1.The clinical expression significance of PD-L1 on the CD4+CD25hi CD127low/- Treg cells in the peripheral blood of colon cancer

Objective To investigate the expression of PD‐L1 in the CD4+CD25hiCD127low/‐ Treg cells in the peripheral blood of colon cancer and the correlation with clinical stage .Methods The expression of PD‐L1 on CD4+CD25hiCD127low/‐Treg cells from 47 samples of the peripheral blood of colon cancer was analyzed by flow cytometry .Results Compared with those from healthy do‐nor ,the proportion of CD4+CD25hiCD127low/‐Treg cells and expression of PD‐L1 on these cells increased significantly (P<0 .01) . Further analysis showed that expression of PD‐L1 increased with the increase of clinical stage of colon cancer (P<0 .01) ,and de‐creased significantly after postoperation (P<0 .05) .Conclusion Increase of proportion of CD4+CD25hiCD127low/‐Treg cells and the expression of PD‐L1 may enhance the suppression of antitumor immunity ,and thereby promote tumor immunoescape .
2.Effect of intrathecal injection of dexmedetomidine on the behavioral activity, pain degree and expression of protein kinase C in the spinal dorsal horn in rat models of chronic neuropathic pain

Objective To analyze the effect of intrathecal injection of dexmedetomidine on the behavioral activity, pain degree and expression of protein kinase C in spinal dorsal horn of rat models of chronic neuropathic pain, and to investigate the analgesic mechanism of dexmedetomidine.Methods 75 healthy male rats were randomly divided into observation group, model group and control group, 25 rats in each group.Chronic sciatic nerve injury model was established in the observation group and model group.After modeling, intrathecal dexmedetomidine intervention was used in the observation group.The model group was treated with saline injection and there was no intervention in the control group.Before the modeling (BM)and at 3(D3), 5(D5), 7(D7), and 14 (D14)days after medicine administration, the behavioral capacity was evaluated by cumulative evaluation method and movement function evaluation, and the assessment of pain degree (mechanical withdrawal method and thermal withdrawal latency pain threshold detection method), PKC staining score (immunohistochemical SABC method), PKC mRNA assay (RT-PCR method) and PKC protein expression (Western blot) were conducted and the data were statistically analyzed.Results ① Before modeling, the behavior, the cumulative scores of motor function, MWT, and TWL showed no significant differences between the different groups (P>0.05).After modeling, the model group and observation group showed that the cumulative scores and motor function scores were increased significantly, MWT and TWL decreased significantly, and the changes in the observation group were significantly lower than those in the model group (P<0.05).After modeling, the cumulative scores, motor function scores, MWT, and TWL were significantly different between the groups (P<0.05).② The expression of PKC was negative in the control group and positive in the model group.In the observation group, after the initial establishment of model, the PKC was strongly positive, and along with the prolonged treatment, the PKC expression intensity was decreased, and only weakly positively expressed at 14 d.③ After modeling, the observation group and model group showed that the PKC mRNA and PKC protein expression levels were significantly higher than that of the control group (P<0.05).With the continuous drug administration, the PKC mRNA and PKC in the observation group were decreasing, and reached a level close to that of the control group at 14 d of drug administration.From the third day after modeling, at the same time points, the amount of PKC expression in the observation group was significantly lower than that in the model group (P<0.05).Conclusions Intrathecal injection of dexmedetomidine can improve the behavior of rat models with chronic neuropathic pain, and reduce the degree of pain.It may be related to the inhibition of protein kinase C expression in the spinal dorsal horn.
3.Clinical study of intraperitoreal immune chemotherapy for colorectal cancer following radical operation

Objective To Research the efficacy and side effects of intra-abdominal chemotherapy immune to the abdominal cavity after colorectal cancer liver metastasis and local recurrence.Methods 172 eases of colorectal cancer patients after radical operation were randomly divided into conventional regulation 86 cases of abdominal chemotherapy group(control group) and the immune abdominal chemotherapy group of 86 cases(observation group).The control group received 5-FU1.0,MMC8mg intraperitoneal chemotherapy The observation group at the grass-rcots level to add IL-2 50 million U.Results Observation group effective rate .was 70.9% higher than 55.8% (P <0.05) ;the obsenation gionp the three-year survival rate 83.7%,dlsease-free survival rate was 44.2% higher than the 62.8%,22.1% (P<0.05) ;observation group three-year recurrence rate of 25.6% and 34.9% lower than the rate of liver metastases in the control group of 37.8%,48.9% (P<0.05) ;two groups of gastrointestinal reaction,ⅠⅡ or degree of bone marrow suppression,liver function mildly abnormal ( elevated alanine amiuotransferase) had no difference.Conclusion For advanced colorectal cancer patients with early immune abdominal chemotherapy,the effective prevention and treatment of abdominal cavity and liver metastasis planting to improve the survival rate,side effects of light,fewer complications,the extension of survival,worthy of clinical application.
4.Extraction and purification of neonatal versus adult rat Schwann cells

BACKGROUND: Schwann cells are the seed cells of neural repair, and it is a key to harvest a large number of Schwann cells with high purity and activity. OBJECTIVE: To compare the in vitro culture, purification, and morphology of Schwann cells between neonatal and adult rats, and investigate a simple and feasible culture method to harvest high-purity Schwann cells. METHODS: Totally 30 Sprague-Dawley rats, comprising 20 neonatal (1-3 days after birth, neonatal group) and 10 adult (weighing 150-200 g, adult group) rats, were included. Following double-enzyme digestion and two incubations, Schwann cells were isolated and purified by differential attachment. Cell morphology and attaching speed were determined through the use of inverted microscope. Cells were counted and cell purity was calculated. Cell proliferative ability was detected by MTT microcolorimetry. Curves of cell proliferation in each group were depicted to determine proliferative speed. Schwann cells were identified by S-100 immunochemistry.RESULTS AND CONCLUSION: Compared with fibroblasts, neonatal rat Schwann cells exhibited faster, while adult rat Schwann cells showed slower, attaching speed. Both neonatal and adult groups yielded over 96% cell purity. MTT microcolorimetry results revealed that Schwann cells proliferated actively in neonatal and adult groups. Cell proliferative curves show that neonatal rat Schwann cells proliferated faster than adult rat Schwann cells (P < 0.05). S-100 immunochemistry results showed positive results in both groups. All these findings suggest that double-enzyme digestion and two incubations followed by differential attachment is a satisfactory method to harvest considerable Schwann cells with high purity and activity. Neonatal rat Schwann cells show stronger proliferative, attaching capacities than adult rat Schwann cells.
5.Clinical Application of MSCT with MPR and MIP Reconstruction in Diagnosis of Spinal Burst Fracture

Objective To investigate the diagnostic value of MSCT with multi-planar reconstruction(MPR)and maximum intensity projection(MIP)in diagnosis of spinal burst fracture.Methods 45 patients(53 vertebras)with vertebral burst fracture were examined by MSCT and processed with MPR and MIP.The imaging features were analyzed comparatively.Results The axial images clearly demonstrated the vertebral body vertically or transversely burst crack in 49 vertebras(92.5%),bony fragment inserted into the spinal canal and stenosis of spinal canal in 34 vertebras(64.2%).The sagittal images showed kyphosis in 28 vertebras(62.2%).The sagittal and coronal images showed decreased height of the vertebral body in 37 vertebras(69.8%)and depressed fracture of vertebral end plate in 19 vertebras(35.8%).Total 44 fractures were located at spinal appendix,39 were showed by axial images,35 by sagittal images and 33 by coronal images.MIP displayed the space changes of bone structures in all cases and rotary dislocation fracture in 6 cases(11.3%).Conclusion MPR and MIP are of significant values in diagnosis and clinical treatment of spinal burst fracture.
6.A Study of the Anatomy of Superior Mesenteric Artery with Multislice Spiral CT Angiography

Objective To measure the angle and distance between superior mesenteric artery (SMA) and abdominal aorta (AA) at the level of left renal vein and duodenum in Chinese.Methods The angle and distance between SMA and AA were measured in 41 patients with normal CT findings in the supine position using 16 slices multislice spiral CT(MSCT)scanner.Results The average angle of 41 normal cases was 47.4??18.3? (mean?SD), and 3 of 41 (7.3%)≤15?,10 of 41 (24.4%)≥70?. At the level of left renal vein and duodenum, the average distance between SMA and AA were (1.3?0.4) cm and (1.4?0.4) cm respectively.Conclusion MSCT allows accurately measure the angle and distance between SMA and AA, it shows significance in the diagnosis of SMA syndrome and nutcracker phenomenon.

Cyclosporine A( CsA) , as an immune inhibitor, is commonly used after organ transplantation.It has been found that the long-term use of CsA produced serious testicular toxicity and affected the fertility of organ transplantation patients.In order to investigate male reproductive damage induced by CsA, this article reviews its impact on reproductive organ development, its damage mechanism on the male reproductive system and drug research for alleviating its reproductive toxicity.It helps to make medical workers pay more attention to reproductive toxicity produced by CsA and make their efforts to develop some special drugs to lessen CsA reproductive toxicity.
9.Detection of differentially expressed genes between high-rate and low-rate metastatic human lung giant cell carcinoma cell strains using cDNA microarray

Objective To analyze the differences in gene expression between human lung giant cell carcinoma cell strains of high metastatic 95D and low metastatic 95C and to screen lung cancer metastasis-associated genes using cDNA microarray.Methods The mRNAs were extracted from human lung giant cell carcinoma cell strains of high metastatic 95D and low metastatic 95C,and reversely transcribed to the cDNAs and labeled by Cy5-dUTP and Cy3-dUTP to prepare the hybridization probes.The mixed probes were hybridized to the cDNA microarray chip.The information was obtained by managing the cDNA microarray chip with Scan Array 3000 scanner and ImaGene3.0 software.A part of genes whose expressions changed in cDNA microarray analysis were further identified by RT-PCR.Results The cDNA microarray analysis showed that the expressions of 466 genes changed,among which 108 pairs of Double Gene had the same GenBank ID and some of the genes,including Fln29,RUVBL2,C14orf3 et al,were further confirmed by RT-PCR.The results of RT-PCR was coincided well with the cDNA microarray results.Conclusion Many different genes are involved in the metastasis of lung cancer.cDNA microarray technique might be a useful method in screening lung cancer metastasis-associated genes.

Objective To investigate the internal quality control ( IQC ) on clinical chemistry , clinical immunology and clinical hematology in mutual recognition laboratories in medical institutions in Beijing.Methods By means of questionnaire survey and on -site investigation, fresh frozen serum and whole blood samples with assigned values by reference method were measured to investigate the status of IQC on clinical chemistry , clinical immunology and clinical hematology in 142 mutual recognition laboratories in medical institutions of Beijing,and results were analyzed.Results 142 copies of questionnaireson clinical chemistry, clinical immunology and clinical hematology were send out and 120, 97, and 101 laboratories returned the questionnaires respectively .The information feedback rate was 84.5%, 68.3% and 71.1%respectively .All the questionnaires were effective .Questionnaires survey results showed that more than 50%laboratories set up quality control goals and the most of the goals were probability for error detection ( Ped) 95%, probability for false rejection(Pfr)5%;About 70% laboratories usecd the same quality control plan for different tests ;The most frequently used quality control rules are 12s/13s/22s.On-site investigation showed that ,take the results of clinical chemistry for example , based on the desirable biological variation and WS/T 403 -2012 , most of the tests can't meet the quality control goalsunder the existing quality controlcondition.Conclusion Clinical laboratories should consider their actual situations , assess their own qualitylevels that they can reach , set reasonable quality standards for themselves , and make appropriateindividualized quality control plan.
