1.Application evaluation of procalcitonin in diagnosis of neonatal infection

Procalcitonin (PCT) has been recognized a marker of infectious diseases for the past few years.The value of PCT is specific elevation in infectious diseases or inflammation caused by bacterial and it has an important role in the identification of bloodstream infections,bacterial and non-bacterial,rational usage of antibiotics and prediction disease prognosis.PCT is a high value diagnostic indicators.However,the PCT do not reflect a clear advantage in the diagnosis of neonatal infection.In this paper,the composition of the PCT,the source in the body of PCT,the metabolism of PCT,laboratory testing methods of PCT and its applications in neonatal infection were reviewed in order to gain a deeper understanding of the value of PCT in the neonatal diagnosis of infectious diseases.

5.Construction and identification of recombinant baculovirus of the novel CEA vaccine

Objective To design a novel efficient CEA vaccine of fission epitopes with the vehicle of HPV16L1,and construct the recombinant baculovirus in Bac-to-Bac baculovirus expression system.Methods The recombinant baculovirus expression vector pFastBac1-V was constructed.HPV16L1 and CEA-PADRE synthesis primers were inserted into bacmid in E.coli DH10Bac with the help of Tn7 transposition system.Then the recombinant baculovirus was got by infecting insect cell sf9.Results HPV16L1 and CEA-PADRE synthesis primers that were cloned to the baculovirus transfer vector pFastBac1 were assessed by restriction analysis and direct sequencing.The recombinant baculovirus of the novel vaccine had been constructed.After infected sf9,the correct fragment was amplified by PCR.Conclusion The baculovirus of CEA vaccine of fussion epitopes with the vehicle of HPV16L1 was constructed.
6.Application of diffusion weighted imaging in severity assessment and prognosis prediction of diffuse axonal injury

Objective To discuss the value of diffusion weighted imaging (DWI) in severity assessment and prognosis prediction of diffuse axonal injury (DAI). Methods A retrospective study was performed on the clinical imaging data and the follow-up results at six months after injury in 29 patients with DAI. The detection rate of DAI lesion by DWI and conventional MRI was compared by means of one-way ANOVA. The correlation between the number of lesion in different areas with GCS and GOS was analyzed with Spearman rank correlation test. Results (1)The average detected DAI lesions were 19.24±5.72 on DWI, 14.41 ±4.50 on FLAIR, 10.58±3.79 on T2WI and 4.83 ±2. 11 on TIWI, with the highest detection number of DAI lesion on DWI (P < 0, 05). (2) There was a significant correlation of the number of central lesions (in corpus callosum, basal ganglia and brain stem) with GCS and COS (P < 0. 05), but there was no correlation of total lesion number or periphery lesion with GCS and COS (P > 0.05). Conclusions DWI is a potentially useful imaging modality in detecting DAI lesion, when the number of central lesion on DWI can be served as an objective marker in severity assessment and prognosis prediction of DAI.
7.High Throughput Screening of Drug Multi Residues in Fish by Quadrupole-Time-of-Flight Mass Spectrometry

A high-throughput method for screening drug multi residues was developed by quadrupole-time-of-flight( Q-TOF) mass spectrometry together with QuEChERS sample preparation technique. It has been used for the determination of 57 drugs, such as tetracyclines ( TCs ) , sulfonamides ( SAs ) , quinolones ( QNs ) , triphenylmethanes (TPMs), estrogens (ETs), androgens (AGs) and glucocorticoids (GCs) in fish. The optimized pretreatment conditions were examined. The target compounds were extracted with acetonitrile under the condition of Na2 EDTA-Mcllvaine buffer, and the usage of clean-up reagents was 25 mg anhydrous magnesium sulfate, 12. 5 mg octadecylsilane and 6. 25 mg N-Propylethylenediamine sorbent for extracted solvent of each milliliter. The positive results acquired by this high-throughput method were confirmed by liquid chromatography-tandem mass spectrometry ( LC-MS/MS) . The detection limit for these 59 drugs was in the range of 0. 5-5. 3 μg/kg. The method is time-saving, convenient, effective and wide coverage. Its sensitivity can meet the requirement of the detection of drug residues in aquatic products.
8.Preadipocyte viability, proliferation, and apoptosis in young rats following dynamic mechanical force stimulation

BACKGROUND:Schwann cells transplantation can change the local micro-environment and help to repair the injured neural tissue, so getting a large number of highly purified and active Schwann cells is the key of the study. OBJECTIVE: To search for a simple and rapid method to extract and purify the Schwann cells.METHODS: Rats were divided randomly into two groups, namely, in vivo pre-degeneration of sciatic nerve resection group and untreated control group, with 20 rats in each group. Under sterile conditions, the rat sciatic nerves were cut off at post-operative 7 days, Schwann cells were extracted by using mixed enzyme digestion and tissue mass transplantation; through low enzyme digestion and twice inoculation to differential adhesion, Schwann cells were purified. Cell morphology was observed under phase contrast microscope and identified by mmunofluorescence staining; cell purity was calculated; MTT method assay was used to determine the capacity of cell proliferation.RESULTS AND CONCLUSION: At 7 days of the culture, the experimental group showed the typical bipolar or bipolar Schwann cells, with connections between cells; in control group, cell processes were shorter and less associated with the surrounding cells. Following S-100 immunofluorescence staining, cells were positive for green expression.Cells proliferated rapidly in the experimental group and formed a swirling shape at 15 days, there were a relatively small number of fibroblasts, at the purity of 96.1%; in the control group, the cells proliferated slowly, with many fibroblasts at a low purity. MTT assay showed that primary cultured Schwann cell proliferated weakly in both groups; compared with the control group, the proliferation of subcultured Schwann cells in the experimental group was markedly increased (P < 0.05 or 0.01), and reached a peak 3 4 days later. The results confirmed that in vivo denaturing, in vitro hybrid enzyme digestion, tissue mass transplantation combined with low enzyme digestion, separation of double-differential adhesion of Schwann cells is a simple and rapid method to extract and purify Schwann cells.

Xihuang pill,a famous anticancer traditional Chinese medicine formula,has curative effect in clinical application.
