1.Yiqihuoxue recipe induces differentiation of rat bone marrow mesenchymal stem cells towards neurons in vitro

BACKGROUND: Previous studies have demonstrated that Yiqihuoxue recipe (Buyanghuanwu decoction) can inhibit cell apoptosis and antagonize free radical injury in cerebral ischemia/reperfusion models. However, there have been few reports regarding the effects of Yiqihuoxue recipe on stem cell differentiation. OBJECTIVE: To observe the effects of Yiqihuoxue recipe on the differentiation of rat bone marrow mesenchymal stem cells (MSCs) towards neurons. DESIGN, TIME AND SETTING: The present controlled observational expedment based on cells was performed at the Henan University of Traditional Chinese Medicine, i.e., the Third-Grade Laboratory of the State Administration of Traditional Chinese Medicine between March 2006 and February 2008.MATERIALS: Twenty clean Sprague-Dawley (SD) rats, aged 1-2 months, weighing 100-150 g, of either gender, were included in this study. The Chinese medicine compound Yiqihuoxue recipe was made in the Manufacturing Laboratory of the First Affiliated Hospital of Henan College of Traditional Chinese Medicine as follows. 60 g Danggui (Radix Angelicae Sinensis), 30 g Taoren(Semen Persicae), 30 g Chuanxiong(Szechwan Lovage Rhizome) were selected and mixed in 720 mL water for extract volatile oil for later use. The remaining gruffs and extract were supplemented with 1 200 g Huangqi(Radix Astragali), 60 g Chishao (Radix Paeoniae Rubra), 30 g Dilong(Lumbricus), 30 g Honghua(Flos Carthami) before 920 mL water was added. After I hour of decoction and filtering, the gruffs were decocted for another 1 hour after adding 8 640 mL water. Then all decocted liquid was merged and condensed to 600 mL and mixed with 3 mL of volatile oil and tween-80. After high-temperature and high-pressure stedlization, the products were preserved for future use. 1 mL of drug was equal to 2.4 g raw herbs. METHODS: Bone marrow mononuclear cells were isolated by density gradient centrifugation. After culture, bone marrow MSCs were amplified and purified. Passage 10 cell suspension was inoculated into a 40 mm-diameter plastic culture dish. Dulbecco's modified eagle's medium supplemented with 0.1 volume fraction of fetal bovine serum and basic fibroblast growth factors was added for 24 hours of culture when adherent cells reached 60%-90% confluence in each group. Thereafter, the "Yiqihuoxue" group was incubated for 5 hours using medium containing DMSO, butylated hydroxyanisole, 13-mercaptoethanol, and Yiqihuoxue recipe; simultaneously, the control group was treated for 5 hours with fluid containing DMSO, butylated hydroxyanisole, and β- mercaptoethanol.MAIN OUTCOME MEASURES: Identification of bone marrow MSCs by flow cytometry; Detection of Nestin-, and neuron specific enolase(NSE)-, and glial fibrillary acidic protein(GFAP)-positive expression by immunohistochemistry. RESULTS: Flow cytometery results demonstrated that cell surface marker CD90 and CD106 expression was positive, while CD45 and CD34 expression was negative. Immunohistochemistry results showed that the numbers of Nestin-positive cells (1 and 3 hours after induction) and NSE- and GFAP-positive cells (5 hours after induction) were significantly greater in the Yiqihuoxue group than in the control group (P < 0.01). CONCLUSION: Yiqihuoxue recipe can induce the differentiation of bone marrow MSCs towards neurons in vitro.
2.Long-term follow-up study of the microsurgical repairment of the defects in leg and foot

Objective To retrospect and analysis the long-term follow-up results of different kinds of flaps in repairment of the defects in legs and feet' Methods Seven different kinds of flaps or musculocutaneous flaps were used to repair for 66 cases of the soft tissue defect in legs and feet;of the areas from 30 cm ? 15 cm to 6 cm? 4 cm, 28 flaps were transplanted with vessel anastomosis;36 flaps were pedicle and 2 flaps were crossed legs. Results Two flaps with vessel anastomosis were necrosis,three were necrosis partly' Others were healed by first intention. 5 cases of the flaps in foot weight carrying areas were ulceration at different times after operation, 1 case was intraltable ulcer. conclusion The chronic ulceration and infection cause the leg and foot vessel injury,local tissue adhesion and dissection difficultly. all these causes result in blood vessel crisis after operation. It's better to use pedical flap to repair this kind injury. The result is good to use medial flap of foot to rapair the soft tissue defect of heavy load region of foot. If using flap of anastomotic vessel to trasplant, it's much better to use flap containing sensative nerve to repair the defect so that it can satisfy the demand of foot heavy load
3.Effects of subclinical hypothyroidism on bone age and height in children

Objective To investigate the effects of pediatric subclinical hypothyroidism on bone age and height and their treatment outcome.Methods Ninety-seven children with subclinical hypothyroidism were served as this study group and 80 children with normal thyroid function as control group.Bone age and height were detected.Patients in study group were treated with thyroid hormone replacement therapy.The efficacy and lipid changes were evaluated before and after treatment.Results The height and bone age in patients of study group were (1.31 ± 0.06) m and (8.29 ± 0.35) years,significantly lower than the control group ((1.41± 0.09) m,(10.02 ± 1.45) years; t =13.532,P < 0.001 and t =14.590,P < 0.001).Thyroid-stimulating hormone showed a significant negative correlation with bone age (r =-0.783,P < 0.001).In a total of 97 children with subclinical hypothyroidism,71 cases were cured,24 cases with improvement and 2 cases with ineffective,and the total effective rate was 97.9%.Before treatment total cholesterol ((3.45 ±0.42) mmol/L),low density lipoprotein cholesterol((2.50 ± 0.35) mtmol/L) in the study group were significantly higher than those of control group((3.33 ± 0.26) mmol/L,(2.24 ±0.22) mmol/L;t =4.378,5.103 ;P =0.036,0.002).After treatment,the level of low density lipoprotein cholesterol in children with subclinical hypothyroidism was significantly lower than before treatment((2.34 ±0.36) mmol/L vs (2.50 ±0.35) mmol/L,t =4.389,P =0.033),but there was no significant difference in terms of other parameters.Contusion Children with subclinical hypothyroidism can lead to lower bone age and shorter height and it may also lead to changes in lipid levels.More attention should be paid on this disease and early intervention should be provided.
4.Abnormalities of sex chromosome in patients with mental disorders

Subject To explore the features of the abnormalities of the sex chromatin and the sex chromosome of the patients with mental disorders. Methods The sex chromatin in the cells of buccal mucous membrane of the 2 784 patients with mental disorders were examined by using a blinding and random way. The chromosome karyotypes in peripheral blood were further examined in those whose measure showed abnormal. The comparison group consisted of 1 069 normal persons.Results Compaired with the control group,the average frequency of Y chromatin in the male patients [(28.0±6.6)%] and the average frequency of X chromatin in the female patients [(22.3±8.0)%] rose significantly ; the average frequency of X chromatin in the female patients of the schizophrenia subgroup and the affective disorder subgroup were higher than that of neurosis and psychogenic mental disorder subgroup. X chromatins of eight male patients were positive and the karyotypes of the abnormal X chromosome were 47,XXY in 3 cases and 47,XXY/46,XY in 5 cases, the rate of 47,XXY was 7.7 , it rose significantly to the control group. X chromatin of two female patients were negative and the karyotypes of the abnormal X chromosome was 45,X/46,XX, the rate of 45,X was 1.2 ; one female patient had double X chromatin and the karyotypes of the abnormal X chromosome was 47,XXX/46,XX,there is no abnormal sex chromatin and sex chromosome in the control group.Conclusions The higher rate of the sex chromatin might be closely associated with the tendency of heredity of mental disorders , and the rate of the lacking X chromosome in females and the increasing X chromosome in males rise in the patients with mental disorders.
5.The performance verification of high-sensitive C reactive protein test reagent

Objective To verify the accuracy,repeatability and consistency with contracting reagent of the high-sensitive C reac-tive protein(hs-CRP)test by using the independently developed reagents.Methods 150 samples were collected,including 130 ser-um samples and 20 plasma samples.4 times of the relative difference of mean value was used as the detection limits,and the regres-sion analysis was performed after excluding 5 sample outliers,then calculated the expected bias at medical decision level with the 95% confidence interval,in order to judge whether the bias was within the allowable range.Results The recovery rate of hs-CRP test reagent were within the allowable range which was 90% -110%.The hs-CRP testing results of plasma and serum samples from the same source were consistent,and the expected bias of medical decision level was in the permissible range.Conclusion The developed reagent used in hs-CRP test have good accuracy,repeatability,and highly consistency with control reagent.
6.A SURVEY OF THE RELATIONSHIP BETWEEN MOTHERS' CONDITIONS AND 365 LOW BIRTH WEIGHT INFANTS

365 low birth weight infants have been born inour Obstetric Department in the past two years. Aretrospective investigation of their morthers' con-ditions during pregnancy and delivery which had aneffect on their babies weight was done. The resultsshowed that incidence of low birth weight infantswas 7. 99% and mortality rate was 15. 07%. The care.X~2 test indicated that hypertention syndrome ofpregnancy and anemia which the mothers sufferedfrom were the main diseases which affected birthweight of infants. The analysis of multiple stepwiseregression showed that the most dangerous factorswere peasant mothers, premature and neonate ab-normity. So we must strengthen perinatal healthcare.
7.Grafting of iliac bone flap with deep iliac circumflex vessel in treatment of femoral neck fracture

Objective To retrospect the results and probe into the clinical applied vallue of the pedicled bone flap with deep iliac circumflex artery to the treatment of the femoral neck fracture and femoral neck fracture Methods Eight cases of femoral neck fracture and 21 cases nonunion treated by bone and periost flaps with deep iliac circumflex vessel35 casses treated by closure reduction and fixed with 3 trianglar nails,34 cases by arthroplastyThe results of 3 kinds of methods were contrasted and anslysed Results Twenty-nine cases treated by iliac bone and periost flaps with deep iliac circumflex artery were fellowed from 3 to 7 years,the fracture healed 35 months averagely. One case was avascular necrosis of femoral head after 9 month after operation,others received good results35 cases were treated by 3 nails fixtion healed 74 months averagely and 6 case had complications 37 casese treated by arthroplasty had complications in 8 patients,4 cases need walking-stich for walk. Conclusion Femoral neck fracture were treated by transplantation of the pedicled bone and periost flaps with deep iliac circumflex vessel is keeping with the pathological and anatomical characteristicsThe function of the hip joint is not weaken as the time goes on when the fracture is healedIt is the best methods to treat the femoral neck fracture now,the arthroplasty has many complication,so the indication must be confined

9.Study on expression and correlation of Rac1 and E-cadherin protein in oral squamous cell carcinoma

Objective To investigate the expression levels of Rac1 and E-cadherin and their correlation in oral squamous cell carcinoma, and to explore their relations to clinical pathological parameters. Methods The expression levels of Rac1 and E-cadherin were detected in 22 samples of oral squamous cell carcinoma and 8 samples of para-carcinoma tissue by using immunohistochemistry technique (SP). The patients were divided into two groups according to the clinicopathological parameters. The differences of Rac1 and E-cadherin expression levels were analyzed between the groups. Results The positive expression rates of Rac1 were 1/8 and 77.3%(17/22) in para-carcinoma tissue and oral squamous cell carcinoma, respectively. The difference was statistically significant (P=0.003). The positive expression rates of E-cadherin were 8/8 and 31.8%(7/22), and there was a difference between them (P=0.002). The Rac1 protein expression rate was increased and E-cadherin was decreased in oral squamous cell carcinoma. There were no significant differences in Rac 1 and E-cadherin between different age and gender groups. There was significant difference in Rac1 expression between patients with metastasis and patients without metastasis (P=0.021). While there was no difference in E-cadherin expression between patients with metastasis and patients without metastasis. No correlation was found between Rac 1 and E-cadherin expressions in oral squamous cell carcinoma. Conclusion The high expression of Rac1 and low expression of E-cadherin may play an important role in the tumorigenesis and progression of oral squamous cell carcinoma.
