1.Effect of intrathecal injection of dexmedetomidine on the behavioral activity, pain degree and expression of protein kinase C in the spinal dorsal horn in rat models of chronic neuropathic pain

Objective To analyze the effect of intrathecal injection of dexmedetomidine on the behavioral activity, pain degree and expression of protein kinase C in spinal dorsal horn of rat models of chronic neuropathic pain, and to investigate the analgesic mechanism of dexmedetomidine.Methods 75 healthy male rats were randomly divided into observation group, model group and control group, 25 rats in each group.Chronic sciatic nerve injury model was established in the observation group and model group.After modeling, intrathecal dexmedetomidine intervention was used in the observation group.The model group was treated with saline injection and there was no intervention in the control group.Before the modeling (BM)and at 3(D3), 5(D5), 7(D7), and 14 (D14)days after medicine administration, the behavioral capacity was evaluated by cumulative evaluation method and movement function evaluation, and the assessment of pain degree (mechanical withdrawal method and thermal withdrawal latency pain threshold detection method), PKC staining score (immunohistochemical SABC method), PKC mRNA assay (RT-PCR method) and PKC protein expression (Western blot) were conducted and the data were statistically analyzed.Results ① Before modeling, the behavior, the cumulative scores of motor function, MWT, and TWL showed no significant differences between the different groups (P>0.05).After modeling, the model group and observation group showed that the cumulative scores and motor function scores were increased significantly, MWT and TWL decreased significantly, and the changes in the observation group were significantly lower than those in the model group (P<0.05).After modeling, the cumulative scores, motor function scores, MWT, and TWL were significantly different between the groups (P<0.05).② The expression of PKC was negative in the control group and positive in the model group.In the observation group, after the initial establishment of model, the PKC was strongly positive, and along with the prolonged treatment, the PKC expression intensity was decreased, and only weakly positively expressed at 14 d.③ After modeling, the observation group and model group showed that the PKC mRNA and PKC protein expression levels were significantly higher than that of the control group (P<0.05).With the continuous drug administration, the PKC mRNA and PKC in the observation group were decreasing, and reached a level close to that of the control group at 14 d of drug administration.From the third day after modeling, at the same time points, the amount of PKC expression in the observation group was significantly lower than that in the model group (P<0.05).Conclusions Intrathecal injection of dexmedetomidine can improve the behavior of rat models with chronic neuropathic pain, and reduce the degree of pain.It may be related to the inhibition of protein kinase C expression in the spinal dorsal horn.

It has been 4 years since Natural Health Products Regulations(NHPRs)was enforced by Health Canada in January of 2004.In the past time,how will it affect the marketed Traditional Chinese Medicines(TCM) in Canadian market? Recently,The two TCM products Carditionic Pill (Fufang Danshen Diwan)and Chaihu Pill from Tasly have been newly certificated by Health Canada recently.Taking them as examples,this article introduces the current status.opportunities and challenges for the regulation of TCM in Canada.and suggests that the domestic TCM corporations push the product registration as soon as possible.
4.Study of a new method on similarity evaluation for chromatographic fingerprint as applied in traditional Chinese medicine

AIM: To develop and perfect the similarity algorithm on chromatographic fingerprints of traditional Chinese medicine(TCM). METHODS: The experimental and simulated data were used to investigate and verify the characteristics on the new and former methods on similarity evaluation for chromatographic fingerprints method. RESULTS: There are many strong points for new similarity algorithm in practice. CONCLUSION: The new method provides more rationality and practicability in TCM quality control and analysis compare to existing methods.
5.Expression and correlation of PTEN and p27 protein in gastric carcinoma

ObjectiveTo investigate the correlation between PTEN and p27 expression in gastric carcinoma. MethodsImmunohistochemistry was applied to examine the expression of PTEN and p27 in 49 cases of human gastric carcinoma,49 cases of tumor-side ward mucosa and 13 cases of normal gastric mucosa.ResultsPositive expressive rates of PTEN and p27 protein in colorectal carcinoma were 61.22 %(30/49)and 57.14 % (28/49),which were lower than those in tumor-side ward mucosa [91.84 %(45/49) and 93.88%(46/49)]and normal colorectal mucosa [100 %(13/13)and 100 %(13/13)](P< 0.05). The expression of PTEN and p27 in gastric carcinoma correlated with each other (r=0.580,P=0.005),and which were associated with degree of differentiation of carcinoma,PTNM stage and lymph node metastasis of the tumors.Conclusion The abnormal expression of PTEN and p27 participate the malignant conversion process of the gastric mucosa cell and the expression levels of PTEN and p27 protein may be used to evaluate the pathologic biology behavior of gastric carcinoma.
6.Clinical features and risk factors of intracranial hemorrhage in patients with cerebral venous sinus thrombosis

Objective To analyze the clinical features,pathogenic factors and sites of thrombus involvement in patients with cerebral venous/venous sinus thrombosis (CVST) complicated with cerebral hemorrhage.Methods Eighty-seven patients with CVST,admitted to our hospital from January 2013 to December 2016,were selected and divided into observation group (n=39) and control group (n=48) according to cerebral hemorrhage.The demographic data,clinical features,pathogenic factors,and location of the involved venous/venous sinus were compared and analyzed between the two groups.The independent influencing factors of CVST combined with intracerebral hemorrhage were assessed using multivariable Logistic regression.Results Percentages of patients with decreased visual acuity (35.9%),epileptic seizure (48.7%),motor/sensory disorders (46.2%),consciousness changes (25.6%) and aphasia (12.8%) of the observation group were significantly higher than those of the control group (12.5%,16.7%,18.8%,8.3%,and 0%,P<0.05).In the aspect of potential pathogenic factors,the proportion of patients at pregnancy/puerperium in the observation group (20.6%) was significantly higher than that in the control group (6.3%,P<0.05).In the aspect of involving venous/venous sinus area,the proportion of patients involved in straight sinus in the observation group (30.8%) was significantly higher than that in the control group (12.5%,P<0.05).Multivariate Logistic analysis showed that pregnancy/puerperium and involvement of straight sinus were the independent influencing factors of CVST combined with cerebral hemorrhage (OR=6.752,P=0.017,95%CI:2.295-16.213;OR=4.573,P=0.029,95%CI:1.316-11.751).Conclusion CVST patients at pregnancy/postpartum or with straight sinus thrombosis are more prone to cerebral hemorrhage,and targeted treatment should be given as soon as possible.

A novel method was developed for the rapid determination of multi-indicators in corni fructus by means of near infrared (NIR) spectroscopy. Particle swarm optimization (PSO) based least squares support vector machine was investigated to increase the levels of quality control. The calibration models of moisture, extractum, morroniside and loganin were established using the PSO-LS-SVM algorithm. The performance of PSO-LS-SVM models was compared with partial least squares regression (PLSR) and back propagation artificial neural network (BP-ANN). The calibration and validation results of PSO-LS-SVM were superior to both PLS and BP-ANN. For PSO-LS-SVM models, the correlation coefficients (r) of calibrations were all above 0.942. The optimal prediction results were also achieved by PSO-LS-SVM models with the RMSEP (root mean square error of prediction) and RSEP (relative standard errors of prediction) less than 1.176 and 15.5% respectively. The results suggest that PSO-LS-SVM algorithm has a good model performance and high prediction accuracy. NIR has a potential value for rapid determination of multi-indicators in Corni Fructus.

Cyclosporine A( CsA) , as an immune inhibitor, is commonly used after organ transplantation.It has been found that the long-term use of CsA produced serious testicular toxicity and affected the fertility of organ transplantation patients.In order to investigate male reproductive damage induced by CsA, this article reviews its impact on reproductive organ development, its damage mechanism on the male reproductive system and drug research for alleviating its reproductive toxicity.It helps to make medical workers pay more attention to reproductive toxicity produced by CsA and make their efforts to develop some special drugs to lessen CsA reproductive toxicity.

10.Dietary and nutritional intervention of Prader-Willi syndrome

Prader-Willi syndrome is a rare and treatable disease, that is also known as hypotonia-dysgnosia-gonadal dysplasia-obesity syndrome.It is the first recognized imprinted condition due to parental chromosome 15 defect.The clinical symptoms of patients with Prader-Willi syndrome change continuously with age because of multiple organ damages, mainly including hypotonia, feeding difficulty in infancy, overeating and progressive obesity from early childhood, gonadal dysplasia, short stature in adulthood, metabolic syndrome and cognitive behavioral disorders, etc.There are consensus clinical diagnostic criteria and molecular genetics diagnostic methods in domestic and international research.However, the individualized endocrine and metabolic treatment are necessary for Prader-Willi syndrome.Life-long dietary treatment and nutritional intervention are very important to comprehensive treatment of patients with Prader-Willi syndrome, which are cheap and convenient to improve their metabolic condition.The dietary and nutritional intervention can effectively reduce obesity and complications, improve the prognosis and the quality of the life of the patients with Prader-Willi syndrome.Now, the age-related nutritional phases in the patients with Prader-Willi syndrome, the principle and applicable approaches of dietary and nutritional intervention for each phase, as well as long-term follow-up of nutritional status were discussed.
