1.Effects of different degrees and duration of hypothermia on traumatic brain injury in dogs

Objective To investigate the effects of different degrees and duration of hypothermia on severe traumatic brain injury in dogs. Methods Thirty-six one-year-old healthy dogs weighing 13-15 kg were used in this study. Traumatic brain injury model was estabhshed according to the method described by Feeney. Orthogonal design was used in this experiment. Two empirical factors: temperature (factor A) and duration (factor B) were used. Body temperature was maintained at 38 ℃ (A1), 31 ℃ (A2) or35 ℃ (A3) for 6 h (B1) or 12 h (B2).The dogs were divided into 6 groups: group A1B1 , A1B2, A2B1, A2B2, A3B1 and A3B2. Blood gas analysis was performed after the target temperature was maintained for the target duration. Neurological deficit was assessed and scored (0 = no deficit, 500 = severe neurological deficit) and blood samples were obtained at 24, 48 and 72 h after brain injury for determination of serum concentrations of neuron-specific enolase (NSE) and myelin basic protein (MBP) and plasma S-100β protein concentration. Brains were removed at the 72 h after brain injury for determination of Bcl-2 and Bax expression and detection of apoptosis in the brain. Results Hypothermia significantly decreased serum NSE, MBP and plasma S-100β concentrations, neuronal apoptosis and NDS scores;up-regulated Bcl-2 expression and down-regulated Bax expression in brain tissue in the 4 hypothermia groups (group A2B1, A2B2, A3B1 and A3B2) as compared with the control groups (group A1 B1, A1 B2). Best neuroprotective effects were observed in group A3 B1 (35 ℃ for 6 h) in terms of serum NSE, NBP and plasma S-100β concentrations, neuronal apoptosis and cerebral Bax and Bcl-2 expression, but there was no significant difference in the NDS scores among the 4 hypothermia groups. Conclusion Hypothermia can provide neuroprotection in a dog model of traumatic brain injury but the neuroprotective effect is independent of the degree and duration of hypothermia.
2.Determination of isoimperatorin in Exocarpium citri grandis from various species and origins by TLC-Scanning

Objective: To determinate the content of isoimperatorin in Exocarpium citri grandis from 10 various species and origins. Methods: The isoimperatorin was determinated at 305nm by TLC Scanning. Results: The content of isoimperatorin in Exocarpium citri grandis was very low, and the law of distribution could not be found. Conclusion: Although the content of isoimperatorin in Exocarpium citri grandis was very low, the method is very sensitive and simple and the result is accurate.

Object To establish the HPLC fingerprint of flavonoids in Exocarpium Citri Grandis (ECG) Methods HPLC fingerprint of ten samples were established. The naringin was used as standard. The relative migration time and the percentage of peak area were used as parameters Results More than 30 peaks occurred on the HPLC fingerprint of flavonoids of ECG. The fingerprints of C. grandis ‘Tomentosa’ and C. grands had obivous differences Conclusion The method is simple and accurate with a good reproducibility. It could be used for quality control and identification of ECG
4.Effects of endovascular cooling in different degrees on severe traumatic brain injury in dogs

Objective To investigate the effects of endovascular cooling in different degrees on severe traumatic brain injury in dogs.Methods Eighteen dogs of both sexes,weighing 12-15 kg,were randomly divided into 3 groups ( n =6 each):hypothermia at 31 ℃ group (group A),hypothermia at 35 ℃ group (group B) and normothermia group (group C).The model of severe traumatic brain injury was established according to the improved Feeney's free-fall method.Six hours of endovascular cooling was performed using an endovascular cooling system in A and B groups.The cerebrospinal fluid (CSF) was collected before traumatic brain injury (T0),and at 24,48 and 72 h after traumatic brain injury (T1-3) for measurement of the concentrations of neuron-specific enolase (NSE),S-100β,myelin basic protein (MBP),aspartate (Asp),glutamie acid (Glu),glycine ( G ly) and gammaaminobutyric acid (GABA).Results The concentrations of NSE,S-100β,MBP,Asp,Glu and Gly in CSF were significantly lower at T1-3,while the concentration of GABA in CSF was significantly higher at T1-3 in groups A and B than in group C (P <0.05 or 0.01).The concentrations of NSE,Asp,Glu and Gly in CSF were significantly lower at T1-3,while the concentration of GABA in CSF was significantly higher at T1-3 in group B than in group A ( P <0.05 or 0.01).Conclusion Endovascular cooling can reduce severe traumatic brain injury in dogs,and the efficacy is better when the temperature is reduced to 35 ℃ than that when reduced to 31 ℃.
5.Correlations between serum macrophage migration inhibitory factor and active rheumatoid arthritis

BACKGROUND: Macrophage migration inhibitory factor (MIF) is mainly concerned with macrophage mobilizing function, as the upper stream cytokine of tumor necrosis factor-α(TNF-α), interleukin-1β (IL-1β), which may have critical effect in the process of the onset of rheumatoid arthritis. OBJECTIVE: To explore the correlations between the change of serum MIF and the activity of rheumatoid arthritis (RA) disease.DESIGN, TIME AND SETTING: Non-randomized control and case study, which was carried out in the Bethune International Peace Hospital of Chinese PLA from September in 2005 to October in 2006.PARTICIPANTS: Sixty RA patients were included in this study, and other thirty healthy subjects were selected as the control group. There were significant differences in age and sex between the two groups. METHODS: Clinical data of sixty RA patients were selected by carrying out retrospective analysis, then on the basis of disease activity score (DAS) accumulated points, they were divided into active and inactive group respectively, who were contrasted with 30 health adults. MAIN OUTCOME MEASURES: ① Morning stiffness (in minutes), joint tenderness index, arthrocele index, semi-quantity rheumatoid factor (RF), C-reactive protein concentration (CRP), erythrocyte sedimentation rate (ESR), and platelet count (PLT) were recorded; ② To compare the level of serum MIF, IL-1β and TNF-α among active group, inactive group, and control group; ③ The correlation analysis was carried out among the level of serum MIF, inflammatory index and clinical observation index.RESULTS: There was significantly increased in serum MIF of patients in the active group compared to of inactive and normal groups (P < 0.05), but there were no significantly differences between inactive and control groups (P > 0.05). There were significant correlations between the serum MIF concentration and active inflammatory index of RA disease, blood sedimentationrate (ESR), C-reactive protein (CRP), platelet counting (PLT), interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), swell joint index (SJI) and tenderness joint, but no significant difference was observed between the serum MIF, age, disease course, morning stiffness and rheumatoid factor (RF).CONCLUSION: The serum MIF concentration is significantly increased in patients with RA, and it may be a useful parameter for monitoring disease activity of RA.
6.Observation of effect of metformin and exercise therapy for IGT.

Objective To observe the effect of metformin and exercise therapy for IGT. Methods 86 patients with IGT were given metformin and exercise therapy,the range of glucose,BMI and symptoms improvement were observed before and after the therapy. Results After 6 months therapy,the fasting blood glucose and 2 h after oral administration of 75 g glucose levels were significantly reduced(P<0.01=,BMI reduced(P<0.05=.72 cases (83.7%)were back to normal glucose tolerance,14 cases maintained IGT(those unable to adhere to exercise and no diet controlled),0 case became DM. Conclusion Metformin and exercise therapy had good efficacy in curing IGT.
7.Effect of Touqing capsule associated with acupuncture at Fengchi on cervical spondylosis

objective To evaluate the clinical effect of touqing capsule associated with Fengchi on the vertebral artery type of cervical spondylosis(CSA)in the attack stage.Methods A total of 100 patients with CSA, according to the doctor order, were randomly divided into a control group and a treatment group respectively, with 50 patients in each. The control group was treated with acupuncture therapy, the treatment group was additionally treated with Touqingcapsule on the basis of the control group, and both groups were treated for 20 days. The hemodynamics was detected by transcranial doppler, the change of TCM syndrome integral was observed before and after the treatment, and the clinical efficacy was evaluated.Results After treatment, the treatment group dizziness (1.10 ± 0.33 vs. 2.01 ± 0.26,t=14.909), neck shoulder ache (0.87 ± 0.16 vs. 1.92 ± 0.69,t=10.266), numbness (0.95 ± 0.14)vs. 1.87 ± 0.16,t=29.844), the total integral (2.16 ± 0.75 vs.3.89 ± 1.14,t=8.756) integral were significantly lower than the control group(P<0.01).Treatment group RVA (40.03 ± 5.89 cm/svs. 34.26 ± 6.26 cm/s,t=4.628), the LVA (42.14 ± 5.42 cm/svs.33.89 ± 6.08 cm/s,t=6.985) rise and PI (0.79 ± 0.14 vs.1.04 ± 0.19,t=7.312) reduction were significantly better than those of the control group (P<0.01). The total effective rate in the treatment group was significantly better than the control group (91.7%(44/48)vs. 74.5%(35/47),χ2=3.863, P=3.863).Conclusion Touqing Capsule associated with Fengchi is effective for CSA. Its functional mechanism is correlated with improvement of haemodynamics of vertebral-basilar artery of the CSA patients.
8.Extraction of prostate cancer stem cells using self-designed magnetic beads

BACKGROUND:Effective sorting of prostate cancer stem cels is the basis of experimental studies in prostate cancer developing. The most common sorting method is magnetic-activated cel sorting. OBJECTIVE:To separate CD133+/CD44+ cels in prostate cancer tissues using self-designed magnetic beads folowed by culture, passage and immunological identification. METHODS:Self-designed magnetic microspheres were applied to establish immunomagnetic beads to sort CD133+/CD44+ cels in prostate cancer tissues. The sorted cels were cultured in serum-free medium. The sphere formation, cel morphology, and proliferation ability after cel passage were statisticaly compared between the sorted cels and the normal tumor cel lines. Immunofluorescence detection was performed to detect the expression of specific antibodies. RESULTS AND CONCLUSION:Self-designed immunomagnetic beads had smal diameter and a high-sorting effect. The sorted cels possessed a high capacity of microsphere formation. After cel culture and passage, the cels highly expressed CD133 and CD44 antigens. The sorted cels with no induction had varying shapes and grew vigorously. After induction with transforming growth factor-β, the cultured cels were noted to have a single shape and grow slowly. The cel proliferation ability of sorted cels in these two groups differed significantly from that of the normal cancer cel lines (bothP < 0.05). In conclusion, the CD133+/CD44+ cels sorted from prostate tumor cels possessed cel morphology and function characteristics of stem cels which can provide a basis for extraction and culture of prostate cancer stem cels. 9.Impact of glucagon-like peptide-1 receptor agonists on nasopharyngitis and upper re-spiratory tract infection among patients with type 2 diabetes:a network meta-analy-sis

Objective:To systematically review the effects of glucagon-like peptide-1 receptor agonists (GLP-1 RAs)on two common respiratory system adverse events (RSAE:nasopharyngitis and upper re-spiratory tract infection)among type 2 diabetes (T2DM).Methods:Medline,Embase,Clinical trials and Cochrane library were searched from inception through May 201 5 to identify randomized clinical trials (RCTs)assessed safety of GLP-1 RAs versus placebo or other anti-diabetic drugs in T2DM.Network meta-analysis within a Bayesian framework was performed to calculate odds ratios for the incidence of RSAE.Results:In the study,50 RCTs were included,including 1 3 treatments:7 GLP-1 RAs (exenati-de,exenatide-long-release-agent,liraglutide,lixisenatide,taspoglutide,albiglutide and dulaglutide), placebo and 5 traditional anti-diabetic drugs(insulin,metformin,sulfonylureas,sitagliptin and thiazo-lidinediones ketones).Compared with insulin,taspoglutide significantly decreased the incidence of naso-pharyngitis (OR =0.67,95%CI:0.46 -0.96).Significant lowering effects on upper respiratory tract infection were found when taspoglutide versus placebo (OR =0.57,95%CI:0.34 -0.99)and insulin (OR =0.39,95%CI:0.23 -0.73).The result from the network meta-analysis based on Bayesian theo-ry could be used to rank all the treatments included,which showed that taspoglutide ranked last with mi-nimum risk on nasopharyngitis and upper respiratory tract infection.Conclusion:Taspoglutide was associ-ated with significantly lowering effect on RSAE.
10.The incidence of the main adverse events of thoracic/abdominal aortic coated graft: a systematic review and meta-analysis

Objective The purpose of this study was to review all published reports on thoracic/ abdominal stent graft and investigate the incidence rates of six main adverse events(MAEs):myocardial infarction,paraplegia,renal failure,respiratory failure,stroke and all-cause mortality.Methods Electronic databases(PubMed、Embase、OVID、ProQuest、Elsevier and The Cochrane Library) were searched from inception through May 2014 to identify studies that assessed the safety of thoracic/abdominal aortic coated stents on MAEs.The incidence rates with 95% confidence intervals(95% CIs) were derived using a random effects model,considering the heterogeneity between the included studies.Subgroup and sensitivity analyses were applied to explore heterogeneity.Results A total of 152 studies were included in the analysis with 264 arms.For thoracic stent graft,meta-analysis yielded a combined estimated incidence rates of 12.2%,and the most common MAEs was all-cause mortality (8.1%),followed by respiratory failure (6.5 %).For abdominal stent graft,meta-analysis yielded a combined estimated incidence rates of 4.6%,and the most common MAEs was all-cause mortality(4.0%),followed by myocardial infarction(2.1%).For thoracic and abdominal stent graft,subgroup analysis stratified by age and proportion of males indicated that middle-aged and females have a higher incidence rates of all-cause mortality.Besides,subgroup analysis stratified by follow-up time indicated that the longer follow-up time,the higher incidence rate of all-cause mortality.Conclusion The current evidence indicates that the incidence of MAEs of thoracic and abdominal stent graft is high,and we should pay more attention to the patients and follow up them as long as possible.
