1.Clinical Observation on Internal-External Treatment of Kidney-tonifying Herbal Medicine for Chronic Aplastic Anemia

[Objective] To observe the therapeutic effect of kidney-tonifying herbal medicine for chronic aplastic anemia (CAA) and its effect in regulating immune function. [Methods] Twenty-nine cases of CAA were randomized into 2 groups: group A (n = 15) was treated with routine western medicine plus Huosui Tablets and kidney-tonifying decoction for oral use, and Huosui Ointment for external application; group B (n = 14) was treated with routine western medicine only. Three months constituted one treatment course and the two groups were treated for over 2 courses. After treatment, the therapeutic effect was assessed and the changes of peripheral blood counting and immune function indexes were compared with those before treatment. [Results] After treatment, the total effective rate was 80.0% in group A and 42.9% in group B, the difference being significant (P 0.05) . [Conclusion] Kidney-tonifying herbal medicine is effective for the treatment of CAA and its therapeutic effect may be related to the regulation of immune function.
2.Zhongshan nasopharyngeal carcinoma (NPC) mortality in 1970-2010

Background and purpose:Zhongshan City is one of the areas with the highest NPC incidence worldwide and nationwide. Although its incidence rate had been analyzed thoroughly, but its mortality rate was not studied further until now. This study was to explore NPC mortality status in Zhongshan City from 1970 to 2010, and to provide scientiifc information for its control and prevention.Methods:NPC death data in Zhongshan City from 1970 to 2010 came from Zhongshan Cancer Registry Ofifce. Such indices as its death number, crude rate, age standardized rate (ASR) and truncated rate etc were calculated and analyzed. Results:There were 5 267 NPC death cases in Zhongshan City from 1970 to 2010, its crude world ASR and truncated rates were 11.03/105, 12.34/105 and 29.31/105 respectively. Its world ASRs increased before 1999 but no statistical signiifcance (P=0.64) and decreased thereafter with obvious statistical signiifcance (P=0.001). But overall, there were a decreasing trend for its ASRs (P=0.001) from 1970 to 2010 in Zhongshan City. Moreover, NPC mortality rate in male was remarkably higher than in female, the death peak age was 55-59 age group, and the different age groups were with different trends fro 1970 to 2010. Conclusion:Although with declining trend from 1970 to 2010, NPC mortality rate in Zhongshan City was at high level worldwide. It suggested that NPC control and prevention should be enhanced.
3.Determination of Prim-O-glucosylcimifugin in Biyan Tablets by HPLC

Objective To establish the determination of prim-O-glucosylcimifugin in Biyan Tablets by HPLC.Methods The HPLC determination was carried out on Diamonsil C18 column(4.6 mm? 250 mm,5 ? m).The mobile phase consisted of methanol-acetonirile-H2O(18 :12 :70) with a flow rate of 1.0 mL/min,and the ultraviolet detection wavelength was set at 254 nm.Results Prim-O-glucosylcimifugin showed a good linearity in the range of 0.1888~ 1.534 ? g with the peak area score,the coefficient was 0.9997,and the average recovery was 101.74 %(RSD=2.97 %).Conclusion The method is accurate and reliable,and can be used for the quality control of Biyan Tablets.
4.A New Phenylated Flavone from Melicope pteleifolia

Objective To study the constituents in Melicope pteleifolia. Methods Plant material was isolated with 80% EtOH. Compounds were separated with chromatographic methods and their structures were elucidated on the basis of spectral analysis (EI-MS, 1H-NMR, and 13C-NMR) and chemical evidence. Results Five compounds were isolated from petrol ether or ethyl acetate soluble fraction. Their structures were identified as 3,5,3'-trihydroxy-4'-methoxy-7-(3-methylbut-2-enyloxy) flavone (pteleifolosin C, 1), 3,7-dimethoxyl kaempferol (kamatakenin, 2), vanillic acid (3), tricosanoic acid tetradecyl ester (4), and p-sitosterol (5), respectively. Conclusion Compound 1 is a new structure named pteleifolosin C. Compounds 2-4 are isolated from this plant for the first time.
5.Determination of Lasiokaurin in the Stems and Leaves of Rabdosia nervosa by HPLC

Objective To develop a HPLC method for the determination of Lasiokaurin in the stems and leaves of Rabdosia nervosa.Methods HPLC was performed on a Kromsil C18 reversed-phase column(250mm?4.2mm,5?m).All of the reference substances and samples were separated with the mobile phase of methanol :water(55:45) under isocratic elution for 30min,flow rate was 1.0ml/min,the detection wavelength was 234nm,and the column temperature was 30℃.Results The content of Lasiokaurin was 0.005mg/g in the stems and 0.372mg/g in the leaves,the content in the stems were almost 1/75 times as much as that in the leaves.The average recovery of Lasiokaurin was 96.21%.Conclusion This developed method,which is simple and with good precision,high sensitivity and selectivity,can be used for quality evaluation of Lasiokaurin in Rabdosia nervosa.
6.Determination of Icariin Content in Qianlie Shule Granule by HPLC

Objective To determine the icariin content in Qianlie Shule Granules (QSG). Methods After proper pre-treatment,HPLC method was performed on a C18 column. The mobile phase consisted of acetonitrile-water (29 ∶71) and the UV d etective wavelength was set at 270 nm. Results The peak of icariin was separated completely. The calibration curve of icariin was linear within 0.16~1.2 mg?mL -1,Y=0.028 7X-0.009 62 (r=0.999 9). Conclusion This method is accurate and su itable for the determination of icariin content.

Objective To study the chemical constituents of Evodia lepta.Methods The chemical constituents were isolated by chromatographic methods and their structures were elucidated by physicochemical characteristics and spectral data.Results Two compounds were isolated and their structures were identified as erythro-3-(1',2',3'-trihydroxy) isopentyl-7-hydroxycoumarin(Ⅰ) and?-daucosterol (Ⅱ).Conclusion CompoundⅠis a new one named evodosin A while compoundⅡis isolated from E. lepta for the first time.
9.Establishment of differential expression profiles from invasive and non-invasive pituitary adenomas

Objective To establish high resolution, reproducible 2-dimensional electrophoresis (2-DE) profiles of invasive and non-invasive pituitary adenoma tissues and to identify differentially ex-pressed proteins between the invasive and non-invasive tissues. Methods The proteome from invasive and non-invasive pituitary adenomas tissues was dissected and analyzed by: (1) immobilized pH gradient two-dimensional polyacrylamide gel electrophoresis, (2) silver staining, (3) imageMaster 2-D software analysis, (4) peptide mass fingerprint based (PMS) on matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS), and (5) database comparison. Results High-resolution 2-D patterns of invasive and non-invasive pituitary adenoma tissues were successfully produced and re-peated 3 times for each sample. An average of 1080±24 and 1035±28 spots were detected for invasive and non-invasive pituitary adenoma tissues, respectively. Additionally, 975±45 and 918±56 spots were found to have an average matching rate of 90.3% and 88.7% for invasive and non-invasive tissues, re-spectively. The spot positional deviation was (1.563±0.259) mm for IEF and (1.088±0.206) mm for SDS-PAGE. A total of 99 spots of differential expression were matched between the invasive and non-in-vasive pituitary adenoma tissues. Thirty differential proteins, some of which were involved in the regula-tion of cells cycle and signal transduction, were initially characterized by PMS. Conclusion The acquisi-tion of well-resolved and reproducible 2-D patterns of invasive and non-invasive pituitary adenoma tissues and the identification of differentially expressed proteins provides a proteome database for invasive pituita-ry adenomas.
10.Study on Chemical Constituents of Citrus mediea var. sarcodactylis from Sichuan Province (Ⅱ)

Objective To investigate the chemical constituents of Citrus medica var. sareodactylis from Sichuan province, and to provide evidence for the development and utilization and quality evaluation of the medicinal material.Methods Various chromatographic techniques were used to purify the components of this herb. Compounds were identi-fied by their physical characteristics and spectral feature. Results Seven compounds were isolated from Citrus medica var. sarcodactylis, and they were identified as sibiricol (Ⅵ), 7-methylesculetin (Ⅶ), bergapten (Ⅷ), sigmasteryl acetate(Ⅸ), 5-methoxyfurfural(Ⅹ), limonin(Ⅺ), daucosterol (Ⅻ). Conclusion Compound Ⅵ, Ⅶ and Ⅸ are isolated from plants of Rutaceae for the first time.
