1.Changes of serum advanced oxidation protein products in type 2 diabetes mellitus patients with diabetic nephropathy

Objective:To study the changes of serum advanced oxidation protein products(AOPP) in patients with diabetic nephropathy(DN) and its correlationship with superoxide dismutase(SOD) and glutathione peroxidase(GPx). Methods: Eighty-five diabetes mellitus (DM) patients were divided into 4 groups: non-DN group(DM, n=25), early-stage DN group(DN3, (n=24), )significant DN group(DN4, n=17) and end-stage DN group(DN5, n=17). The concentration of serum AOPP was measured spectrophotometerically, serum SOD was determined by xanthine oxidase test and GPx by DTNB reaction test. Purpose The aim is to investigate the effects of BSP on immunological function in normal and immunosuppressed mices.Methods Thymus and spleen indexes, peripheral blood WBC number,the lymphocyte proliferation response, IL-2 production and serum and splenocyte hemolysin contents were measured after intraperitoneal injection of BSP in normal and immunosuppressed mices.Results (1)BSP 100 mg/(kg*d)×10d significantly increased the thymus and spleen indexes and peripheral blood WBC number in immunosuppressed mice.The thymus and spleen indexes in normal mice was also increased. In addition,BSP markedly improved T,B lymphocyte proliferation responses and IL-2 production in normal and immunosupressed mices.(2) BSP improved the serum and splenocyte hemolysin contents in normal and immunosuppressed mice. Conclusion It was suggested that BSP was a kind of immunomodulator, and could improve the immunological function of normal and immunosuppressed mices.
4.Primary epldidymul rhabdomyosarcoma; one ease report

Objective To discuss the clinical features of primary epididymal rhabdomyosarcoma and review the diagnostic and therapeutic advances. Methods One 16-yesr-old male presented with right scro-turn swelling mass for 4 months. Physical examination revealed an ovoid and solid mass of 4.5×3.5×3.0 cn in right scrotum. The mass was hard and its surface was smooth without tenderness. Transillumination test was negative. There was no palpable inguinal lymph node. Pre-operative diagnosis was inflammatory nodule of right epididymis. Results Right epididymectomy was performed under caudal anesthesia une-ventfully. The pathological study showed that the neoplasm was composed of small round shape ceils with small and anachromasis nucleus. The immunohistochemical study showed that these cells were positive for myoglobin, aetin and desmin, but negative for smooth muscle actin. Electron microscopy showed that paral-leling thin filaments were found with no asrcomere-like structure in the intracytoplasm. The pathologic diag-nosis was right epydidymal rhabdomyosarcoma. The patient refused further therapy. A mass was found with rapid growth accompanied with pain in the right scrotum 2 months after the first operation and was clinically diagnosed as tumor recurrence. This patient had then undergone right radical orchiectomy. Frozen section analysis revealed negative surgical margin. The pathologic diagnosis was embryonal rhabdomyosarcoma re-currence. Patient underwent subsequent chemotherapy with ifosfamide, etoposide and vincristine. After one year follow-up, he was generally well without evidence of recurrence. Conculsions Primary embryonal rhabdomyosarcoma is a rare tumor with aggressive growth patterns. The diagnosis depends on the pathologi-cal examination because the clinical presentation is not specific. Combination therapy with radical orchiecto-my, chemotherapy and radiotherapy provides patients an excellent long-term prognosis.

Objective To study the effects of choline chloride on activities of myosin adenosine triphosphatase (mATPase) of muscle fibres and muscle atrophy in tail-suspended rats. Method Twenty-four adult female Sprague-dawley rats were averagely divided into 3 groups matched for body mass:control group(CON),tail suspension group(TS) and tail suspension group associated with choline chloride(TS+Cch).Weightlessness was simulated by 14 d tail suspension of rats;gastric lavage with choline chloride was given to 1/2 of the rats during tail suspension (15 mg/kg body weight.i.g.).Activities of myosin adenosine triphosphatase (mATpase) of intrafusal and extrafusal fibres in soleus (SOL) muscle were determined by Ca 2+-ATPase method.Paraffin sections of the soleus muscle were prepared by routine histochemistry method. Objective To study the relationship of serum leptin and inflammation with nutrition status in continuous ambulatory peritoneal dialysis(CAPD) patients.Methods 30 CAPD patients and 30 healthy controls were enrolled in the study.Leptin was measured by ELISA,inflammatory factors C reactive protein(CRP),IL-6,TNF-?.These patients were classified to three nutritional categories based on SGA normalnutrition,mild-to-moderate malnutrition,and severe malnutrition.Results CAPD group had significantly higher serum leptin,peritoneal dialysis fluid leptin,insulin,CRP,IL-6 and TNF-?,lower serum Alb than the control group.IL-6 was significantly positive correlation with TNF-?.Serum leptin was no correlation with Alb,CRP,IL-6 and TNF-?.Conclusion Serum leptin and inflammation are closely related to the occurrence of malnutrition in CAPD.

Objective To investigate the roles of oxidative stress and inflammatory reaction in pathogenesis of endothelial dysfunction and atherosclerosis in patients with chronic renal failure(CRF).Methods 63 CRF patients were divided into non-dialysis group and hemodialysis group and 27 healthy volunteers were involved in the study.The levels of plasma malondialdehyde(MDA) were determined by thiobarbituric acid reaction,the activity of glutathione peroxidase(GSHPx) was mearsured by spectrophotometry,plasma nitric oxide(NO),high-sensitive C-reactive protein(hs-CRP),endothelin-1(ET-1) and tumor necrosis factor(TNF)-? were determined by sensitive enzyme-linked imunosorbent assay(ELISA).Results Levels of plasma MDA,hs-CRP,IL-6,TNF-?and ET-1 in CRF patients were higher than those in healthy controls,while NO,GSHPx were lower than those in healthy controls and hemodialysis aggravated these changes.Multivariate stepwise regressive analysis showed that ET-1,vWF and NO were related to MDA,TNF-?,hs-CRP,Cr(R2=0.502,0.631,0.476,respectively,P

Objective To investigagte the effect of high dose folic acid(FA) combined with vitamin B_(12) to homocysteine plasma level in maintenance hemodialysis(MHD) patients.Methods 52 MHD patients were randomly divided into 4 groups,receiving of FA 0,15,30,60mg/d orally for 3 months and those who receiced folic acid were given vitamin B_(12) 500?g/d,im.The levels of plasma FA,vitamin B_(12) and total homocysteine(tHcy) at 0,1,2,3,6 month were detemined by radioimmunoassay and flurescence polarization immunoassay respectively.Results Levels of tHcy in MHD patients were significantly higher than those in healthy controls.Both high dose and low dose FA could lower tHcy plasma concentrations,but had no significant difference in homocysteine reduction ratio,and could not normalize it.Three doses of FA combined with vitamin B_(12) treatment could effectively increase the plasma FA and vitamin B_(12) concentrations.Conclusion tHcy is elevated in the majority of MHD patients.Low dose of FA combined with vitamin B_(12) can decrease tHcy plasma levels,but cannot normalize it.
10.EFFECTS OF BROWN SEAWEED POLYSACCHARIDE ON LYMPHOCYTE FUNCTION AND CYTOKINES PRODUCTION

Objective: To investigate the effects of BSP on lymphocyte function and cytokines production in normal and immunosuppressed mice.Methods: The lymphocyte proliferation response, T lymphocyte subpopulations and IL 2 production were measured after intraperitoneal injection of BSP in normal and immunosuppressed mice, and the activity of IL 1 and TNF secreted by mouse peritoneal macrophages were detected after BSP supplementation in vitro.Results: (1)BSP〔(100 mg/(kg?d)?10 d)〕markedly improved T,B lymphocyte proliferation responses and IL 2 production in normal and immunosuppressed mice, and significantly increased the number of L 3T + 4 cells in thymus and spleen of immunosuppressed mice, and the ratio of L 3T + 4/Lyt 2 + cells was also increased.(2)BSP distinctly enhanced the activity of IL 1 and TNF production by mouse peritoneal macrophages in vitro.Conclusion: BSP can improve the immunological function of normal and immunosuppressed mice.
