BACKGROUND:Stem cel transplantation is increasingly hoped to promote osteoblast differentiation and inhibit osteoclast proliferation in the treatment of osteoporosis.
OBJECTIVE: To study the therapeutic effect of exogenous adipose-derived stem cel (ADSC) transplantation on osteoporosis in ovariectomized rats.
METHODS:Thirty Sprague-Dawley female rats were equivalently randomized into sham, model, ADSC transplantation groups. Rats in al groups except the sham group underwent bilateral ovariectomy to make osteoporosis models. Surrounding adipose tissues instead of the ovary were removed in the sham group. After modeling, rats were given 2×106ADSCs at passage 4via the tail vein in the transplantation group and the same volume of normal saline in the model group, once a week. After 6 weeks, levels of serum calcium, phosphorus, and alkaline phosphatase as wel as bone mineral density and histomorphometry indicators were detected in rats.
RESULTS AND CONCLUSION:Compared with the sham group, the trabecular bone volume fraction was significantly decreased in the model group (P < 0.01), but remarkably increased after ADSC transplantation (P < 0.05). After modeling, the bone trabecular absorption surface percentage and rate of bone trabecular formation were elevated significantly (P < 0.05 orP < 0.01), while these increases were improved by ADSC transplantation (P < 0.05). Additionaly, the levels of serum calcium and alkaline phosphatase and bone mineral density were significantly decreased after modeling, but were increased after ADSC transplantation. In contrast, the serum level of phosphorus was significantly increased in the model group (P < 0.05) but decreased markedly in the ADSC transplantation group (P < 0.05). To conclude, ADSC transplantation can reduce the loss of bone mass in osteoporosis rats by ovariectomy.

Objective The purpose of this study was to compare the whole body oxygen consumption determined by reverse Pick method and indirect calorimetry in mechanically ventilated patients after cardiac surgery. Methods Eight NYHA II-III patients (2 male, 6 female ) aged 43-68 yr, undergoing aortic valve replacement, mitral valve replacement or coronary artery bypass grafting(CABG) under CPB. Swan-Ganz catheter was placed via right internal jugular vein. Anesthesia and operation were carried out uneventfully. The patients were placed in ICU after operation and mechanical ventilation was continued (IPPV, FiO2 40%-50% , PEEP 5 cm H2O) . PaCO2 was maintained at 35-45 mm Hg by adjustment of VT and RR. T was maintained at 37.0 1C+0.5C .Total body oxygen consumption was measured by the reverse Fick method and indirect calorimetry simultaneously at 2h and 6h after operation. Results The mean oxygen consumption (VO2 ) value determined by indirect calorimetry( 162 + 30 mlmin-1m-2) was significantly higher than that determined by reverse Fick method (127+23 ml min-1m-2)(P

Objective To study the pretreatment with prostaglandin E1 ( PGE1 ) on acute superior mesenteric ar-tery ( SMA) ischemia reperfusion injury of intestinal cell in rats. Methods 18 healthy male SD rats were randomly divided into control group, sham operation group and experimental group. IRI of SMA model was made by clamping the SMA for one hour and two hours after reperfusion in the control group and the experimental group, respectively. PGE1(20 μg/kg) was injected from the tail vein in the control group and the experimental group. Character of pa-thology of small intestine was examined. The expression of Bax and Bcl-2 in small intestine cells and change of IF-ABP and DAO in serum were detected. Results Pathologic changes showed that there was no change in the control group;while in the sham operation group, chorionic epithelium mucosa ulceration and hemorrhage and necrosis oc-curred more seriously than that in the experimental group. Expression of Bax and Bcl-2 was higher in the sham op-eration group and the experimental group than that in the control group(P<0. 05), and it was obviously lower in the experimental group than in the sham operation group(P<0. 05). The content of IFABP and DAO in blood ser-um:it was higher in the sham operation group and the experimental group than in the control group ( P<0. 05 ) , and it was lower in the experimental group than in the sham operation group(P<0. 05). Conclusion PGE1 can relieve the alvine necrosis caused by rats' mesenteric reperfusion injury after acute artery ischemia and thus protect damaged mucosa of small intestine.

Objective To observe the E cadherin gene abnormality in non small cell lung cancer(NSCLC). Methods E cadherin gene mutations in exons 6, 7, 8 and 9 in 24 cases of non small cell lung cancer were studied by PCR SSCP and DNA sequence analysis. Results Abnormality of the E cadherin gene was found in exon 9 by means of insertion, deletion and mutation in 2 cases. Conclusion Abnormality of E cadherin gene is present in NSCLC by means of insertion and deletion in extracelluar matrix. The main cause of the decrease in epithelial adhesion may be resulted from the abnormality gene change of the E cadherin.

BACKGROUND: Schwann cells are the seed cells of neural repair, and it is a key to harvest a large number of Schwann cells with high purity and activity. OBJECTIVE: To compare the in vitro culture, purification, and morphology of Schwann cells between neonatal and adult rats, and investigate a simple and feasible culture method to harvest high-purity Schwann cells. METHODS: Totally 30 Sprague-Dawley rats, comprising 20 neonatal (1-3 days after birth, neonatal group) and 10 adult (weighing 150-200 g, adult group) rats, were included. Following double-enzyme digestion and two incubations, Schwann cells were isolated and purified by differential attachment. Cell morphology and attaching speed were determined through the use of inverted microscope. Cells were counted and cell purity was calculated. Cell proliferative ability was detected by MTT microcolorimetry. Curves of cell proliferation in each group were depicted to determine proliferative speed. Schwann cells were identified by S-100 immunochemistry.RESULTS AND CONCLUSION: Compared with fibroblasts, neonatal rat Schwann cells exhibited faster, while adult rat Schwann cells showed slower, attaching speed. Both neonatal and adult groups yielded over 96% cell purity. MTT microcolorimetry results revealed that Schwann cells proliferated actively in neonatal and adult groups. Cell proliferative curves show that neonatal rat Schwann cells proliferated faster than adult rat Schwann cells (P < 0.05). S-100 immunochemistry results showed positive results in both groups. All these findings suggest that double-enzyme digestion and two incubations followed by differential attachment is a satisfactory method to harvest considerable Schwann cells with high purity and activity. Neonatal rat Schwann cells show stronger proliferative, attaching capacities than adult rat Schwann cells.
