1.Culture of rabbit periosteal osteoblasts using modified enzymatic digestion combined with explant method

BACKGROUND:Periosteal osteoblasts possess strong reproductive activity,as well as osteoblastic differentiation potential,which is an ideal seed cell if can shorten the culture time.OBJECTIVE:Modified enzymatic digestion was used to culture rabbits'osteoblasts.and to study the adherence and proliferation of osteoblasts on the sudace of sandblasting titanium.MEITHODS:Periostea were harvested from the theanteromedial surface of the proximal tibia of male,Japanese white rabbits,and cultured as follow:①Routine method:Digested with 0.25%trypsinase at 37 ℃ for 30 minutes,followed by digestion with 0.1%type I collagenase at 37 ℃ for 30 minutes,vibration.removed trypsinase and dried.After 2 hours,DMEM containing 15% fetal bovine serums were added.②Modified method:30 minutes culture of type I collagenase was prolonged to 1 hour.The osteoblasts were identified by alkaline phosphatase staining and calcium node staining.The adherence and proliferation of osteoblasts cultured on sandblasting surface were measured by scanning electron microscopy and MTT.RESULTS AND CONCLUSION:Five days after culture.the periosteal steoblasts crawled out from tissues,gathered as monolayer with tdangle or polygon at after 25 days of modified culture.After 1 month of culture,superposition growth of calcium nodus appeared.The cultured cells possessed the morphological characteristic and biological behavior of osteoblasts.which were positive to alkaline phosphorase and calcium node staining.The time of cells cultured with routine method covered flask delayed 12 days than modified method.The osteoblasts were inseted into sandblasting titatium with pseudopodium.However,the adherence and proliferation of osteoblasts cultured on sandblasting surface had no obviously difference between two culture methods.The results suggested that modified enzymatic digestion can sho~en the culture time without effect on adherence and proliferation of osteoblasts.
2.Effect of fluoride therapy on remineralization and whiting of bleached enamel

Objective To evaluate the effect of fluorin dentifrice on the microhardness and whiting of dental enamel bleached with different concentration bleachers.Methods 6%carbamide peroxide(CP)and 30%CP were selected as bleachers.Twenty undefected molars were subjected to 3 groups.Groups A and B.which composed of 8 teeth each,were tested and group C was the control one.Group A(buccal surface)and group B(buccal surface)were treated with 6%CP.Group A (lingual surface)and group B(lingual surface)were treated with 30%CP.They were bleached 30 min every day for two weeks.The teeth in group B were brushed with fluorin dentifrice for 15 min after bleached every time and then all tested samples were kept in the artificial saliva at 37℃.Vickers microhardness of all teeth and color measurement of groups A and B were made before and at the end of bleaching procedure.Results The difference of microhardness values between the bleached and control samples were not statistically significant(P>0.05).Brushed with fluorin agent significantly increased the hardness of enamelin group B(P<0.05).The color change was not significant between bleached samples.Conclusion Using fluoride in the interphase of bleaching makes this therapy safer,and does not affect the whitening effect.
3.Value of transcranial Doppler ultrasonography on diagnosing the cerebral vasospasm following subarachnoid hemorrhage

Objective To explore the value of transcranial Doppler ultrasonography (TCD) on diagnosing the cerebral vasospasm (CVS) following subarachnoid hemorrhage (SAH). Methods TCD was used to detect and monitor the cerebral blood flow status to estimate CVS following SAH in 30 cases, then the results of TCD were analyzed by combining with clinical manifestation and SAH severity on CT. Results 17 patients (56.7%) had the evidence of CVS confirmed by TCD, 10 of them were nonsymptomatic and 7 were symptomatic. 5.Psychological stresses of government employees, businessmen, and police officers

Objective To explore stress response of government employees,businessmen and police officers.Methods A total of1547 government employees,8672 businessmen and1555 police officers were recruited and completed a questionnaire survey concerning psychological stress.t or F test was used for data analysis.Results There were significant differences of stress response among government employees,businessmen and police officers ( P < 0.01).Male or female government employees showed different stress response,physiology,emotion and behavior (all P<0.01).Government employees from various age group showed difference in emotion and behavior ( both P < 0.01).Businessmen of various gender and age were found difference of stress response at all dimensions ( P < 0.05 ).Police officers of different gender showed different behavior ( P < 0.0l ).ConclusionIt may be necessary to carry out occupation-specific stress management for individuals.
6.Prospectively longitudinal study of the relationship between infancy abuse and antisocial personality disorder

ObjectiveTo investigate the relationship of antisocial personality disorder(ASPD) and infancy abuse.MethodsWith prospectively longitudinal research design,76 samples at infancy were initially recruited.Infancy abuse was measured by Child Protective Service records and reconfirmed by one hour family interview.56 remained samples at young adulthood were measured for antisocial personality disorder,depression,and substance abuse by utilizing Structured Clinical Interview-Ⅰ/Ⅱ (SCID-Ⅰ/Ⅱ) of DSM-Ⅳ.Longitudinal data was analyzed by multiple imputation and hierarchal regression.Results 33.9% samples met at least one item of ASPD,38.0% were diagnosed with substance abuse and 32.4% with depression.Gender was correlated with ASPD symptoms (F=1.40,P =0.001 ).Infancy abuse significantly predicted ASPD (β =0.28,P=0.03 ).Further analysis demonstrated that infancy abuse was associated with ASPD with controlling substance abuse and depression (β =0.19,P =0.04 ).ConclusionInfancy abuse,with fundamental long-term effects on personality,is a risk factor for the etiology of antisocial personality disorder.The underlying mechanism might involve biological alternations.It is necessary to establish national early-life abuse monitoring and prevention system.
7.Tanshinone IIA protects against triptolide-induced liver injury via Nrf2/ARE activation.

The aim of this study is to investigate the protection effect of tanshinone IIA (Tan) against triptolide (TP)-induced liver injury and the mechanisms involved. Acute liver injury was induced by intraperitoneal injection of TP (1 mg x kg(-1)) in mice. The activities of AST, ALT and LDH in serum and the levels of GSH, GST, GSH-PX, SOD, CAT and MDA in liver tissue were detected. The histopathological changes of liver tissues were observed after HE staining. Nrf2 translocation in liver tissue was detected by Western blotting, and real-time PCR was used to measure the expression levels of GCLC, NQO1 and HO-1 mRNA. The results showed that pretreatment with Tan significantly prevented the TP induced liver injury as indicated by reducing the activities of AST, ALT and LDH (P < 0.01). Tan pretreatment also prevented TP-induced oxidative stress in the mice liver by inhibiting MDA and restoring the levels of GSH, GST, SOD and CAT (P < 0.05). Parallel to these changes, pretreatment with Tan could attenuate histopathologic changes induced by TP. Furthermore, the results indicated that Tan pretreatment caused nuclear accumulation of Nrf2 as well as induction of mRNA expression of antioxidant response element (ARE)-driven genes such as GCLC, NQO1 and HO-1. These results indicated that Tan could protect against TP-induced acute liver injury via the activation of Nrf2/ARE pathway.
8.Analysis of Clinical Characteristics and Misdiagnosis of 178 Patients with Pulmonary Embolism

Objective The study aimed to analyze the clinical data of patients with pulmonary embolism and summarize the clinical characteristics of pulmonary embolism,in order to improve the correct diagnostic rate and the prognosis. Methods Data of 178 patients who were cured in our hospital from January 2011 to December 2014 were analyzed retrospectively,including relative risk factors,clinical behaviors,examination data,misdiagnosis and prognosis. Results Patients with pulmonary embolism had related basic diseases and risk factors. Their clinical symptoms,signs and ordinary detection methods indicated pneumatorexis and cardiovascular injury,but lacked of specificity. The misdiagnosis rate of the first diagnosis was 48.88%,including 22 cases of the misdiagnosis to COPD, 18 cases to pneumonia and 16 cases to coronary heart disease. The misdiagnosis rate of elderly patients was 56.36%, while that of the young and middle-aged patients was 36.80%,the difference was statistically significant,P < 0.05. Computed tomography pulmonary angiography(CTPA)could accurately observe the pathological changes of pulmonary embolism,and all the patients were diagnosed by CTPA in the research. 11 cases(10%)of elderly patients died,while 2 cases(2.9%)happened in young and middle-aged patients. There was no statistical significant difference(P > 0.05). Conclusion Although the related risk factors of pulmonary embolism in patients existed,the misdiagnosed rate of the disease was high and prognosis was poor due to many confusing basic diseases. And the symptoms,signs and ordinary inspection of the patients lacked specificity. CTPA,being utilized to diagnose pulmonary embolism reliably,should be promoted.
9.Research progress of lactoferrin as drug carriers.

Lactoferrin (Lf) is one of the food protein belonged to the innate immune system. Apart from its main biological function of binding and transport of iron ions, lactoferrin also has many other functions and properties such as antibacterial, antiviral, antiparasitic, catalytic, anti-cancer, anti-allergic and radioprotecting. Lf is usually used as additives of food and cosmetics. The research of lactoferrin has been increasingly reported, and the application of lactoferrin as a drug carrier has drawn extensive attention over the recent year. In this paper, researches of lactoferrin as drug carriers are classified and summarized in brain targeting, liver tumor targeting, lung tumor targeting and oral delivery systems according to their different characteristics.
