1.Analysis on clinical and cell immunophenotype of primary extra-nodular non-Hodgkin′s lymphoma in head and neck

Objective:To report the characteristic of clinical manifestation and cell immunophenotype in cases with primary extra-nodular non-Hodgkin′s lymphoma(PE-NHL) in head and neck.Methods.One hundred and fifteen patients with PE-NHL in head and neck were reviewed,which were diagnosed by pathology in the pas ten years.The features of clinical manifestation and cell immunophenotype in these patients wre analyzed.Results:Twenty to forty years old was most common (42.0%) ages range in the group of patients with PE-NHL.Nasal cavity was the most commonly involved site (36.6%,42/115), next the PE-NHL respectively occurred in nasopharynx(26.1%,30/115),tonsilla(20.1%,24/115) and oral cavity (8.7%,10/115).Only in a few cases the PE-NHL occurred in oroharynx,laryngopharynx and larynx.The patients mainly showed the symptoms of primary lesion located.The lump,ulcer or both signs were observed on primary position,the lump was the most common sign.Cell immunophenotype resulte displayed 51.2% of the PE-NHL originating from T cells,48.8% originating from B cells.Most of nasal PE-NHL originated from T cells ,while most of tonsilla and oral PE-NHL originated from B cells.About half of nasopharyngeal PE-NHL originated from T cells,another half origiated from B cells.Conclusion:The PE-NHL in head and neck mostly occurred in nasal cavity ,then occurred in nasopharynx,tonsilla and oral cavity.The clinical manifestation in the patients with the PE-NHL lacked characteristic symptoms.Sometimes these PE-NHL were confused with other malignant tumours in primary position,which were easily made a misdiagnosis.Therefore discrimination and diagnosis have to be made by pathology and immunohistochemistry.
2.Time course and distribution of myocyte apoptosis induced by acute ischemia in rats

AIM: Myocyte apoptosis in rats can be induced by acute ischemia, but time course and distribution of myocyte apoptosis were unclear.METHODS:DNA agarose gel electrophoresis and TdT-mediated dUTP nick end-labling(TUNEL) assay were performed to evaluate apoptosis in mycardium exposed to 45 minutes, 2 hours, 6 hours, 12 hours ischemia and sham-operated rats in vivo.RESULTS: DNA ladders were clearly visible in agarose gel of DNA from ischemic myocardium exposed to 2 hours, 6 hours and 12 hours ischemia, and DNA ladders became more apparent with increasing duration of ischemia. TUNEL positive cells with apoptotic morphologic characters were present in above ischemia time, and apoptotic index increased with increasing ischemia time. Epigenetics refers to non-coding sequence changes such as DNA methylation, histone modifications, chromosome remodeling and non-coding RNA regulation.Histone modifications include acetylation, phosphorylation, methylation, ubiquitination and ADP ribosylation.The combinations of different histone modifications, known as "histone code", are dynamic during development and differentiation and play important roles in the regulation of gene expressions in spatial-temporal manners.The modification on a particular residue in a histone affects not only the modifications at different residues in its own protein but also other histones.The histone modifications is a complicated network and the regulation remains elusive.
4.Bioavailability and pharmacokinetics of two sustained release formulations of mizolastine in healthy volunteers

Purpose The aim of this study was to compare the pharmacokinetic(PK) profile and relative bioavailability of two sustained release tablets containing 10 mg mizolastine in healthy, young Chinese volunteers. Methods A single oral dose of mizolastine was given under fasting conditions to volunteers aged from 21 to 24 years in this open-label, randomized, crossover study. A ten-day washout period was applied between each of the two formulations. Plasma samples were obtained before dosing and at predetermined time points after dosing up to 48 hours and were analyzed for plasma concentration with a high-performance-liquid chromatography-UV method. PK parameters representing the extent and the rate of absorption of mizolastine were obtained. An analysis of variance, 90% confidence intervals, and two one-sided tests were employed for statistical analysis of relative difference between the two formulations. Results According to the pharmacokinetic and statistical analysis, parameters were not statistically different between the two formulations except the peak concentration (cmax). The point estimates of the ratios of AUC0→t, AUC0→∞ of mizolastine were (101.26 + 9.82) % and (102.52 + 8.61)% with 90% confidence intervals (CIs) of 95. 5% - 106. 5% and 97. 7% -106.9% respectively, comprised in the stipulated 80% - 125% range; for cmax values, the ratios was 82.17% - 15.32% with the 90% CIs of 71. 7% - 91.1%, fell in the recommended range of 70% -143%. Conclusions The results indicate that there is no statistically significant difference in PK parameters except cmax between the two sustained release tablets of mizolastine. The 90% CIs of AUC0→t,AUC0→∞ and cmax are within the predefined range. Thus, the two sustained release tablets of mizolastine are considered bioequivalent and generally well tolerated.
5.Expression of 14-3-3 sigma in pancreatic carcinoma and its clinical significance

Objective To explore the overexpression of the 14-3-3 sigma (14-3-3σ) gene effects proliferation in pancreatic cancer cells.Methods The 14-3-3σ protein level in 46 pancreatic cancers and 9 normal pancreases specimens were analyzed by immunohistochemistry.The mRNA and protein expression of 14-3-3σ in pancreatic carcinoma cell lines (BxPC3,CAPAN-1,PANC-1,AsPC-1,SW1990,MiaPaCa-2 and CFPAC-1) were detected by RT-qPCR and Western blot respectively.A recombinant plasmid of pEGFP-14-3-3σ was constructed and transfected into the pancreatic cancer cell PANC-1 by liposome,and the expression of 14-3-3σ was detected by Western blot and real time fluorescence quantitative PCR.Cell proliferation activity was determined by MTS assay.Results The 14-3-3σ protein level was higher in pancreatic cancer tissue than in normal pancreatic tissue.mRNA and protein expression of 14-3-3σ was highest in BxPC3,high in AsPC-1,SW1990 and CFPAC-1,low in PANC-1 and Capan-1,and lowest in MiaPaCa-2.The successfully constructed pEGFP-14-3-3σ was confirmed by RT-qPCR and Western blot.MTS assay showed cell proliferation activity was significantly enhanced by overexpression of the 14-3-3σ gene compared to negative and blank control cells.Conclusion The expression of 14-3-3σ was higher in pancreatic cancer compared with normal pancre atic tissue,and the 14-3-3σ gene enhanced the cell proliferation activity of PANC-1.Therefore,14-3-3σ may play an important role in pancreatic cancer development.
6.Physical and chemical properties of acetylated low molecular weight heparin and its antineoplastic effect in vitro

Aims To synthesize acetylated low molecu-lar weight heparin( ALMWH) and to detect its physico-chemical properties and antineoplastic activity. Meth-ods LMWH was prepared by degradation of UFH with sodium periodate oxidation and sodium borohydride re-duction, then the LMWH was acetylated by acetic an-hydride where N, N′, -dicyclohexylcarbodiimide ( DCC ) and 4-dimethylaminopyridine ( DMAP ) were used as catalysts. X-ray diffraction(XRD)and Differ-ential Scanning Calorimetry ( DSC ) of ALMWH were obtained. The antiproliferative activity and anti-inva-sive activity were determined on MDA-MB-231 and MCE-7 human breast cancer cells. Results XRD a-nalysis showed that the LMWH degraded from UFH and ALMWH synthesized by acetylation of LMWH be-longed to amorphous structure, however, their DSC curves were significantly different. Compared with LM-WH, ALMWH had more powerful capacity for binding water and lowering anticoagulant activity, more signifi-cantly ALMWH exhibited stranger anti-proliferative and anti-invasive activity than LMWH, especially when it was used in low concentrations. Conclusion The syn-thesized ALMWH possesses a low anticoagulant activi-ty, certain anti-proliferative, anti-invasive and anti-metastatic activity. This study provides a basic method for screening of antineoplastic drug with low toxicity.
7.Inhibition of MEK sensitizes human breast carcinoma cells to endoplasmic reticulum pathway's apoptosis

Aim To investigate the inhibition of MEK/ERK pathway affecting the sensitivity of human breast carcinoma cells SK-BR-3 to endoplasmic reticulum(ER) stress-induced apoptosis and wish to find new targets for human breast carcinoma chemotherapy.Methods Different concentrations(0,1.5,3,6,9 and 12 ?mol?L-1) tunicamycin(TM) treated human breast carcinoma cells SK-BR-3 for 48 h,then propidium iodide(PI) staining measured apoptotic cells in Flow Cytometry(FCM).Different times(0,6,12,24 and 36 h) of TM(3 ?mol?L-1) treated SK-BR-3 cells,Western blot measured proteins GRP78,ERK1/2 and pERK expression.MEK inhibitor U0126(20 ?mol?L-1) pretreated cells for 1 h before treatment with TM(3 ?mol?L-1) in different concentrations and times,measured above identical indexes and compared with their diversities of treatment with U0126 or not.Results TM induced apoptotic cells
8.Inhibitory effects of heparan sulfate proteoglycan on transplanted breast cancer in C_3H mice

Aim To observe the anti-tumor activity and the mechanism of heparan sulfate proteoglycan(HSPG) on C3H mice transplanted tumors.Methods Tumor model was established and randomly divided into five groups.HSPG groups(5,10,50 mg?kg-1),positive group and control group,intraperitoneal injection was performed once a day for 22 days and the volume of tumors was measured.Mice were treated on 24th day,then tumor weight was examined,thymus index,and spleen index were calculated,the apoptosis was determined by TdT-mediated Dutp nick end labeling(TUNEL) assay in situ,the expression of vascular endothelial growth factor(VEGF) was detected by immunohistochemistry.Results The tumor volume in HSPG groups was reduced without the decrease of thymus index,spleen index.TUNEL assay in situ showed numerous heavy blue apoptosis cells in the HSPG groups significantly higher than in control groups.The tumors in HSPG groups showed significantly lower VEGF expression than those in control group.Conclusion HSPG had significant anti-tumor effects on C3H mice transplantable breast cancer.The mechanisms may be associated with the effects of inducing tumor cell apoptosis and inhibiting the VEGF expression.
9.Effects of cisplatin combined with heparanase inhibitor on proliferation and invasion of human nasopharyngeal carcinoma cells.

This study is to investigate the effects of cisplatin combined with heparanase inhibitor OGT2115 on proliferation, invasion and migration of human nasopharyngeal carcinoma cells CNE-2 and to provide a new target for the treatment of metastasis of nasopharyngeal carcinoma. MTT assay was used to detect the cell viability of CNE-2 after exposure to different concentrations of DDP (2, 4, 8, 16 and 32 micromol x L(-1)), different concentrations of OGT2115 (0.4, 0.8, 1.6, 3.2 and 6.4 micromol x L(-1)), and DDP combined with OGT2115. Transwell assay was applied to analyze the effects of drugs on invasion and migration of human nasopharyngeal carcinoma cells. Wound healing assay was performed to detect cell migration and heparanase activity was analyzed by ELISA. MTT results showed that DDP can inhibit the proliferation of CNE-2 cells in a time and concentration-dependent manner, with an IC50 of 24.03 micromol x L(-1) at 24 h (P < 0.05), low concentration of DDP has almost no inhibitory effect on cell invasion and migration. DDP combined with OGT2115 can significantly inhibit cell invasion and migration. Inhibition of heparanase can significantly enhance anti-invasion and anti-proliferation of DDP.
10.Clinical studies of risperidone in the treatment of the mental symptoms after frontotemporal brain contu-sion

Objective To examine the effect of risperidone on the mental symptoms after frontotemporal brain contu?sion. Methods Sixty cases with mental symptoms after frontotemporal brain contusion were recruited from Jan 2009 to Dec 2011 and were randomly divided into control group and treatment group. The patients in the control group were giv?en vitamin B1 60mg/d, while the patients in the treatment group were given risperidone 1mg/d. Both groups were treated for 1 month. Positive and negative symptom scale (PANSS) and symptom scale (TESS) were used to evaluate the efficacy and adverse reactions of treatment. Results The PANSS score was significantly lower in the treatment group than in con?trol group at 1, 2, 3 and 4 weeks following treatment(difference between groups:F=48.12 ,P<0.0001;Time difference:F=290.99 ,P<0.0001; Interaction between group and time: F=11.91,P<0.0001 ). After time-adjustment, the PANSS score was significantly lower in the treatment group than in control group at 2, 3 and 4 weeks following treatment. In the course of treatment, the patients in both groups had varying degrees of headache, nausea, weight gain and Beckoning. These side effects were alleviated through symptomatic treatment. Conclusion Risperidone can significantly improve psy?chiatric symptoms in patients with frontotemporal brain contusion with satisfactory safety.
