1.Establishment of rabbit femoral 3D biomechanical models
Chinese Journal of Tissue Engineering Research 2008;12(26):5189-5192
BACKGROUND: The image edge of rabbit femur is identified using self-programmed Matlab program. But owing to the complexity of the model, can we set up a femoral structure model after the solid model of the femur created in UG three-dimensional (3D) software is put into ANSYS software? OBJECTIVE: To explore the method of constructing 3D biomechanical model of rabbit femur.DESIGN: Randomized controlled observation.SETTING: Biomechanics Laboratory of Beijing Institute of Technology.MATERIAL: One 1-year-old female New Zealand white rabbit weighing 2.6 kg. ACTIS 400/225 industry CT scanner (BIR,USA).METHODS: The experiment was performed at the Biomechanics Laboratory of Beijing Institute of Technology from July to transformed into *.bmp format. Photoshop image processing software was applied for some essential processing on the original image, such as contrast control, smoothing and noise attenuation to strengthen the differentiability and analyzability of the image. In addition, the magnetic noose tool was used to outline the contour line of inside and outside boundary, and obtain the inside and outside contour line of every layer of the femur. The rabbit femur was scanned by industry CT starting from the upper extreme of the femur, vertical to femoral ordinate axis line (slice space=I ram). The scanning slice space was adjusted to 2 mm from the 43rd layer below small trochanter to the 53ra layer of middle femur because the shape between these layers was relatively regular. Totally 87 slices of layer images were scanned 9.9 cm in length. The image was put into the computer and processed to get the boundary contour line; the contour line coordinate for modeling was obtained through imported from UG into ANSYS software. A solid model of the femur was produced automatically. Finally, the solid model with cavity was obtained using subtraction option of Boolean in ANSYS.MAIN OUTCOME MEASURES: Establishment of rabbit femoral structure model. connected by the multiple valid points was formed. The 2nd, 3rd, 4th until the 87th layers were read in turn, and the inside and automatically divide the mesh to produce finite element meshes and 42 221 nodes and 27 768 elements were obtained. CONCLUSION: The 3D model of rabbit femur actually simulated the anatomic appearance of the femur. This model lays a foundation for identifying the optimal parameter of vibration that can improve bone mineral density by finite element analysis.

Objective To study the effect of vibration as a countermeasure against bone loss. Methods Volunteers were divided into vibration-group(VIB)and control-group(CON).Vibration experiments were performed in VIB for 3 months(10 min/d,5 d/week).Bone mineral density(BMD)of both VIB and CON was observed after 3 months. Results BMD in VIB increased obviously after vibration treatment(+1.29% for vertebrae and +1.65% for femur). The effectiveness rate was 80%.Conclusion Whole body vibration is effective in countermining bone loss.It has a broad prospect in treating osteoporosis and counteracting bone loss caused by weightlessness.

Objective To investigate the clinical significance of MTAP, CDKN2A and CDKN2B gene expression in diffuse large B-cell lymphoma (DLBCL). Methods MTAP, CDKN2A and CDKN2B gene expression were assessed by Real-time quantitative PCR in 40 cases of DLBCL and 19 cases of reactive hyperplasia. The clinical and follow-up data were also collected. Results Comparing with reactive hyperplasia, MTAP, CDKN2A and CDKN2B gene expression were decreased in DLBCL group (P = 0.024,0.044 and 0.047, respectively). Low-expression of all the three genes were associated with advanced Ann Arbor stage (P=0.004, 0.001 and 0.027, respectively). No obvious difference were observed according to gender, age, the number of the extra-nodal infiltration, ECOG score, bone marrow involvement and serum LDH level (P >0.05). MTAP and CDKN2A gene expression were associated with B symptoms (P =0.003 and 0.028, respectively) and IPI scores (P =0.001 and 0.011, respectively). With regard to survival rates,MTAP, CDKN2A and CDKN2B gene expression were significantly associated with OS (P =0.022, 0.019 and 0.042, respectively). Conclusion MTAP, CDKN2A and CDKN2B gene expression in DLBCL were decreased and related to disease progression and prognosis. They could be considered as biomarkers to evaluate biological behavior and clinical outcome of DLBCL patients.

Objective To evaluate the efficacy of PAD regimen (bortezomib, doxorubicin, dexamethasone) and VAD-like T regimen (vincristine, doxorubicin/doxorubicin derivatives, dexamethasone combined with thalidomide) in the treatment of patients with newly diagnosed multiple myeloma (MM). Methods The efficacy of 54 patients with MM who received VAD like-T regimen and 72 patients with MM who were treated with PAD regimen, including complete remission (CR) rate, very good partial remission (VGPR) rate, overall response rate (ORR), overall survival (OS), progression-free survival (PFS) and adverse events, were retrospectively analyzed. Results The CR rate of PAD group was higher than that of VAD-like T group [31.5 % (23/72) vs. 9.3 % (5/54), χ2=0.30, P=0.002]. The VGPR rate and ORR of PAD group were not statistically higher than those of VAD-like T group [16.7 % (12/72) vs. 16.6 % (9/54), P=0.180; 82.2 %(65/72) vs. 81.5 % (44/54), P=0.190, respectively]. Median PFS of PAD group was significant longer than that of VAD-like T group [(38.2±2.2) months vs. (28.0±7.6) months, P=0.017]. The 3- and 5-year OS rates of PAD group were higher than those of VAD-like T group, but there were no significant differences between two groups (P>0.05). In terms of the adverse events, the incidence of peripheral neuropathy in PAD group was significantly higher than that of VAD-like T group [31.5 % (23/72) vs. 14.5 % (8/54), P=0.03]. Conclusions Compared with PAD protocol, the CR and median PFS of VAD-like T regimen are poor, however, VGPR,ORR, PFS and 5-year OS are similar between the two groups, and VAD-like T regimen is safer with low incidence of peripheral neuropathy. VAD-like T regimen as the first-line treatment is effective and well-tolerated, especially for newly diagnosed MM patients not suitable for transplantation and bortezomib.

To investigate the expression of vaginal Th1 and Th2 cytokines in rats with experimental vaginal candidiasis under different immune conditions, ICR murine vaginal candidiasis model was established and immno-suppressed murine models of vaginal cadidiasis were established in estrogen-treated mice. Non-estrogen-treated mice were used as controls. The mRNA level of Th1 (IL-2)/Th2 (IL-4, IL-10, TGF-beta1) cytokines in murine vaginal tissues was determined by RT-PCR. The cykotine in local tissues was increased to different extent under normal immune condition. IL-2 mRNA was increased during early stage of infection, while IL-10 was increased transiently during late stage of infection. TGF-beta1 production was found to be increased persistently. At same time, the expression of IL-2 mRNA was suppressed in immno-suppressed group, and the level of IL-4, IL-10, and TGF-beta1 were higher than the normal immunity group to different degree during infection. The high level of IL-2 mRNA during early stage of infection was associated with clearance of mucosal Candidia albicans (C. albicans), and its expression suppressed leading to decreased clearance of mucosal C. albican in immuno-suppression. The over-expression of IL-4 and IL-10 could significantly enhance the susceptibility to C. albicans infection in mice.
