1.The Changes of Serum Estradiol and TNF-Alpha Levels in Endometriosis Patients with Pelvic Pain

Objective To investigate the changes of serum estradiol (E2) and tumor necrosis factor (TNF)-αin endome-triosis (EMS) patients with pelvic pain, and their correlation thereof. Methods The serum levels of E2 and TNF-α were mea-sured with ELISA in 113 EMS patients with pelvic pain ( 37 cases in mild pain group, 41 cases in moderate pain group and 35 cases in severe pain group) and 30 healthy women without EMS (control group). Results There were no significant differences in the serum levels of E2 and TNF-α between mild pain group and normal control group. There were significantly higher levels of E2 and TNF-αin moderate and severe pain groups than those of mild pain group and normal control group. The serum levels of E2 and TNF-αwere significantly higher in severe pain group compared with those of moderate pain group (P<0.05). There was no correlation between serum levels of E2 and TNF-αin mild pain group. The serum levels of E2 and TNF-αwere positive-ly correlated between moderate pain group and severe pain group (P<0.05). Conclusion The increased E2 level can play a role in chronic pelvic pain of EMS by inducing macrophage to release TNF-α.

3.Surgical cooperation of percutaneous nephrolithotomy under flexible ureteroscope in the treatment of staghorn calculi

ObjectiveTo summarize the experience of percutaneous nephrolithotomy under flexible ureteroscope in the treatment of staghorn calculi.Method The clinical data of 30 patieits with staghorn calculi undergoing percutaneous nephrolithotomy under flexible ureteroscope were reviewed for summarizing the surgical cooperation experience.Results All operations were successful,with the clearance rates of stage one and stage two calculi 86.0% and 95.0%,respectively.There occurred one case of massive hemorrhage,but no cases of kidney perforation,peritoneal perforation,liquid pneumothorax or organ damage.Conclusion Well-prepared surgical cooperation,full mastery of surgical procedures and skilled cooperation can increase surgical efficiency and safety by shortening surgical duration.
4.Synergistic effect ofRhBMP-2 and bFGF on ectopic osteogenesis in mice

Objective:To investigate the synergistic effect and mechanism of the combined application of recombinant human bone morphogenetic protein-2(rhBMP-2) and basic fibroblast growth factor (bFGF).Methods:24KM male mice were randomly divided into6 groups with4 mice in each group, namely,GroupA(control group),GroupB(only treated with collagen),GroupC(treated with 2 ng bFGF+collagen),GroupD(treated with4 μg rhBMP-2+collagen),GroupE(treated with4 μg rhBMP-2+2 ng bFGF+collagen) andGroupF(treated with4 μg rhBMP-2+4 ng bFGF+collagen). The composites were implanted into the intermuscular septum of hind legs mice; whereas in control group, intermuscular septum of mice was separated and no implantation was performed. General observation, detection of concentration of calcium content, micro computed tomography (Micro-CT), three-dimensional reconstruction scan, measurement of bone mineral density(BMD), bone volume fraction(BVF) and trabecular thickness(Tb.Th), as well as histological observation withHE staining andALP andCD34 immumohistochemical staining were performed.Results:Ectopic osteogenesis was found inGroupsD,E andF mice.The difference in concentration of calcium contentswas statistically significant betweenGroupsD andE(P<0.05), but insignificant betweenGroupsE andF(P>0.05).Micro-CT and three-dimensional reconstruction revealed continuous newborn bone substance in external surface of ectopic bone formation, and the center of bone formation did not show obvious substantial filling by bone substance.The differences in BMD,BVF andTb.Th were statistically significant betweenGroupsD andE orF(P<0.01 or <0.05). HE staining showed that inGroupsD,E andF, newborn bone substance was mainly located at the edge of ectopic bone formation, and the bone formation inGroupsE andF was better than that in GroupD.ALP andCD34 immumohistochemical staining revealed the positive expression mainly at the edge of ectopic bone formation, and area of positiveexpression inGroupsE andF was larger than that inGroupsD.Conclusions:rhBMP-2 possesses the capacity to induce ectopic osteogenesis independently, but bFGF does not have this ability; the combined application of rhBMP-2 and bFGF can enhance the synergetic effect on inducing ectopic osteogenesis.
5.Seroepidemiological investigation of measles among residents in Quzhou City

Objective:
To investigate the measles antibody level among residents in Quzhou City, Zhejiang Province in 2018, so as to provide the evidence for improving the measles control strategy.
Methods:
The permanent residents aged 0 to 59 years were randomly sampled from 10 townships ( streets ) in Kecheng District and Changshan County of Quzhou City. Residents' demographics and vaccination of measles-containing vaccine ( MCV ) were collected, and serum anti- measles IgG antibody was detected using enzyme-linked immunosorbent assay ( ELISA ). The positive rate, protective rate and geometric mean concentration (GMC) of anti-measles antibody were estimated.
Results:
A total of 606 residents were tested, with a male to female ratio of 0.83∶1. The subjects had a median age ( interquartile range ) of 17.36 ( 29.07 ) years, and 399 residents ( 65.84% ) had a vaccination history of MCV. The positive rate, protective rate and GMC of anti-measles IgG antibody were 94.88%, 48.68%, and 784.51 ( 95%CI: 731.14-841.40) mIU/mL, respectively. The positive rate of anti-measles IgG antibody was higher in men than in women ( 97.08% vs. 93.07%, χ2=4.968, P=0.026 ), and the protection rate was lower in men than in women ( 44.16% vs. 52.41%, χ2=4.089, P=0.043 ). The protective rate and GMC of anti-measles IgG antibody showed a“U”-shaped distribution with age, and a low protective rate was seen in residents aged 10 to 39 years ( 23.53% to 46.67% ), which the GMC of anti-measles IgG antibody that did not reach the protective level. A total of 233 residents at age of 15 years and below had with a history of two-dose MCV vaccination, and the positive rate ( χ2trend=7.260, P=0.007 ), protective rate ( χ2trend=12.756, P<0.001 ) and GMC ( rs=-0.289, P<0.001 ) of anti-measles IgG antibody presented a tendency towards a reduction with time <1 to 11 years after vaccination of the last dose of MCV.
Conclusions
A high positive rate of anti-measles antibody was detected among residents in Quzhou City in 2018; however, the protection rate of anti-measles antibody was low among residents at ages of 10 to 39 years. The coverage of MCV vaccination is recommended to be improved among residents at ages of 10 to 39 years in Quzhou City.
6.Sulforaphane protects PC12 cells against oxidative toxicity

Objective To investigate the effect of sulforaphane on 1-methyl-4-phenylpyridinium (MPP +)-induced cell viability loss in cultured PC12 cells and to explore the possible mechanism.Methods MPP + induced damage in PC12 cells was prepared as oxidative damage model.Sulforaphane (0.5,1.O,2.5,5.0 and 10.0 μmol/L) was added in each group cell growth medium.Subsequent experiments were divided into 4 groups:(A) normal control group,(B) sulforaphane group,(C) MPP+ injury group,(D)sulforaphane pretreatment + MPP+ injury group.Cell viability was detected by MTT assay,and the sulforaphane pretreatment PC12 cell viability was observed in different concentrations.Flow cytometry was used to detect changes in the rate of apoptosis in different packet PC12 cells,and protein expression levels of nuclear factor erythroid2-related factor 2 (Nrf2),heme oxygenase (HO-1) and human NAD (P) H dehydrogenase,quinone 1 (NQO1) were detected by Western blot when the PC12 cells were incubated with sulforaphane (2.5 μmol/L) and (or) MPP+ (500 μmol/L) for 24 h in vitro.Results Compared to control group (cell survival rate was 98.70%),the survival percents of PC12 cells were significantly decreased in MPP+-treated group (58.16%).A significant difference was showed between group A and C (F =21.83,P < 0.05),and the cell survival rate in group D was significantly improved.Compared to control group,the rate of apoptosis in MPP+ injury group was increased,and the rate of apoptosis after pretreatment of the sulforaphane was significantly reduced.Compared to MPP+ injury group,the levels of Nrf2,HO-1 and NQO1 protein expression were significantly increased in sulforaphane pretreatment group.Conclusion Sulforaphane have a protective effect against MPP+-induced PC12 cell model damage,and the protective effect may be achieved by activating the Nrf2-antioxidant response element pathway.
7.Relationship between blood pressure variability and left ventricular diastolic function in patients with ;essential hypertension

Objective:To observe the relationship between dynamic blood pressure variability (BPV) and left ventric‐ular diastolic function in patients with essential hypertension (EH) .Methods :A total of 140 newly diagnosed EH pa‐tients were selected .According to total coefficient of variation (CV) of 24h systolic blood pressure BPV ,they were divided into high CV group (n=70 ,CV>12.16% ) and low CV group (n=70 ,CV≤12.16% ) .Another 70 healthy subjects with corresponding gender and age were enrolled as healthy control group simultaneously .Echocardiography was used to measure mitral early diastolic peak flow velocity (E) ,late diastolic peak flow velocity (A) and decelera‐tion time of E peak (DT);meanwhile ,tissue Doppler mode was used to record early diastolic peak velocity (Em) and late diastolic peak velocity (Am) .All above indexes were compared among all groups .Results:Compared with healthy control group ,BPV significantly rose in EH patients (P<0.05 or < 0.01) .All BPV indexes in high CV group were significantly higher than those of low CV group except daytime mean diastolic blood pressure variability (dDBP‐BPV) and nighttime mean diastolic blood pressure variability (nDBP‐BPV) , P<0.05 all .Compared with healthy control group ,there were significant reductions in E and Em/Am ,and significant rise in DT and E/Em in EH patients ,P<0.05 or <0.01 ;compared with low CV group ,there was significant rise in E/Em [(10.32 ± 3.20) vs .(14.22 ± 2.20) ,P=0.033] in high CV group .Conclusion:Left ventricular diastolic dysfunction is more severe in hypertensive patients with higher dynamic blood pressure variability .
8.Volume changes of whole brain gray matter in pediatric patients with Tourette syndrome: evidence from voxel-based morphometry

ObjectivesTo identify the related abnormalities of gray matter in pediatric patients with Tourette syndrome (TS) by using the optimized voxel-based morphometry (VBM).Methods Three dimensional T1WI was acquired in 31 TS children (28 boys,3 girts,mean age 8 years,range 4-15 years) and 50 age- and sex-matched controls on a 1.5 Tesla Philips scanner. Images were pre-processed and analyzed using a version of VBM 2 in SPM 2.The whole brain gray matter volume was compared between the study and control group by using t-test.Multivariate linear regression analysis was used for analyzing the correlation between the change of grey matter volume within each brain region (mm3 ) and YGTSS score and course of disease of TS patients.Statistical analyses were performed by using SPSS 13.0.ResultsUsing VBM,significant increases in gray matter volumes in left superior parietal lobule, right cerebellar hemisphere and left parahippocampal gyrus were detected in TS patients,and the volume changes were 4059,2126 and 84 mm3 ( t =3.93,3.71,3.58,P < 0.05 ) respectively.Compared to the control group,decreased grey matter volumes were found in medulla and left pons,and the volume changes were 213 and 117 mm3( t =3.53,3.48,P < 0.05 )respectively.Tic severity was not correlated with any volume changes of gray matter in brain (P > 0.05,a small volume correction,KE ≥ 10 voxel).Tic course was negatively correlated with the gray matter volume of left parahippocampal gyrus ( Beta =- 0.391,P =0.039 ).ConclusionsUsing VBM technique,the gray matter abnormalities can be revealed in TS patients without obvious lesions on conventional MR imaging.The increasing volume of temporal and parietal lobes and cerebellar may be an adaptive anatomical change in response to experiential demand. The gray matter volume of the parahippocampal gyrus may be used as one potential objective index for evaluating the prognosis of TS.
9.The influence between managements in emergency room and outcome of severe traumatic brain injury

Objective To assess the influence between managements in emergency room(ER) andoutcome of severe traumatic brain injury (TBI),in order to provide inference for treatment.Methods A retrospective analysis was performed in severe TBI patients and recorded next indexes.(1) The managements in ER,including endotracheal intubation and oxygenation,fluid resuscitation,and mannitol intake.(2) The vital signs arriving at ICU,including systolic pressure and blood oxygen saturation.(3) Prognostic indicators including inhospital mortality and days during ICU,the scores of Glasgow outcome scale (GOS) at discharge and 6 months after injury.Results In 140 severe TBI patients,65 patients (46.4%) died during ICU.The mortality of patients with endotracheal intubation [65.0% (39/60)] was significantly higher than that without endotracheal intubation [32.5%(26/80)](P< 0.01).The mortality in whether fluid resuscitation and using mannitol had no significant difference [44.7% (46/103) vs.51.4% (19/37),49.2% (31/63) vs.44.2% (34/77)] (P >0.05).In days during ICU,there was no significant difference among the three treatment measures (P> 0.05).In GOS grade at discharge and 6 months after injury,the proportion of 4 and 5 grade were 8.3% (5/60) and 25.0% (15/60) in patients with endotracheal intubation,while 27.5% (22/80) and 52.5% (42/80) in patients without endotraeheal intubation (P < 0.01).In fluid resuscitation and using mannitol patients,there were no significant difference(P > 0.05).Conclusion Treating severe TBI patients in ER,endotracheal intubation should be carefully chosen,fluid resuscitation and mannitol may not be given.
10.Quantification of fat content of lower limbs using iterative decomposition of water and fat with asymmetry and least squares estimation?quantitative fat imaging in Duchenne muscular dystrophy

Objective To evaluate the fatty infiltration of lower limbs (pelvis, thighs and calf) by using iterative decomposition of water and fat with asymmetry and least squares estimation‐quantitative fat imaging (IDEAL‐IQ) in Duchenne muscular dystrophy (DMD) patients, and correlate muscle fat fraction(FF) with clinical assessments. Methods The prospective study method was applied, twenty‐one patients with DMD were diagnosed by genetic analysis and/or muscle biopsy and twenty‐one age‐matched healthy control subjects, were evaluated using IDEAL‐IQ, calculating fat fraction (FF) of muscles in the lower limbs. The clinical data of patients, such as age, height, weight, course of disease, time to walk 10 m, Medical Research Council Scale score, were collected. The age and height between DMD group and control group were compared by independent sample t test. The weight, BMI and FF values between two groups were compared by independent samples nonparametric tests (Mann‐Whitney U tests). K related samples nonparametric test (Friedman test) was used for comparison of FF values between pelvic muscles, thighs muscles and calf muscles. The relationships between FF values and clinical assessments were investigated using Spearman correlation test. Results The mean FF of pelvic muscles [13.1(6.9, 33.8)] was significantly higher than that of the thighs muscles [8.3 (23.4, 4.5)] and calf muscles [8.3 (23.4, 4.5)] (all P<0.05) in DMD group. The FF of all the pelvic muscles in DMD were significantly higher than that of the control group muscles (all P<0.05). The gluteus maximus had the greatest mean FF values [34.7 (26.4, 63.7)] in pelvic muscles. The FF of all the thigh muscles in DMD were significantly higher than that of the control group muscles, except for the sartorius, adductor longus and gracilis (Z=0.69, 1.67 and 1.22, respectively, P=0.489, 0.094 and 0.222, respectively). The adductor magnus had the greatest mean FF values [27.2 (12.4, 58.5)] in thigh muscles. The FF of all the calf muscles in DMD were significantly higher than that of the control group muscles, except for the tibialis anterior and tibialis posterior (Z value was1.77 and 1.85, respectively, P value was 0.076 and 0.064, respectively). The gastrocnemius lateral had the greatest mean FF values [7.4 (4.6, 20.9)] in calf muscles. The mean FF value of pelvic muscles (r=0.679, 0.704,?0.761, 0.755, respectively, all P<0.001), thighs muscles (r=0.801, 0.813,?0.738, 0.706, respectively, all P<0.001) and calf muscles (r=0.502, 0.682,?0.637, 0.664, respectively, all P<0.05) were significantly correlate with age, course of disease, %MRC and time to walk 10 m. Conclusions The fat infiltration of lower limbs in DMD patients showes a distinct involvement pattern. IDEAL‐IQ technique is able to quantitatively and objectively evaluate the degree of fat infiltration of lower limbs in DMD, and it has great value in clinical application.
