Objective:To prepare and identify the monoclonal antibody against hPPAR?2.Methods:cDNA of hPPAR?2 was obtained from pMD18-T/hPPAR?2.hPPAR?2 was expressed in prokaryocyte and purified successfully.The anti-hPPAR?2 monoclonal antibodies (mAbs) 1B4,3H2,3H10,10D6 and 10D8 were produced by immunization with purified hPPAR?2.The specificity of the antibodies was identified by ELISA,Western blot assay and immunocytochemistry.Results:Five novel murine monoclonal antibodies only against hPPAR?2 were harvested,and they were all specific to hPPAR?2.Conclusion:The antibodies obtained provide more useful tools for following research and settling the basis for screening new drugs and mechanism.

OBJECTIVE:To detect and analyze the safety signals of pemetrexed in post-marketing,and provide references for rational drug use in the clinic. METHODS:A form of data mining techniques was used to detect the safety signals of pemetrexed in American adverse events reporting system and the analysis of signal effect factors i.e.,gender,age,dosage and indication was per-formed by SPSS 19.0. RESULTS:173 signals related to pemetrexed were excavated in 5 397 235 reports,and the number of report-ed adverse event in blood-lymphatic system was the most(491)and the number of signals in gastrointestinal system was the most (17). There was no statistical significance in the distribution of blood-lymphatic system and gastrointestinal system in gender(P=0.075) and age (P=0.726) compared to other system adverse event,but there was statistical significance in dosage (P=0.007) and indication(P=0.001). CONCLUSIONS:Adverse events induced by pemetrexed in blood-lymphatic system and gastrointestinal system should be monitored at the same time,especially combined with non-steroidal anti-inflammatory drugs.

Objective To investigate the effect of the inhibitor SB203580 of p38 mitogen-activated protein kinase (MAPK) signal transduction pathway on biopterin (BH 4)/nitric oxide (NO) expression and elucidate the potential mechanism of MAPK in biopterin-mediated NO induction after endotoxic shock. Methods A total of 56 male Wistar rats were randomly divided into normal control group (n=8), endotoxic shock group (n=32) and SB203580 treatment group (n=16). After animals were sacrificed, tissue samples from the liver, the lungs as well as the kidneys were harvested to determine the expressions of guanosine triphosphate cyclohydrolase I (GTP-CHI) and inducible nitric oxide synthase (iNOS) mRNA and observe the changes of BH 4 and NO levels in blood and tissues. Results With endotoxin challenge, GTP-CHI mRNA expression and BH 4 levels were significantly elevated in various tissues and maintained at high levels up to 24 hours. Similarly, the iNOS mRNA expression and NO levels in the tissues significantly increased too, especially in the liver and the lungs. Objective To investigate the value of fused 3D-SPACE and 3D-TOF images in diagnosis of intracranial neurovascular compression syndrome.Methods 26 patients with intracranial neurovascular compression syndrome were examined using 3D-SPACE sequence and 3D-TOF sequence.After 3D-SPACE and 3D-TOF were fused by different times,the quality of image as well as the ability of j udging the relationship between nerves and blood vessels was analyzed and compared between the original images and the fused images.Results In this study,the fused images by adding one 3D-SPACE sequence and one 3D-TOF sequence showed the highest accurate than others(2 1 cases),and the j udgement of the relationship between the responsible blood vessels and nerve accor-ded with the surgical results to the highest extent (P<0.05).Conclusion The fusion image of 3D-SPACE sequence and 3D-TOF se-quence can improve the diagnostic ability of the intracranial neurovascular compression syndrome,and the appropriate proportion of the fusion image can show the relationship between the nerve and blood vessel.

Objective To study the curative effect and practicability of traditional anterior cervical surgery for cervical spondylosis.Methods retrospective lyunaly2ed,104 cage8 of anterior cervical sugery from June 1997 tO October 2005 Among them,there were 17 patients with radculopathy,31patients with cervical myelopathy,56 patients with combination of myelopathy and radieulopathy.all cases were treated by traditional anterior trephiement to excise prominent uncleus+interbedy fusion with auto-ilium graft.Results The resuhs were evaluated by Odom.The cases were all followed up.13 excellent,68 good,23 fair,0 poor.Excellent/good rate is 77.9%.The average medical coat of one patient is 4200 RMB.Conclusion The traditional Anterior cervical surgery for cervical mydopathy is safe、effective、practical.

ObjectiveBy the study of histological feature of the regenerated nerve at different time after operation,to explore the mechanism of the peripheral nerve regenerationwhen the distal nerve stump is repaired by a smaller proximal end with artery sleeve briding.MethodsFour Wistar rats were served as the donors of arteris for bridging.Sixteen Wistar rats were randomized into 2 groups (n =8 per group).In control group,the right sciatic nerves were cut off and received epineurial suture.In experimental group,the right sciatic nerves received artery sleeve bridging in different size.HE staining was taken to observe the histological changes of the regenerated nerve at 7,14,21 and 28 days after operation.ResultsThe new regenerated axons need about 21 days to pass the bridging gap.Twenty-eight days after operation,the number of the regenerated axons in distal segment was greater than that of the proximal in the experimental group.The regenerated axons were matured and more regular.Conclusion The functional reserve of repaired nerve is exists when the proximal end is smaller than the distal in size by means of amplification effect.And the method of artery sleeve bridging provides a better situation for functional reserve.
