1.The effect of rhynchophylline on NR2B mRNA expression in nucleus accubems and amygdaloid of amphetamine-dependent rats
Chinese Pharmacological Bulletin 2003;0(09):-
Aim To study the effect of rhynchophylline on NMDA receptor 2B subunit(NR2B) mRNA expression in the nucleus accumbens(NAc) and amygdala(Amy) of amphetamine-induced condictioned place preference(CPP) rats.Method 56 male SPF grade Sprague-Dawley rats were randomly divided into control group,amphetamine-induced model group, model group with ketamine, model group with low, middle, high dose of rhynchophylline, and control group with rhynchophylline(n=8 for each group) The time of rats' staying in white compartment was measured in the 5th day, then NR2B mRNA expression in NAc and Amy of each rat was detected with in situ hybridization technology. Results The rats exhibited CPP after treated with 2 mg?kg-1?d-1 amphetamine for 4 d.Ketamine and rhynchophylline eliminated CPP effect, and the effect of rhychophylline showed a dose-dependence.In amphetamine-induced model group,NR2B mRNA expression in NAc and AMY of rats was significantly increased.Ketamine, middle and high dose of rhynchophylline blocked the increase of NR2B mRNA expression induced by amphetamine.Rhynchophylline had no effect on the basal level of RN2B mRNA expression in normal rats. Conclusion Inhibition of NR2B mRNA expression in NAc and AMY may contribute to the antagonistic effect of rhychophylline on amphetamine-induced CPP.

Regulation of gene expression is one of the most importa nt responses of cells to DNA damage induced by radiation. A novel expressed seq u ence tag (EST) fragment had been cloned from human embryo lung cells induced by 50cGy radiation and named RIG1. To clone the full-length cDNA of RIG1, a non-c loned cDNA library of human embryo lung cells induced by low dose irradiation ha d been established. This library was used as template in enchanced nest RACE PCR and biotin-labeled probe was used for further purification. The 3′ flanking s equence of this EST was cloned and sequenced with this set of technology. It was illuminated by homology analysis that this 3′ flanking sequence and the origin al EST are well aligned with a BAC clone of 20th chromosome and the predic ted exons sequence of this chromosome is well consisten ce with the real EST. Thus the RIG1 can be roughly located in 20th chromos ome. By use of the exons sequence predicted from chromosome sequence by GENSCA N, full-length of RIG1 gene has been cloned. Chromosome location of RIG1 gene i s further determined by this successful verification of Bioinformatics predictio n by experiment. By the same step, genome sequence of RIG1 has been determined. Therefore,by the combined use of Bioinformatics analysis, the full-length cDNA sequence and genome sequence of RIG1 gene are obtained and the predicted protei n sequence is determined.

Objective To test the effect of bone marrow mesenehymal stem cells (MSCs) transplantation on oxidative stress and the development of pulmonary emphysema in rats. Methods SD rats (n=26) were randomly divided into three groups:normal control group (group A, n=8),emphysema group (group B, n=8) and emphysema+MSCs transplantation group (group C, n=10).Rat models of emphysema was established by exposing rats to cigarette smoking for 14 weeks. Then rats of group C received MSCs transplantation. At the 14th and 28th days after 4 course of MSCs transplantations, one rat in group C was sacrificed at each time point and their lungs were preserved in frozen sections. Survival of MSCs in the lung tissues were observed by fluorescence microscopy. Eight weeks after transplantations, lung sections were stained by hematoxylin and eo?sin (HE) to observe the morphological alterations.Mean linear intercept (MLI) and mean alveolar numbers (MAN) were also measured. Serum and lung malondialdehyde (MDA) levels and superoxide dismutase (SOD) activity were also examined. Re?sults At the 14th day and 28th day after transplantations of MSCs, MSCs successfully localized to lung and survived in rat models of emphysema. Emphysematous changes of lung tissues were observed in both group B and group C. MLI was higher while MAN was lower in group B and C than those in group A (P<0.05). MLI and MDA levels in serum and lung were high?er while MAN level and SOD activity were lower in group B than those in group C (P<0.05).MDA levels in serum and lung was higher while SOD activity was lower in group B and C than those in group A (P<0.05). Conclusion MSCs transplanta?tions can effectively alleviates pulmonary emphysema in rat models which might through reducing oxidative stress .
