1.Inhibitory effect of ultrasound-mediated drug loaded carboxymethyl hexanoyl chitosan nanodroplet on ovarian cancer cells

Objective To evaluate the inhibitory effect of phase change doxorubicin loaded carboxymethyl hexanoyl chitosan nanodroplet on ovarian cancer cells,and the effect of its ultrasound image in vitro.Methods The carboxymethyl hexanoyl chitosan synthesized through the acylationreaction with carboxymethyl chitosan and hexanoic anhydride.The drug loaded carboxymethyl hexanoyl chitosan nanodroplets were prepared by ultrasonic emulsification.The surface morphology,particle diameter and electric potential were characterized.Ultrasound imaging of the nanodroplet was evaluated in vitro.The encapsulation efficiency was determined by ultraviolet spectrophotometry.The survival rate of ovarian cancer cell was detected using CCK-8 reagent.The statistical analysis was performed.Results The drug loaded carboxymethyl hexanoyl chitosan nanodroplet was successfully prepared which showed regular morphology in microscope,the mean diameter of (458.33± 43.50)nm.The encapsulation efficiency was (52.06 ± 10.14)%.The nanodroplet could enhance ultrasonic imaging.The survival rate of ultrasound combined with drug loaded nanodroplet group ([62.54± 3.60]%) was lower than those of the free drug group ([75.55±7.21]%) and drug loaded nanodroplet group ([76.18±4.94]%),ultrasound group ([89.90±0.83]%;P<0.05).Conclusion Ultrasound-mediated drug loaded nanodroplet can inhibit ovarian cancer cells,and has the potential for application in the clinical diagnosis and treatment.
2.Diagnostic efficacy of lung ultrasound and X-ray for neonatal respiratory distress syndrome and transient tachypnea

Objective To compare the efficacy of lung ultrasound and chest X-ray for diagnosis of neonatal transient respiration syndrome (TTN) and respiratory distress syndrome (RDS).Methods Totally 120 infants with respiratory distress underwent lung ultrasound and chest X-ray within 6 h after birth.Taking clinical diagnosis as the standard,the sensitivity,specificity and accuracy of lung ultrasound and X-ray for diagnosis of TTN and RDS were calculated.Results According to clinical diagnosis,there were 75 infants of TTN and 34 of RDS.The sensitivity,specificity and accuracy of lung ultrasound in diagnosis of TTN was 96.00% (72/75),88.89% (40/45) and 93.33% (112/120),respectively,higher than those of chest X-ray (85.33% [64/75],84.44% [38/45] and 85.00% [102/120]).Lung ultrasound showed 85.29% (29/34) of sensitivity,95.35% (82/86) of specificity and 92.50% (111/120) of accuracy for diagnosis of RDS,whereas chest X-ray showed 88.23% (30/34) of sensitivity,89.53% (77/86) of specificity and 89.17% (107/120) of accuracy.Conclusion Lung ultrasound is accurate for the diagnosis of TTN and RDS.
3.The effect of ultrasound monitoring of inferior vena cava collapse index guiding fluid replacement on circulation in elderly patients during induction of general anesthesia

Objective:To investigate the effect of ultrasound monitoring of inferior vena cava collapse index (IVC-CI) guiding fluid replacement on circulation in elderly patients during induction of general anesthesia.Methods:A total of 71 elderly patients who underwent elective surgery under general anesthesia and tracheal intubation at Hunan Provincial People′s Hospital from April 2021 to September 2022 were randomly divided into control group (35 cases) and observation group (36 cases) using a random number table method. Before anesthesia, both groups of patients underwent IVC ultrasound examination and calculated the IVC-CI value. For patients with IVC-CI≥40%, the observation group was given 8 ml/kg of crystal solution before anesthesia induction, while the control group was not treated. The incidence of hypotension, the use of vasoactive drugs, and the total infusion volume from anesthesia induction to skin incision were recorded in two groups. Mean arterial blood pressure (MBP), heart rate (HR), oxygen saturation (SpO 2), cardiac index (CI), and cardiac volume variability (SVV) before anesthesia (T 0), 5 min after induction (T 1), 1 min after tracheal intubation (T 2), 5 min after tracheal intubation (T 3), 10 min after tracheal intubation (T 4), and 1 min before skin incision (T 5) were recorded and compared between the two groups. Results:The incidence of hypotension (27.8% vs 60.0%) and utilization rate of vasoactive drugs (25.0% vs 48.6%) in the observation group were lower than those in the control group, and the total infusion volume during anesthesia induction was higher than that in the control group, with statistical significance (all P<0.05). SVV, CI and MBP at T 1, T 3, T 4 and T 5 were significantly different from those at T 0 in the control group ( F=3.85, 14.66, 3.96, all P<0.05). SVV, CI and MBP at T 1, T 3, T 4 and T 5 in the observation group were significantly different from those at T 0 ( F=3.51, 13.20, 4.35, all P<0.05). There was no significant difference in SVV, CI, MBP, HR and SpO 2 between 2 groups (all P>0.05). Conclusions:For the elderly patients with preoperative IVC-CI≥40%, pre-filling with 8ml/kg crystal solution before anesthesia induction can significantly reduce the incidence of hypotension and the utilization rate of vasoactive drugs in the elderly patients during anesthesia induction.
4.Inhibitory Effect of Gancao Fuzitang on Bone Destruction in Collagen-induced Arthritis Mice by Regulating NF-κB Signaling Pathway

ObjectiveTo explore the mechanism of Gancao Fuzitang (GCFZ)in inhibiting the bone destruction of collagen-induced arthritis (CIA) model in mice. MethodThirty male DBa/1J mice were randomly divided into normal group, CIA group, low-dose GCFZ group (GCFZ-L, 2.4 g·kg-1), high-dose GCFZ group (GCFZ-H, 4.8 g·kg-1), and methotrexate group (MTX, 1 mg·kg-1), with six mice in each group. The CIA model was induced by secondary immunization method. The arthritis index of mice in each group was observed and recorded, and the histopathological changes in ankle joint were observed by hematoxylin-eosin (HE) staining. The damage to ankle cartilage was detected by safranin O-fast green staining. Micro-CT scanning was used to detect the bone destruction of ankle joint, and the expression of nuclear factor-κB p65 (NF-κB p65), p-NF-κB p65, inhibitory-κB kinase α/β (IKKα/β), and p-IKKα/β was observed by immunohistochemical staining. ResultCompared with the normal group, the CIA group showed manifest joint swelling and increased arthritis index score (P<0.01). Compared with the CIA group, the groups with drug intervention could inhibit joint swelling and reduce arthritis index score (P<0.05, P<0.01). As revealed by HE staining and safranine O-green staining, compared with the CIA group, the groups with drug intervention could inhibit synovial invasion and reduce the destruction of articular cartilage. Micro-CT scanning analysis showed that compared with the CIA group, the GCFZ-H group and the MTX group showed reduced bone destruction scores (P<0.01). The immunohistochemical results showed that compared with the normal group, the CIA group showed increased optical density values of NF-κB p65, p-NF-κB p65, IKKα/β, and p-IKKα/β(P<0.01). Compared with the CIA group, the GCFZ-H group and the MTX group showed reduced optical density values of NF-κB p65, p-NF-κB p65, IKKα/β, and p-IKKα/β(P<0.05,P<0.01). In the GCFZ-L group, only the NF-κB p65 optical density value decreased(P<0.01). ConclusionGCFZ may inhibit bone destruction in CIA mice by regulating the NF-κB signaling pathway.
