECPGN and 13 cases of acute poststreptococcal infection endocapillary proliferative glomerulonephritis ( APS-ECPGN) for comparison. Renal biopsy tissue were fixed in formalin and embedded in paraffin, stained with HE, PAS and PAM-Masson. Immunohistochemical study with EliVision method was performed. Three cases were submitted for electron microscopy. Results There were 7 males and 2 females ( M ∶ F=7 ∶ 2) of HBV-ECPGN. The median age was 10. 3 years. Serum C4 deposition ratio HBV-ECPGN was significantly greater than APS-ECPGN group (P<0. 05). There was an average of 11. 2 weeks of HBV-ECPGN kidney disease duration, which was significantly lon-ger than an average of 3. 8 weeks of APS-ECPGN group (P<0. 05). There was no disease relapse in all cases during 53. 55 months follow-up. C4d deposit was significantly stronger in all HBV-ECPGN cases compared with control group (APS-ECPGN cases). There were no significant differences in deposit of IgG, IgM, IgA, C3d and C1q between the two groups. HBsAg deposit in juxtaglomerular sites was identified in all cases. Conclusions Serum C4 decrease is more common in HBV-ECPGN than APS-ECPGN. Which may be associated with HBV infection, there is longer disease duration of HBV-ECPGN. C4d deposit is significantly stronger than control group, suggesting pathogenesis of HBV-ECPGN and APS-ECPGN is different. HBsAg deposit may be closely related to the pathogene-sis of HBV-ECPGN. HBsAg deposit in juxtaglomerular sites may be characteristic of HBV-ECPGN.

Basic & Clinical Medicine 2006;0(06):-
Objective To explore the chromosome features of pancreatic cancer cell lines from Chinese patients. Methods G-band Chromosome karyotyping was performed on pancreatic cancer cell lines of PC1, PC2, PC3, PC4 and PC7 that were established in our laboratory. The results of cytogenetic analysis were confirmed by fluorescence in situ hybridization using Chromosome 3, 13 18 and 20 paint probes. Results All the 5 cell lines were hypotriploid and showed a modal number of 58 for PC1, 56 for PC2, 61 for PC3, 53 for PC4 and 54 for PC7 with different proportion of complex chromosome rearrangements including dicentric chromosomes, double-minute chromosomes, ring chromosomes, acentric fragments or complex chromosome translocation, etc. Conclusion Hapotriploid accompanied with complex numerical and structural chromosomal rearrangements is the main cytogenetic marker of chromosomal instability in pancreatic cancer cell lines. Molecular cytogenetic techniques have to be used beside conventional G-band karyotyping for accurate identifying abnormal chromosomes of pancreatic cancer cell lines.

Objective To prospectively evaluate the correlation between a single breath-hold three-dimensional (3 D) and several breath-hold two-dimensional (2D) delayed enhancement MR imaging sequences in the assessment of myocardial infarction size and the differences on image quality. Methods Fifteen patients with myocardial infarction underwent MR scan by using a single breath-hold 3D inversion-recovery fast low-angle shot (FLASH) sequence and several breath-hold 2D turbo-FLASH as the reference standard. Paired-samples t test was used to compare the ratio of the infarction areas in two sequences. Two-way ANOVA was used to assess the contrast-to-noise ratio (CNR) on 3D,2D magnitude-reconstructed and 2D phase-sensitive (PS) images. P value less than 0.05 was considered to indicate a significant difference. Meanwhile subjective visual evaluation was also performed to compare the image quality. Results The infarction ratio determined by 3D MR imaging 31.28% was no significant difference with that of 2D MR imaging 30.91% and had a good correlation (t = -0.505,P =0.621,r =0.990). The CNR was significantly higher in 3D and 2D magnitude-reconstructed groups than in 2D-PS group (3D and 2D magnitude-reconstructed images, 2D P5 image, 43.43±20.67 and 34.10±14.29, 7.59±2.59, respectively) (F = 24.376, P < 0.01). However, the contrast between normal and infracted myocardium was the highest in 2D-PS group with subjective visual evaluation (scores of 3D, 2D magnitude-reconstructed and 2D-PS images, 2.33, 2.13 and 2.73, respectively). On the other hand, the background noise on 2D-PS images was the highest in 3 groups (scores of 3 D, 2 D magnitude-reconstructed and 2 D-PS images, 2.67, 2.53 and 1.20, respectively). Conclusion The myocardial infarction ratio obtained with 3D MR imaging sequence is accurate, and the image quality is good.
