1.Effects of recombinant human bone morphogenetic protein 2 on integrin beta 1 expression during mandibular distraction

BACKGROUND:Integfin levels can affect cellular function,regulate bone formation and bone resorption.However,this process need activated by other factors,whether recombinant human bone morphogenetic protein 2 (rhBMP-2)has this effect is poorly understood.OBJECTIVE:To study the effects of the different concentration of rhBMP-2 on the expression of inergrin β_1 during the mandibular dfScraction in rabbits.DESIGN,TIME AND SETTING:The randomized,controlled.animal grouping experiment of histological observation was performed at the Animal ExperimentaI Center and Immunohistochemistry Center of Liaoning Medical University from May 2006 to May 2007.MATERIALS:A total of 45 Japanese white rabbits were used.rhBMP-2 was supplied by Beijing Academy of Military Medical Sciences.METHODS:The rabbits were prepared for mandibular distraction models and randomly assigned into control,1.5 mg rhBMP-2 and 3.0 mg rhBMP-2 groups.with 15 animals in each group.Collagen sponge and collagen binding proteins were implanted into mandibles of rabbits,respectively,and distraction 0.4 mm per time,twice per day for 5 days.The mandibles were lengthened 4 mm.MAIN OUTCOME MEASURES:The expression of integrin 131 was detected using immunohistochemistry at days 1,3,7,14,and 28 after distraction.RESULTS:All rabbits were included in the final analysis.The integdn β_1 mainly expressed at 7 days after distraction.At the same time point.when concenrtation rhBMP-2 was increased,the positive integrin β_1 rate and positive stained area percentage were increased(P<0.05).Compared to the same concentration,the positive integrin β_1 rate and positive stained area percentage were gradually increased as time prolonged (P<0.05).CONCLUSION:The integdn β_1 expression can be promoted by rhBMP-2 at the anaphase of mandibular distraction osteogenesis in a concentration-dependent manner.
2.Abnormalities of sex chromosome in patients with mental disorders

Subject To explore the features of the abnormalities of the sex chromatin and the sex chromosome of the patients with mental disorders. Methods The sex chromatin in the cells of buccal mucous membrane of the 2 784 patients with mental disorders were examined by using a blinding and random way. The chromosome karyotypes in peripheral blood were further examined in those whose measure showed abnormal. The comparison group consisted of 1 069 normal persons.Results Compaired with the control group,the average frequency of Y chromatin in the male patients [(28.0±6.6)%] and the average frequency of X chromatin in the female patients [(22.3±8.0)%] rose significantly ; the average frequency of X chromatin in the female patients of the schizophrenia subgroup and the affective disorder subgroup were higher than that of neurosis and psychogenic mental disorder subgroup. X chromatins of eight male patients were positive and the karyotypes of the abnormal X chromosome were 47,XXY in 3 cases and 47,XXY/46,XY in 5 cases, the rate of 47,XXY was 7.7 , it rose significantly to the control group. X chromatin of two female patients were negative and the karyotypes of the abnormal X chromosome was 45,X/46,XX, the rate of 45,X was 1.2 ; one female patient had double X chromatin and the karyotypes of the abnormal X chromosome was 47,XXX/46,XX,there is no abnormal sex chromatin and sex chromosome in the control group.Conclusions The higher rate of the sex chromatin might be closely associated with the tendency of heredity of mental disorders , and the rate of the lacking X chromosome in females and the increasing X chromosome in males rise in the patients with mental disorders.
3.STRUCTURE ELUCIDATION OF GLYCAN OF A GLYCOCONJUGATE SPPA-1 ISOLATED FROM SPIRULINA PLATENSIS

AIM To elucidate the structure of the glycan of SPPA-1, a glycoconjugate isolated from Spirulina platensis. METHODS Methylation analysis, GC/MS, and 1D, 2DNMR techniques were used to determine the structures of the glycoconjugate (SPPA-1). RESULTS SPPA-1 was only composed of α-D-glucose and shown to be a (1→4) linked α-D-glucan to which a few glucosyl side chains are attached at O-6 of the glucosyl residues of the main chain. CONCLUSION The glycan of SPPA-1 is a new glucan.
4.Isolation,purification,physico-chemical properties and biological activities of a polysaccharide SPPC-1 from Spirulina Platensis

A polysaccharide SPPC 1 was isolated from spirulina platensis by acetone fractional precipation, free protein was removed by trichloroacetic acid(TCA). Purification of the polysaccharide by Sephadex G 75 colume and CM Sephadex C 50 colume.The homogeneity of this polysaccharide was proved by HPLC and CE. molecular weight was 1.62?10 6. GC showed that the glycoconjugate was composed of Rha,Xyl,Man and Glc. Molar ratio is Rha:Xyl:Man:Glc=6.24:0.37:0.13:0.71. IR, 1HNMR spectrum indicated the presence of ? linkage glycoside. ESR experiment showed that SPPC 1 could eliminate O . 2 radicals.
5.Expression of JNK/SAPK in lung and bronchus of radon-exposed mice

Objective To study the expression of JNK/SAPK(c-Jun NH2-terminal kinase/stress activated protein kinase)in lung and bronchus of radon-exposed mice.Methods Male BALB/c mice were exposed to radon and its progeny with the cumulative dose of 0.02,30 or 60 working level month(WLM),respectively.The expression levels of JNK/SAPK in lung and bronchus were determined with Real-Time PCR,Western blot and immunohistochemistry methods.Results The JNK mRNA levels in lung tissues of mice exposed to radon of 30 and 60 WLM were higher than those of the control by 3.56 and 2.96 times,respectively.The relative expression levels of JNK and phospho-JNK proteins were higher than those of the control by using Western blot and immunohistochemistry methods.Condusiom Expose to the radon and its progeny might activate the JNK/SAPK intracellular signaling pathway.
6.Radon induced pulmonary lesion and expression of P53 and Bcl-2,Bax in mice

Objective To establish the mice model of lung injury induced by radon,and to observe the changes of pulmonary lesion at different doses and to analyze the influence of radon exposure on P53 and Bcl-2、Bax expression in lung tissue.Methods Mice were exposed to radon of 30 and 60 WLM,respectively.Apoptosis was detected by terrainal deoxynucleotidy transferse-mediated dUTP-biotin nick end labeling(TUNEL).The expressions of P53,Bcl-2 and Bax protein were observed by immunohistochemistry and Western blot.Results Compared with those in the control group,the apoptotic indexes increased significantly in the 30 and 60 WLM groups(t=18.11,-10.30,P<0.05).The protein expression of P53 was significantly increased(t=-11.08,P<0.05).The expression levels of P53 and Bax were remarkably inereased(t=-7.00,-2.52,P<0.05),while the expression of Bcl-2 protein was significantly decreased in the 60 WLM group(t=4.36,P<0.05).But Bcl-2,Bax expression was decreased in both 30 WLM and 60 WLM groups(t=2.78,4.07,P<0.05).Conclusions Radon could induce pulmonary lesion of mice.It may be involved in the regulation of apoptosis of pulmonary lesion by the P53,Bcl-2、Bax pathway.
7.An Experimental Study on the Early Changes in ~(18)F-FLT and ~(18)F-FDG Uptake of Tumor Cells after Chemotherapy

Purpose:The efficacy of evaluation of changes of tumoral uptake of 3'-deoxy-3'-[~(18)F] fluorothymidine (FLT) was comparatively analyzed with that of ~(18)F-FDG at early stage after anticancer chemotherapy.Materials and Methods:Cells derived from human lung adenocarcinoma were incubated with cisplatin (CDDP),5-fluorouracil(5-FU),doxorubicin (Dox),for 1,4,24 and 72h.The doses(CD-DP: 67 μM; 5-FU 1,540 μM;MTX: 440 μM;) were determined corresponding to a estimated 10% - 95% proliferation inhibition.The cells were allowed to recover before FLT or FDG being added into the culture media for 60 min.Cell counts,viability,estimated by MTT method,were used to evaluate the cytotoxic effects of chemotherapy.Results: FLT uptake was increased significantly at 1 and 4 h after treatment with 5-FU( 145 ± 12%,150 ± 14%,P <0.01).decreased at 24 h and 72 h.In contrast,FLT accumulation was significantly reduced at cytostatic concentrations of CDDP at different time.The uptake of FDG did not change significantly at early time points after treatment,but decreased at 72 h.Conclusion: The tumor cell uptake of FLT revealed specific changes depending on the auti-cancer drug used at much earlier time than FDG after chemotherpay.
9. A case of maternal 45, X/46, XX mosaicism detected by non-invasive prenatal testing

Objective:
To report on a case of maternally derived 45, X mosaicism detected by non-invasive prenatal testing (NIPT).
Methods:
Fetal sex chromosomal abnormality was detected by NIPT. Maternally derived 45, X mosaicism was confirmed by chromosome karyotype analysis. Fetal sex chromosome aneuploidy was detected by amniotic fluid chromosome microarray analysis.
Results:
A maternal 45, X mosaicism was diagnosed. The fetus was confirmed to be normal.
Conclusion
Maternal 45, X masaicism can be diagnosed by NIPT.
10. Perianal swabs surveillance cultures of Carbapenem-resistant Enterobacteriaceae(CRE) can be hints for CRE bloodstream infection in patients with hematological diseases

Objective:
To analyze the hints role of surveillance cultures of Carbapenem-resistant Enterobacteriaceae (CRE) by perianal swabs in patients with hematological diseases, and seek risk factors of CRE bloodstream infection.
Methods:
The resistance of CRE from 2 914 patients with hematological diseases who cultured perianal swabs, CRE bloodstream infection and risk factors were analyzed during January 2016 to December 2017.
Results:
In this study, perianal swabs from 2 914 patients with hematological diseases were cultured, 74 patients were CRE positive, and bloodstream infection with CRE was found in 13 of these patients.
